1.Clinical study on the relationship of the expressions of proliferating cell nuclear antigen and endocranium invasion in the convex meningiomas

Objective To evaluate the relationship of the expressions of proliferating cell nuclear antigen (PCNA) and endocranium invasion in the convex meningiomas.Methods Convex meningioma Simpson I level excises 51 patients were selected.The dural mater 15 mm around the rumors was resected,stochastically divided into four sections (0-1 mm,2-5 mm,6-10 mm,>10 mm)for pathological inspection.Analysed statistically the study results based on microscopic observation of the HE staining slides and immunohistochemistry of proliferating cell nuclear antigen(PCNA).Results PCNA LI of the group with the endocranium invasion was 10.32±6.74,the other was 2.16±4.93.The atypical meningioma had a high endocranium invasion degree.Conclusion Pathologic grading of meningioma and the expressions of PCNA by immunohistochemical staining method are the factors affecting endoeranium invasion of the convex meningiomas.
2.Preliminary study of MR elastography for differentiating hepatic focal benign and malignant tumors

Objective To explore the diagnostic value of MRE for differentiating hepatic benign and malignant tumors.Methods 36 patients with liver tumor (a total of 39 lesions,including 20 hepatocellular carcinomas,7 hemangiomas,5 cholangiocellular carcinomas,3 metastases,2 hepatic angiomyolipomas,1 carcinosarcoma,1 castleman’s disease)and 9 healthy volunteers were evaluated with MRE.The elastogram were generated with FUNCTOOL post processing program.The mean value of elasticity of hepatic malignant tumors,hepatic benign tumors,hepatic parenchyma around the malignant tumors,hepatic parenchyma around the benign tumors and the normal liver of healthy volunteers were measured and compared.Results The mean value of elasticity of malignant tumors [(7.39±1.70)kPa]was significantly higher than these of benign tumors [(4.11±0.37)kPa,P < 0.001],peripheral parenchyma around the malignant tumors [(3.50±0.73)kPa,P < 0.001],peripheral parenchyma around the benign tumors [(2.61±0.45)kPa,P < 0.001] and normal liver of healthy volunteers [(2.38±0.24)kPa,P <0.001].The mean value of elasticity of parenchyma around the malignant tumors [(3.50±0.73)kPa]was significantly higher than that of hepatic parenchyma around the benign tumors [(2.61 ± 0.45)kPa,P <0.001].The mean value of elasticity of hepatic parenchyma around the benign tumors [(2.61±0.45)kPa]was slightly higher than normal liver of healthy volunteers [(2.38±0.24)kPa],and there was no significant difference between the two (P >0.05).A cutoff value of 5.08 kPa can accurately differentiate malignant tumors from benign tumors and normal liver parenchyma.Conclusion MRE could be used in diagnosis of hepatic focal tumors,which is helpful for differentiating benign and malignant liver tumors.
3.Analysis of the Regularity of Chinese Patent Medicine Prescriptions for the Qi Deficiency and Blood Stasis Syndrome

OBJECTIVE:To investigate the regularity of Chinese patent medicine prescriptions for Qi deficiency and blood sta-sis syndrome (QDBSS),and to provide reference for studying QDBSS diagnosis standard and new TCM for related symptoms. METHODS:Chinese patent medicine for QDBSS were collected from Chinese Pharmacopoeia (2015 edition) and Chinese Mate-ria Medica Preparation(1993 version). The regularity of Chinese patent medicine prescriptions and symptom association rule were analyzed by using TCM inheritance auxiliary system (V2.5). RESULTS:Among the 171 symptoms,chest tightness,shortness of breath,chest pain,fatigue and palpitation were common symptoms,and three groups of symptoms were included as chest tight-ness-shortness of breath-chest pain-palpitation-irregularly pulse;mouth and eye deviation-numbness of hands and feet-hemiple-gia-stiff tongue-spasm;hypomenorrhea-irregular menstruation-abdominal mass-dysmenorrheal. One hundred and thirty-one Chinese patent medicines included 304 ingredients,top 5 in the list of frequency were Astragalus membranaceus,Salvia militiorrhiza,An-gelica sinensis,Ligusticum chuanxiong,Panax ginseng. A. membranaceus- A. sinensis,L. chuanxiong-A. sinensis,S. militiorrhi-za-A. membranaceus,L. chuanxiong- A. membranaceus and L. chuanxiong-S. militiorrhiza were frequently used herbal combina-tions. A. membranaceus-A. sinensis-L. chuanxiong-S. militiorrhiza-P. ginseng-Carthamus tinctorius-Panax notoginseng was the core combination. A new prescription for QDBSS was formed (P. ginseng-Radix Paeoniae Rubra-borneol-Gastrodia elata-artificial moschus-Myristica fragrans). CONCLUSIONS:QDBSS has common symptoms which exist in many kinds of diseases,such as chest tightness,chest pain and shortness of breath;it also has the unique symptoms among various diseases. Invigorating Qi and promoting blood circulation is the basic treatment for the QDBSS. It is of significance to pay attention to"supplement by passing, combination of pass and supplement"and combine regulating Qi-flowing method with enriching blood method or aromatic warming method for QDBSS therapy according to the differ-ence of Zhengxie shengshuai.
4.Establishment of a cell line expressing the CD36 on human platelets and its application to the detec-tion of anti-CD36 antibodies

