To optimize the immunization strategy against HIV-1, a DNA vaccine was combined with a recombinant vaccinia virus (rTV) vaccine and a protein vaccine. Immune responses against HIV-1 were detected in 30 female guinea pigs divided into six groups. Three groups of guinea pigs were primed with HIV-1 DNA vaccine three times, boosted with rTV at week 14, and then boosted with gp140 protein at intervals of 4, 8 or 12 weeks. Simultaneously, the other three groups of animals were primed with rTV vaccine once, and then boosted with gp140 after 4, 8 or 12 weeks. The HIV-1 specific binding antibody and neutralizing antibody, in addition to the relative affinity of these antibodies, were detected at different time points after the final administration of vaccine in each group. The DNA-rTV-gp140 immune regimen induced higher titers and affinity levels of HIV-1 gp120/gp140 antibodies and stronger V1V2-gp70 antibodies than the rTV-gp140 regimen. In the guinea pigs that underwent the DNA-rTV-gp140 regimen, the highest V1V2-gp70 antibody was induced in the 12-week-interval group. However, the avidity of antibodies was improved in the 4-week-interval group. Using the rTV-gp140 immunization strategy, guinea pigs boosted at 8 or 12 weeks after rTV priming elicited stronger humoral responses than those boosted at 4 weeks after priming. In conclusion, this study shows that the immunization strategy of HIV-1 DNA vaccine priming, followed by rTV and protein vaccine boosting, could strengthen the humoral response against HIV-1. Longer intervals were better to induce V1V2-gp70-specific antibodies, while shorter intervals were more beneficial to enhance the avidity of antibodies.

Objective To establish a suitable evaluation index system to track implementation effect of clinical research program. Methods Delphi method was used to creat the evaluation index system. The weighted average method was adopted to determine the weight of each index. Results After two rounds of expert consultation, twenty seven evaluation indices were selected, including three first-class indices, eight second-class indices and sixteen third-class indices, and the weight of each index was determined. Conclusions The evaluation index system reflects the purpose of tracking clinical research to a certain extent. This index system is simple and easy to be used.

The primary processing is important links and closely related to the quality of traditional Chinese medicinal materials, and is not only cleaning of remove the non-officinal parts, drying for termination the physiological status of organisms, but also retaining the most active substances, decreasing the toxic components, and promoting the transformation among chemical ingredients through primary processing. So the traditional primary processing endows with characters, quality, specifications and properties of traditional Chinese medicine, and embodies some important science truth. The traditional primary processing method and technology systems are derived from the long-term practices and experiences, which are distinctive, colorful, diverse, and scientific, which are helpful to development and utilization of traditional Chinese medicine resources. This paper systemically expounds the research status of the Chinese medicine processing method, summarizes the problems in the primary processing of traditional Chinese medicinal materials research, and prospects its bright future.

The stability of melittin in different solvents (water, deoxygenated water, physiological saline, PBS, 50% ethanol, ethanol, glycerol)was studied and the results showed that the stability of melittin is not influenced by light, temperature and pH in 50% ethanol, which melittin can be completed dissolved when compared with ethanol and glycerol, in such, 50% ethanol was chosen as solvent storage when measured content of melittin. Then the effect of different concentrations of PBS, the pH of PBS and rat skin ho- mogenates were tested, and the results showed that melittin was degraded rapidly at low concentration solution and low ionic strength. Increasing pH of PBS and rat skin homogenate can accelerate the degradation of melittin. These researches provide an experimental ba- sis for further study of melittin.

Objective:Based on the investigation on the advantages of Traditional Chinese Medicine(TCM)participation in the field of basic public health services, exploring how TCM participates in public health. Methods:It searched for policies related to basic public health services and conducted a questionnaire survey on 2 995 people working in public health-related jobs in Beijing to conduct an empirical study on the advantages of Chinese medicine and the current status of participation.Results:2 982 valid questionnaires were collected, and practitioners in the field of public health services in Beijing had a high level of attention and recognition towards the participation of TCM.TCM is generally considered to have obvious advantages in basic public health services such as responding to epidemics, preventing and treating chronic diseases, and managing the health of Children 0-6 Years and the elderly. However,TCM shows overall shortcomings in emphasis, talent allocation, infrastructure construction, public perception and resource balance. Conclusion:To play the ascendant role and enhance the service capacity of TCM,basic public health services can be further developed by optimizing mechanisms related to the participation of TCM in disease prevention and control, chronic disease management, children's,and elderly health management.Basic public health services can enhance the service capacity of TCM from several aspects such as increasing financial support, strengthening the cultivation and management of TCM talents, and emphasizing grassroots and balanced resource allocation of TCM.
