1.Studies on the effects of EGF and EGFR on the injury of mice intestines induced by neutron and ?-ray irradiation

Objective To study comparatively the changes in epi th elial growth factor (EGF) and its receptor (EGFR) in injured intestine induced b y neutron and ?-ray irradiation in mice and their significance. Method s 350 male BALB/C mice were irradiated with neutron and ?-rays, and t hey were sacrificed at 6 and 12 hours and 1, 2, 3, 4, 5, 7, 10, 14, 21 and 28 da ys, respectively, after irradiation. Immunohistochemical method was employed to assess EGF and EGFR in the intestinal tissue of the mice. Results After the neutron radiation with 2.5Gy dosage, the expressions of EGF and E GFR in the cytoplasm of mucosa epithelial cells and crypt cells were obviously u p-regulated within 1 day, decreased after 1~2 days, increased again on 3~7 days , reached the peak value at the 5th day, and returned to normal values in 14 day s. Whereas EGF and EGFR were increased at 6 hours and progressively decreased fr om 12 hours up to 4 days after 4.0 and 5.5Gy neutron and 12Gy g-ray radiation . They were increased progressively within 3 days, reaching peak value on the 3r d day, and returned to normal values 5 days after 5.5Gy g-ray irradiation. Conclusions The expressions of endogenous EGF and EGFR showed diffe rent regularities after neutron and g-ray radiation, and they were involved in the pathologic courses of radiation damage and recovery of the intestine.

Bone growth factors play an important role in the process of bone metabolism and perform a variety of physio -logical functions such as regulating bone growth , differentiation and remodeling .Meanwhile , these cytokines are associated with orthopedic disorder and therapy .The curative effect of the overturned sartorius iliac flap in repair of acetabular defect in developmental dysplasia of the hip proves to be good , but mechanism between the overturned flap and the surrounding bone tissue is unclear .This review briefly summarizes the functions and applications of several major cytokines in bone for -mation and remodeling , which may provide new ideas and methods for illuminating the mechanism of these cytokines during the repair of the acetabular defect .
3.Determination of Contents and Related Substances in Loxoprofen Sodium Tablets by RP-HPLC

OBJECTIVE:To improve the determination method for the contents of main components and related substances in Loxoprofen sodium tablets. METHODS:RP-HPLC method was adopted. The determination was performed on Inspire C18 column with mobile phase consisted of acetonitile-0.01 mol/L potassium dihydrogen phosphate(containing 0.2% triethylamine,phosphoric acid adjusted to 3.0±0.1,62 : 38,V/V)at the flow rate of 1.0 mL/min. The column temperature was 40 ℃,and the detection wave-length was set at 221 nm. The sample size was 20 μL. RESULTS:The peak of loxoprofen sodium was well separated with the peak of its related substances(R>1.5). The linear range of loxoprofen sodium ranged 30.0-90.0 μg/mL(r=0.9998). The detection lim-it of loxoprofen was 0.3 μ g/mL. RSDs of precision,stability and repeatability tests were <1.0% . The average recovery rates ranged 99.00%-99.87%(RSD=0.33%,n=9). CONCLUSIONS:This method is accurate,simple,rapid and suitable for the quali-ty control of Loxoprofen sodium tablets.
4.Expression and significance of TNF-α in intestine tissue of mice irradiated by neutron and γ rays

Objective To study the expression of tumor necrosis factor alpha (TNF-α) in the intestine of mice irradiated by neutron and γ rays.Methods 350 male BALB/c mice were irradiated with neutron and γ rays of different doses, and sacrificed at 6 and 12hours, 1, 2, 3, 4, 5, 7, 10, 14, 21 and 28 days after irradiation.The TNF-α in the mice intestinal tissue was detected by means of immunohistochemistry and image analysis.Results In normal control mice, TNF-α was expressed in the cytoplasm of macrophages in intestinal villus interstitium, submucosa and lymph tissue.After 2.5Gy neutron radiation, TNF-α was decreased progressively within 2 days, increased obviously in macrophages and crypt cells during 3rd~7th day, reached the peak at 5th day and recovered to normal level at 14th day and TNF-α was decreased progressively within 4 days after 4.0 and 5.5Gy neutron and 12Gy ray irradiation.TNF-α was increased obviously in 6~12 hours, decreased at 1st day, increased at 2nd~5th day, peaked at 3rd day and recovered at 10th day after 5.5Gy ray irradiation.Conclusion Neutron and ray radiation induce different expression profile of endogenous TNF-α in small intestine, which may be related with the pathologic courses of irradiation-induced damage and repair of intestine.
5.Comparison of epidemiological situation in chronic kidney disease between urban and rural areas in Guangxi province

