3. Mechanism of the cardiovascular action of salusin-a within caudal ventrolateral medulla in rats
Chinese Pharmaceutical Journal 2012;47(18):1473-1477
OBJECTIVE: To determine the cardiovascular actions of salusin-α in the caudal ventrolateral medulla (CVLM) in anesthetized rats. METHODS: Sixty-one anesthetic male SD rats were employed in the present study. The dose-dependant effects of salusin-α (0.04-4 pmol) on blood pressure and heart rate in the CVLM were determined by unilateral microinjection of salusin-α or artificial cerebrospinal fluid (aCSF) into the CVLM in 25 rats. In the other 36 rats, a CSF, kynurenic acid (Kyn), atropine, hexametho-nium (Hex) in the CVLM or a CSF/muscimol in the RVLM were given in advance to determine the mechanism of the cardiovascular actions of intra-CVLM salusin-α. RESULTS: Unilateral microinjection of salusin-a into the CVLM produced a dose-dependent hypotension and bradycardia. Prior administration of Kyn (1 nmol) or Hex (120 pmol) into the CVLM did not alter the hypotension and bradycardia induced by intra-CVLM salusin-α (P>0.05). But prior administration of atropine into the CVLM or pretreatment with muscimol within RVLM almost completely abolished the hypotension and bradycardia evoked by intra-CVLM salusin-α (P<0.05). CONCLUSION: Microinjection of salusin-α into the CVLM produces significantly hypotension and bradycardia, which probably originates from suppressing the activities of presympathetic neurons in the RVLM.
4.Adenoviral-mediated somatostatin receptor subtype 2 gene transfer inhibits pancreatic cancer xenografts.
Chinese Journal of Practical Internal Medicine 2002;0(08):-
Objective To construct recombinant adenovirus carrying the gene of human somatostatin receptor subtype 2(SSTR2),to investigate its effect on growth of human pancreatic cancer xenografts in nude mice, and to further elucidate the underlying mechanisms. Methods The recombinant adenovirus coding for human SSTR2(Ad-SSTR2)and reporter gene LacZ(Ad-LacZ)were generated by site-specific recombination from Jun 1,2004 to Dec 10,2005.Human pancreatic cancer cell line was implanted subcutaneously into nude mice;normal saline(control group),Ad-LacZ(reporter gene control group)and Ad-SSTR2(experimental group)were injected into pancreatic cancer xenografts, respectively. The tumor volume and weight were measured.RTPCR and Western blot were used to determine the expression of SSTR2 after pancreatic cancer xenografts were infected with Ad-SSTR2.The expression level of ERK2 and ras proteins were assessed by Western blot. Results The virus titer of Ad-SSTR2 and Ad-LacZ was 6.0?1012pfu/L and 6.5?1012pfu/L,respectively.SSTR2 mRNA and protein were detected after Ad-SSTR2 infected pancreatic cancer xenografts.Growth of pancreatic cancer was significantly inhibited in the experimental group as compared with the control group. The growth inhibitory rate was 48.2%(P

Objective: To study the correlation of blood stasis syndrome or its accompanied syndromes with Gensini score in patients with coronary heart disease (CHD) in stable condition. Methods: The syndrome types of traditional Chinese medicine (TCM) and blood stasis score in 131 CHD patients confirmed by coronary angiography were recorded. Gensini score was calculated according to the coronary pathological characteristics showed by angiography. The correlations of blood stasis syndrome and its accompanied syndromes with coronary lesion and Gensini score were analyzed. Results: Among the TCM syndrome types, blood stasis, turbid phlegm and qi deficiency were the most common syndromes, revealed in 85 patients (64.9%), 83 patients (63.4%) and 85 patients (64.9%), respectively. The coronary lesion length and Gensini score in the patients with blood stasis syndrome were much higher than those in the patients with non-blood stasis syndrome (P<0.05 or P<0.01). In the subtypes of blood stasis, the coronary lesion length and Gensini score in the patients with blood stasis accompanied by turbid phlegm syndrome were higher than those in the patients with non-blood stasis syndrome (P<0.05). And in the patients whose blood stasis syndrome score was more than 9 points, the coronary lesion length was higher than that in the patients whose blood stasis syndrome score was less than 9 points (P<0.05). Besides, with bivariate analysis, the blood stasis syndrome score showed no correlation with Gensini score (Pearson correlation coefficient was 0.104, P=0.241). Conclusion: Blood stasis syndrome is the most common TCM syndrome in CHD patients in stable condition. The blood stasis syndrome score is proportional to coronary lesion length, and reflects the severity of coronary lesion.

Objective To investigate the clinical symptoms and imaging features of Budd-Chiari syndrome with hepatic vein obstruction (HVBCS) and the reasons of mistreatment. Methods Thirteen patients with HVBCS were misdiagnosed and mistreated as inferior vena cava (IVC) obstruction, including 8 patients treated with stent implantation in IVC once and 5 patients with balloon dilatation. After analysis of the clinical symptoms, signs and imaging features; hepatic vein obstruction was further confirmed by digital subtraction angiography (DSA)in all patients. Results All patients had variable degrees of portal hypertension and no apparent symptoms of IVC obstruction. CT or / and MRI showed obvious caudate lobe enlargement and DSA showed IVC narrowing with external compression. All patients were undertaken hepatic vein angiography including 4 with PTV and 9 with hepatic vein stent implantation. All patients' clinical symptoms and signs completely disappeared or markedly improved after the procedure. Conclusion The stenosis of IVC in HVBCS,caused by compression compensatory hypertrophy of hepatic caudate lobe can be cured by hepatic vein angioplasty which is the most correct and effective method.

Objective To study anti-radiation behavior of plasma sprayed boron carbide coatings. Methods The anti-radiation capacity of 16Mn steel which was coated with 0.1 mm plasma sprayed boron carbide were studied. The irradiation beams were 6,10,15 MY X-ray and 6,9,12,15 MeV electron emitted by accelerator, X-ray emitted by 60Co machine, fast neutron, and X-ray emitted by kilovoltage X-ray ma-chine. Results Anti-radiation capacity of plasma sprayed boron carbide coatings was not found for X-ray beams emitted by accelerator and 60Co machine. For electron beams, the anti-radiation capacity were found. The deeper of location, the stronger was anti-radiation. However, the anti-radiation capacity was not good. For fast neutron, the anti-radiation capacity was good. For X-ray emitted by kilovoltage X-ray machine, the anti-radiation was good, and only 0.1 nun plasma sprayed boron carbide had 15% attenuation. Conclusions The plasma sprayed boron carbide coatings have the anti-radiation capacity for X-ray emitted by kilovoltage X-ray machine in medical field.
