1.Comparison between the effects of oxaliplatin or irinotecan combined with fluoropyrimidine in the treatment of advanced gastric cancer

Objective To retrospectively observe and compare the efficacy and safty of irinotecan or oxaliplatin combined with fluoropyrimidine in the first line treatment of advanced gastric cancer. Methods 71 patients were divided into two groups. The irinotecan group consisted of 37 patients. Irinotecan 180 mg/m2 was injected in 90 minutes on d1. And the oxaliplatin group consisted of 34 patients. Oxaliplatin 85 mg/m2 was injected on d1. All the groups combined fluoropyrimidine. 5-Fluorouracil (5-Fu) 400 mg/m2 and leucovorin 200 mg/m2 were injected on d1,2 and 5-Fu 600 mg/m2 was 22 h continuous injected on d1,2, every 2 weeks was a cycle. It could also take capecitabine 1.0 g/m2 or tegafur-gimeracil-oteracil potassium 25 mg/m2 Bid for d1~14 orally, every 3 weeks was a treatment. Results 65 patients were evaluated. The response rates were 32.35% and 22.58%, the disease con-trol rates were 76.47% and 70.97%, and the median progression free survival (PFS) were 5 (3.962~6.038) months and 4 (2.730~5.270) months in irinotecan group and oxaliplatingroup, respectively, with no significant difference (P>0.05). Toxicity was tolerable, major adverse events were neutropenia, nausea and vomiting in two group. The incidence rate of diarrhea (45.95%) was higher in irinotecan group, and the peripheral nerve toxicity was 29. 41% in oxaliplatin group. Conclusion The effects of irinotecan or oxaliplatin combined with fluoropyrimi-dine in the treatment of advanced gastric cancer are comparable, and their toxicities are tolerable.
2.Peripheral Blood Motilin,Neuropeptide Y and Leptin Levels in Patients with Functional Dyspepsia

Background:The pathophysiology of functional dyspepsia(FD)is complicated and unclarified yet. Gastrointestinal hormone dysfunction may contribute to the development of FD. Aims:To investigate the correlation of different subtypes of FD with gastrointestinal hormone motilin(MTL),neuropeptide Y(NPY)and leptin(LEP). Methods:A total of 57 FD patients fulfilling Rome Ⅲ criteria were recruited and divided into epigastric pain syndrome(EPS)group(n = 24)and postprandial distress syndrome( PDS) group( n = 33 ). Ten healthy volunteers were served as controls. Fast and postprandial levels of MTL,NPY and LEP in peripheral blood were detected by radioimmunoassay. Results:Peripheral levels of fast MTL in EPS and PDS groups[(182. 90 ± 108. 57)pg/ mL and(145. 21 ± 67. 18)pg/ mL vs.(224. 47 ± 64. 55)pg/ mL,P < 0. 05],fast NPY in EPS group[(57. 40 ± 28. 75)pg/ mL vs.(90. 75 ± 49. 57)pg/ mL,P < 0. 01], and fast and postprandial NPY in PDS group[fast level:(38. 25 ± 20. 66)pg/ mL vs. (90. 75 ± 49. 57)pg/ mL,P <0. 01;postprandial level:(30. 26 ± 15. 12)pg/ mL vs.(65. 23 ± 54. 42)pg/ mL,P < 0. 01]were significantly lower than those in control group at same time points,especially the PDS group. Fast and postprandial levels of NPY were significantly lower in PDS group than in EPS group at same time points( P < 0. 05). No significant differences were observed in peripheral LEP levels among the three groups at any time points(P > 0. 05). Conclusions:The pathogenic mechanism of FD is related to the level of gastrointestinal hormones,and is different in EPS and PDS subtypes. Reduced MTL and NPY levels might be involved in the pathogenesis of PDS.
3.The expression of HGF and c-Met in gastric carcinoma

