1.Umbilical cord mesenchymal stem cells undergoing hypoxic preconditioning can differentiate into vascular endothelial-like cells

BACKGROUND:Incidence of diabetic lower extremity vascular disease is increasing, so how to improve blood vessels of the lower limbs and increase angiogenesis becomes the research focus. Umbilical cord mesenchymal stem cells have been employed clinical y via local intramuscular injection, but the specific therapeutic effect and mechanism are not clear.
OBJECTIVE:To investigate the effects of hypoxic preconditioning and cobalt chloride medium on the differentiation of umbilical cord mesenchymal stem cells into vascular endothelial-like cells in vitro.
METHODS:Umbilical cord mesenchymal stem cells were isolated and cultured, and then treated with different concentrations of cobalt chloride. Enzyme linked immunosorbent assay was used to detect levels of basic fibroblast growth factor and vascular endothelial growth factor gene in cellsupernatants, and 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide to detect cellproliferation. The safety of umbilical cord mesenchymal stem cells before and after cobalt chloride induction was detected using chromosome. The umbilical cord mesenchymal stem cells were cultured in Dulbecco’s modified Eagle’s medium/Ham’s nutrient mixture F-12 containing 10μg/L vascular endothelial growth factor, 10μg/L basic fibroblast growth factor and 10%fetal bovine serum, which were induced to differentiate into vascular endothelial-like cells. Endothelial-like cellphenotype CD31 and von Wil ebrand factor were identified before and after induction, through the observation of three-dimensional model of angiogenesis, the angiogenic capacity of umbilical cord mesenchymal stem cells was determined.
RESULTS AND CONCLUSION:Umbilical cord mesenchymal stem cells strongly expressed the surface marks. After the cobalt chloride induction, the proliferation of umbilical cord mesenchymal stem cells was positivelycorrelated with the time of induction. Based on the levels of vascular endothelial growth factor and basic fibroblast growth factor, the optimal concentration of cobalt chloride was 200 μmol/L. Chromosome detection showed the stability of cells after cobalt chloride induction. After induction, CD31 and von Wil ebrand factor were strongly expressed. Three-dimensional observation showed umbilical cord mesenchymal stem cells could be induced to form the lumen-like structure with different diameter sizes, and umbilical cord mesenchymal stem cells could be induced to differentiate into endothelial-like cells, and have a angiogenic capacity.

2.Vascularization of acidic fibroblast growth factor/partially deproteinised bone in repair of early-stage avascular necrosis of the femoral head in rabbits

BACKGROUND:Acidic fibroblast growth factor (aFGF) possesses good effects on vascularization and osteogenesis.But whether aFGF can promote the vascularization in animals with early-stage avascular necrosis of the femoral head (ANFH) remains unclear.OBJECTIVE:To investigate the vascularization of aFGF composited by partially depreteinized bone (PDPB) in repair of early-stage ANFH.DESIGN,TIME AND SETTING:A randomized,controlled,animal experiment was performed in the College of Life Science,Nanhua University between January 2008 and January 2009.MATERIALS:Ribs from healthy,adult,New Zealand rabbits were prepared into PDPB by a series of physico-chernical methods including degreasing,deproteinization,partial decalcification and freeze drying,aFGF diluted with sterile distilled water was composited by PDPB particles to prepare artificial composite bone.METHODS:A bone window was made at the juncture of femoral head and femoral neck bilaterally in 24 healthy,adult,New Zealand rabbits.Rabbit models of bilateral ANFH were established by removing approximately 50% of cancellous bone and perfusion with 95% ethanol.Successful bilateral ANFH models were randomly divided into a blank group,a simple PDPB group,and an artificial composite bone group.PDPB and artificial composite bone were implanted into the PDPB and artificial composite bone group accordingly.The blank group did not receive any implantation.MAIN OUTCOME MEASURES:At 2,4,and 8 weeks after surgery,ink-injected specimens were prepared for microvessel count and microvessel area analysis.RESULTS:Microvessel number and microvessel area were least in the blank group,followed by simple PDPB group,and lastly the artificial composite bone group.There was significant difference in microvessel number and micrevessel area between artificial composite bone group and blank,simple PDPB groups (P<0.05).CONCLUSION:Tissue-engineered artificial bone composited by aFGF and PDPB promotes vasculadzation in repair of earlv-staqe ANFH in rabbits.
3.Research on the design of management system for pathological sample library

Objective:To establish the pathological sample database and standardize pathology specimen management processes to provide a platform for the study of pathology samples. Methods: Using J2EE, JSP, and component object model technology, the system establish the pathological sample library work platform for the pathology network resources, documentation, and intelligent. Results:The system established pathology samples and workflow management system architecture for the collection, storage and analysis of pathology specimens. Conclusion:The system designed to enhance the authenticity and credibility of the sample test results, thereby improve the accuracy of the sample analysis and provides a good reference for information technology-related departments.
4.The effect of ventricular electrical activity on the precordial cardioelectrical field and the ECG manifestation

Through the simulation based on a two-dimensional whole-heart electrophysiological model, the effect of ventricular electrical activity on the precordial cardioelectrical field as well as the ECG manifestation have been discussed. Such effect can vary with the changed excitation sequence of ventricles and the changed active and passive electrical properties of cardiac tissues. As the result, many abnormal ECG waves can form. By carefully analyzing such effect, the correlation among ECG waveforms recorded at different lead locations can be revealed and the significance of ECG manifestation can be fully explored.As a case of analyzing, some ECG waves at certain right chest lead locations can get a proper explanation.
