Based on the current situation and the application situation of Intranet and Internet technologies of Gansu Provincial Maternity and Child-care Hospital,the paper takes advantage of interconnection and interworking technologies of Intranet and Internet,constructs the day parting appointment registration and examination system in a variety of ways,introduces the overall design,deployment and implementation of the system,and summarizes the application experience.This system saves greatly the time for the queuing of patients,relieves the pressure of waiting for treatment,and improves the visiting experience of patients.
2.Deoxyadenosine antagonizes MTX-induced suppression of inflammatory bone destruction

Objective:To study the effects of deoxyadenosine(dAdo) on methotrexate (MTX) induced suppression of inflammatory bone destruction.Methods:The culture system of whole bone marrow cells (WBMCs) was utilized to evaluate osteoclastogenesis.TRAP staining and μCT analysis were utilized to evaluate osteoclastogenesis and bone destruction in adjuvant arthritis rats.Results:In the bone marrow culture system,MTX-induced suppression of osteoclastogenesis was abrogated by the addition of dAdo.dAdo canceled MTX-induced suppression of osteoclastogenesis in the rats with arthritis,and significantly abolished the therapeutic effects of MTX on inflammatory bone destruction in the rats.Conclusion:The accumulation of dAdo may be one cause of the losing effectiveness of MTX in bone destruction.
3.Diagnostic Significance of BAT in Anaphylaxis to Non-ionic Contrast Media.

OBJECTIVE:
To investigate the diagnostic significance of basophil activation test (BAT) in anaphylaxis to non-ionic contrast media through testing the content of CD63, mast cell-carboxypeptidase A3 (MC-CPA3), and terminal complement complex SC5b-9 of the individuals by testing their levels in the normal immune group and the anaphylaxis groups to β-lactam drugs and non -ionic contrast media.
METHODS:
The CD63 expression of basophilic granulocyte in blood was detected by flow cytometry. The levels of MC-CPA3 in blood serum and SC5b-9 in blood plasma were detected by ELISA.
RESULTS:
The CD63 expression of basophilic granulocyte in blood, the levels of MC-CPA3 and SC5b-9 of anaphylaxis to non-ionic contrast media and β-lactam drugs were significantly higher than that in normal immune group (P < 0.05).
CONCLUSION
There is activation of basophilic granulocytes, mast cells and complement system in anaphylaxis to non-ionic contrast media. BAT can be used to diagnose the anaphylaxis to non-ionic contrast media.

4.Preliminary studies on the nutrient characters of different kinds of Chinese goldthread soil.

OBJECTIVETo study the changes of nutrient contents in Chinese goldthread soil before and after planting Coptis chinensis.
METHODThe soil in Shizhu, Chongqing, was analyzed for nutrient contents.
RESULT AND CONCLUSIONThe results indicated that the preplanted soil was strongly acidic and the levels of organic matter, total nitrogen and potassium were high. However, the content of total phosphorus was very low; so were the contents of other eleven nutrients. Variations of nutrients except zinc and molybdenum were large. After four years of planting C. chinensis, the soil pH was increased. Contents of organic matters, manganese, zinc and boron were increased, while the contents of iron and copper were decreased. In general, the variance coefficient of all nutrient elements and the difference of soil fertility were decreased.

5.Quantitative study of effects of cinnamaldehyde on levels of endotoxin in root canals in rats' periapical periodontitis model.

OBJECTIVETo provide experimental evidence for the exploitation of cinnamaldehyde as a kind of root canal disinfectant through studying the effect of cinnamaldehyde on endotoxin in root canals.
METHODSThis experimental model of periapical periodontitis was established with Wistar rats. The 75 rats were divided randomly into 3 groups: Group of cinnamaldehyde, group of formaldehyde cresol formocresol, group of physiological saline. The level of endotoxin was measured by quantitative chromogenic tachypleus amebocyte lysate method before and after sealing the drugs in the root canal.
RESULTSThe level of endotoxin in the group of cinnamaldehyde and formaldehyde cresol formocresol decreased obviously (P < 0.05), and the difference between them was of no significance (P > 0.05), the group of physiological saline was of no significant difference (P > 0.05).
CONCLUSIONCinnamaldehyde can decrease the level of endotoxin obviously.

6.Synchronous Ectopic Pancreatoblastoma in a Child: A Case Report.

Pancreatoblastoma is a rare primary pancreatic neoplasm of children that may arise in any portion of the pancreas. We report a case of a 3-yr-old boy who presented to with abdominal pain our hospital and a progressive bulge in his right abdomen. Biochemical evaluation and serum levels of tumoral markers were within reference limits. On the computed tomography, two tumors were found. One located in the head of the pancreas; however, a laparotomy revealed that the head of pancreas was compressed but normal. The other was in the left abdomen near the spleen and the tail of the pancreas. The diagnosis of two synchronous pancreatoblastoma originating from the omentum was confirmed by pathology. Therefore, a pancreatoblastoma should be considered when a large well-defined, lobulated, and heterogeneous mass is identified in the pancreas of children. In addition, an ectopic pancreatoblastoma should be considered when identified within or near the ectopic pancreatic tissue.