Objective To establish a cell line stably expressing the human CD36 by using TA clo-ning and cell transfection technology and to analyze its application to the detection of anti-CD36 antibodies. Methods Total RNA was isolated from human platelets and then used to synthesize complementary DNA ( cDNA) . Sequence of the gene encoding CD36 on human platelets was obtained by PCR amplification. The recombinant vector was transformed into TOP10 E. coli after TA cloning. The positive recombinant pcDNA3. 1/V5-CD36 plasmid was screened out by blue-white selection and then sequenced. The correctly constructed plasmid coated with Effectene? Transfection Reagent was transferred into HEK293T cells. Fluo-rescence-activated cell sorting was performed to screen out the cell line that could stably express the CD36 on human platelets. The transfected cell line-based flow cytometry analysis and antibody capture assay ( ACA) were established and used for antibody detection in nine serum samples positive for anti-CD36 antibodies. Results The HEK293T cell line stably expressing the recombinant CD36 was successfully established. Compare with the monoclonal antibody immobilization of platelet antigens assay ( MAIPA) , anti-CD36 anti-bodies could be easily identified in nine serum samples by using the transfected cell line-based flow cytome-try analysis and ACA. Conclusion This study suggests that the HEK293T cells stably expressing the re-combinant CD36 could be used in flow cytometry analysis and ACA for the detection of anti-CD36 antibod-ies. It also paves the way for further researches on the mechanism of CD36 in other diseases.
5.Total hip arthroplasty in treating acetabular protrusion secondary to rheumatoid arthritis

