1.Cluster ensemble algorithm based on dual neural gas applied to cancer gene expression profiles.
Journal of Biomedical Engineering 2015;32(1):93-98
The microarray technology used in biological and medical research provides a new idea for the diagnosis and treatment of cancer. To find different types of cancer and to classify the cancer samples accurately, we propose a new cluster ensemble framework Dual Neural Gas Cluster Ensemble (DNGCE), which is based on neural gas algorithm, to discover the underlying structure of noisy cancer gene expression profiles. This framework DNGCE applies the neural gas algorithm to perform clustering not only on the sample dimension, but also on the attribute dimension. It also adopts the normalized cut algorithm to partition off the consensus matrix constructed from multiple clustering solutions. We obtained the final accurate results. Experiments on cancer gene expression profiles illustrated that the proposed approach could achieve good performance, as it outperforms the single clustering algorithms and most of the existing approaches in the process of clustering gene expression profiles.

Objective To discuss the value of percutaneous compression plate (PCCP) in treating intertrochanteric fractures. Methods A retrospective study was done on 57 patients with intertrochanteric fractures treated by PCCP from January 2008 to January 2009 to evaluate the operative effect. Results All the fractures were healed in 11-15 weeks after the operation, with no incision infection, hematoma, internal fixation failure or fracture collapse. Two surgical incisions were for 2 cm, with no blood transfusion found in any patient. According to Harris score, the postoperative excellent rate reached 92.6%. Conclusion PCCP can attain similar effect wiht DHS and now is the choice of treatment in treating intertrochanteric fracture in the elderly patients.

Objective To observe the antioxidative dosage-effect relationship of Marigold lutein, and provide experimental data for clinical use. Methods The mice were randomly divided into seven groups:blank control group, model control group, 1 mg/kg lutein group, 5 mg/kg lutein group, 25 mg/kg lutein group, 125 mg/kg lutein group and 625 mg/kg lutein group. The mice in blank control group were dealt with saline solution by intraperitoneal injection, the others were dealt with D-galactose (120 mg/kg) by intraperitoneal injection for seven weeks to make oxidative damage model, meanwhile the mice were given corresponding dose of the drug. Subsequently, the level of malondialdehyde (MDA) and activity of superoxide dismutase (SOD) in the serum were measured, and the antioxidative dosage-effect relationship was observed. Results The 1, 5, 25 mg/kg lutein reduced the MDA level and increased SOD activity, and the 125, 625 mg/kg dose of lutein did not show significant antioxidant activity. Conclusion Lutein has significant antioxidant activity in mouse dealt with D-galactose within the dose range of 1-25 mg/kg. The results suggest that the clinical dosage range of lutein should be kept within reasonable limits.

Objective To observe the antioxidative dosage-effect relationship of grape seed proanthocyanidin extract (GSPE). Methods The mice were randomly divided into seven groups:blank group, model group, 5 mg/kg GSPE group, 15 mg/kg GSPE group, 45 mg/kg GSPE group, 135 mg/kg GSPE group and 405 mg/kg GSPE group. The mice in blank group were dealt with saline solution by intraperitoneal injection, the others were dealt with D-galactose (120 mg/kg) by intraperitoneal injection for seven weeks to make oxidative damage model. Meanwhile, the mice were given corresponding dose of the drug. Subsequently the level of malondialdehyde (MDA) and activity of superoxide dismutase (SOD) in the serum were measured to observe the antioxidative dosage-effect relationship of GSPE. Results The 45, 135, 405 mg/kg GSPE group reduced the MDA level, and the 15, 45, 135, 405 mg/kg GSPE group increased the SOD activity. Conclusion GSPE has significant antioxidant activity on mice dealt with D-galactose above the dose of 15 mg/kg, suggesting that the clinical use of GSPE should guarantee a certain dose to play a good antioxidant effect.

Using the immunohistochemical and immunoelectron microscopic methods, the localization and pattern of vasoactive intestinal polypeptidergic nerves in the wall of cerebral vessels were investigated. The results revealed that the VIP-like immunoreactive (VIPLI) fibres were located in the adventitia or at the adventitia-media border. No synaptic contacts were identified among the nerve terminals or between nerve terminal and smooth muscle cell. VIPLI terminals directly apposed to the smooth muscle cells with a distance of 100 nm.The observations mentioned above indicate that there is peptidergic innervation in the wall of the cerebral vessels besides traditional adrenergic and cholinergic innervations. The regulatory function of VIP-containing nerves to the cerebral vessels may be performed by affecting the smooth muscle directly through the nonsynaptic release. In addition, present study identified and discussed the distribution and function of substance P(SP) in the cerebral vessels.

Objective To observe the effect of different doses of esketamine on early mood in pa-tients with preoperative anxiety after thoracoscopic surgery. Methods Eighty patients with preoperative anx-iety before thoracoscopic surgery,37 males and 43 females, aged 25-64 years,BMI 18-26 kg/m2,ASA physical statusⅠ-Ⅲwere selected.According to random number table, the patients were randomly divided into four groups:control group(group C),esketamine 0.1 mg/kg group(group E1),esketamine 0.2 mg/kg group(group E2),and esketamine 0.3 mg/kg group(group E3),20 patients in each group. In groups E1,E2,and E3,the patients were slowly injected with corresponding doses of esketamine intrave-nously 30 minutes before the end of the operation, and the patients in group C were given 10 ml of normal saline at the same time. The 7-item generalized anxiety disorder scale(GAD-7)score and hospital anxiety and depression scale(HADS)score were evaluated 24 hours before surgery,24,48,72,and 120 hours af-ter surgery, respectively.Peripheral venous blood(5 ml)was collected at the same time points, and serum brain-derived neurotrophic factor(BDNF)concentration was detected by ELISA.NRS scores at rest and ex-ercise were recorded 12 and 24 hours after surgery. The number of remedial analgesia and the number of ef-fective compressions of analgesia pump within 48 hours after surgery were recorded. Results Compared with group C,the scores of GAD-7 and HADS in groups E1,E2,and E3 were significantly decreased 24,48,and 72 hours after surgery(P<0.05).And compared with groups E1 and E2,the scores of GAD-7 and HADS in group E3 were significantly lower(P<0.05).The serum BDNF level in groups E1,E2,and E3 were significantly increased compared with group C 24,48,and 72 hours after surgery(P<0.05).Compared with group E1 and E2,the serum BDNF level in group E3 was significantly higher(P<0.05).Compared with group C,the NRS scores at rest 12 and 24 hours and the scores at exercise 12 hours after surgery in groups E1,E2,and E3 were significantly decreased(P<0.05).And compared with groups E1 and E2,the scores in group E3 were significantly lower(P<0.05).The number of postoperative remedial analgesia and the number of effective compressions of analgesia pump in groups E1,E2,and E3 were signif-icantly decreased compared with group C within 48 hours after surgery(P<0.05).And compared with groups E1 and E2,the number in group E3 was significantly lower(P<0.05).Conclusion Single intra-venous injection of esketamine 0.1,0.2,and 0.3 mg/kg can improve early postoperative mood of patients with anxiety before thoracoscopic surgery, and esketamine 0.3 mg/kg has better anti-anxiety effect.
