Objective To study the effects of activation and inhibition agents of protein kinase C on the function and expression of P glycoprotein in drug resistant gastric cancer cells. Methods Immunohistochemical staining and FACS were used to determine the expression of P glycoprotein on drug resistant gastric cancer cells SGC7901/VCR and its parent cells SGC7901, which were treated or not treated by activation or inhibition agents of protein kinase C(PMA or H 7). Double labeled immunoflurescent and laser scan confocal microscopy were used to detect the coexpression of protein kinase C ? and P glycoprotein on drug resistant gastric cancer cells. Rohdamin 123 was used as fluorescent probe to observe the effects of PMA and H 7 on the function of P glycoprotein and drug accumulation in SGC7901/VCR. Results P glycoprotein had positive staining on SGC7901 cells and much stronger staining on SGC7901/VCR cells. Protein kinase C ? and P glycoprotein exhibited coexpression on SGC7901 cells and much higher coexpression level on SGC7901/VCR cells. When treated by PMA, SGC7901/VCR cells showed time dependent decreasing of drug accumulation, enhancing function and decreasing expression of P glycoprotein. When treated by H 7, SGC7901/VCR cells showed time dependent increasing of drug accumulation, weakening function and increasing expression of P glycoprotein. Conclusion Protein kinase C may play a role in multidrug resistance of gastric cancer cells via modulating the expression and function of P glycoprotein.

OBJECTIVE:To investigate the satisfaction degree of outpatients on policy of separation of clinical pharmacy from 4 third grade hospitals in Shanghai, and provide reference for promoting medical reform policy. METHODS:Parts of outpatients in the 4 new suburb third grade hospitals were investigated by the questionnaire and interview in Shanghai city, and the data was sta-tistically analyzed by Excel software and SPSS 19.0 software. RESULTS:Totally 400 questionnaires were sent out and 368 were valid with effective recovery of 94.3%. The awareness rate of medical separation policy was 81.3% in the outpatients;55.5% of outpatients thought that medicine cost savings effect was not obvious;the overall satisfaction of patients was 91.6%,and the high-est of satisfaction was the service attitude and the lowest was medical expenses. CONCLUSIONS:Policy of separation of clinical pharmacy has reduced the medical expenses, guided the triage treatment of patients partly and improved the satisfaction of patients. It is suggested to shorten the waiting time by optimizing examination process, strengthen price regulation in drug, standardize pro-duction and distribution process, improve use rate of essential medicines and promote clinical rational drug use for further improve-ment of satisfaction.

Chinese Journal of Endocrinology and Metabolism 2013;(5):427-429
[Summary] Type 2 diabetic rat model accompanied by insulin resistance was established by high fat/high glucose diet and streptozotocin.Following metformin treatment for 4 weeks, realtime PCR and Western blot were used to detect the expressions of SIRT3 mRNA and protein in skeletal muscle tissue, respectively. The results showed that insulin sensitivity, superoxide dismutase (SOD),and glutathione (GSH) levels were significantly reduced in diabetic group compared with the control group (P<0.05),while methane dicarboxylic aldehyde (MDA) level was increased (P<0.05),along with the decreased expressions of SIRT3 mRNA and protein in skeletal muscles (P<0.01).After metformin treatment, the insulin sensitivity,SOD,GSH,and SIRT3 mRNA and protein levels were significantly increased (P< 0.05 or P< 0.01),while MDA level was decreased (P < 0.05),suggesting that metformin may ameliorate insulin resistance via upregulating SIRT3 expression in skeletal muscles of diabetic rats.

A comprehensive evaluation of zinc and iron nutritional status was conducted in 203 children aged 2-7 years with anorexia. 147 of them took 3-7 mg of zinc sulfate?kg-1?d-1 for 60 days. The purpose of the study were to explore the relationship between zinc supplementation and the nutritional status of iron.Nutritional survey by 3-day-record method pre-and post zinc treatment showed that the average intakes of energy, protein, zinc, iron before treatment were 994kcal/d, 30.8g/d, 4.9mg/d, 7.6mg/d and were only 71%, 68%, 49%, 76% of RDA respectively. After treatment they reached 91%, 89%, 70%, 113% of RDA.There were significant positive correlations between zinc contents in serum or hair and hemoglobin (HB) or serum ferritin (SF) of the children with anorexia. After administration of zinc , the iron status of the children improved in some and fell in others. There were significant differences in some indices between the two groups before treatment. By stepwise discriminant analysis, a discriminant equation was obtained;Y = 3.591 - 0.175X1- 0.0152X2- 0.144X3+ 0.706X4(X1 = age, X2 = hair zinc,X3 = Hb, X4 = growth)If Y0, iron status may improve.
