National Journal of Andrology 2014;20(5):423-429
OBJECTIVETo improve the sample collection methods and bacteriologic localization patterns in male genital tract infection, and to investigate the influence of specimen collection and pathogen isolation on the diagnosis and treatment of prostatitis.
METHODSWe collected the samples of the initial urinary stream, the third portion of the urinary stream, expressed prostatic secretion (ESP), and semen from 200 adult males with chronic prostatitis-like symptoms, inoculated them quantitatively in culture media for isolation of microorganisms, and evaluated their laboratory diagnostic significance according to the count of colonies and distribution of the isolates.
RESULTSA total of 468 strains of microorganisms were isolated from the samples, including 414 strains of bacteria spp (88.5%), 12 strains of fungi spp (2.6%), 40 strains of mycoplasma spp (8.5%), and 2 strains of chlamydia spp (0.4%). Pathogens were isolated from the ESP in 66 cases (33.0%), from the semen in 34 cases (17.0%), and from both the ESP and semen in 100 cases (50.0%). Only 1 species of pathogen was found in the ESP samples of 36 cases (18.0%), in the semen samples of 20 cases (10%), and in both the ESP and semen samples of 39 cases (19.5%); 2 species in the ESP samples of 30 cases (15.0%), in the semen samples of 14 cases (7.0%), and in both the ESP and semen samples of 60 cases (30.0%); and 3 species in both the ESP and semen samples of 1 case (0.5%).
CONCLUSIONMultiple microbial infection (MMI), multi-organ infection (MOI) and drug-resistance strains infection are common in patients with prostatitis-like symptoms, frequently leading to missed diagnosis and misdiagnosis in clinic and laboratory, and affecting the effect of antimicrobial therapy. MMI and MOI can be diagnosed and differentially diagnosed with the improved sample collection methods and bacteriologic localization patterns.

Journal of Chinese Physician 2017;19(2):195-200
Objective To explore the Cep70 by adjusting the stability of acetylated alpha tubulin, participate in breast cancer drug resistance mechanisms. Methods (1) In order to induce taxol drug resistance cell line Michigan cancer foundation-7 (MCF-7)/pac,high-dose shock treatments taxol MCF-7 was used for 6 months, until the cells can grow in 3.5 μmol/L of paclitaxel.(2) The 3-(4,5-dimenthylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium (MTS) method was used to detect inhibition rate by taxol to MCF-7 and MCF-7/pac cell.(3) Immunofluorescence and Western blot were used to test acetylated alpha-tubulin and Cep70 expression levels in MCF-7 and MCF-7/pac cells.(4) Chemical intervention was used to acetylate apha-tubulin expression,Western blot and polymerase chain reaction (PCR) were used to detect the change of acetylated alpha-tubulin and Cep70 in MCF-7 and MCF-7/pac groups.Flow cytometry and Western blot were used to detect the change of cell cycle. Results (1) IC50 of MCF-7 and MCF-7/pac was 22.47 μ mol/L and 31.38 μmol/L,respectively.(2) Immunofluorescence and Western blot results showed that the expression of acetylation of alpha-tubulin in resistant MCF-7 cell/pac was obviously decreased.(3) Real time polymerase chain reaction (RT-PCR) and Western blot showed Cep70 expression was consistent of acetylation of alpha-tubulin.(4) After incubation with paclitaxel for 24 hours, the expressions of acetylation of alpha-tubulin and Cep70 in MCF-7 and MCF-7/pac were increased, but the extent of MCF-7 cell was much higher.Instead, incubation with nocodazole after 24 hours, the acetylation of alpha-tubulin and Cep70 in MCF-7 and MCF-7/pac cells were obviously lowered.(5) After paclitaxel intervention, compared to the same group MCF-7 cells, the G2 phase ratio in MCF-7/pac cells was lower. In addition, given nocodazole after the intervention, compared to the same group MCF-7 cells, the ratio of G2 phase in MCF-7 cell/pac was significantly decreased. Conclusions Cep70 decreased the expression of the acetylated alpha-tubulin,reduced the stability of microtubules, which could be an important mechanism of taxol drug resistance.

