1.Experimental study on cAMP analogue 8-CPT-cAMP inducing apoptosis in multiple myeloma cells

Background and purpose:Despite the high remission rate in patients with multiple myeloma (MM) after the standard regimen, but often relapsed and resistant. It has been shown that modulation of cAMP can induce cell cycle arrest and apoptosis in a variety of tumor cells, which has become an interesting approach to cancer therapy. This study aimed to investigate possible effects of cyclic adenosine monophosphate (cAMP) analogue 8-(4-chlorophenylthio) adenosine 3’, 5’-cyclic monophosphate (8-CPT-cAMP) on multiple myeloma cells, provide direction to develop new drugs for the treatment of MM. Methods:The myeloma cell line U266 cells were treated with 8-CPT-cAMP of different concentrations. The proliferation of U266 cells was evaluated through cell counting kit (CCK-8) assay, lfow cytometry was used to analyze the changes of cell were distribution, apoptosis rate as well as mitochondrial transmem-brane potential (ΔΨm) in U266 cells before and after the treatment. Meanwhile, real-time quantitative polymerase chain reaction (RT-PCR) and Western blot assay were used to detect expression levels of apoptosis regulators including caspase-8, caspase-9, Bcl-2 and Bax genes in U266 cells before and after the treatment. Results:The U266 cells were treated 5 days with 8-CPT-cAMP of different concentration, it was shown that 8-CPT-cAMP could signiifcantly inhibit cell growth of U266 cells in a concentration and time dependent manner, the IC50 of 8-CPT-cAMP was reduced obvious prolonged reaction time, and reached to 58.52μmol/L in the iffth day. The cell cycle of U266 cells was stopped in G0/G1 stage as the progress of concentration. It was showed statistical signiifcant difference associated with the cellular proliferation inhibition rate and apoptosis rate in different concentration and control (P<0.05). Meanwhile, 8-CPT-cAMP could induce mitochondrial transmembrane potential collapse in U266 cells. Compared with control groups, the levels of Bcl-2 mRNA transcripts and protein in U266 cells were reduced in 8-CPT-cAMP treated groups (P<0.05), while the levels of caspase-9, Bax mRNA transcription and the expression of Bax protein were increased in treatment groups, but the caspase-8 mRNA had no statistical signiifcant difference with controls. Conclusion:8-CPT-cAMP can inhibit the proliferation and promote apoptosis of myeloma cells, which might be mediated by caspase via mitochondrial pathway.
2.Sequence analysis of rDNA ITS of de-virus seedlings of Pseudostellaria heterophylla

Objective To study rDNA ITS sequence similarities and differences between the de-virus seedlings and outside seedlings of Pseudostellaria heterophylla. Methods The sequences of ITS region (including ITS1, ITS2, and 5.8S) from nine de-virus seedlings and four outside seedlings were studied by the method of DNA sequence analysis. Results There were the same sequences in 5.8S region between the de-virus seedlings and outside seedlings of P. heterophylla. Variable sites lay in the initial position of ITS1 and termination of ITS2 basically. The de-virus seedlings from Liyang (Jiangsu Province) and Xuancheng (Anhui Province) had more variable sites. Phylogenetic dendrogram based on ITS was constructed by using UPGMA methods, which showed the inherent relation at the level of molecular biology. Conclusion The analysis of rDNA ITS of P. heterophylla has further clarified the inherit stability of de-virus seedlings at the molecular level.
3.Investigation of Drinking Water Quality in Zhejiang Provinice,China,2002—2006

Objective To know the drinking water safety of water plants in Zhejiang and to provide the scientific data for government decision-making.Methods By the nonrandom sampling,the water plants were selected,103 in 2002,102 in 2003, 138 in 2004,419 in 2005,and 555 in 2006.The quality of tap water,product water and source water was determined according to the related standards.Results The eligible rate of product water and tap water in 2002 was 85.4%and 81.7%respectively,in 2003 it was 83.3%and 90.2%respectively,in 2004 it was 70.9%and 69.9%respectively,in 2005 it was 62.5%and 73.5% respectively,in 2006 it was 36.8%and 48.4%respectively.The eligible rate of source water was 68.1%.Conclusion The main problem in drinking water safety in Zhejiang is that the water supply systems in the rural areas are not good enough,no water treatment and disinfection is still common.
4.Clinical observation of phacoemulsification and intraocular lens implantation combined with trabeculectomy in cataract and glaucoma patients