Objective To investigate the relationship between HGF/c-Met and biological behavior and prognosis of gastric carcinoma.Methods The expression of HGF.c-Met.microvessel density and microlymphatic densitv in 58 cases of gastric carcinoma and 27 cases of gastric ulcer were evaluated by immunohistochemistry,using monoclonal antibodies for HGF,c-Met,CD34 and Lyve-1 respectively.Results HGF and c-Met protein were overexpressed in gastric carcinoma,and were significantly higher than those in the mucosa of gastric ulcer(P<0.01).The microvessel density and microlymphatic density of gastric carcinoma were significantlv different between those with and those without lymphatic metastasis and distant metastasis(all P<0.05):The positive expression of HGF and c-Met in those with lymph node metastasis(86%,80%)was significantly higher than those without lymph node metastasis(57%,36%),P<0.05.That with distant metastasis(100%.94%)was higher than that without(71%,60%),P<0.01.The 5 year survival rate in patients with positive HGF and c-Met expression was much lower than that with negative expression(P=0.019,P<0.01),Multivariate analysis indicated HGF and c-Met expression,depth of invasion.lymph node metastasis.distant metastasis,microvessel density and microlymphatic density were all independent prognostic factors of gastric carcinoma.ConclusionsExpression of HGF and c-Met closely correlated with carcinogenesis,development and prognosis of gastric carcinoma.
4.Comparative study of the adjuvanticity of type-A,B and C CpG-ODN in mice

Objective To compare the adjuvanticity of type-A,B and C CpG-ODN in mice.Methods Three types of CpG-ODN were identified through in vitro stimulation of murine splenocytes with various CpG-ODN.BALB/c mice were immunized with HBsAg,together with different types of CpG-ODN,and antigen-specific IgG.IgG1 and IgG2a titers were measured 4 weeks later by indirect ELISA.Resuits Compared with the control group.all 3 types of CpG-ODN enhanced humoral immune response to HBsAg.However,type-B and C CpG-ODN induced much higher levels of antigen-specific IgG and IgG2a than type A CpG-ODN.Type-C CpG-ODN induced a similar TH 1-biased immune response as type-B CpG-ODN,revealed by decreased IsG1 to IgG2a ratio.In contrast,although type-A CpG-ODN increased IgG titers,it did not switch the balance between TH1 and TH2 immune responses.Conclusion All 3 types of CpG-ODN can enhance the humoral immune response to vaccines,but their aaiuvanticity could be mediated through different mechanisms.
6.Construction of a Saccharopolyspora erythraea mutant producing erythromycin B as its major fermentation product

Objective:To obtain a large amount of erythromycin B and to investigate the activity site in eryK. Methods:The key sequence of the BC loop region in eryK gene was knocked out and the eryK gene with 101 bp deleted was amplified by overlapping PCR,and cloned into vector pWHM3 to construct recombinant plasmid. The Saccharopolyspora erythraea mutant AK17 was constructed through chromosomal homologous recombination technique.Results and Conclusions:The S.erythraea mutant AK17 was constructed. The results of TCL and MS analysis showed that the major fermentation product of AK17 is erythromycin B.
7.Prediction of miRNA regulating the potential cancer-promoting gene CCL18 in cutaneous malignant melanoma and correlation analysis between CCL18 mRNA and miRNA expression

