1.The characteristics of epithelial-mesenchymal transition in human prostate cancer stem progenitor cells

Objective To implore the characteristics of epithelial-mesenchymal transition(EMT) in human prostate cancer stem progenitor (S/P) cells isolated from LNCaP cell lines.Methods The S/P cells were obtained through florescence-activated cell sorting (FACS).Western blot and immunofluorescence assay were used to detect the S/P cells' EMT markers expression,such as E Cadherin,N Cadherin,Vimentin and Snail.Soft agar assay was used to detect the tumorigenesis ability of S/P cells.Cell migration assay was used to detect the migration ability of S/P cells.Results Compared with non S/P cells,the expressions of EMT markers,such as N Cadherin,Vimentin and Snail,were increased in S/P cells,while the expressions of epithelial marker and E Cadherin were decreased in S/P cells.After cultured for three weeks,S/P cells and non S/P cells both clonally grew.The colony numbers were (18.34±1.21) and (82.27±7.54),respectively (t =8.617,P =0.001).After cultured for 48 hours,the migration cells number was (25.33±5.13) in non S/P cells and (74.33±7.64) in S/P cells (t =7.953,P =O.001).Conclusions Human prostate cancer S/P cells isolated from LNCaP cell line have some characteristics of EMT,such as stronger tumorigenesis and migration ability,which could promote tumor invasion and metastasis.
2.Influence of Yifeiqinghua Granules on Expression of Vascular Endothelial Growth Factors and Its Receptor KDR in Lewis Lung Cancer Mice

Objective To clarify the mechanism of Yifeiqinghua granules on tumor angiogenesis and tumor inhibition by observing its influence on the expression of vascular endothelial growth factor (VEGF) and its receptor KDR in Lewis lung cancer mice. Methods Eighty C57BL/6 inbred mice subcutaneously planted with Lewis tumor cell suspension 0.2 mL were randomly divided into 8 groups:blank group, control group, low-, medium- and high-dose of Yifeiqinghua granules groups, Gefitinib group, Gefitinib plus medium-dose of Yifeiqinghua granules group and CTX group, 10 mice in each group. Drugs were administrated from the second day. CTX group was administered on third day and seventh day, other groups were administered for 14 days. The mice were killed to take the tumor on the 15th days. VEGF and KDR expression in tumor tissue was measured by Western Blot. Results Compared with the blank group, the expression of VEGF was significantly decreased in the low- and medium-dose of Yifeiqinghua granules groups (P<0.01). Gefitinib group’s VEGF value was lower than the blank group (P<0.05). Compared with the blank group, the expression of VEGF was significantly decreased in the high-dose of Yifeiqinghua granular group, Gefitinib plus medium-dose of Yifeiqinghua granules group and CTX group (P<0.001). Compared with the blank group, the expression of KDR was significantly decreased in the low-, medium- and high-dose of Yifeiqinghua granules groups, Gefitinib group and CTX group (P<0.05). Conclusions The possible mechanism of Yifeiqinghua granules in treating non-small cell lung cancer is reducing the expression of VEGF and KDR, thus play an inhibitory effect.
3.The Applied Study of Postprocessing for ~(31)P MRS of Human Liver in Vivo

Objective To investigate the best protocol of liver 31 P MR spectroscopy . Methods The 31 P MRS was performed using 1.5T MR scanner,heart/liver surface coil and prone position with Resp/Trigger gate in the normal volunteer and the cases with liver diseases including hepatic cirrhosis, hepatocellular carcinoma and benige hepatic tumour. The age ranged From 15 to 58 years old. The related data were processed and compared using several kinds of protocol methods.Results By using the edit skills with Filter, Zero-filling and Curve fitting under the type of J-coupling peak, the curves corresponding with the request and precise data had been obtained for 31 P MRS of the liver. By the simple protocol methods, the 31 P MRS, concentration and ratio of the materials in the liver could be obtained in all cases.Conclusion The process of MR spectroscopy is easy to apply. The requested curves and the accurate data will be obtained by using the editing skill of edit.
4.Study on adscription of plasma effective constituents of rat after administrated with alkaloid fraction of Nelumbo nucifera

Objective: To study the adscription of plasma effective constituents of rat after oral administration of alkaloid fraction of Nelumbo nucifera.Methods: Based on the established HPLC analytical method of plasma effective constituents,analysis and comparison were carried out among HPLC profi les of plasma samples obtained after oral administration of alkaloid fraction of Nelumbo nucifera,blank plasma,alkaloid fraction of Nelumbo nucifera.Results: Sixteen compounds were detected under the method,three of them were architypes of compound contained in alkaloid fraction of Nelumbo nucifera,and the others were metabolites.Through the comparison of the UV spectra,seven metabolites remain essential structure because the max wavelength was same to the architypes’,the other six metabolites’ structure changed greatly.Conclusion: Through the study of the adscription of plasma effective constituents of rat after oral administration of alkaloid fraction of Nelumbo nucifera,the structure of metabolites can be conjectured,which may be helpful to study the ADME and mechanism of action.
5.Characteristics of CD8+ stem memory T cells in patients with chronic HIV-1 infection during antiretroviral therapy