Objective To investigate the clinical outcomes of total hip arthroplasty in treatment of severe acetabular protrusion secondary to rheumatoid arthritis.Methods From January 2011 to November 2014,eighteen patients (20 hips) with severe acetabular protrusion secondary to rheumatoid arthritis were treated with total hip arthroplasty (THA),including 6 males (6 hips) and 12 females (14 hips).The age ranged from 37 to 68 years (average,45.8±8.3 year).According to the classification of Sotelo-Garza and Charnley,there were 15 cases (17 hips) of type Ⅱ (acetabular protrusion 6-15 mm) and 3 cases (3 hips) of type Ⅲ (acetabular protrusion > 15 mm).During the surgery,the femoral heads were moved out through retrograde method after the osteotomy of femoral neck by posterolateral approach.All patients underwent THA with autogenous bone graft by impacting technique for reconstruction of acetabula,and the cementless cups were planted with press-fit skills (Tantalum trabecular metal cup in 16 cases and Titanium porous coating cup in 4 cases).The hip function was evaluated by using Harris hip score.Radiographic measurement was performed with X-ray films to assess the restoration of the hip rotation center,graft incorporation and sign of prostheses loosening in follow-up duration.Results The mean operation duration and intraoperative blood loss were 89.5±8.1 min (range 55-131 min) and 295±10.9 ml (range 165-480 ml) respectively.The average amount of bone graft was 33.7±6.8 cm3 (range 32.5-61.2 cm3) in reconstruction of protrusion acetabulum.The mean follow up duration was 4.5± 1.7 years (range 2.5-6.0 years).The horizontal distance between the center of femoral head and the Kohler's line was increased from preoperative 8.87±3.9 mm (range-11.11-12.78 mm) to 23.55 ± 2.5 mm (range 21.23-25.57 mm) postoperatively,and the vertical distance between the center of femoral head and the line joining the bilateral ischial tuberosities was decreased from preoperative 87.45 ± 3.5 mm (range 74.75-96.97 mm) to postoperative 77.83±4.1 mm (range 68.22-90.40 mm) with statistically significant difference.The bone incorporation was achieved at 6 months postoperatively in radiological film.No loosening of acetabular components was found in final fellow-up.At last follow-up,Harris hip score was significantly improved from 55.3±9.5 points preoperatively to 92.2±12.7 postoperatively (t=51.569,P=0.000).The range of flexion of the hip significantly improved from 41.5°±6.7° preoperatively to 102.3°±14.5° at finial follow-up (t=64.865,P=0.000),and the range of extension of the hip improved from 8.2°±3.5° preoperatively to 29.8°±6.6° at finial follow-up (t=51.161,P=0.000).The discrepancy was significantly decreased from 23.7±0.82 mm preoperatively to 4.7±0.39 mm postoperatively (t=19.761,P=0.000).Mild thigh pain was reported in two hips,which typically occurred only with prolonged activity without disabling and not requiring medication.Conclusion Total hip arthroplasty with impaction bone grafting and cementless modular cup can recover the acetabular center of rotation and achieve satisfactory short-term outcomes in treating patients severe protrusion acetabula secondary to rheumatoid arthritis.
6.Cementless total hip replacement with femoral calcar reconstruction for unstable intertrochanteric fractures combined with femoral head necrosis

Objective To explore the feasibility and therapeutic effects of total hip replacement (THR) for intertrochanteric fractures combined with late necrosis of the femoral head.Methods From June 2010 to October 2015,8 patients underwent THR for intertrochanteric fractures combined with necrosis of the femoral head.They were 6 males and 2 females,with an average age of 53.5 years(from 45 to 67 years).According to the Evans-Jensen classification,2 fractures were type ⅠB,5 type ⅡA and one type ⅡB.All the 8 fractures were complicated with late femoral head necrosis,5 cases of which were ischemic.According to the Ficat classification,the ischemic necrosis was type Ⅲ in one and type Ⅳ in 5 cases.In the other 3 cases,the necrosis secondary to the hip osteoarthrosis was type Ⅲ according to the Tonnis classification.The necrotic femoral head was removed after osteotomy of the femoral neck via the posterolateral approach;after reduction of the femoral calcar,the intertrochanteric fracture was reduced and fixated in a press-fit manner through antegrade implantation of a biological stem prosthesis.Functional assessment of the affected hip was carried out at the final follow-up using Harris scoring.Results The 8 patients were followed up for an average of 25.5 months (from 12 to 45 months).After operation,biological press-fit and initial stability of the femoral stem were achieved in all the patients.The X-ray films showed bone ingrowth fixation in all the 8 hips at 3 months postoperatively.The Harris hip scores at the final follow-up averaged 92.3(from 89 to 98).The patients showed fine subjective satisfaction.Follow-ups revealed no infection,prosthetic failure,osteolysis,dislocation or articular instability.Conclusion In treatment of intertrochanteric fractures combined with necrosis of the femoral head,THR can lead to fixation and reduction of the intertrochanteric fracture and replacement of the necrotic femoral head at one stage,promoting functional recovery of the affected hip.
7.Construction of eukaryotic expression vector for human platelet CD36 gene 220C>T and 429+4insg variants and analysis of their expressions in HEK293T cells.

OBJECTIVE:
To construct eukaryotic expression vectors for human platelet CD36 gene 220 C>T and 429+4insg variants and analyze their expressions in HEK293T cells.
METHODS:
RNA was isolated from human platelets and reversely transcribed into cDNA. Sequences of 220C>T and 429+4insg variants were derived by PCR amplification. The target sequence was ligated into a pcDNA3.1/V5-His-TOPO vector by TA cloning, which was transformed into TOP10 E. coli. Positive plasmids were screened by blue-white selection. After sequencing, plasmid DNA carrying 220C>T or 429+4insg variant was used to transfect HEK293T cells with the help of effectene. Expression of CD36 protein was then analyzed by flow cytometry and Western blotting.
RESULTS:
An eukaryotic expression vector pcDNA3.1/V5-His-CD36 (220C>T/429+4insg) was constructed by TA cloning. After transfected into HEK293T cells, the 220C>T and 429+4insg variants resulted in CD36 deficiency in HEK cells, which was confirmed by flow cytometry and Western blotting.
CONCLUSION
The 220C>T and 429+4insg variants can cause CD36 deficiency in human platelets. This system may be used for assessing the effect of 220C>T, 429+4insg, and other variants on the expression of CD36.

