1.Rosiglitazone inhibition of A375 human malignant melanoma cell invasion

Objective To explore the effect ofrosiglitazone (RGZ), a ligand of peroxisome proliferatoractivated receptor γ (PPARγ), on the invasiveness of A375 cells in vitro and its mechanism of action.Methods A375 human melanoma cells were cultured in vitro and treated with different concentrations of RGZ. The proliferation of the cells, mRNA expression levels of matrix metalloproteinase (MMP) 2 and tissue inhibitor of metalloproteinase (TIMP) 2, protein expression of MMP2 in A375 cells were detected by MTT assay, semi-quantitative RT-PCR and Western-blot, respectively. The invasiveness of A375 cells was detected by Matrigel invasion assay. Results MTT assay showed that the proliferation of A375 cells was inhibited by (4.86±0.31 )% and (5.15±0.52)% under the 24-hour treatment with RGZ of 10 and 25 μmol/L, respectively, and no evident cytotoxity was observed for RGZ. Compared with untreated A375 ceils, a significant decrease was observed in the mRNA and protein expression of MMP2 in A375 ceils treated with RGZ of 10 and 25 μmol/L (all P < 0.05), along with an increase in the mRNA expression of TIMP2 (both P < 0.01 ).The count of A375 cells transmigrating through matrigel was 154.1±7.7 and 87.3±8.1 under the treatment with RGZ of 10 and 25 μmol/L, significantly lower than that of those without treatment (210.7±14.9,both P < 0.01 ). Conclusions RGZ could inhibit the transmigration of A375 ceils, likely by down-regulating the mRNA and protein expression of MMP2.
2.Laparoscopic surgery in the treatment of early uterine malignancies: a report of 8 cases

Objective To explore the feasibility of laparoscopic operation as treatment for early malignant uterine tumors Method Six patients with endometrial carcinoma underwent laparoscopic extensive total hysterectomy and bilateral adnexectomy, and 2 with cervical cancer underwent additional pelvic lymphadenectomy Results The mean operating time of the 6 laparoscopic extensive total hysterectomy and bilateral adnexectomy was 220 min, and mean blood loss was 200 ml The mean operating time of 2 cases underwent additional pelvic lymphadenectomy was 240 min No organ trauma occurred, and the average post operative hospital stay was 8 days Conclusion These results suggest that laparoscopic operation is safe and feasible in treating early stage malignant uterine tumors
3.Evaluation of dose delivery stability of a new linear accelerator TrueBeam

Objective:To study the long-term stability of 6MV photon beam on TrueBeam, which is a new linear accelerator introduced by Varian Medical System. Methods:QA BeamChecker Plus (QABC+, Standard Imaging Inc., Middleton, USA) was used to measure 6MV X-ray on TrueBeam linac as daily quality assurance (Daily QA). Dosimetric parameter, including output Constancy, flatness and symmetry, were employed to evaluate the dose delivery stability of TrueBeam linac. Results: Daily QA results of 18 months showed the dose output delta was (0.0%±0.7%), only once out of tolerance of 2%. The constancy of Flatness was excellent with the delta of (0.1%±0.1%). The symmetric of axial and transverse varied within 1%. Conclusion:The output constancy of TrueBeam with 6MV is very stable, as well as flatness and symmetry. It’s stable and reliable to implement Truebeam linear accelerator in clinical.
4.Temperament of children with vocal fold nodules

Objective:To examine the temperatment of children with vocal fold nodules.Method:To compare the temperatment dimension and temperatmental types of 42 children with vocal fold nodules with 46 vocally normal children, using Chinese children's Temperament Problem Screening system(CCTPSs).Result:The children with vocal fold nodules differed significantly from the comparasion group in their temperament dimension's adaptability,intensity of reaction, mood value, persistency and temperatmental types.Conclusion:There are more difficult and slow-to-warm-up children in patients with vocal fold nodules than vocally normal children.
5.Effects of endotoxin on NF-κB mRNA expression and aldosteron secretion in rat hepatic stellate cells

Objecfive To investigate the effects of endotoxin on nuclear factor-κB p65(NF-κB p65)mRNA expression and ahtosteron secretion in rat hepatic stellate cells(HSCs).Methods Cultured rat HSCs(HSC-T6)were divided into endotoxin-treated group and control group.Cells in endotoxin-treated group were exposure to 1 mg/ml.endotoxin.Aldosteron secretions of HSCs were determined by radioimmunoassay,and NF-κB p65 mRNA expressions of HSCs were detected by one-step RT-PCR.Results At 6,12,24 and 48 h,aldosteron secretions in endotoxin-treated group were significantly hisher than those in the control group(t=3.063,4.577,6.847 and 9.317,P<0.05),and the expressions of NF-κB p65 mRNA in endotoxin-treated group were also higher than those in control group(t=5.155,6.095,7.875 and 9.313,P<0.01).Aldosteron secretions and NF-κB p65 mRNA expressions in HSCs displayed a positive correlation(r=0.886,P<0.01).Conclusion Endotoxin can up-regulate the aldosteron secretion and NF-κB p65 mRNA expression in rat HSCs,which may be one of the mechanisms of liver fibrosis induced by endotoxin.
6.Effects of endotoxin on aldosteron secretion and nuclear factor-κB P65 mRNA expression of hepatic stellate cell

