Objective: To observe changes of variation of red blood cell distribution width-coefficient (RDW-CV), levels of matrix metalloproteinase (MMP)-2, MMP-9 and tissue inhibitor of metalloproteinases (TIMP)-1 in patients with essential hypertension (EH) of different risk stratification.Methods: A total of 105 EH patients treated in our department from Oct 2015 to Sep 2016 were regarded as EH group.According to hypertension risk stratification, they were divided into low risk group (n=34), medium risk group (n=38) and high-and extremely high risk group (n=33).Another 105 subjects with corresponding age and gender were selected as healthy control group during the same period.RDW-CV, serum levels of MMP-2, MMP-9 and TIMP-1 were measured and compared among all groups.Multivariate Logistic regression analysis was used to analyze relationship among above indexes and hypertension.Results: Compared with healthy control group, there were significant rise in RDW-CV [(12.57±1.46) vs.(14.54±1.82)], serum levels of MMP-2 [(121.71±18.86)ng/ml vs.(155.43±40.81)ng/ml], MMP-9 [(109.72±21.80)ng/ml vs.(191.23±53.05)ng/ml] and TIMP-1 [(59.42±9.41)ng/ml vs.(83.64±15.82)ng/ml] in EH group, P<0.05 or <0.01.Compared with low risk group, there were significant rise in RDW-CV [(13.35±1.54) vs.(14.43±1.17) vs.(15.90±1.81)], serum levels of MMP-2 [(131.21±35.24) ng/ml vs.(152.16±33.15)ng/ml vs.(184.16±37.14)ng/ml], MMP-9 [(163.95±38.61) ng/ml vs.(198.70±43.52)ng/ml vs.(232.83±54.12)ng/ml] and TIMP-1 [(73.15±13.12)ng/ml vs.(83.78±10.22) ng/ml vs.(94.27±16.77)ng/ml] in medium risk group, high-and extremely high risk group, and those of high-and extremely high risk group were significantly higher than those of medium risk group, P<0.05 or <0.01.Multivariate Logistic regression analysis indicated that RDW-CV, MMP-2, MMP-9 and TIMP-1 were independent risk factors for hypertension (OR=2.248~2.725, P<0.05 or <0.01).Conclusion: RDW-CV,MMP-2,MMP-9 and TIMP-1 are independent risk factors for hypertension, active monitoring and intervention should be given for these risk factors.

Objective To evaluate the prospects of recombinant 38000 protein of Mycobacterium tuberculosis in tuberculosis epidemic investigation and subunit vaccine preparation.Methods Physicochemical characteristics of recombinant 38000 protein was detected by P I, peptide-mapping analysis and circular dichroism,guinea pig skin test,MTT stain,and peripheral blood macrophage phagocytosis were used to investigate the roles of recombinant 38000 protein in the cell immunity.Results Recombinant 38000 protein was acidic protein,its P I, was 4 67.The number of alkaline amino acid correspond with theoretic number;The secondary structure of recombinant 38000 protein was composed of ?-helix(32 6%),?-turn(31 6%) and random coil(35 8%) Recombinant 38000 protein could induce DTH in guinea pig sensitized by Mycobacterium tuberculosis Recombinant 38000 protein enhanced phagocytosis of macrophage in mice . PBMC from 30 8% healthy donors and 25% tuberculosis patients were stimulated by the recombinant 38000 protein.Conclusion Recombinant 38000 protein may be used as diagnostic reagent and as an candidate in development of subunit vaccine.

Objective To compare expression of extracellular proteins of virulent H37Rv and attenuated H37Ra in order to search differential proteins,to provide a train of thought for studing M.TB toxicity further.Methods Extracellular proteins were extracted from H37Rv and H37Ra which were inoculated and cultured on Suton's medium for three weeks.The first dimensional electrophoresis was performed on immobilized pH gradient rod gels(pH 3～10).Then the proteins in the rod gels were separated using SDS-PAGE gels.The silver-stained gels were dried and scanned with image scanner.The 2D image analysis was performed with image Master 2D Elite 3 10.Results The most protein spots deriving from extracellular proteins of H37Rv and H37Ra strains were in acidic range.In the basic range(pI more than 9 0),the number of protein spots belong to extracellular proteins of H37Rv and H37Ra was few.Three protein spots belong to low molecular range in H37Rv strain.However,absent in H37Ra strain.Conclusions Two-dimensional gel electrophoresis is useful to separate protein in Mycobacterium tuberculosis.

Objective To investigate the effects of substance P (SP) on the expression of apoptosis-associated genes in fibroblasts derived from pathological scars. Methods Fibroblasts derived from keloid, hypertrophic scar and normal skin were cultured separately in media containing SP and SP receptor antagonist. PCNA, bcl-2 and bax protein in fibroblasts were assessed by means of immunohistochemistry. Results SP enhanced PCNA and bcl-2 expression in all three kinds of fibroblasts, whereas, bax expression was inhibited significantly. SP inhibited the expression of bax in keloid scar fibroblasts (KSFB) more remarkably than that in hypertrophic scar fibroblasts (HSFB) or normal fibroblasts (NFB), and the effect was stronger on HSFB than on NFB. SP receptor antagonist could inhibit those effects of SP totally or partially. Conclusion SP may play an important role in the formation of pathological scars by modulating the expression of apoptosis-associated genes, which is mediated by SP receptor.