ObjectiveTo investigate and compare the prevalence,awareness and risk factors of chronic kidney disease (CKD) between urban and rural population in Guangxi province in order to provide information for prevention and treatment of CKD.MethodsBy a stratified multistage random sampling method,18 to 74 years old residents in Guangxi province were surveyed. They wereevaluatedbyquestionnaire,urinaryalbumin/creatinineratio, hematuria (microscopic examination of centrifuged urine sediment),kidney B-mode ultrasound,and abnormal results were reviewed 3 months later.Estimated glomerular filtration rate(eGFR) was calculated with the simplified MDRD equation modified by a Chinese coefficient.The risk factors associated with CKD were also investigated.Results There were no significant differences between urban and rural residents in the prevalence of albuminuria (5.22% vs 5.47%) and hematuria (1.07% vs 1.11%)(all P>0.05).The prevalence of renal lithiasis in rural residents was significantly higher than that in the town(10.54% vs 6.95%)(P<0.05).The decreased renal function between urban and rural residents(3.87% vs 4.04%,P>0.05) had no significant difference.The prevalence of CKD was 9.58% in urban and 9.42% in rural(P>0.05).The prevalence of albuminuria according to the age distribution was different between urban and rural,which increased along with the age in urban but showed two peaks(30-40 years old and 60-74 years old) in rural.Based on logistic regression analysis,the risk factors for albuminuria were diabetes,hyperuricemia,the history of cardiovascular disease,chronic tonsillitis and HBsAg positive.The risk factors for kidney function decline were age,hyperuricemia,hypertension,diabetes,renal lithiasis and history of cardiovascular disease.The awareness rate of CKD in urban was significantly higher than that in rural (14.45% vs 6.27%,P<0.05).Conclusions The prevalenceof CKD has no significant difference between urban and rural in Guangxi province.The awareness rate of CKD in urban is significantly higher than that in rural.It is needed to enhance the prevention and treatment of CKD in rural.

Objective To study changes and significance of endothelin(ET) in rat cerebral concussion.Methods 80 Wistar male rats were used for animal model of cerebral concussion,which were sacrificed on 1,3,7,14 and 30 days after injury and the brain tissue were taken off. The expression of ET was studied in the course of cerebral concussion by means of immunohistochemistry.Results Typical clinical manifestation was observed in the 100 g group in which the pathological changes included cerebral vascular constriction and dilatation,congestion and edema of cerebral tissue,neuronal degeneration,necrosis,and obviously decreased even disappeared Nissl bodies.Increased expression of ET was observed on the first day,the positive area was seen in the plasma of endothelial cells in cerebral cortex,hippocampus,cerebellum and thalamus.ET expression peak occurred on the 7th day,the positive area was also found in the plasma of Purkinje cells in the cerebellum.Decreased ET expression was found on 14th day and returned to normal level on the 30th day.Conclusion The main pathological changes of cerebral concussion contained blood circulation disorder,and degeneration and necrosis of substantial cells.ET was involved in the brain tissue injury during the pathological process of cerebral concussion and might be related to regulation of cerebral vascular reaction,and neuron degeneration and necrosis.
8.The short-term therapeutic effects of DNA immunoadsorbent combined with medication on patients with ;severe systemic lupus erythematosus

