2.Effects of propofol on noradrenaline release from the locus coeruleus in rabbits

Objective To investigate the effects of propofol on the release of noradrenaline ( NE) from the locus coeruleus in the brain of rabbits trying to elucidate the central mechanism of the cardiovascular inhibition induced by propofol.Methods Nine healthy male New Zealand rabbits weighing 2.0-2.5 kg were used in this study. A trocar (0.8 mm in diameter) was inserted into locus coeruleus using the stereotactic technique and fixed. Four days later push-pull perfusion of the brain was performed. 37℃ artificial cerebrospinal fluid (aCSF) was infused through the trocar at 0.1 ml?min-1 . A loading dose of propofol 2 mg?kg-1 was given i.v. followed by continuous infusion at 150 ?g?kg-1?min-1 for 30 min. The experiment was concluded at 20 min after propofol infusion. The perfusate having passed through the locus coeruleus was collected before and every 10 min during and after infusion. The NE concentration of the perfusate was measured by high performance liquid chromatography. Results The NE concentration of the perfusate from locus coeruleus significantly decreased after the loading dose and during the infusion of propofol and reached its bottom level at 10 min after loading dose. The maximal decrease was 75.5% [from (15.9 ? 3.2) pg??l-1 to (3.9?0.5) pg ? ?l-1]. Conclusion Intravenous propofol decreases the NE concentration in locus coeruleus. The cardiovascular inhibition induced by propofol may partly be explained by this central mechanism.
3.Application of multiple gene methylations in plasma for diagnosis of lung cancer

[ ABSTRACT] AIM: To determine the aberrant methylation status in the gene promoter regions of CDH13, RASSF1A, DLEC1, SEPT9 and RUNX3 by detecting the plasma specimens and the value of their combined detection for di-agnosis of lung cancers.METHODS:Nest methylation specific PCR ( nMSP) was used to detect the promoter methylation status of the 5 genes in the plasma from 106 normal controls, lung cancer tissues, lung benign tissues and the plasma from 106 patients with lung cancers.Multiple displacement amplification ( MDA) was used to amplify modified genomic DNA to solve the problem of insufficient of plasma DNA template.RESULTS: The positive rates of promoter methylation of CDH13, RASSF1A, DLEC1, SEPT9 and RUNX3 in the lung cancer tissues were 51.9%, 44.3%, 54.7%, 36.8%, 24.5%, respectively, and those in the plasma were 46.2%, 41.5%, 50.9%, 31.1%, 19.8%, respectively.The re-sults of the Kappa consistency check showed that the lung cancer tissues and the plasma had obviously coherence in the methylation status of the 5 gene promoter regions.Combination of DLEC1, CDH13, RASSF1A, and SEPT9 had a higher di-agnostic efficiency than the others, as their ACC value was 0.8208 and youden index was 0.6415 ( with the sensitivity of 81.13% and the specificity of 83.02%) .CONCLUSION:Combination detection of promoter methylation of lung cancer-related genes in the plasma is expected to apply to the early diagnosis of lung cancer.
4.Analogy of CRISPR-Cas systems to mammalian immune systems and research progress in their functions

The immune system of bacteria against phage shares a lot of similarity with that of mammals, especially the adaptive immune system.The elements and components of the bacterial adaptive immune system———clustered regularly interspaced short palindromic repeats ( CRISPR ) and the mammalian adaptive immune system have a lot of parallel mechanisms.We could acquire new understanding about the immune function of CRISPR systems through that analogy.In recent years, researchers have found CRISPR-Cas system can play significant roles in regulating bacterial growth and metabolism.These researches have revealed new functions of CRISPR beyond immunity.The ability of CRISPR to affect gene expression has attracted increasing attention.Further studies are needed to shed light on the complicated functions of CRISPR.
5.Topping-off technique for the protection of degenerative intervertebral disc