8.Effect of resilience intervention on rehabilitation of traumatic brain injury: a systematic review

ObjectiveTo systematically review the effect of resilience intervention on rehabilitation of traumatic brain injury. MethodsThe literatures related to resilience intervention on rehabilitation outcome of patient with traumatic brain injury were retrieved from PubMed, ScienceDirect, CNKI, Wanfang data and VIP databases through subject headings retrieval. The retrieval time was from establishment to December 31th, 2021. After filtering, the research, country, research object, research design, intervention frequency, intervention effect, rehabilitation impact and influencing factors were extracted from the literature, and the methodological quality was evaluated using the PEDro scale. ResultsA total of six valid literatures were obtained, which mainly focused on the research of resilience intervention on physical function, post-traumatic stress disorder, psychological function and social adaptation of patients with traumatic brain injury. ConclusionResilience intervention could reduce the patients' physical symptoms and post-traumatic stress disorder, and promoted the recovery of psychological functions, allowing them to maintain an optimistic attitude, and adopt flexible and effective coping style to adapt to the society.
9.Improvement of Anti-CD36 Antibody Detection via Monoclonal Antibody Immobilization of Platelet Antigens Assay by Using Selected Monoclonal Antibodies

Antibodies against human CD36 are responsible for several immune-mediated disorders. The detection of anti-CD36 antibodies using the standard monoclonal antibody (mAb) immobilization of platelet antigens (MAIPA) assay is hampered by a high frequency of false-negative results, most likely due to competitive inhibition of the mAb used as the capture antibody. We generated a panel of mouse mAbs against CD36 and seven hybridomas (GZ-3, GZ-13, GZ-70, GZ-143, GZ-413, GZ-507, and GZ-608), which were selected for MAIPA assays, as they reacted with mouse and human CD36. Fourteen anti-CD36 sera were assayed; all of which showed a positive reaction in a PakPlus (Immucor GTI Diagnostics, Inc., Waukesha, WI, USA) ELISA-based screening (optical density: 0.257–2.292). When the reference anti-CD36 mAb FA6-152 was used in the MAIPA assay, only 6/14 (42.9%) sera displayed a positive reaction. In contrast, anti-CD36 antibodies were detected in 13/14 (92.9%) sera when GZ-70 and GZ-608 mAbs were used. This significant improvement resulted in the identification of anti-CD36 antibodies by an antigen capture assay. Since patient’s platelets possibly carrying rare native antigens are used, this method will facilitate the identification of new platelet antibodies against CD36 that are involved in immune-mediated thrombocytopenia and other diseases, such as transfusion-related acute lung injury.
10.Research progress in hip-preserving modalities for early osteonecrosis of the femoral head in young and middle-aged patients

Osteonecrosis of the femoral head (ONFH) is one of the common and difficult-to-treat orthopedic diseases caused by a variety of factors that lead to abnormal blood flow to the femoral head, which in turn leads to deformation and collapse of the femoral head and eventually results in severe hip joint dysfunction. The key to the treatment is early diagnosis and correct treatment according to the stage classification and active prevention of further aggravation of ONFH aiming to delay or avoid hip replacement surgery in young and middle-aged patients. At present, there are various non-surgical and surgical hip-preserving modalities for early ONFH, designed to slow down the progression of the disease, prevent the femoral head from collapsing and stop the mild collapse. In recent years, with the emergence and development of bone reconstruction biomaterials, artificial bone reconstruction after scraping of ONFH lesions has shown great potential in the treatment of early ONFH. The authors review the research progress in hip-preserving modalities for early ONFH in young and middle-aged patients from non-surgical and surgical perspectives, hoping to provide a reference for clinical treatment of early ONFH.
