1.TIMING OF MEIOTIC PROGRESSION AND MORPHOLOGICAL CHANGES IN HUMAN OOCYTES

Objective When cumulus-enclosed oocytes are isolated from ovarian follicles and cultured in vitro,they undergo germinal vesicle breakdown(GVBD) and extrude the first polar body,then remain in meiotic arrest.In this study,we examined the timing of progression of human oocytes through meiosis using in vitro maturation conditions.Measurements made include nuclear changes,oocyte diameter and the perivitelline space(PVS).Methods Oocytes were collected from polycystic ovary patients by transvaginal immature follicle puncture(TV-IMFP) and matured in vitro.At 4,8,12,16,20,24,and 28?h from the onset of culture,oocytes were put into culture droplets and analyzed under light microscope to measure their diameter and the PVS width,then fixed and stained to evaluate the chromosomal status.Results About 50% of the oocytes underwent GVBD before 10?h culture,maximum chromatin condensation peaked at 8?h to 12?h,the majority of oocytes reached metaphase Ⅰ at 20?h,and soon anaphase and telophase Ⅰ progression occurred from 20?h to 24?h.Finally about 70% of oocytes extruded the first polar body by 28?h.With the maturation progressing,oocyte diameter did not change significantly;however,the perivitelline space width enlarged from 5.27+/-0.88??m to 17.18+/-1.26??m during this time.Conclusion The results presented here delineate the timing of nuclear events in the human oocyte during maturation in vitro with this culture system. The size of PVS is related to human oocyte meiotic progression.Thus,the size of the perivitelline may be a useful indication of the maturation state of the ooeyte.
2.Ancient clinical application of herb-participated moxibustion on umbilicus.

In order to further improve the curative effect of the herb-participated moxibustion on umbilicus, we collected a wide range of literature on herb-participated moxibustion on umbilicus and then systematically arranged them to analyze and summarize the technology and operating methods of herb-participated moxibustion on umbilicus. We also briefly discussed issues on application of medicine, locust tree skin and ginger, the method of kneading dough for medical uses, and the appropriate size of moxa cone and its using frequency in order to form clear concepts and standardized operations to provide theories and operational basis for the clinical application of modern herb-participated moxibustion on umbilicus.

3.Studies on genetic relationship of Dioscorea.

Based on the results of the morphologic studies on genus Dioscorea, the paper summarized the entire chemical constituent that isolated from this genus and analyzed it with the methods of chemotaxonomy. The rules of the chemical constituent and pharmacodynamic effects were analyzed. Seventeen species which belong to Sect. Stenophora Uline of Dioscorea contain steroidal sapogenin. Other species with different main components such as polysaccharide and tannin have have different effects. This chemotaxonomic view point will conduce to establish a phylogeny of the genus Dioscorea.

6.Applying Six Sigma management to shorten the waiting time of outpatients accepting radiography

Objective To evaluate the efficacy of Six Sigma management for shortening the waiting time of outpatients accepting radiography and improving patient satisfaction. Methods Six Sigma management was applied to identify the key points critical to quality in the process of radiography,analyze the factors influencing the waiting time,and take targeted measures to modify the process. Results The waiting time of patients accepting radiography was reduced by an average of 25.2min and the rate of dissatisfaction was decreased by 18%. Conclusion The application of Six Sigma management can effectively shorten the waiting time of outpatients accepting radiography and increase patient satisfaction.
7.Study on application of hospital-school combined three-dimensional teaching mode in the operation nursing practice teaching