Journal of Chinese Physician 2017;19(2):186-190
Objective To reveal acetylated a-tubulin acts as pressure sensors, sensing the changes in extracellular matrix to impact on the invasion and metastasis of breast cancer. Methods The acetylated alpha microtubule protein expressions were detected, and its relationship with invasion and metastasis in breast cancer clinical specimens and different molecular subtypes of breast cancer cell lines were analyzed. The expression of acetylated a-tubulin was interfered through chemical methods, and cell morphology and the change of the invasion and metastasis ability were detected under the condition of different matrix hardness. Results The expression of acetylated a-tubulin was highest in basal-like breast cancer tissue and cell lines, and was lowest expressed in the luminal B breast cancer tissue and the breast cancer cell lines. The expression of acetylated a-microtubule was positively correlated with the occurrence of breast cancer.Under the condition of soft substrate cultivation, cell polarization was declined, becoming the circular or oval shape. The acetylated a-tubulin caused reduction in cell polarity, accompanied with less invasion and metastasis ability. Conclusions The acetylated a-tubulin acts as pressure sensors, sensing the sclerosis of extracellular matrix in the process of tumorigenesis, promoting invasion and metastasis of breast cancer.

Chinese Journal of Medical Ultrasound (Electronic Edition) 2014;(10):809-815
ObjectiveTo explore the contrast-enhanced ultrasonography features of postpartum placenta implantation and its clinical value in diagnosis of postpartum placenta implantation.MethodsBetween April 2009 and May 2013, twelve patients with clinically suspected placenta implantation underwent contrast-enhanced ultrasonography in Hubei Maternal and Child Health Hospital. The location and extent of intrauterine lesions and perfusion features of contrast agent were observed. The contrast-enhanced ultrasonography features of postpartum placenta implantation were analyzed and compared with postoperative pathological diagnosis.ResultsIn twelve patients with placenta implantation, eleven showed enhancement. Only one lesion showed no enhancement, which was diagnosed as placental degeneration and calcification on postoperative pathology. One case was placenta adhesions conifrmed by postoperative pathological diagnosis, which presented with a hyperenhanced lesion in uterine cavity with a clear boundary to myometrium. Six cases were placenta implantation confirmed by postoperative pathological diagnosis, which presented with a residual placenta surrounded with extremely thin or evendiscontinuous uterine serosa. Five cases were placenta penetration confirmed by postoperative pathological diagnosis, which presented with a residual placenta penetrating the uterine serosa and contrast agent spilling out. All cases in this study were cured by operation.ConclusionsContrast-enhanced ultrasound could diagnose placenta implantation via the difference of tissue perfusion. The lesion of placenta implantation showed enhancement after injection of contrast agent. If discontinuous uterine serosa and contrast agent spilling was observed, placenta penetrating uterine serosa should be considered. Contrast-enhanced ultrasound has important clinical value in diagnosing postpartum placenta implantation and observing postoperative recovery processes of uterine rupture.

Journal of Chinese Physician 2001;0(10):-
Objective To investigate the relationship between helicobacter pylori (H. pylori) infection and reflux esophagitis (RE). Methods H.Pylori infection rates of RE in patients with simple chronic gastritis(CG),which was confirmed by gastrospcope combined with pathological diagnosis were researched. The relationship between HP infection and RE classifications using gastroscope and pathology was explored. Results ⑴H.pylori infection was found in 29(27.1%) in 107 cases of RE and 43(40.2%) in 107 cases of simple CG patients respectively,HP infection positive in RE patients less than that of simple CG patients.RE of class Ⅰ,Ⅱand Ⅲ using gastroscopy classification were 62.1%, 10.3% and 27.6% in H. Pylori-positive cases respectively, while they were 56.4%, 6.4% and 37.2% respectively in H. Pylori-negative ones. However, mild, moderate and severe RE identified by pathohistology were 72.4%, 13.8% and 13.8% in H.Pylori-positive cases respectively, and they were 57.7%, 17.9% and 24.4% respectively in H.Pylori-negative ones. Conclusions All above results suggested that H.Pylori possibly has prevention role in some extent to pathogenesis of RE. RE more commonly was seen in H.Pylori-negative cases.RE could inhibit the H.Pylori survive,So that, the RE was occurred frequently in H.Pylori-negative patients. The inflammatory extent of RE is not serious in H.Pylori-positive patients.

Journal of Interventional Radiology 1992;0(01):-
As a result of the increasing use of radiologic contrast medium in patients, especially the elderly in severe or clinical condition with attendant comorbidities, such as diabetes mellitus, renal failure, cardiac failure, and volume depletion. Contrast induced nephropathy(CIN)is now the third most common cause of hospital-aquired renal failure and accounts for approximately 11% of the cases. This article mainly introduces the evaluation of laboratory investigations providing insights into the pathophysiology of this disorder and the examination of the important clinical data of CIN including risk factors and diagnosis. The last section deals with renal protection and preventive strategies.
