BACKGROUND:Bionic structure is one of the most important goals of scaffold design in tissue engineering. However, the majority of the bionic scaffolds do not have the capacity of sustained release of growth factors.
OBJECTIVE:To fabricate a novel biomimetic nanofibrous heparin/gelatin scaffold and investigate the characterization of the scaffolds including the microstructure, the mechanical strength andin vitro recombinant human bone morphogenetic protein-2 (rhBMP-2) release.
METHODS:Heparin-conjugated gelatin was prepared by chemical crosslinking and mixed with gelatin at different ratios. Nanofibrous composite scaffolds with different ratios of heparin-conjugated gelatin (0, 20% and 40%) were fabricated by using a thermaly induced phase separation combined with particle leaching technique. The amount of conjugated heparin in the scaffolds, scaffold microstructures and mechanical strength were detected and observed. The release kinetics of rhBMP-2 from the scaffolds with different concentrations of heparin conjugated gelatin was detected using ELISA method.
RESULTS AND CONCLUSION:The composite scaffolds possessed abundant porosities, good mechanical properties, and biomimetic nano-structures. The macroporous size was 250-420 μm. When the ratio of heparin-conjugated gelatin was 0, 20% and 40%, the content of heparin in the scaffolds was 0, (12.82±2.49) and (25.08±3.69) mg/g, respectively, with significant differences; the elastic modulus of the composite scaffolds was (299±38), (286±40) and (236±33) kPa, respectively, with insignificant differences (P > 0.05). With the increase in the content of heparin-conjugated gelatin, the rhBMP-2 adsorption capacity of the nanofibrous heparin/gelatin scaffold is improved, thereby significantly reducing the burst release of rhBMP-2 and prolonging the releasing time.

Objective:This study was aimed to investigate the influence of CDK7 siRNA on the sensitivity of endometrial carci-noma cell line HEC-1-A to cisplatin (DDP)-based chemotherapy. Methods:Different CDK7 siRNA fragments were synthesized based on the designs of the CDK gene sequence and were transfected into HEC-1-A. Real time reverse transcription polymerase chain reac-tion (RT-PCR) and Western blot analysis were employed to demonstrate the effects of transfection. The best CDK7 siRNA was chosen to specifically silence CDK7 expression in HEC-1-A.The sensitivity of the cells to DDP therapy before and after transfection was deter-mined by methyl thiazol tetrazolium (MTT) cytotoxicity assay, flow cytometry, and Hoechst/PI double-staining fluorescence microsco-py. Results:A total of four different CDK7 siRNA segments were designed and successfully transfected into HEC-1-A cells. The inter-ference effect in each group was confirmed by real time RT-PCR and Western blot assays. CDK7-423 was determined as the best per-forming CDK7 siRNA (over 70%) to transfect into HEC-1-A cells. MTT cytotoxicity test showed that IC50 of DDP decreased to a range from 45.122 μg/mL and 3.200 μg/mL after inhibition of CDK7 expression. DDP toxicity to the endometrial carcinoma cells sig-nificantly increased (P<0.05). Flow cytometry revealed that the average cell apoptosis rate significantly increased after the inhibition of CDK7 expression (11.66%to 37.57%, P<0.05). Similar results were observed using Hoechst/PI double-staining fluorescence microsco-py, and the number of apoptotic corpuscle demonstrated an apparent increase in the low CDK7-expressing group compared with the pa-rental cells. Conclusion:After the downregulation of CDK7 expression by CDK7 siRNA transfection, DDP chemotherapy sensitivity and apoptosis of endometrial carcinoma cells significantly increased. Further research is anticipated on the use of CDK7 as a new treat-ment target for endometrial carcinoma.

