Neural stem cells were labeled with superparamagnetic iron oxide (SPIO) and tracked by MRI in vitro and in vivo after implantation. Rat neural stem cells were labeled with SPIO combined with PLL by the means of receptor-mediated endocytosis. Prussian blue staining and electron microscopy were conducted to identify the iron particles in these neural stem cells. SPIO-labeled cells were tracked by 4.7T MRI in vivo and in vitro after implantation. The subjects were divided into 5 groups, including 5 x 10(5) labeled cells cultured for one day after labeling, 5 x 10(5) same phase unlabeled cells, cell culture medium with 25 mug Fe/mL SPIO, cell culture medium without SPIO and distilled water. MRI scanning sequences included T(1)WI, T(2)WI and T(2)*WI. R(2) and R(2)* of labeled cells were calculated. The results showed: (1) Neural stem cells could be labeled with SPIO and labeling efficiency was 100%. Prussian blue staining showed numerous blue-stained iron particles in the cytoplasm; (2) The average percentage change of signal intensity of labeled cells on T(1)WI in 4.7T MRI was 24.06%, T2WI 50.66% and T(2)*WI 53.70% respectively; (3) T2 of labeled cells and unlabeled cells in 4.7T MRI was 516 ms and 77 ms respectively, R(2) was 1.94 s(-1) and 12.98 s(-1) respectively, and T(2)* was 109 ms and 22.9 ms, R(2)* was 9.17 s(-1) and 43.67 s(-1) respectively; (4) Remarkable low signal area on T(2)WI and T(2)*WI could exist for nearly 7 weeks and then disappeared gradually in the left brain transplanted with labeled cells, however no signal change in the right brain implanted with unlabeled cells. It was concluded that neural stem cells could be labeled effectively with SPIO. R2 and R(2)* of labeled cells were increased obviously. MRI can be used to track labeled cells in vitro and in vivo.

7.Experimental study on antagonism of tetrandrine in acute renal injury induced by gentamicin in guinea pig.
Chinese Journal of Applied Physiology 2003;19(3):278-281
AIMTo investigate the antagonism of tetrandrine in acute renal injury induced by gentamicin in guinea pig.
METHODSThe experimental guinea pigs are divided into 4 groups by randomized blocks, they were group 1 (control), group 2 (tetrandrine), group 3 (gentamicin) and group 4 (gentamicin with tetrandrine). After 10 days, set apart urinary specimen for determination the activity of NAG, reserve renal specimen for observing the renal histology and measuring expression of actin and TGF-beta1.
RESULTSThe renal histology showed that the injury of group 3 was the most grievous, group 4 was lighter than group 3. In addition, the apoptosis was obviously less than group 3. The activity of urine of NAG in group 4 was lower than group 3. The levels of expression of actin and TGF-beta1 in group 4 were more than group 3.
CONCLUSIONWhen kidney is damaged by gentamicin, tetrandrine is able to reduce the activity of urinary NAG and inhibit the renal tubular epithelial cell apoptosis. Besides, tetrandrine can prevent breaking actin and evoke the endogenous transmission growth factor beta1 tune-up, the above results can indicate that tetrandrine has antagonism to acute renal injury induced by gentamicin in guinea pig.

OBJECTIVETo discuss the therapeutic effects of closed or mini-open assisted reduction and percutaneous pinning for the treatment of two-part proximal humeral fracture.
METHODSTwenty-eight patients with two-part proximal humeral fractures were treated with closed or mini-open assisted reduction and percutaneous pinning from November 2005 to November 2006. There were 12 males and 16 females. The mean age was 54.7 years, ranging from 31 to 73 years. The two-part proximal humeral fractures of 28 patients were all fresh fractures. According to Neer classification, there were 18 patients with two-part surgical neck fractures and 10 patients with impacted valgus two-part greater tuberosity fractures.
RESULTSThe average operation time was 42 minutes and the wound healed at the primary stage. The X-ray showed all fracture had good reduction with almost no anterior angulation. The average neck shaft angle was changed from 175 (ranged, 160 degrees to 200 degrees) preoperatively to 136 degrees (ranged, 128 degrees to 142 degrees) postoperatively, and the displacement of greater tuberosity was reduced to less than 3 mm postoperatively in impacted valgus two-part greater tuberosity fracture group. The average clinical follow-up time was 10.3 months (ranged, 6 to 13 months). All fractures healed and had no avascular necrosis of the humeral head. The wires were removed 8 to 10 weeks post-op. According to the shoulder score of Constant-Murley and ASES,the mean ASES score was 91.2 points (ranged, 63 to 100 points) and Constant-Murley score was 90.4 points (ranged, 67 to 100 points). After the treatment, 12 cases got an excellent result, 14 good and 2 moderate.
CONCLUSIONClosed or mini-open assisted reduction and percutaneous pinning is not as easy as it looks and must be applied with fluoroscopy. But it is the effective method for managing the certain types of proximal humeral fracture with the advantage of less injury and faster recovery. The bone condition of proximal humerus is the key factor to the stability of fixation.

BACKGROUND:Femoral neck fracture is mainly fixed by three inverted triangle cannulated screws. Scholars have proposed to add a cannulated screw to enhance the fixation strength of femoral neck fracture of Pauwels III type based on three cannulated screw fixation, but the stability is not verified. OBJECTIVE:To analyze the biomechanical stability and stress of the three and four cannulated screws for the treatment of the Pauwels III femoral neck fractures. METHODS:The CT imaging results of the fourth generation of artificial bone sawbones were imported into the Mimics software wherein a three-dimensional finite element model of the proximal femur was prepared and introduced in the 3-matic software. Models of middlesegment of femoral neck with Pauwels III fractures were established. Cannulated screw models were established with UG 8.0 software and introduced in the fractures models. Finally, finite element models of Pauwels III femoral neck fractures fixed with three and four screws were established. In the same condition, an axial load of 411 N was applied on the femoral head with Abaqus software. The displacement of two markers of the broken ends and internal fixation system Von Mises stress distribution were compared between the two models. RESULTS AND CONCLUSION:(1) The displacement was 0.42 mm in three screws model, and 0.17 mm in the four screws model. (2) Von Mises stress peak was 547 MPa and 27.8 MPa in both models. The peak value was lower in models of fourscrews than that of three screws. Stress concentration position was at the fracture site in both models. The stress range of models of four screws was more extensive and scattered. (3) Finite element analysis results demonstrated that four-screw implantation for Pauwels III femoral neck fractures had strong anti-shearing force and biomechanical stability. Clinical advantages need further clinical comparative study.