AIM: To evaluate the clinical effects of dual-incision phacoemulsification and intraocular lens implantation combined with trabeculectomy in cataract and glaucoma patients.
METHODS: To observe the visual acuity, intraocular pressure and complications for 3-12mo after surgery as a retrospective study of 65 patients ( 70 eyes ) with cataract and glaucoma.
RESULTS: The visual acuity of 2 eyes was lower than 0.1, 6 eyes were ranged from 0. 1 to 0. 3, 60 eyes were from 0. 4 to 0. 8, 2 eyes were over 1. 0. The postoperative intraocular pressure of 26 eyes were effective controlled. The postoperative shallow anterior chamber occurred in 1 eye.
CONCLUSION: It is an ideal treatment to take dual-incision phacoemulsification and intraocular lens implantation combined with trabeculectomy in cataract and glaucoma patients and it shows better effects than normal operation method.
5.Improving iron deficiency anemia with orange juice and iron carried in chocolate

BACKGROUND: Iron deficiency anemia (IDA) is one of the highest incidence nutritional-deficiency diseases all over the world; especially infants and children are the main group. IDA presently becomes one of the most important nutritional problems to be solved.OBJECTIVE: To observe the effect of chocolate carrier and orange juice on recovery of IDA model rats.DESIGN: Randomized controlled animal study.SETTING: Laboratory of Nutrient and Food Hygiene, School of Public Health and Tropical Medicine, Southern Medical University.MATERIALS: The experiment was carried out in the Laboratory of Nutri ent and Food Hygiene, School of Public Health And Tropiacal Medicine, Southern Medical University from March to June 2006. A total of 60 healthy SD rats of clean grade were provided by Animal Center of Southern Medical University (certification: 2002-009 2005A047). METHODS: ① Establishment of IDA models: Among them, 20 rats of half genders were randomly selected toregard as control group, and other 40 were regarded as model group. Rats in control group were fed with rou tinefeed and drank freely. Rats in model group were fed with AOAC-modi fied low-dosage iron feeds to establish IDA models by blooding at caudal vein. Three weeks later, average concentration of ferrohemoglobin in model group was decreased to about 90 g/L, and this suggested that model estab lishment was successful. Ten rats of half genders in each group were ran domly sacrificed. Pre-experiment and 3 weeks of post-experiment, rats were weighed to measure concentration of ferrohemoglobin with hemoglobin cyanide (HiCN) technique, red blood cell count (RBC, direct method), serum iron (microparticle chemiluminescent immunoassa y and related kit) and concentration of serum transferrin receptor (STFR, ELISA method and related kit). ② Recovery test: Other 10 rats in control group were regarded as normal control group, and they were fed with routine feed and drank freely. The rest 30 rats of half genders in model group were randomly di vided into 3 subgroups: model control group, FeSO4 group and chocolate & orange juice group with 10 in each group. Rats in model control group were perfused with distilled water everyday; rats in FeSO4 group were per fused with FeSO4, and rats in chocolate & orange juice group were per fused with chocolate carrier and orange juice. The iron volume in the last two groups was 6 mg/(kg·d). At 40 days after intervention, the experiment was stopped. Concentration of ferrohemoglobin, RBC, serum iron, concentration of STFR and activity of plasma-protein aconitase were measured with atom-trapping atomic-absorption spectrophotometry; meanwhile, biological utilization rate of chocolate carrier & orange juice was calculated.MAIN OUTCOME MEASURES: ① Contents of herrohemoglobin, RBC,serum iron and STFR before experiment and after modeling; ② contents of ferrohemoglobin, RBC, serum iron, STFR and activity of plasma-protein aconitase before recovery test and at 40 days after experiment; ③ Related biological utilization rate.RESULTS: All 60 rats were involved in the final analysis without any loss. ① Comparison of blood index after modelling: Content of ferrohemoglobin, RBC and content of serum iron were lower in model group than those in control group (P < 0.01), but content of STFR was higher than that in control group (P < 0.01). ② Comparison of blood index and activity of plasma-protein aconitase in liver before recovery test and at 40 days after experiment: At 40 days after intervention, concentration of ferrohemoglobin,RBC and content of serum iron were higher in FeSO4 group and chocolate & orange juice group than those in model control group (P< 0.01); however, content of STFR was lower than that in model control group (P < 0.01).At 40 days after intervention, activity of plasma-protein aconitase in FeSO4 group and chocolate & orange juice group was higher than those before recovery test (P < 0.01). ③ Related biological utilization rate of chocolate carrier plus orange juice: Biological utilization rate of FeSO4 was regarded as 100%, and biological utilization rate of chocolate carrier plus orange juice was increased remarkably (106.7%).CONCLUSION: Chocolate carrier plus orange juice can improve IDA function and wildly use on treating IDA because of its good absorption. It is characterized by well biological utilization rate and good taste; therefore,it is a hot topic for trophology and foods produce presently.
6.Application of whole brain angiography via radial approach in young patients for the diagnosis of ischemic cerebrovascular disease

