1.OBSERVATION OF ULTRASTRUCTURE AND DRUG SENSITIVITY OF IN VITRO CULTURED EXOERYTHROCYTIC FORM OF PLASMODIUM BERGHEI

The infrastructure of in vitro cultured exoerythrocytic stage (EE) of Plasmodium ber-ghei (P.b.)was observed under transmission electron microscope (TEM).The drug sensitivity of EE was also measured in vitro.The EE was cultured in monolayer host cell,fixed and embeded in situ.The ultrathin sections were prepared and examined by routine methods.The TEM pictures showed that the fine structure of in vitro cultured EE was similar with that of EE grown in rat hepatocytes in vivo,as described by Meis et al.The parasite was found within a parasitophorous vacuole,with nucleus,mitochondria,endoplasmic reticuia and Golgi apparatus were observed inside the parasite.After 24h cultivation,the EE was incubated for 48h in medium containing a serial concentrations of primaquine or chlo-roquine,then examined under light microscope.The percentage of abnormal parasites was calculated.The preliminary results showed that the sensitivity of cultured P.b.EE to primaquine and chloroquine was significantly different.At the same concentration of 1?10-5 mol/L,the percentage of abnormal parasites was 38.5?3.9% and 5.7?1.9% ,respectively These results demonstrated that the in vitro cultivation system of P.b.EE would have potential utility in antimalarial drug research.(Figs.1-6)
2.Effect of cordycepin on inhibiting delayed type hypersensitivity

Objective:To investigate the effect and the mechanism of cordycepin on inhibiting delayed type hypersensitivity.Methods:The mouse model of delayed type hypersensitivity was established by sensitizing the skin of Kunming mice with 2,4-dinitrochlorobenzen(2,4-DNCB). There are two experimental groups respectively treated with different dosages of cordycepin:12 mg/kg and 16 mg/kg and one negative control group treated with normal saline. All the mice were intraperitoneally injected at 24 h interval(the last interval was 4 hours). We calculated the differences of the thickness and the weight between the left and right earlaps of every mouse and the edema rate, then used statistical methods to compare the edema rate and the spleen index among all the groups. After HE staining, the pathological changes of inflammatory earlap and spleen were observed and compared among these three groups.Results:Cordycepin alleviated the erythema, edema and the infiltration of peripheral mononuclear cells in the earlaps, so lowering the differences of the thickness and weight between left and right earlaps in the experimental groups. But it showed no significant effects on the spleen index and the spleen pathological changes. 3.Intra-hospital transport nursing of patients with severe inhalation pulmonary injury treated with extracorporeal membrane oxygenation

Objective To explore the nursing experience of the safe intra-hospital transport of patients with the severe aspiration of the inhalation injury. Method The nursing measures for the intra-hospital transport of 2 cases of smoke pot inhalation injury caused by extracorporeal membrane oxygenation treatment were taken, including disease risk assessment, preparation for transport, organization of a transport team, effective vital signs monitoring during transport, extracorporeal membrane oxygenation (ECMO) pipeline monitoring and nursing and observation and nursing of complications. Result No emergency was found during the transport of patients and both of them were safely transported. Conclusion Such nursing measures as pre-transport assessment and preparation and bettering predictive nursing for the patients with severe inhalation pulmonary injury treated with extracorporeal membrane oxygenation are key to the safety during intra-hospital transport.
4.Response surface optimization of microwave-assisted extraction for HPLC-fluorescence determination of puerarin and daidzein in Radix Puerariae thomsonii

Microwave-assisted extraction was optimized with response surface methodology for HPLC-fluorescence determination of puerarin and daidzein in Radix Puerariae thomsonii.The optimized extraction procedure was achieved by soaking the sample with 70% methanol(1∶15,v/v)for 30 min,and then microwave irradiation for 11 min at a power of 600 W.Coupling the extraction process with HPLC-fluorescence presented good recovery,satisfactory precision,and good linear relation.Compared with a method from the Chinese Pharmacopoeia,the proposed method enables higher extraction efficiency and more accurate analytical results.It can be of potential value in quality assessment of Radix Puerariae thomsonii medicinal materials.
5.Reduction in stimulation of bendazac lysine eyedrops by cold storage

Objective To evaluate whether the stimulation on eyes of bendazac lysine eyedrops could be reduced by cold storage.Methods Mate design scheme and double-blind clinical trial were performed. 160 healthy eyes was divided into two groups: the test group was given bendazac lysine eyedrops which was stored in refrigerator at 4℃ over night, and the control group was given it which was stored at room temperature.Results ln the control group and the test group, the occuring rate of stimulation was 19.30% and 6.25%. The cases of B、C stimulation degrees were 31 and 10; the average stimulation degrees of each group were 1.21 and 1.07, the scoring change for eye stimulation were 1.57±1.50,0.79±1.40,all respectively. The statistical difference was significant (P<0.05 or P<0.01).Conclusion The eye stimulation of bendazac lysine eyedrops can be reduced by cold storage.
6.Irradiation injury temporarily induces enhancement of APN/CD13 peptidase activity on aorta-gonads-mesonephros (AGM)-derived stromal cells.

This study was designed to investigate the expression of aminopeptidase N (APN)/CD13 on intraembryonic AGM stromal cells, and the change of its enzymatic activity after irradiation injury. The expression of APN/CD13 on AGM stromal cells was assayed by RT-PCR and immunihistochemistry. After the stromal cells in AGM region were irradiated with 8.0 Gy of (60)Co gamma-rays, APN/CD13 enzymatic activity was measured by spectrophotometer at different time points. The result showed that AGM stromal cells strongly expressed APN/CD13. The enzymatic activity of APN/CD13 decreased temporarily after irradiation injury, then increased to higher level 4 h after irradiation, and it returned to the pre-irradiation level 24 to 48 h after the irradiation. The enzymatic activity of APN/CD13 was temporarily enhanced after irradiation injury, which might be one of the compensatory mechanisms that promote the hematopoietic recovery after irradiation.

8.Research advance on frozen injury and protective mechanisms of cryopreservated platelets.

Platelets are important component in the blood which play vital role in physiological hemostasis. In clinic, platelets are usually transfused to patients suffered from idiopathic thrombocytopenic purpura,thrombocytopenia after chemotherapy and radiotherapy,or hemorrhagic thrombocytopenia caused by other reasons. Storage time for platelet concentrates is limited to five days at room temperature, leading to an increasing number of scrap rate, therefore the prolonging the storage time is important for clinical application. Storage at -80°C is an ideal long-term cryopreservation method for platelet concentrates. In this article, platelets cryopreservation,the mechanism of frozen injury and the protective mechanisms of platelets were reviewed.

9.Study of denoising of simultaneous electroencephalogram-functional magnetic resonance imaging signal based on real-time constrained independent components analysis.

Simultaneous recording of electroencephalogram (EEG)-functional magnetic resonance imaging (fMRI) plays an important role in scientific research and clinical field due to its high spatial and temporal resolution. However, the fusion results are seriously influenced by ballistocardiogram (BCG) artifacts under MRI environment. In this paper, we improve the off-line constrained independent components analysis using real-time technique (rt-cICA), which is applied to the simulated and real resting-state EEG data. The results show that for simulated data analysis, the value of error in signal amplitude (Er) obtained by rt-cICA method was obviously lower than the traditional methods such as average artifact subtraction ( <0.005). In real EEG data analysis, the improvement of normalized power spectrum (INPS) calculated by rt-cICA method was much higher than other methods ( <0.005). In conclusion, the novel method proposed by this paper lays the technical foundation for further research on the fusion model of EEG-fMRI.
