1.Clinical application and analysis of minimally invasive techniques combined with anterolateral thigh flap in repairing skin and soft tissue defects of leg and foot

Objective To investigated the efficacy of minimally invasive technique combined with anterolateral thigh flap in repairing skin and soft tissue defects of leg and foot.Methods Anterior thigh flap was used to repair 20 cases of skin and soft tissue defects of leg and foot,the lesions defect area was 3.9 cm × 7.4cm-11.4 cm × 20.6 cm.Given the appropriate minimally invasive techniquesurgical treatment,the effect of follow-up was observed after operation.Results The supply valve area healed.The postoperative flaps of 20cases of patients all survived,no vascular crisis occurred.The color of skin flap was normal,close to normal skin color,and the elasticity of the skin was good,no crack,no fat,no pigmentation and other complications occurred,and no effect on shoes and ankle joint activities.Excellent and good rate of overall assessment flap was 90%(excellent in 12 cases,good in 6 cases);excellent and good rate of Mazur ankle function evaluation was 95%(excellent in 12 cases,good in 7 cases).Conclusion The minimally invasive technique combined with anterolateral thigh flap is a more ideal method to repair the leg and foot soft tissue defects.
2.Influence of ischemia/reperfusion on characteristics of membrane potassium channels of human renal tubule epithelial cells

Objective:To observe the influence of ischemia/reperfusion(simulated by anoxia) on the characteristics of K~(+) channels in renal tubular epithelial cells.Methods: Healthy renal tubule epithelial cells were divided into normal control group(cultured in serum-free RPMI 1640 medium),anoxic group(treated with antimycin A + serum-free RPMI 1640 medium),and oxidative stress group(H_(2)O_(2)+ serum-free RPMI 1640 medium)(n=15).The whole-cell patch clamp technique was used to examine the characteristics of K~(+) channels in renal tubular epithelial cells.Results: The transmembrane K~(+) current in tubular epithelial cells was voltage-dependent and could be blocked by K~(+) channel blocker Ba~(2+).The transmembrane potential levels in anoxic group and oxidative stress group were remarkably higher than that in normal control group(P
3.The clinical analysis of 17 cases with lymphomatous infiltration of the lung.

Objective To discuss the clinical manifestation and diagnosis of lymphomatous infiltration of the lung.Methods Clinical informations of 17 cases of lymphomatous infiltration of the lung were assessed.Results of 78 patients with malignant lymphoma,17 patients were infiltrated in lung.3 cases were Hodgkin's disease,14 cases were non -Hodgkin's lymphoma.All cases were advanced patients.Survival time of 1 patient was more than 5 years.Survival time of 1 patient which was the shortest was 2 months.Conclusion Patients with lymphomatous infiltration of the lung are not rare.The clinical symptoms and signs are not typical,image diagnosis is the main part in diagnosis of lymphomatous infil- tration of the lung.
4.Expression and clinical significance of stromal cell-derived factor-1 and its receptor CXCR4 in leukemia

Stromal cell-derived factor-1(SDF-1)is an early cell growth factor which belongs to a subfamily of chemokines.The CXCR4 is SDF-1 receptor.The SDF-1 interacting with CXCR4 plays important roles in physiological and pathological processes such as hematopiesis, stem cell homing,angiogenesis and leukemic cells infiltrating.To study the expression of CXCR4 in leukemia and the relationship between CXCR4 and leukemic cells infiltration can add indicators for leukemic MIC diagnosis of immunology indexes and have great significance to take molecule targeted therapy for preventing leukemia from infiltration or relapse as well.

As the most popular simplified model of the optical imaging system, the acquisition of the Gaussian point spread function (PSF) parameter is one of the hotspots and key points on which people do research in the field of image restoration. Based on the idea by which there exists deterministic mathematical relationship between Gaussian OTF feature points as well as its parameter and the frequency representation of the image in an existed literature, we proposed an automatic, accurate, stable, and improved approach. This method is able to give prominence to the related calculation feature by a Gaussian convolution and degeneration operation and finally realize the automatic esti mation of PSF parameter of a microscopic image. Experiments have proved that a good restoration result can be achieved utilizing the estimated PSF by the present method, which is of considerable application and reference value in restoration of other sorts with Gaussian approximate PSF model or 3D microscopic image restoration.