Objective To assess the feasibility and safety of transradial puncture route for digital subtraction angiography (DSA)in young patients with ischemic cerebrovascular disease. Methods One hundred young patients with cerebrovascular disease who underwent whole brain angiography at the Department of Neurology,the 81 st Hospital of PLA were enrolled. They were divided into either a radial group (n =50)or a femoral group (n = 50)using a computer random number method. The time of puncture,success rate of puncture,success rate of selective angiography,exposure time,and incidence of complications were compared and analyzed. Results (1)The puncture time of the radial group was significantly longer than that of the femoral group,they were 3. 00 ± 0. 50 min and 1. 50 ± 0. 25 min respectively. There was significant difference (t = 18. 97,P < 0. 05). There were no significant differences in the success rate of puncture (98% vs. 100%),success rate of selective angiography (100% vs. 100%),and exposure time (5. 3 ± 2. 2 vs. 4. 8 ± 1. 7 min)between the two groups (all P > 0. 05). (2)The incidence of complications of the radial group (4%,n = 2)was significantly 1ower than that of the femoral group (18%,n = 9 ). There was significant difference between the two groups (χ2 = 5. 01,P < 0. 05 ). Conclusion Transradial route puncture for whole brain DSA in young patients with cerebrovascular disease is safely and feasible.
7.Effects of Salidroside on expression of substance P and neurokinin-1 receptor of bone marrow cells in bone marrow depressed anemia mice

Objective:To observe the effects of salidroside on the expression of substance P (SP)and neurokinin-1 receptor (NK-1R)of bone marrow cells(BMCs)in bone marrow(BM)depressed anemia mice,and to explore its roles for hematopoietic regulation.Methods:Automatic blood cell analysator was used to detected the changes of peripheral blood cells in each group ,immuno-histochemistry and reverse transcription-polymerase chain reaction ( RT-PCR) were used to analyze the expressions of SP and its receptor NK-1R of BMCs in each group respectively.Results: Peripheral blood testing results showed that the number of white blood cells (WBC),red blood cells(RBC)and hemoglobin(HB)of model group were significantly decreased when compared with control group.Compared with model group ,low-dose,middle-dose and high-dose salidroside obviously elevated the number of white blood cells ,and middle-dose salidroside obviously elevated the number of platelets.Immunohistochemistry studies showed that the expression of SP and its receptor NK-1R of BMCs was found in each group.Compared with control group , the expression of SP of BMCs was decreased obviously in model group(P<0.05),and the expression of NK-1R of BMCs was decreased slightly in model group (P>0.05),and the expression of SP and its receptor NK-1R of BMCs was increased significantly in low-dose,middle-dose and high-dose salidroside groups (P<0.05).RT-PCR data showed that the expression of SP mRNA of BMCs was increased obviously in model group ,low-dose,middle-dose and high-dose salidroside groups when compared with control group (P<0.05).The expression of NK-1R mRNA of BMCs was un-detected in control group and model group.The expression level of NK-1R mRNA of BMCs was elevated gradually with the increase of salidroside dosage in low-dose,middle-dose and high-dose salidroside groups.Conclusion: The mRNA and protein expressions of SP and its receptor NK-1R of BMCs were up-regulated significantly by salidroside in dose-dependent manner.These data suggest that salidroside could participate in the recovery of hematopoietic function of BM depressed anemia mice by increasing the expression of SP and its receptor NK-1R of BMCs.
8.Therapeutic effect of external - route microsurgery for rhegmatogenous retinal detachment

AlM: To study the therapeutic effect of external-route microsurgery forrhegmatogenous retinal detachment.
METHODS: ln 55 patients ( 55 eyes ) with rhegmatogenous retinal detachment, drainage of subretinal fluid, examination of locating the holes, sclera cryotherapy, scleral buckling, and vitreous cavity injection of filtrated air were performed under surgical microscope.
RESULTS:The retinal reattachment occurred in 50 cases after the primary surgery. The final rate of reattachment was 91% during 6 - 12mo follow - up. The retinal reattachment occurred in 1 case ( recurrent retinal detachment) after the secondary surgery and in 4 cases ( recurrent retinal detachment ) after vitrectomy. The eyesight was improved with different degrees in 55 cases.CONCLUSlON: The external- route microsurgery for rhegmatogenous retinal detachment is simple, safe and effective.

AIM To study the effects of homoharringtonine(HHT) on inhibition of proliferation and induction of apoptosis of nasopharyngeal carcinoma cells CNE 2Z. METHODS The inhibitory rates of the proliferation and IC 50 were detected by MTT method. The apoptosis was analyzed by flow cytometry (FCM), agarose gel electrophoresis and Hoechst 33258/PI fluorescence staining. RESULTS After cells were treated with HHT of different concentrations for 24, 48 and 72 h,respectively,the inhibitory rates of the proliferation rised with concentration and time. Effects of mannan-oligosaccharide (MOS) on the growth of Lactobacillus were investigated by pure culture of three swine originated Lactobacillus strains or co-culture with swine pathogenic E. coli.