Objective To explore the effect of application of hospital-school combined three-dimensional teaching mode in the operation nursing practice teaching.Methods 129 students were collected by cluster sampling,then based on class unit,they were randomized into the experimental group (69 students) and the control group (60 students).The experimental group used hospital-school combined threedimensional teaching mode and the control group used traditional teaching mode.Then,both groups had finished an operation nursing comprehensive theoretical examination and a self-report questionnaire for mastery of basic knowledge of operation nursing.Besides,the experimental group had a questionnaire survey of cognitive evaluation for the new teaching mode.Results The average score of operation nursing comprehensive theoretical examination of the experimental group was (77.81 ±9.87),higher than the control group (70.35±12.37); The self-report score for mastery of basic knowledge of operation nursing of the experimental group was (3.94±0.45),higher than the control group (3.68±0.46).The students of the experimental group also considered that the new teaching mode improved their comprehensive knowledge and innovative abilities,critical thinking ability,humanistic care spirit,communication ability,team cooperation ability and innovation ability,etc.Conclusions The effect of application of hospital-school combined three-dimensional teaching mode in the operation nursing practice teaching was good,but it still had some limitations which needed us to pay attention to and to perfect.
8.Micro-PET Observation of the Effect of Electroacupuncture on Cerebral Hippocampus Glucose Metabolism in SAMP8 Mice with Senile Dementia

Objective To observe cerebral hippocampus glucose metabolism using 18F-FDG micro-PET as an imaging technique in a normal group of mice, and model and electroacupuncture groups of SAMP8 mice with senile dementia (rapid aging mice). Method Four 6-month-old SAMR1 mice were picked at random out of the normal group and every four 6-month-old SAMP8 mice, out of the model and electroacupuncture groups respectively. Each of them was anesthetized by inhalation of 2% isoflurane and then given a bolus injection of radioactive tracer 18F-FDG 14.8~16.5 MBq through the tail vein. After one hour of the uptake, a PET scan was performed for 10 min. Hippocampus 18F-FDG uptake rate per gram of brain was calculated in every group of mice and the uptake rates were compared between the groups.Result The 18F-FDG uptake rate per gram of brain tissue was higher in the electroacupuncture group of mice than in the normal and model groups.Conclusion Electroacupuncture can markedly increase cerebral 18F-FDG uptake and the uptake rate per gram of brain tissue in SAMP8 mice. It may play a neuroprotective role through its influence on cerebral glucose metabolism.
9.Establishment of orthotopic liver transplantation model in pigs without veno-venous bypass

Objective To summarize the experience of orthotopic liver transplantation in pigs without veno-venous bypass.Method In general,Bama miniature pigs were used as both the donors and the recipients.Suprahepatic inferior vena cava and portal vein anastomosis was performed with running prolene sutures.After completion of the portal vein anastomosis,the graft was reperfused.Infrahepatic inferior vena cava anastomosis and hepatic artery anastomosis were performed in a similar fashion.Finally,the common bile duct was reconstructed.Result For all of the transplant procedures,the average cold ischemic time was 356.3 ± 66.4 min and anhepatic time 22.5 ± 2.6 min,and the average operative time was 185.7 ± 24.8 min.During the anhepatic phase,the central venous pressure (CVP) and the mean arterial pressure (MAP) were significantly lower than those at baseline (P< 0.05).Heart rate (HR),on the other hand,was increased significantly during the anhepatic phase (P<0.05).By the time the portal vein and the hepatic artery were reperfused,and CVP and MAP were gradually elevated,and HR gradually reduced.All receptors were successfully extubated and awake after surgeries.On the third postoperative day they began to eat.All receptors survived during the intraoperative period,and the survival rate was 93.8% (15/16) on the fifth postoperative day.One receptor was died on the third postoperative day due to abdominal infection.Conclusion This model has satisfactory stability and reproducibility.Without using any vasoactive substances,to maintain the MAP beyond 50 mmHg in the anhepatic phase and the short anhepatic time are important to perform successful liver transplantation.
10.Isolation and Identification on a Thermoacidophilic Fungus of High-producing Cellulase and the Characteristics of its Enzyme

A novel Aspergillus terreus strain M11 was isolated from the compost containing cellulose and identified. The isolate grow best at 45℃ and pH2.0. It was found that the activity of the CMCase was up to 3.680IU/mL with high heat stability and the optimal reaction conditions of the CMCase were at 60℃ and pH2.0.