Objective To study the effects of endotoxin on aldosteron secretion and nuclear factor-κB P65(NF-κB P65)mRNA expression in rat hepatic stellate cell(HSC).Methods Cultured rat HSC were treated with endotoxin of different concentrations. Aldosteron secretion of HSC was detected by radio-immunoassay. NF-κB P65 mRNA expression of HSC was determined by semiquantitative reverse transcription-polymerase chain reaction(RT-PCR).The data were analyzed by variance analysis,t-test and Pearson linear regression analysis. Results Aldosteron secretions of HSC groups treated with 0.01,0.1,1.0 and 10.0 mg/L endotoxin[(4.95±0.35),(5.52±0.32),(6.04±0.60)and(5.16±0.46)μg/L, respectively] were all significantly higher than that in control group(3.655±0.51)μtg/L(t=2.9745,5.8725,6.8465 and 3.2065,respectively;all P<0.05).NF-κB P65 mRNA expressions of HSC groups treated with 0.01,0.1,1.0 and 10.01 mg/L endotoxin (0.82±0.06、1.07±0.07,1.23±0.06 and 0.96±0.05.respectively)were also significantly higher than that in control group 0.43±0.04(t=5.4776,6.8084,7.9382 and 7.5136,respectively;all P<0.01).Both aldosteron secretion and NF-κB P65 mRNA expression in HSC groud treated with 10.0 mg/L endotoxin were significantly lower than those in HSC group treated with 1.0 mg/L endotoxin(t=4.3865,3.7246;both P<0.05).In these treated HSC,aldosteron secretion was positively correlated with NF-κB P65 mRNA expression(r=0.886,P<0.01).Conclusions Aldosteron secretion and NF-κB P65 mRNA expression in rat HSC could be up-regulated by stimulation with endotoxin, which shows a certain dose-response relationship. This may be one of the important factors of hepatic fibrosis development.
7.Hospital Infection Prevalence Rate:Investigation and Analysis

OBJECTIVE To understand the status quo for hospital infection hospital infection to provide a scientific basis. METHODS A cross-sectional survey was taken combined with the hospital bedside investigation and records investigation. RESULTS In 1033 cases,the prevalence rate was 3.87%,and the infected sites were the respiratory site,superficial incision,skin and soft tissue. Utilization rate of anti-bacterial drugs was 60.31%,the pathagen detection rate before treatment was low only 7.73%. CONCLUSIONS Prevalence rate survey method is simple and reliable,it may be the basic reflect of hospital infection. Further strengthening the management of invasive operations,regulateing the rational use of antibiotics status quo,improving the detection rate of pathogens and reduceing preventive medication and antibiotic usage are all evitable.
8.Determination of Solubility and Apparent Oil and Water Partition Coefficient of Saponin H1

OBJECTIVE:To determine the solubility and apparent oil and water partition coefficient (lg P) of saponin H1,and provide reference for dosage form design and druggability research of saponin. METHODS:HPLC-ELSD was conducted to deter-mine the equilibrium solubility of saponin H1 in different organic solvents and pH buffer solutions;shaking flask was applied to de-termine lg P value of saponin H1. RESULTS:The equilibrium solubility of saponin H1 in water,methanol and ethanol at 25 ℃ was 0.09175 g/L,96.51 g/L and 46.89 g/L,respectively. The solubility increased apparently at high pH within pH range at 7.6-10.0;the lg P value was between 0.695-0.773 in buffers within pH range at 6.0-8.0. CONCLUSIONS:Saponin H1 and the oleanolic acid type saponins belong to low solubility, low transmission components, so it is not suitable for use as a conventional oral formulation is developed.
9.Diagnosis of congenital esophageal atresia in neonates with high frequency ultrasound combined with esophageal filling

Objective To investigate the diagnostic value of high frequency ultrasound combined with esophageal filling in neonates with congenital esophageal atresia.Methods Fifty-eight newborns who were suspected with congenital esophageal atresia underwent ultrasound,the surgical results considered as the gold standard,then the ultrasound features of the congenital esophageal atresia were summarized.ResultsSeven cases were the type Ⅰ of the congenital esophageal atresia,1 case belongs to type Ⅱ,40 cases were considered as type Ⅲ,4 cases were classified type Ⅳ and 6 cases were type Ⅴ,the diagnostic accuracy of ultrasound in congenital esophageal atresia was 96.55% and the classification accuracy was 89.65%.Conclusions High frequency ultrasound combined with esophageal filling has a high accuracy and can be used as a new diagnostic method in diagnosing neonates with congenital esophageal atresia,and it′s worthy of further research and promotion.
10.Osteogenic induction of human bone marrow mesenchymal stem cells cultured in complex medium

BACKGROUND:Studies have shown that the number of osteoblasts is often decreased after osteoporosis, and osteoblast replacement therapy becomes a new target for the treatment of osteoporosis. OBJECTIVE:To observe the osteogenic differentiation of human bone marrow mesenchymal stem cels cultured in dexamethasone, vitamin C and beta-glycerophosphate. METHODS:Mesenchymal stem cels were isolated and purified from adult bone marrow using human lymphocyte separation medium. The expression of cel surface markers was detected by flow cytometry. Cel ultrastructure was observed by transmission electron microscope. Then, the bone marrow mesenchymal stem cels were cultured in osteogenic induction medium containing dexamethasone, vitamin C andβ-glycerophosphate, and RT-PCR was used to detect the bone morphogenetic protein-2 mRNA expression after osteogenic induction. RESULTS AND CONCLUSION:A large number of adherent cels were visible as fibrous growth at 2 weeks after culture and strongly expressed CD44, CD29, but did not express CD34, CD45. These cels could be induced to differentiate into osteoblasts, and express bone morphogenetic protein-2 mRNA. Alizarin red staining and alkaline phosphatase staining were positive for the cels. These findings suggest that human bone marrow mesenchymal stem cels cultured in dexamethasone, vitamin C and beta-glycerophosphate can differentiate into osteoblasts, and has a potential for the treatment of osteoporosis.