7.Objectification on Metabolomics of Different Traditional Chinese Medicine Syndromes in Patients with Coronary Heart Disease and Angina Pectoris

This study was aimed to investigate potential biomarkers in different traditional Chinese medicine (TCM) syndromes of angina pectoris of coronary heart disease (CHD) by using metabolomic technology,and to explore the objective law of different TCM syndromes of CHD.Endogenous metabolites in serum and urine from the healthy group,and patients with DHD angina pectoris patients of the syndrome of Qi deficiency and blood stasis,as well as the syndrome of Qi deficiency,blood stasis and phlegm turbidity were detected by the liquid chromatography-mass spectrometry (LC-MS).Metabolic profiles were analyzed by the principal component analysis (PCA) and the orthogonal partial least squares discriminant analysis (OPLS-DA).The results showed that in PCA,the healthy group,Qi deficiency and blood stasis group,as well as the Qi deficiency,blood stasis and phlegm turbidity group can be obviously distinguished.Potential biomarkers in the Qi deficiency and blood stasis syndrome contained aspartyl methionine and cysteine sulfinic acid.Potential biomarkers in the Qi deficiency,blood stasis and phlegm turbidity syndrome contained hippuric acid,amino glucose,fructosamine and triglyceride.The objective performance of Qi deficiency syndrome was the absence of biotin,lysyl tyrosine,phosphatidylglycerol and glycocholic acid.It was concluded that through the new metabolomic technology,different endogenous metabolites in the syndrome of Qi deficiency and blood stasis,as well as the syndrome of Qi deficiency,blood stasis and phlegm turbidity of patients with DHD angina pectoris were detected.It provided ideas for the clinical practice of prevention,diagnosis and treatment of different TCM syndromes.
8.A novel biomimetic nanofibrous heparin/gelatin scaffold with sustained-release recombinant human bone morphogenetic protein-2:preparation and characterization

BACKGROUND:Bionic structure is one of the most important goals of scaffold design in tissue engineering. However, the majority of the bionic scaffolds do not have the capacity of sustained release of growth factors.
OBJECTIVE:To fabricate a novel biomimetic nanofibrous heparin/gelatin scaffold and investigate the characterization of the scaffolds including the microstructure, the mechanical strength andin vitro recombinant human bone morphogenetic protein-2 (rhBMP-2) release.
METHODS:Heparin-conjugated gelatin was prepared by chemical crosslinking and mixed with gelatin at different ratios. Nanofibrous composite scaffolds with different ratios of heparin-conjugated gelatin (0, 20% and 40%) were fabricated by using a thermaly induced phase separation combined with particle leaching technique. The amount of conjugated heparin in the scaffolds, scaffold microstructures and mechanical strength were detected and observed. The release kinetics of rhBMP-2 from the scaffolds with different concentrations of heparin conjugated gelatin was detected using ELISA method.
RESULTS AND CONCLUSION:The composite scaffolds possessed abundant porosities, good mechanical properties, and biomimetic nano-structures. The macroporous size was 250-420 μm. When the ratio of heparin-conjugated gelatin was 0, 20% and 40%, the content of heparin in the scaffolds was 0, (12.82±2.49) and (25.08±3.69) mg/g, respectively, with significant differences; the elastic modulus of the composite scaffolds was (299±38), (286±40) and (236±33) kPa, respectively, with insignificant differences (P > 0.05). With the increase in the content of heparin-conjugated gelatin, the rhBMP-2 adsorption capacity of the nanofibrous heparin/gelatin scaffold is improved, thereby significantly reducing the burst release of rhBMP-2 and prolonging the releasing time.
9.Statistical approaches for evaluating and dealing with center effects in multi-center clinical trial

To search the statistical approaches for evaluating and dealing with the center effects in multi-center clinical trial. Breslow-Day test was used for evaluating the differences of the virtual rating among the centers whose responses were binary, Cochran-Mantel-Haenszel test for dealing with the center effects whose responses were binary or ordinal, and logistic regression for evaluating and dealing with the center effects simultaneously. The results showed that Breslow-Day test is invalid in evaluating the center effects whose response is ordinal, and Cochran-Mantel- Haenszel test can not be used for dealing with the effects of other covariates meanwhile.

The transmembrane potential levels in anoxic group and oxidative stress group were remarkably higher than that in normal control group (P<0.05), while their time constants (r) were smaller than that of normal control group (P<0.05), with no significant difference between the former 2 groups. Conclusion: There are voltage dependent K+ channels on tubular epithelial cells. Their expression increases obviously after ischemia-reperfusion and the activities of the channels also increase obviously.
