1.Effects of down-regulation of PI3K/Akt signaling pathway on growth of laryngeal cancer cells

Objective To investigate the effect of potassium channel blocker tetraethtylamine (TEA) on the proliferation and apoptosis of human laryngeal carcinoma cells (Hep-2) through the PI3K/Akt signaling pathway.Methods TEA acted on HEp-2.The methyl thiazolyl blue (MTT) method was used to determine the Hep-2 cell activity and the proliferation inhibition rate of TEA on Hep-2 was calculated;the Hoechest33258 staining was used to detect cell apoptosis,and the Hep-2 apoptosis rate was detected by flow cytometry (FCM) assay.Results After inoculation by 5,10,20 mmol/L TEA,the Hep-2 cell proliferation inhibition rate reached 12.573%,31.385% and 56.132%,respectively.After 96 h Hep-2 cell action,the cellular apoptosis rates were (41.64±2.67)%,(58.76±4.32)% and (72.65±6.54)% respectively,the inhibition rate and apoptosis rate in the TEA treating group were significantly higher than those in the non-TEA treating group,the differences were statistically significant (P<0.05).Conclusion TEA can inhibit the proliferation and in vivo growth of human laryngeal carcinoma cells and promotes the apoptosis of laryngeal cancer cells.

We report the case of a patient with catecholaminergic polymorphic ventricular tachycardia resulting
from a RYR2 mutation who had not only typical electroencephalogram changes, but also epileptiform
discharges in electroencephalogram. We believe the changes were closely related to the RYR2 mutation.

Objective To evaluate the effects of 131 adrenoceptor anfisense on blood pressure and?1 adrenoceptor mRNA and protein levels in 2 kidney 1 clip(2K1C)rats.Method 2KIC hypertensive rots were produced by clipping renal artery of SD rats.Liposome/AS-ODNs 2.0 were tested intravenously in rats with 2KIC hypertension.Animals were divided into 5 groups(n=18 in each group):?1-AS-ODN group,?1-IN-ODN group,2K1C group,Sham group and SD group.Blood pressure was measured by tail-cuff method,the levels of myocardial?adreneceptor mRNA and protein were tested by RT-PCR and binding assay.Results On the basis of the magnitude and duration of hypotension,?1-AS-ODN decreased blood pressure by 39 mmHg at the most for 4 weeks.Compared with the 2KIC group,?1-AS-ODN did not significantly change the levels of myocardial?1 adrenoceptor mRNA but significantly decreased the levels of myocardial?1 adrenoceptor protein at 2,7,30 days (P

To facilitate the expression of GL 7 ACA acylase gene in a recombinant E coli , a fragment of the gene, in which the signal peptide was deleted by PCR method, was inserted into a prokaryotic expression vector, pET 28a By colony PCR method screening, a recombinant plasmid pET ACY was obtained and then transformed into the expression host BL21 (DE3) The influences of induction conditions such as IPTG concentration, the time of induction and the induction temperature on the expression of the recombinant protein were investigated Under optimal condition, the enzyme activity could reach 266 U/L Finally, the recombinant GL 7 ACA acylase can be easily isolated to a purity of about 80% by a simple anion ion exchange chromatography with enzyme activity recovery of 50%

Fibroblast growth factor-21 (FGF-21) is an important metabolism regulator, however, whether FGF-21 has effects on cardiovascular remains unclear. In this study, H2O2-induced injury in H9c2 cells was used as a cell model, the anti-apoptosis potential and mechanism of FGF-21 against oxidative injury were evaluated by MTT assay, flow cytometry assay and real-time PCR. The results showed that FGF-21 could increase the cell survival of H2O2-induced injury in H9c2 cells and prevent H9c2 cells from oxidative stress-induced apoptosis. Furthermore, FGF-21 can elevate SOD activity and regulate Bcl-2/Bax expression in H9c2 cells. The results suggest that FGF-21 have protective effect against the H2O2-induced apoptosis in H9c2 cells.

7.Treatment of intertrochantetic hip fractures with improved technique of Gamma nail in the elderly patients

Objective To explore the clinical outcome of improved technique of Gamma nail in the treatment of intertrochanteric hip fracture of the elderly patients. Methods From March 2002 to October 2006.39 patients with intertrochanteric hip fracture were operated by improved technique of Gam-ma nail.There were 18 males and 21 females at average age of75.7 years(67_98 years).There were 6 patients with type A1 fracture,24 with type A2 fracture and 9 with type A3 fracture according to AO/ASIF classification.Of all.36 patients(92.3%)had osteoporosis.The operation improvements included the following points:(1)The patients were placed at the lateral decubitus position with the fractured limb on the uppermost,with flexion of knee and hip of 60°.The normal hip and knee were flexed as possible.(2)One-off indirect traction reduction was used after general anesthesia. no requirement of continuous mechanical traction.(3)C-arm image intensifier was employed to obtain normal and lateral projections.Results Of all,35 patients were followed up for a mean period of3 years and 2 months, ranging from 6 months to 5 years and 2 months.Operation data showed incision length of(4.3±1.2)cm,mean opera-tion time of(46±10)minutes,intraoperative bleeding volume of(65±26)ml and intraoperative X-ray exposure of(3.0±2.1)times.Postoperative recovery data showed survival in one-year follow up,with ambulation time of(10.5±3.6)days and fracture union time of(10.9±2.1)weeks.Mean Parker's score wag(6.9±3.2)points 6 months after operation. Conclusions Improved technique of Gamma nail can shorten operation duration,reduce operative trauma and bleeding,reduce X-ray exposure and im-prove success rate of surgery.as facilitates early pest-operative recovery and reduces the perioperative mortality rate of the elderly.

Silk fibroin is a natural polymer with certain water solubility, structural modification, good biocompatibility and biodegradability, which can be used as a drug delivery carrier material. As a promising drug delivery system, drug-loaded silk fibroin nanoparticles can control drug release, reduce toxicity and improve therapeutic effects. In this paper, the basic characteristics of silk fibroin, the preparation methods of drug-loaded silk fibroin nanoparticles and the application of silk fibroin in nanoparticulate drug delivery systems are reviewed, and on this basis, the further development of drug-loaded silk fibroin nanoparticles is prospected.

10.Apolipoprotein E and serum lipid levels for clinical diagnosis of early cerebral infarction

Objective To analyze the diagnostic value of apolipoprotein E and lipid levels in patients for early diagnosis of cerebral infarction.Methods A total of 80 patients with cerebral infarction were enrolled in this study as the study group from June 2015 to October 2016.Meanwhile,80 healthy subjects were selected as the control group.The serum apolipoprotein E and blood lipid were compared between the two groups.Results The difference of apolipoprotein E levels between the two groups was statistically significant (P < 0.05).Apolipoprotein E combined with serum total cholesterol and low density lipoprotein cholesterol for the diagnosis of cerebral infarction had a sensitivity of 37.5 %,specificity accuracy of 100%,and diagnostic efficiency of 60%.Conclusion Early cerebral infarction patients diagnosed with apolipoprotein E combined with lipid levels had a good clinical value.