Objective To express TmpA recombinant antigen of Treponema pallidum in E.coli and to develop an Enzyme linked Immunoassay (EIA) based on the recombinant antigen for diagnosing syphilis. Methods The target TmpA gene was amplified by polymerase chain reaction (PCR). The PCR products of the gene were inserted into pBluescript T vector, and then expressed in E.coli, using pQE 30 system. Then the recombinant antigen was purified by an affinity chromatography and used for the development of EIA. Results The antigenicity of the recombinant antigen was identified by western blotting (WB) and EIA. The sensitivity and specificity of EIA were 100%(10/10) and 100% (20/20), respectively. The positive rates of anti TmpA antibodies were 91.67% (11/12) for the patients with Ⅰ phase of syphilis and 100% for the patients of Ⅱ and late stage of the disease. Conclusions The TmpA recombinant protein can be used to diagnose syphilis since 97.2% (35/36) of patients with syphilis were positive for the anti TmpA antibody by EIA.

Objective To observe the clinical efficacy and adverse reactions of desogestrel ethinylestradiol in the treatment of dysfunctional uterine bleeding ( adolescent Dub ) .Methods 86 cases with adolescent functional bleeding were divided into the treatment group and control group ,43 cases in each group .The control group received conjugated estrogen(Premarin) treatment,the treatment group was given desogestrel ethinylestradiol treatment .The hemostatic effect and drug adverse reaction of the two groups were observed .Results The cure rate of the treatment group was 65.1%,that in the control group was 23.3%,there was significant difference between the two groups (χ2 =15.28,P<0.01).The hemostatic time of the treatment group was (33.24 ±12.85)h,which was shorter than (50.31 ± 16.32)h of the control group (t=5.39,P<0.05).The incidence rate of adverse reactions between the two groups had no significant difference (χ2 =0.49,P>0.05).Conclusion Desogestrel ethinylestradiol in the treatment of ado-lescent functional bleeding has exact curative effect ,it has low incidence of adverse reactions and better acceptance .

Objective:The non targeted high-throughput urine metabolomics technology was used to study the pathogenesis of APP/PS 1 double transgenic mice and the mechanism of action of Gouteng san.Methods:5-month-old APP/PS 1 double transgenic mice were test with Morris water maze for spatial learning ability.Then we employed the non targeted high-throughput urine metabolomics technology to study the pathogenesis of APP/PS1 double transgenic mice based on the metabolic network.The focus investigation of the key pathways and the observation of the treatment by Morris water maze and metabolic level have been used after spatial learning ability damaged confirmed.Results:The comparison between APP/PS1 double transgenic mice and normal mice suggested that a significant longer was existed in former,which was call-back by Gouteng san.With the non targeted high-throughput urine metabolomics analysis and pathway focused analysis,we found certain signals from metabolic profiling,which was identified to be 6 biomarkers associated with learning and memory function by mass spectrometry analysis or authoritative database.Respectively,they were taurine,pteroylglutamic acid,neopterin,glutaurine,2-oxoglutarate and dihydroneopterin.They were mainly related to taurine metabolism and folate metabolism and represented an effective callback.Conclusion:Gouteng san possess a favorable effect on learning and memory ability of APP/PS1 double transgenic mice,6 biomarkers may be a potential target for the pathogenesis of APP/PSI double transgenic mice and provide experimental basis for the study of Gouteng san.

Objective:To investigate the correlation of serum hs-CRP,D-dimmer and Lp-PLA2 with vulnerable plaque of coronary artery atherosclerosis in patients with coronary heart disease.Methods:Selected 106 cases of patients in our hospital from January 2014 to December 2015,all taken coronary angiography and intravascular ultrasound.Divided into three groups according to the results of the examination,the levels of serum hs-CRP,D-dimmer and Lp-PLA2 were examined and compared,and the correlation with fiber cap thickness,plaque eccentricity index,and vascular remodeling index were tested by Pearson correlation analysis.Results:Serum levels of hs-CRP,D-dimmer and Lp-PLA2 of vulnerable plaque group and stable plaque group were significantly higher than that of the control group (P＜0.05),and the serum levels of hs-CRP,D-dimmer and Lp-PLA2 of vulnerable plaque group were significantly higher than that of stable plaque group (P＜0.05);hs-CRP was negative correlated with the thickness of fibrous cap (r=-0.712,P＜0.05),and positive correlated with eccentric plaque index and vascular remodeling index (r=0.813,0.756;D-,P＜0.05),D-dimmer was negative correlated with the thickness of fibrous cap (r=-0.654,P＜0.05),and positive correlated with eccentric plaque index and vascular remodeling index (r=0.912,0.853,P＜0.05);Lp-PLA2 was negative correlated with the thickness of fibrous cap (r=-0.796,P＜0.05),and positive correlated with eccentric plaque index and vascular remodeling index (r=0.836,0.729,P＜0.05).Conclusion:Hs-CRP,D-dimmer and Lp-PLA2 have high correlation with vulnerable plaque in coronary artery disease,can be used as reference indexes for assessing the instability of coronary atherosclerotic plaque.