Purpose To investigate the clinicopathologic features and prognostic factors of apocrine carcinoma ( AC) of breast. Meth-od Clinical data of 70 ACs and 283 invasive carcinomas, not otherwise specified were collected. Differences between the prognostic outcomes of the two groups were compared, and the relationship between clinicopathological characteristics and prognosis was also ana-lyzed. Results The mean age of the patients with AC (56. 17 ± 12. 41 years) was older than those with invasive carcinoma not other-wise specified (52. 77 ± 11. 07 years) (P=0. 039). The patients with AC had a lower frequency of axillary nodal metastasis, a lower frequency of ER and PR positivity comparing to invasive carcinoma not otherwise specified ( P<0. 05 ) . No significant differences in the overall survival (P=0. 221) and disease-free survival (P=0. 378) periods of the two groups were observed. Kaplan-Meier surviv-al analysis showed tumor size, lymph node metastasis, pathological stage, lymph node tissue infiltration were related with prognosis of patients with AC ( P<0. 05 ) . In multivariate analysis, lymph node metastasis was associated with a worse prognosis ( P<0. 05 ) . Conclusions Although AC and invasive carcinoma not otherwise specified have different clinicopathologic characteristics, the prog-noses of patients with these diseases are similar. Lymph node metastasis could be used as an independent factor for predicting the prog-nosis of patients with AC, early diagnosis and early treatment is the key to improve its prognosis.

Objective To investigate new methods of osteonecrosis of femoral head (ONFH) with retrospective analysis of the clinical cases with technique of rotary cutting pressure-relief,impacted graft,and tantalum rod supporting treatment of early-stage femoral head necrosis.Methods From June 2010 to June 2014,32 cases (38 hips,23 males and 9 females) with ONFH were treated with technique of rotary cutting pressure-relief,impacted graft,and tantalum rod supporting,with the mean age of 24 ～ 52 (36.2 ± 3.8) years old.According to the Association Research Circulation Osseous (ARCO) classification,there were 11 hips on Ⅱ A stage,12 hips Ⅱ B,8 hips Ⅱ c,7 hips Ⅲ A.The averaged Harris score of all cases was 65 ～ 81 (72.5 ± 5.8) points.All cases were cured with minimally invasive technique,rotary cutter pressure-relief in femoral head,thoroughly remove sequestrum and implant allogeneic bone.Postoperative patients laid in bed for 1 month,after 1 month hold crutch gradually began to functional training.Postoperative 1 month,X ray film was reviewed every 3 months.According to the clinical symptoms,imaging findings,Harris scoring system was graded to evaluate the effectiveness.Results All the cases were followed up for average of 12 ～32 (20.2 ± 2.6) months,31 hips Kept hips succeed,hips pain; lame line symptoms were reduced or disappeared; x-ray showed femoral head necrosis area formed new bone,Harris scoring was upgraded to 75 ～ 95 (87.7 ± 6.4) points ; and seven hips failed with operation of total hip arthroplasty.The overall good rate was promoted from 26.3％ to 84.2％.Conclusions The technique treating for early-stagefemoral head necrosis with minimally invasive,rotary cutting pressure-relief,impacted graft,and tantalum rod supporting had a good short-term effects.

Objective To investigate the efficacy of photodynamic therapy(PDT) for tumor diagnosis.Methods A total of 1 400 patients with the cervix of uterus disease,56 patients with gullet disease,133 patients with stomach diseases,37 patients with urinary bladder tumor and 14 patients with brain tumor underwent diagnosis with PDT,combination of He-cd laser,Ar+ laser,and KTP laser.Results All of the patients could be explicitly diagnosed by this method,which indicated that PDT helped greatly in early tumor diagnosis. Conclusion PDT can be an effective diagnostic method for tumors of early stage,and should be widely applied clinically.

AIM To establish an HPLC method for the simultaneous content determination of swertiamarin,gentiopicrin,swertianolin,taxifolin,silychristin,silydianin and silybin in Dangfei Liganning Capsules (Silybi Fructus and Swertiae Herba).METHODS The analysis of 75％ methanol extract of this drug was performed on a 30 ℃ thermostatic Kromasil C18 column (4.6 mm × 250 mm,5.0 μm),with the mobile phase comprising of methanol-0.1％ formic acid flowing at 0.9 mL/min in a gradient elution manner,and the detection wavelengths were set at 254 nm and 288 nm.RESULTS Seven constituents showed good linear relationships within their own ranges (r ＞ 0.999 0),whose average recoveries were 96.78％-100.45％ with the RSDs of less than 2.0％.CONCLUSION This accurate and reproducible method can be used for the rapid quality control of Dangfei Liganning Capsules.