BACKGROUND:Topping-off technique can be used for fixation treatment through the combination of fusion and interspinous dynamic device, in order to prevent or slow down the adjacent lumbar segment degeneration.
OBJECTIVE:To obverse the protective effect of Topping-off technique (posterior lumbar interbody fusion
procedure combined with the fixation of dynamic interspinous device Coflex) for the degenerative intervertebral disc. METHODS:A total of 32 patients with degenerative lumbar diseases who had been treated with Topping-off technique were included in this study. The Oswestry disability index, the Japanese Orthopaedic Association scores, range of motion for Coflex implanted segment and during the relative signal intensity of the Coflex implanted segment in MRI image were recorded and calculated preoperatively and the entire fol ow-up period.
RESULTS AND CONCLUSION:Al patients were fol owed-up for 20.6 months averagely. Up to the last fol ow-up, the Oswestry disability index and Japanese Orthopaedic Association scores were significantly improved when compared with those before treatment (P<0.001). There was no significant difference in the range of motion for Coflex implanted segment before and after treatment (P=0.19). The relative signal intensity of the Coflex implanted segment was significantly improved when compared with that before treatment (P<0.01). The clinical application of the Topping-off technique showed a protective effect on the intervertebral disc.

7.OBSERVATION ON SPECIFIC IgG4 ANTIBODY OF SCHISTOSOMIASIS PATIENTS BEFORE AND AFTER TREATMENT

Objective] To observe the alteration of specific IgG4 antibody of schistosomiasis patients before and after treatment. [Methods] ELISA. [Results] The SEA IgG4 and AWA IgG4 positive rates of 27 schistosomiasis cases were 96 3% and 100%, respectively,their average OD values were 1 62 and 0 72. 6 months post treatment 18 cases were followed up, the positive rates were 94 4% and 100%, respectively, their average OD values were 1 06 and 0 56, respectively. 12 months post treatment all cases were followed up, the positive rates of SEA IgG4 and AWA IgG4 were 96 3% and 92 6%, resepectively ,their average OD values were 0 99 and 0 58, respectively. [Conclusion] No obvious changes were found in the SEA IgG4 and AWA IgG4 positive rates of 27 schistosomiasis cases before and after treatment, whereas the antibody level of specific IgG4 was decreased.
8.Association of oxygen saturation of arterial hemoglobin with late-onsethypertension in Chinese Han population resided in the Daxinganling area

Objective To identify the association of oxygen saturation of arterial hemoglobin (SaO2) with late-onset hypertension in the Chinese Han population located in the Daxinganling area.Methods A total participants were selected by convenience sampling methods from the Daxinganling area.All data were collected from each person by the questionnaire record of physical examinations as well as biochemical index measuring.SaO2 was noninvasively measured with finger pulse oxymetry,the reported SaO2 was the average of three readings taken 10 seconds apart.Results There were significant differences for SaO2 within the population of individuals,the mean SaO2 values was 97.71%±6.14%,with range from 88% to 100%.There was association of SaO2 with sex,BMI and age.SaO2 level declined with BMI and age increasing.Particularly,it was found that the risk increasing to hypertension was marked association with SaO2 rapid drop.During the period from 40-50 years of age,SaO2 declined from 97.85% to 97.64%,The risk to hypertension increased more than 10 times(P<0.001).That implicated hypoxia mightinvolve in the etiology of hypertension.Conclusions The preliminary results demonstrated the rapid decline of SaO2 with lapse of age may be one of the major risk factors to hypertension,it may be helpful to explain late-onset hypertension to some extent at least.

10.Clinical application of interventional MR with optical navigating

Objective To introduce the procedures and to evaluate the application value of the optical navigating interventional MR-guided percutaneous biopsies and therapies. Methods Thirty-five of 54 patients (33 males, 21 females) underwent percutaneous biopsies and 19 of 54 patients underwent percutaneous therapies under optical navigating interventional MR-guidance. The age ranged from 9 months to 78 years (mean 38.8 years). All patients were performed under a 0.23 T open MRI system (Proview, Philips Medical System) with optical navigating system (iPath 200, Philips Medical System). The needle and bone biopsy drill were MR-compatible (Daum, Germany). Results Punctures were accurate in all patients (54/54). 33 of 35 biopsies got the pathological diagnosis. All therapies (19/19) got the definitive curative effect. All patients had no obvious complications, such as severe hemorrhage, viscera or nerve injury.Conclusion iPath 200 interventional MR-guided percutaneous biopsies and therapies are accurate and safe techniques.