7.Optimization of conditions for supercritical fluid extraction of flavonoids from hops (Humulus lupulus L.).

Waste hops are good sources of flavonoids. Extraction of flavonoids from waste hops (SC-CO(2) extracted hops) using supercritical fluids technology was investigated. Various temperatures, pressures and concentrations of ethanol (modifier) and the ratio (w/w) of solvent to material were tested in this study. The results of single factor and orthogonal experiments showed that at 50 degrees C, 25 MPa, the ratio of solvent to material (50%), ethanol concentration (80%) resulted in maximum extraction yield flavonoids (7.8 mg/g). HPLC-MS analysis of the extracts indicated that flavonoids obtained were xanthohumol, the principal prenylflavonoid in hops.

8.Construction of an adeno-associated virus vector expressing CTLA-4Ig and its expression in the transplanted liver allografts.

OBJECTIVESTo construct a recombinant adeno-associated virus vector pSNAV expressing CTLA-4Ig and to demonstrate its expression in transplanted liver allografts and to see if a long term inhibitive effect of CTLA-4Ig could be obtained though its use.
METHODSAfter AAVCTLA-4Ig and PUC18 were cut with BamHI, CTLA-4Ig cDNA was inserted into the plasmid PUC18 by T4DNA ligase and PUC18-CTLA-4Ig was constructed. The obtained PUC18-CTLA-4Ig and pSNAV cut with Kpn I and EcoR I, CTLA-4Ig cDNA was inserted into plasmid pSNAV to construct the recombinant vector pSNAV-CTLA-4Ig, which was transfected into BHK-21 packaging cells by lipofectine-mediated transfection. Then the BHK-21 cell line was infected with HSV1-rc to produce a large amount of pSNAV- CTLA-4Ig. The specificity of the expressed product was identified by digestion with BamHI, PCR and sequence determination. The titer of the virus was detected. The product was infused into rats liver allografts via portal vein and its expression in the transplanted livers was detected immunohistochemically.
RESULTSRecombinant adeno-associated virus vector pSNAV-CTLA-4Ig was generated and purified into 8.5 x 10(11)/ml. Agarose gel analysis of PCR products verified the presence of CTLA-4Ig. Digestion with BamHI and sequence determination confirmed that pSNAV-CTLA-4Ig was constructed. Expression of CTLA-4Ig in the transplanted livers was detected successfully.
CONCLUSIONPrepared pSNAV-CTLA-4Ig was constructed correctly and can express CTLA-4Ig effectively. Besides this, it can express CTLA-4Ig in rat liver allografts. It may be used in the study of transplant tolerance.

9.Construction of PCI-neo mammalian expression vector system containing murine 4-1BBL gene and its stable expression in hepatocellular carcinoma cell line.

OBJECTIVETo study the construction of the PCI-neo mammalian expression vector system containing murine 4-1BBL gene and its stable and effective expression in rat hepatocellular carcinoma cell line CBRH7919.
METHODSThe murine full-length 4-1BBL cDNA was obtained and subcloned into the PCI-neo mammalian expression vector. The recombinant named as PCI-neo-4-1BBL was identificated by restriction enzyme digestion and sequencing. Subsequently, PCI-neo-4-1BBL was transfected into CBRH7919 with lipofectamine reagent, then G418-resistance clone was acquired and named as PCI-neo-4-1BBL-CBRH7919+. The stable expression of 4-1BBL mRNA in PCI-neo-4-1BBL-CBRH7919+ was detected by RT-PCR.
RESULTSThe fragment of 980 bp (4-1BBL) and 5.4 kb (PCI-neo) was shown after PCI-neo-4-1BBL had been digested by EcoR I and Not I and agarose gel electrophoresis. The DNA sequencing of 4-1BBL, proved to be identical to the data of 4-1BBL in Genebank, showed stable and effective expression in PCI-neo-4-1BBL-CBRH7919+.
CONCLUSIONThe PCI-neo mammalian expression vector system containing murine 4-1BBL has been constructed successfully, which shows stable and effective expression in CBRH7919.

10.Drilling and evaluation of emergency rescue against mass casualties at general hospitals in Tianjin

Objective To understand the ability and level of emergency rescue at general hospitals in Tianjin city. Methods Such actions as formulating plans and examination forms, establishing assessment indicators and evaluation criteria, and simulation exercises were performed to evaluate the capacity of 28 general hospitals in terms of their organizational structure, emergency response, event reporting, and summary assessment. Results The emergency response assessment system consisted of 4 level-1 indicators, 19 level-2 indicators and 58 level-3 indicators. 28 hospitals were found high in their overall emergency response capacity, but some were found with setbacks. For example, the " organizational structure" scored the highest in 4 first-level indicators, up to 88. 91% , while " incident report" scored the lowest, down to 67. 99% . Among level-2 indicators, professional emergency professional procedures and initial reporting scored the lowest. Conclusions In order to further improve the ability of medical institutions to respond to emergency events, the hospitals are recommended to enhance their backup resources for emergency response, their staff′s awareness of first aid knowledge and first aid skills, as well as their timeliness of initial reports and the completeness of progress reports.
