The traditional culture methods and the molecular biology methods were used to study the rhizosphere bacterial diversity between fusarium wilt resistant and susceptible watermelon. The results showed that the diversity and the equality of cultured rhizosphere bacteria of resistant watermelon were higher than those of the susceptible watermelon. The reason was that the cultured rhizosphere bacterial di- versity index H′ and 1/D of the resistant watermelon were higher than those of the susceptible watermelon and that the cultured rhizosphere bacterial equality index E of the resistant watermelon were higher than those of the susceptible watermelon. The dominant cultured bacterial genotypes were different between re- sistant and susceptible watermelon. The genotype 1 is the dominant genotype of resistant watermelon, con- sists 51.1%. The genotype 7 is the dominant genotype of susceptible watermelon, consists 58.7%.

Objective To investigate the relationship between the residual forward blood flow and the therapeutic outcome in patients with acute cerebral infarction after arterial thrombolysis treatment. Methods During the period from January 2009 to December 2013 at authors’ hospital, a total of 40 patients with acute anterior circle cerebral infarction were treated with arterial thrombolysis. The clinical data were retrospectively analyzed. The patients were divided into group A (n=23) when residual forward blood flow and/or collateral circulation were presented, and group B (n=17) if no residual forward blood flow and/or collateral circulation were detected. The NIHSS scores and hemorrhagic transformation state were determined 14 days after the thrombolysis treatment, and the results were compared between the two groups. Results In group A, 21 cases (91.3%) showed complete or partial re- canalization of the infracted vessels, and asymptomatic hemorrhagic transformation was seen in one case. The pre-treatment NIHSS score was 12.69 ± 3.88 and the post-treatment NIHSS score was 6.05 ± 3.25. In group B, complete or partial re-canalization of the infracted vessels was seen in 15 cases (88.2%), asymptomatic hemorrhagic transformation was found in one case and symptomatic hemorrhagic transformation occurred in one case, who died of massive cerebral hemorrhage seven hours after the thrombolysis treatment. The pre-treatment NIHSS score was 13.51 ± 4.19 and the post-treatment NIHSS score was 8.68 ± 5.16. The differences between the two groups were statistically significant (P < 0.05). The obvious effect rate in group A and group B was 68.1% and 43.8% respectively, and the effective rate in group A and group B was 86.3% and 56.3% respectively, indicating that the clinical outcome of group A was much better than that of group B. Conclusion The therapeutic effect of arterial thrombolysis for acute cerebral infarction patients is not only closely related to the time window but also to the residual forward blood flow. The more the residual forward blood flow there is, the better the therapeutic result with less risk of hemorrhagic transformation will be.

Microbiology 1992;0(03):-
The sfa1 gene encoded a bifunctional enzyme with the activities of both alcohol dehydrogenase and glutathione-dependent formaldehyde dehydrogenase in Saccharomyces cerevisiae. The gene disruption cassette produced by PCR using the same long oligonucleotides which comprise 19 or 22 nucleotides complementary to sequences in the templates(pUG6 and pUG66 marker plasmid)at 3' end and 45 nucleotides at 5' end that annealed to sites upstream or downstream of the genomic target sequence to be deleted. After two linear disruption cassettes with a Cre/loxP mediated marker were transformed into the cells of Saccharomyces cerevisiae YS-1,the positive transformants were checked by PCR to correct the integration of the cassette and concurrent deletion of the chromosomal target sequence.Once correctly integrated into the genome, the select marker can be efficiently rescued by transformating the plasmid pSH47 into YS-1 and inducing the Cre expression with a Cre/loxP-mediated marker removal procedure. The expression of the Cre recombinase finally resulted in the removal of the marker gene, leaving behind a single loxP site at the chromosomal locus. The diploid mutant YS-1-sfa1 was generated, which could enhance the output of ethanol with 8.0% by shaking culture in flask compared with the original strain YS-1.

In this paper, the number of rhizosphere and non-rhizosphere microbial organisms of fusarium wilt resistant and susceptible watermelon under soil culture and soilless substrate culture was studied by traditional culture methods. The results showed that, the number of rhizoshpere microorganisms was significantly higher than non-rhizosphere, and the number was changed with the stage of watermelon grow, the number was the lowest in seedling stage and increased with the watermelon grow, and achieved highest at the flowering and fruiting stage, decreased with the watermelon ageing. The fusarium wilt resistant of watermelon was correspondence with number of rhizosphere bacteria; the number of rhizosphere bacteria of resistant watermelon was higher than that of susceptible watermelon in each stage under soil culture and soilless culture. The fusarium wilt resistant of watermelon is no correspondence with number of rhizosphere fungi and actinomycete. The number of non-rhizosphere microbial organisms was changed in a small range in the whole growing stage. The non-rhizosphere bacteria have no significant change in the whole stage under soil culture and increased quickly under soilless substrate culture and decreased at the later stage. The non-rhizosphere fungi and actinomycete reached highest at the later stage under soil culture or soilless sub-strate culture.

Objective:To investigate the clinical features of direct and indirect carotid cavernous fistulas (CCFs).Methods:Patients with CCF treated in the Affiliated Hospital of Xuzhou Medical University from January 2010 to August 2020 were enrolled retrospectively. Relevant clinical data were collected, including the main clinical manifestations, neuroimaging features, and treatment methods. The clinical features of direct and indirect CCFs were compared. Results:A total of 31 patients were enrolled in the study, 29 (93.5%) had ocular symptoms, of which conjunctival hyperemia and edema ( n=24, 77.4%), exophthalmos ( n=19, 61.3%) and orbital murmur ( n=18, 58.1%) were most common. There were 23 patients (74.2%) in direct CCF group and 8 (25.8%) in indirect CCF group. The former had more history of head trauma (78.2% vs. 12.5%; P=0.002), more flow volume (high-flow CCFs: 100% vs. 37.5%; P<0.001) and more likely to cause orbital murmur (69.6% vs. 25.0%; P=0.043). Endovascular embolization was safe and effective. The common methods of endovascular embolization were EVAL glue combined with coil embolization ( n=18, 66.7%) and detachable balloon embolization alone ( n=6, 22.2%). Conclusion:Ocular manifestations are most prominent in patients with CCFs. Direct CCF is more common, usually with a history of head trauma, and the clinical and imaging features are more typical. Interventional embolization is the preferred treatment option for patients with CCF.
