1.Difference of T Lymphocyte Subsets Expression in Blood of Children with Bronchiolitis or Asthma

Objective To understand the difference of helpful T lymphocyte subsets in patients diagnosed with bronchiolitis or asthma.Methods There were 34 patients in bronchiolitic group including 20 males and 14 females,the age of the patients was 1-15(6.65?4.22) months.They were confirmed by indirect immunofluorescence of nasopharyngeal secretion and (or) virus IgM antibody detected in plasma.All of the patients were infected with virus,23,4,4,and 3 patients were infected with respiratory syncytial virus,parainfluenza virus,influenza virus and adenovirus,respectively.All patients showed wheeze and small to medium blistering sound to different extend in both lungs,and all patients got fever at various degrees (20 patients blow 38 ℃,12 patients at 38-39 ℃ and 2 patients above 39 ℃).The patients′ capillary white cell counts were relatively normal when they were admitted to the hospital.There were 21 patients in asthmatic group including 12 male and 9 female,the age of the patients was 3-13 (8.05?2.85) years.Blood cells of 34 patients with bronchiolitis and 21 patients with asthma were collected by flow cytometry,and their peripheral T lymphocyte subsets were analyzed.The percentages of CD4+ cells secreting IFN-? or IL-4 were counted.They represented the percentage of helpful T lymphocyte Th1 and Th2,respectively.Results The percentage of CD4 positive cells and the ratio of CD4/CD8 were significantly higher in the bronchiolitic group than that in the asthmatic group (Pa=0.002).There was no significant difference of the percentage of Th1 cells between 2 groups.The percentage of Th2 cells in the bronchiolitic group,however,was significantly lower than that of the asthmatic group (P=0).Thus the Th1/Th2 ratio was significantly higher in the bronchiolitic group (P=0.031).Conclusions Compared with asthmatic patients,the patients with bronchiolitis show relative Th1 predominant response.Therefore,the treatment strategy should be different for these 2 diseases.
2.cDNA cloning and sequence analysis of ribosomal protein S7 gene from giant panda

Objective To explore the structure of ribosomal protein S7 ( RPS7) gene of giant panda ( Ailuropoda melanoleuca) and investigate its homologies with other already reported sequences,including Homo sapiens,Mus musculus,Rattus norvegicus and Bos taurus. Methods The cDNA of RPS7 was cloned from the giant panda by RT-PCR. The sequence data were analyzed by GenScan software. Blast 2. 1 was used to study the homology of the obtained RPS7 sequence with the gene sequences of other species; Open reading frame ( ORF) of the DNA sequence was searched using ORF finder software; Protein structure of the RPS7 sequence cloned was deduced using Predict Protein software. Results The full length of the sequence fragment was 589 bp containing an ORF of 585 bp. The deduced protein sequence showed that the protein was composed of 194 ami- no acids and its estimated molecular weight was 22. 126 85 ?103 with a pI of 10. 09. There were 7 different pat- terns of functional sites: one N-glycosylation site; two cAMP and cGMP-dependent kinase phosphorylation sites; four casein kinase C phosphorylation sites; one casein kinase Ⅱ phosphorylation site; two N-myristoylation sites; two amidation sites and one ribosomal protein S7e signature site in the RPS7 protein. Further analysis indicated that the sequence of RPS7 and the protein encoded were highly homologous to some mammals reported.Conclusion The complete coding sequence of RPS7 gene has been cloned through RT-PCR successfully, which is the first report on the RPS7 gene from the giant panda.
3.Establishment and Noninvasive Echocardiographic Assessment of Mouse Myocardial Infarction Model

Objective To establish a standardized mouse myocardial infarction model,and non-invasively evaluate its cardiac function by using the echocardiography.Methods A total of 20 wild male mice were enrolled into this study.The left anterior descending coronary was ligated through the left fourth intercostal space after intubation.The echocardiographic assessment of myocardial infarction and heart function was performed 3 days preoperatively and 2 weeks postoperatively.The shortening fraction(FS),end systolic dimension(ESD),and end diastolic dimension(EDD) was recorded and analyzed.Results The acute and one-week mortality rates were 20%(4/20) and 40%(8/20),respectively.The echocardiography showed that the myocardial infarction model was successfully established in 10 mice.The heart function of the mice was markedly impaired.The FS significantly decreased from(50.23?8.36)% to(25.41?0.97)%(t=16.18,P
4.THE EFFECT OF HEPARIN AND LOW MOLECULAR WEIGHT HEPARIN ON THE RECEPTOR OF AGE ON HUMAN MONOCYTE SURFACE

Radioactive ligand receptor binding assay was used to study the binding effect of heparin and low molecular weight heparin (LMWH) on the receptor of advanced glycation end products (AGE) on monocyte surface in normal individual and chronic renal failure (CRF) patients undergoing hemodialysis. The results showed that heparin caused a dose dependent inhibition of receptor binding to AGE, however LMWH did not have such inhibitory effects. The results suggest that heparin could interrupt AGE clearance and degradation, and LMWH had not these effects.
5.Clinical features of OSAHS closely associated with supine position