Objective To explore the miRNA regulating the potential cancer-promoting gene CCL18 in cutaneous malignant melanoma.Methods Bioinformatics analysis was conducted by using online software miRanda and TargetScan,so as to predict the miRNA targeting CCL18 gene.Three kinds of C CL18 3'UTR dual-luciferase reporter vectors,including mutant 3'UTR vector (mutant 3'UTR group),wildtype 3'UTR vector (wild-type 3'UTR group) and empty vector (blank control group),as well as miRNA vectors carring selected miRNAs were constructed according to human gene sequence analysis,and then were used to co-transfect 293T cells.After 48-hour treatment,the cells were lysed for detection of luciferase activity.Real-time fluorescence-based quantitative PCR was performed to measure the expression of CCL 18 and selected miRNA in 14 fresh malignant melanoma tissue specimens and 14 paracancerous normal skin tissue specimens (control tissues),and their correlations were analyzed.Results Online software analysis showed that some miRNAs were identified to target the 3'UTR of CCL18 gene,including miR-183,miR-128 and miR-33a.Luciferase reporter vectors and miRNA vectors were constructed successfully.As luciferase activity assay showed,when miR-183 and miR-128 were bound to the CCL18 3'UTR,the luciferase activities were significantly higher in their mutant 3'UTR groups (11.63 ± 0.42;8.80 ± 0.49) than in their wild-type 3'UTR groups (4.86 ± 0.39;5.01 ± 0.54;both P < 0.05) and blank control groups (2.41 ± 0.13;2.39 ± 0.05;both P < 0.01),while there were no significant differences between miR-33a-hinding mutant 3'UTR group (6.41 ± 0.47) and miR-33a-binding wild-type 3'UTR group (6.16 ± 0.22,P > 0.05).Real-time fluorescence-based quantitative PCR revealed higher mRNA expression of the CCL18 gene (3.52 ± 1.68),but lower expression of miR-183 (0.49 ± 0.32),miR-128 (0.30 ± 0.20) and miR-33a (0.46 ± 0.40) in the malignant melanoma tissues compared with the control tissues.The mRNA expression of the CCL18 gene was negatively correlated with the expression of miR-128 (rs =-0548,P < 0.05),but showed no significant correlations with the expression of miR-183 and miR-33a (both P > 0.05).Conclusion miR-128 may play a role in regulating the potential malignant melanoma-promoting gene CCL18.
8.Epidemiological investigation and analysis of children with strabismus and amblyopia in No. 1 primary school of Jiajiang County, Leshan, Sichuan Province

AIM: To understand the epidemiology and related factors of strabismic amblyopia of students of primary school, and to provide guidances for the prevention and control strategy.
METHODS: A total of 600 cases of primary school students of Leshan City, Jiajiang County were given vision, oblique incidence and ocular and other screening. The prevalence rate of poor eyesight of strabismus, amblyopia prevalence rate of different sexes, ages were compared, and the degree of amblyopia and strabismus of children with different types of amblyopia and whether or not had stereoscopic vision were counted.
RESULTS: The prevalence rate of amblyopia and strabismus prevalence rate were respectively 4. 0% and 2.5%;With the growth of all age, low vision of students was significantly decreased, the difference of comparison of low vision rate of each age had statistical significance (P<0. 05), but different ages, strabismus prevalence of different sexes, amblyopia prevalence were compared, the difference had no significant differences ( P>0. 05 );Ametropic amblyopia was the main type, accounting for 55. 6%, and the degree of amblyopia mainly was light, moderate; ametropic amblyopia, most of ametropic amblyopia and strabismus had stereo vision, but there were no stereopsis of most of the strabismic amblyopia and all esotropia.
CONCLUSION:Ametropic is mainly type of amblyopia, the prevalence of relationship between the incidence of strabismic amblyopia of primary school students and sexes is not obvious, but the oblique amblyopia treatment effect, such as the establishment of stereoscopic vision and the age, eye position has a close relationship, should be early discovered, early treatment.

10.Pharmacokinetics of praziquantel injection in healthy buffaloes

Objective To investigate the pharmacokinetics and relative bioavailability of praziquantel injection in buffaloes in contrast to praziquantel tablet. Methods A single oral administration of praziquantel tablet at a dose of 20 mg/kg or intramus-cular administration of praziquantel injection at a dose of 10 mg/kg was performed in six healthy adult buffalos according to a two-period crossover design. The praziquantel concentration in plasma was determined by a high performance liquid chromatography (HPLC)method. The pharmacokinetic parameters were calculated by non-compartmental analysis. Results The main pharma-cokinetic parameters of praziquantel tablet were as follows:Tmax=(0.60±0.29)h,Cmax=(0.57±0.37)μg/ml,T1/2β=(0.70±0.42) h,AUC=(0.80±0.70)(μg/ml)·h. The main pharmacokinetic parameters of praziquantel injection were as follows:Tmax=(0.65± 0.49)h,Cmax=(3.82 ± 1.17)μg/ml,T1/2β=(1.00 ± 0.73)h,AUC=(1.61 ± 0.89)(μg/ml)·h. The relative bioavailability of pra-ziquantel injection was 402.5%in contrast to praziquantel tablet. Conclusion The praziquantel injection has pharmacokinetic characteristics of rapid absorption,high bioavailability and extensive distribution,and the clinical recommended dosage of pra-ziquantel injection is 10 mg/kg.