Objective To investigate the characteristics of CD8+ stem memory T cells (CD8+Tscm) in patients with chronic HIV-1 infection before and after antiretroviral therapy (ART) and to analyze their associations with progression of HIV-1 infection.Methods Thirty-six patients with chronic HIV-1 infection and 20 healthy subjects were enrolled in this study.Flow cytometry was performed to detect the percentages and absolute numbers of CD8+Tscm in patients with chronic HIV-1 infection before and after antiretroviral therapy (ART) as well as in healthy subjects.Correlation analysis was used to demonstrate the relationships between CD8+Tscm and markers for progression of HIV-1 infection (CD4+T cell count, HIV-1 viral load and level of activated T cells).Results The percentages and the absolute numbers of CD8+Tscm in patients with chronic HIV-1 infection had no significant change before and after ART.They were respectively positively correlated with the percentages and the absolute numbers of CD4+Tscm.The percentage of CD8+Tscm was proportional to the percentage of CD8+ central memory T cells (CD8+Tcm), but was inversely proportional to the percentage of CD8+ effector memory T cells (CD8+Tem).In addition, the percentages of CD8+Tscm in patients with HIV-1 infection were negatively correlated with the viral loads before ART.Conclusion CD8+Tscm are responsible for maintaining the homeostasis of other CD8+T cell subsets.CD8+Tscm play an important role in inhibiting viral replication.
6.A new aporphine alkaloid in leaves of Nelumbo nucifera

Objective To study the chemical constituents in the leaves of Nelumbo nucifera. MethodsCompounds were repeatedly purified by chromatography and structures were elucidated by physicochemical properties and spectroscopic analyses. Results Nine alkaloids were isolated and identified as 2-hydroxy-1-methoxy-6-methyl-6a,7-dehydroaporphine (1),armepavine (2),dehydroroemerine (3),dehydronuciferine (4),2-hydroxy-1-methoxyaporphine (5),liriodenine (6),pronuciferine (7),roemerine (8),and nuciferine (9). Conclusion compound 1 is a new aporphine alkaloid named nelumnucine.
7.The regulation of TGF-β1 in cardiac death caused by variation of SCN5A gene

The mechanism of sudden cardiac death caused by variation in SCN5A is still unclear. Recently, the converging evidences suggest that the dysfunction of regulation mediated by transforming growth factor-β1 in cardiac fibration and reconstruction of cardiac iron channel could be main reason of SUNDS caused by variation of SCN5A. The resent progress of the mechanism of transforming growth factor-β1 in sudden cardiac death caused by variation of SCN5A gene is reviewed in this paper, hoping to provide reference for the research and practice of sudden cardiac death in forensic medicine.
8.Application of stone basket in ureteroscopic holmium laser lithotripsy for treatment of upper ureteral calculi

Objective To investigate the application effect of stone basket in ureteroscopic holmium laser lithotripsy for treatment of upper ureteral calculi. Methods The clinical data of 96 patients with upper ureteral calculi were retrospectively analyzed, all patients underwent ureteroscopic holmium laser lithotripsy. Forty-eight cases used the stone basket in the operation process (observation group), and 48 cases did not use the stone basket in the operation process (control group). The operation time, length of stay, success rate of lithotripsy, stone residual rate and incidence of postoperative complication were compared between 2 groups. Results The patients of 2 groups successfully completed surgery. There were no statistical differences in operation time, length of stay and incidence of postoperative complication between 2 groups (P>0.05). The success rate of lithotripsy in observation group was significantly higher than that in control group: 97.92% (47/48) vs. 75.00% (36/48), and the stone residual rate was significantly lower than that in control group:4.17%(2/48) vs. 18.75%(9/48), and there were statistical differences (P<0.05). Conclusions The stone basket in ureteroscopic holmium laser lithotripsy for treatment of upper ureteral calculi can thoroughly remove stones. It reduces the incidence of residual stones, does not affect the safety of the operation, and has good clinical value.
9.Study on extracting purified active components of Picria fel-terrae, Scutellaria baicalensis and Phellodendron chinense with macroporous resins

Object To study the application of macroporous resins to the purified active components of Picria fel-terrae Lour., Scutellaria baicalensis Georgi, and Phellodendron chinense Schneid. Methods With the purified active components of Chinese materia medica (CMM) as a standard, we selected the suitable macroporous resins and studied the optimum technological parameters of the adsorption and elution. Results The suitable three types of macroporous resins which were used to the purified active components of CMM were HPD500 for P. fel-terrae., HPD300 for S. baicalensis, and HPD100 for P. chinense. In the absorption course, when the ratios between the medicinal materials amount and the volume of macroporous resins were 1.5 for P. fel-terrae, 0.5 for S. baicalensis and 1 for P. chinense, the purified active components of CMM appeared leaking, and were no more absorbed when the ratios were 11 for P. fel-terrae, 2 for S. baicalensis and 4.75 for P. chinense. In the elution course, the optimum alcohol concentrations were 50% for P. fel-terrae, 30% for S. baicalensis and 50% for P. chinense. Conclusion It is obviously different to refine the active components of CMM, while using the diverse types of macroporous resins.
10.Selective arterial embolization for the treatment of arterial hemobilia after biliary tract surgery:an analysis of therapeutic results

Objective To explore the feasibility and efficacy of selective arterial embolization for the treatment of hemobilia occurred after biliary tract surgery.Methods The clinical data of 16 patients with massive hemobilia after biliary tract operation,who were treated in our hospital during the period from March 1989 to August 2009,were retrospectively analyzed.Conservative treatment of hemobilia was initially adopted in all patients,and,if it failed,interventional management was carried out.Emergent artery angiography was performed in all patients,which was followed by selective arterial embolization of the bleeding arteries with Gelfoam particles and coils.Results Of the total 16 cases,15 received selective arterial embolization therapy and the remaining one died.No re-bleeding occurred during a follow-up period of 8 months to 3 years.Conclusion For the treatment of arterial hemobilia occurred after biliary tract surgery,selective arterial embolization with Gelfoam particles and coils is a safe,mini-invasive and effective therapy with few complications.