OBJECTIVE To study the clinical features of OSAHS patients closely associated with supine position. METHODS The clinical data of 105 patients with OSAHS diagnosed by overnight polysomnography(PSG)were retrospectively studied. The patients were divided in two groups: the positional patients(PP)group, those patients who had a apnea-hypopnea index(AHI) that was at least two times higher than the lateral AHI, and the non-positional patients(NPP)group, those patients in whom the AHI in the supine position was two times less than that in the lateral position, and the two groups were compared with age, BMI and the severity of OSAHS. RESULTS The difference of age and BMI were not significant between the two groups[(40.4?10.5)year versus(37.0?10.6) year,(28.2?3.8)kg/m2 versus(29.1?3.0)kg/m2]. There was significantly different in AHI between two groups[(36.8?25.4)/h versus (51.2?29.0)/h ]. AHIsupine and AHInot-supine was significantly different in mild and moderate OSAHS patients [(23.9?13.8) /h versus(12.4?10.1)/h ]. CONCLUSION Mild and moderate OSAHS patients are mainly PP. Positional therapy should play an important role in treatment of mild and moderate patients.

7.Establishment of CD200 stably-expressed U937 cell line

Objective:To establish a human monocyte cell line U937 stably expressing CD200.Methods:The eukaryotic expression vector pcDNA3-CD200 containing human CD200 cDNA and vector pcDNA3 were transferred into human monocyte U937 cell line by electrotransfer.These two cell lines were selected by G418, and the selected cell lines were subcultured. U937 cell line was enclosed as control group.The expression of CD200 mRNA and protein was detected by RT-PCR and flow cytometry.Results:After G418 selection,U937 cell line in control group was died, and the cell lines transferring pcDNA3 and recombined pcDNA3-CD200 were subcultured over 30 generations.The CD200 mRNA expression in pcDNA3-CD200 group was confirmed with RT-PCR.The CD200 expression in U937-pcDNA3-CD200,U937-pcDNA3 and U937 were 77.20%,3.20% and 2.10%,compared with other two groups (F=133 996.40,P0.05).Conclusion:Our study provides foundation for mechanism of action for CD200 in future.The human monocyte series U937 cell line that expresses CD200 protein stably is established successfully by gene transfection method.
8.Biological characteristics of bone marrow derived mesenchymal stem cells from patients with coronary artery disease

Objective To establish a method for the isolation and culture of bone marrow derived mesenchymal stem cells (MSCs) from patients with coronary artery disease and investigated its biological characteristics. Methods Bone marrow was extracted from patients' sternum when they were operated for coronary artery bypass graft. MSCs were separated from bone marrow by density gradient centrifugation in Percoll solution and seeded in culture medium supplemented with 10% fetal bovine serum. The morphology and growth characteristic of MSCs were observed in primary and passage generation culture with inverted microscope. The cell phenotype and cell cycle of MSCs were analyzed by flow cytometry. The growth curves were compared between different passages and ages. Results Most adherent MSCs showed spindle morphology whatever passages they were. The MSCs could be expanded for 15 passages. The proliferative potential of MSCs over the fifth passage or from old patients decreased gradually. The MSCs in primary and passage were positive for CD29 and CD44, but negative for CD34 and CD45. Analysis for cell cycle showed most MSCs in G_0/G_1 phase. Conclusion The method of isolation and culture of sternum bone marrow derived MSCs from patients with coronary artery disease is feasible, the MSCs have self-renewal capacity and might be ideal for cellular cardiomyoplasty.
9.The ReIationship of Mean PIateIet VoIume and Sudden SensorineuraI Hearing Loss

Objective Our aim was to investigate the association between sudden sensorineural hearing loss (SSHL)and mean platelet volume (MPV)values,in order to evaluate the etiology of SSHL.Methods This study included 40 patients with SSHL in our ward between Jan.2012 and Jan.2014 and 30 age- and sex-matched healthy subjects were included as controls.The correlation among the levels of the number of platelets,mean plate-let volume(MPV),platelet distribution width (PDW)was evaluated in the two groups.ResuIts The platelet count was significantly lower in the SSHL than the control group(212.0±47.9 versus 237.8±46.2 ×109/L).MPV and PDW were significantly higher in SSHL(10.9±0.8 versus 10.3±0.8 fL;12.9%±2.0% versus 12.0%±1.3 %). The SSHL patients were divided two subgroups:the mild-moderate subgroup (hearing loss 25~70 dB)and the se-vere subgroup(hearing loss≥70 dB).The MPV values were insignificantly different between the two subgroups(11. 0±1.1 fL versus 10.8 ±1.0 fL).Compared to the control group,the two subgroups had both significant differ-ences in MPV.ConcIusion High MPV values are a risk factor for thrombotic vascular diseases.Considering the in-creased MPV levels,SSHL appears to be characterized by ischaemic or thrombotic events.
