1.The serum level of leptin before and after rosiglitazone administration in type 2 diabetes

Objective To observe leptin level before and after rosiglitazone administration and to explore the possible mechanism of rosiglitazone's improving insulin sensitivity.Methods A total of 30 patients newly diagnosed with type 2 diabetes were separated into three groups at random,and treated with rosiglitazone(4mg/d),metform(750mg/d) and gliclazide(160mg/d),respectively.Serum leptin,insulin and GHbA1c were measured after 12 weeks.Results After 12 weeks' treatment,the fasting glucose and GHbA1c in each group declined sharply,while free insulin did not change.The leptin level in rosiglitazone group decreased and insulin sensitivity was improved after administration,but there was no change in metformin and sulfonylureas groups.Conclusion Leptin may play a role in rosiglitazone's mediating the insulin-sensitizing effects.
2.The effect of adenover-mediated VEGF gene on surival of a random skin flap in the rat

Objective To investigate the effect of adenovirus-medicated gene therpy with vascular endothelial growth factor(VEGF)delivered into the subdermal space to treat compromised skin flap in the rat.Methods Thir- ty sprague-dawley rats weighing 350~400g were used in this study.The rats was randomly divided into three groups of ten rats each.A dorsal flap(8cm?2cm)in full thickness with the pedicale located at the leve of the iliaccrest was designed,each,animal received recipient bed subfascial injections of adenovirus encoding VEGF(Ad-VEGF)or green fluorescent protein(Ad-GFP)as treatment control.Another set of animals(n = 10)received NG was designated as control.The flap were elevated as originally designated and sutured back to its bed.The survival rate of the flap was evaluated on day seven after operation.Results The survival rates of the flap in Ad-VEGF group increased signifi- cantly as compared with those of the Ad-GFP group(P
3.Correlative factors analysis and prevention of perimenopausal symptoms

Objective To investigate the incidence,correlative factors and prevention of perimenopausal symptoms.Methods 146 perimenopausal women were investigated by using Kupperman menopausal index(KMI),life event scale,improved social support scale and self-made inventory.Results We got 112 available data,the incidence of perimenopausal symptoms was 75.0%,58.0%was mild,15.2%was moderate,1.8%was sevcre.The main correlative factors of perimenopausal symptoms were inhabited environment,self emotional controllability,history of gynecologic diseases,history of depression,dysmenorrhea,premenstrual syndrome,menopause,score of life eventscale,improved social support scale.Conclusions The incidence of perimenopausal symptoms is high.It is also related to factors of society,family and mentality,except hypogonadism.Preventing perimenopausal symptoms according to its cirrelative factors may decrease its incidence and enhance the life quality of women.

Humanized medical service should take Benevolence as the goal and run throughout the whole process of diagnosis and treatment of rheumatic immune diseases.Through the discussion on improving doctors' medical skills and paying attention to patients' mental health and physiological needs in the department of rheumatology and immunology,this paper put forward that it should improve the patient's disease control rate,prolong the survival,improve the quality of life and build a harmonious relationship between doctors and patients through the humanized treatment program,health education and follow-up.
6.Effect of angiotensin( 1 -7) on left ventricular hypertrophy and myocardial fibrosis in 2K1C and INAC hypertensive models

Objective To determine the effect of angiotensin( 1 -7) [Ang( 1 -7) ]on the left ventricular hypertrophy and myocardial fibrosis in a pressure-overload hypertensive models,infra-nephric aorta coarctation ( INAC) ,and a renin-angiotensin-aldosterone system ( RAAS) activation hypertensive model,two kidney one clip ( 2K1C) in the early and late stage. Methods Totally 120 health male Wistar rats were randomly and equally divided into 6 groups,that is,sham INAC group,INAC group,INAC Ang( 1 - 7) group,sham 2K1C group,2K1C group and 2K1C Ang( 1 -7) group. INAC and 2K1C hypertensive models was established to cor- responding groups and osmotic minipump implantation was carried out to the rats to deliver the Ang( 1 - 7) to their body homogenously and to examine the parameters representing the blood pressure,including left ventricu- lar hypertrophy ( LVW,LVI,CA) and myocardial fibrosis ( CVF and PVCA) . Results Left ventricular hy- pertrophy ( LVH) was observed in the early stage and developed in the late stage both in the 2K1C and INAC groups. Ang( 1 -7) decreased LVW,LVI,CA both in the early and late stage in the 2K1C model( P 0. 05) . Myocardial fibrosis happened and developed progressively in the 2K1C hypertensive model ( P 0. 05) . Natural killer T (NKT) cells,a subset of lymphocytes that bridge innate and adaptive immune systems,involve in processes of infection immunity,tumor immunity,transplantation immunity and autoimmunity.A significant progress has been made in the mechanisms of origin,selection,differentiation and maturation of NKT cells.However,some viewpoints are still controversial,and need to be further intensively investigated.The potential therapeutic applications of functional NKT cells have been suggested in the prevention and the treatment of various diseases.
9.Determination of Stilbence Glucoside in Polygonum Multiflorum by HPLC

Objective:To determine the content of stilbene glucoside in polygonum multiflorum by HPLC. Methods: An Agilent Eclipse Plus C18(100 mm ×4.6 mm,3.5 μm)column was adopted with acetonitrile-water(18∶82) as the mobile phase at a flow rate of 1. 0 ml·min-1 . The detection wavelength was set at 320nm and the injection volume was 8 μl. Results:The linear range of stilbene glucoside was 38-237. 5 μg·ml-1(r=0. 999 4)with the average recovery of 98. 32%(RSD=1. 59%). Conclusion: The method is simple, sensitive and specific in the content determination of stilbene glucoside in polygonum multiflorum.
10.Expression and significance inflammatory of cytokine mediated by Gli1 in acute pancreatitis

Objective:To investigate the expression and the significance of inflammatory cytokine IL-6 through regulating Gli1 in acute pancreatitis.Methods: In this study,C57 mice were randomly divided into three groups:control group,model group,inhibitor group.caerulein intraperitoneal injection induce acute pancreatitis model.Use HE staining and amylase to testify the model successfully.Use RT-qPCR,Western blot to detect the expression of Gli1 in the pancreas,liver,lung,kidney and intestine and ELISA method to detect inflammatory cytokines IL-6.Results: Compared with control group,the expression of Gli1 is higher in model group,then the expression of IL-6 increases in inhibitor group which uses Gant61 to suppress Gli1 compared with model group.Conclusion: Gli1 may involved in the process of the distant tissue injury and repair in acute pancreatitis and through regulate its downstream cytokines like IL-6 to play a protective role in acute pancreatitis.