Objective To evaluate the short-term therapeutic effects of DNA immunoadsorbent (IA) combined with glucocorticoid and immune depressant on patients with severe systemic lupus erythematosus(SLE). Methods 32 patients with severe SLE were selected to undergo DNA IA treatment combined with glucocorticoid plus cyclophosphamide therapy, and each patient received IA therapy 3 times, once 2.5 hours, with an interval of 24-48 hours to take another two times of IA. The changes in SLE disease activity index(SLEDAI)score, health status evaluation indexes〔 physiologic functional( PF) and emotional health( MH) scores〕,renal function indexes〔 blood urea nitrogen(BUN)and serum creatinine(SCr)〕 were observed; and anti-double stranded DNA antibody( ds-DNA), immunoglobulin (IgA, IgG, IgM), complements(C3 and C4)and high-sensitivity C-reactive protein (hs-CRP) were examined before and after IA treatment for 2 weeks. Results Two weeks after the combination therapy, the SLEDAI score, BUN, SCr, dsDNA, IgA, IgG, IgM, hs-CRP were significantly lower than those before treatment 〔SLEDAI score : 14.38±3.85 vs. 15.69±1.40, BUN (mmol/L): 11.22±4.78 vs. 16.31±7.90, SCr (μmol/L): 127.02±38.17 vs. 167.25±45.63, dsDNA( U/L): 1.36±0.12 vs. 1.43±0.18, IgA( g/L): 2.41±0.73 vs. 2.59±0.86, IgG( g/L): 16.82±4.83 vs. 21.01±4.84, IgM( g/L): 1.64±0.45 vs. 1.75±0.58, hs-CRP( mg/L): 14.41±2.20 vs. 14.94±2.60, P<0.05 or P<0.01〕; PF score, MH score, complement C3 were increased〔 PF score : 71.19±17.53 vs. 56.66±22.41, MH score : 74.01±15.72 vs. 61.50±17.98, C3( g/L): 0.56±0.09 vs. 0.52±0.10, all P<0.05〕; clinical symptoms were improved significantly, and no significant adverse reactions were found. Conclusion IA combined with medical treatment has shown that it has significant therapeutic effect for treatment of patients with severe SLE, and it may decrease the levels of dsDNA, IgA, IgG, IgM,hs-CRP, and increase the level of complement C3.
9.Venetoclax combined with azacitidine in treatment of myelodysplastic syndromes complicated with monoclonal globulinemia of unknown significance: report of 1 case and review of literature

Objective:To investigate the therapeutic effect of venetoclax combined with azacitidine in treatment of myelodysplastic syndromes (MDS) complicated with monoclonal globulinemia of unknown significance (MGUS).Methods:The clinical data of a patient with MDS complicated with MGUS in the Second People's Hospital of Shenzhen in December 2020 were retrospectively analyzed, and the literatures were reviewed.Results:According to results of bone marrow smear, cytogenetics, and next-generation sequencing, the patient was diagnosed as MDS and MGUS complicated with ASXL1, RUNX1, EZH2, STAG2 mutations as well as t(11;14). No response was observed after 2 courses of azacitidine and 1 course of azacitidine plus HAG. Later the patient achieved complete remission and negative RUNX1 and STAG2 mutations after a course of venetoclax combined with azacitidine. Meanwhile, M protein exhibited a decrease more than 50%. To date, the patient was still in complete remission.Conclusions:The regimen of venetoclax combined with azacitidine shows a significant efficacy and good tolerance to patient with co-occurrence of MDS and MGUS with t(11; 14).
10.Study on the value of HPV DNA detection in the screening of cervical lesions

Objective To investigate the effect of human papilloma virus (HPV) DNA detection on cervical cancer and its precancerous lesions screening.Methods A total of 2 852 patients participated in cervical screening in Sichuan Provincial People's Hospital from January 2014 to December 2016 were enrolled.The HPV DNA testing and pathological biopsy were conducted in all patients,and the pathological examination results were defined as the gold standard.The influences of age,regions of residence (rural and urban) and obesity on the positive rate of HPV DNA were analysed.The results of HPV DNA testing in different pathological types were compared,and the sensitivity,specificity,positive predictive value,negative predictive value of of HPV DNA testing were analysed,as well.Results The positive rate of HPV DNA in the 18-<35 age group was significantly higher than that in the 35-<50 age group and 50-58 age group,the differences were statistically significant (P<0.05).There was no statistically significant difference in positive rate of HPV DNA between the 35-<50 age group and 50-58 age group (P>0.05).The positive rate of HPV DNA of patients in urban areas was significantly lower than that of those patients in rural areas (P<0.05),while no statistically significant difference was found in the positive rate of HPV DNA between patients with normal weight and patients whose weight exceeded the standard (P>0.05).Among patients with positive HPV DNA testing results,the detection rate of cervical cancer and its precancerous lesions based on pathological examination was 1.60 % and 28.60 %,respectively.The sensitivity,specificity,positive predictive value,negative predictive value and missed diagnosis rate of HPV DNA testing was 87.79%,86.98 %,30.20 %,99.11% and 12.21%,respectively.Conclusion HPV DNA detection has a high sensitivity and specificity in cervical screening.
