1.Determination of Nitrite in Water by High Performance Liquid Chromatography

ve To establish a method of high performance liquid chromatography (HPLC) for determi-nation of nitrite in water. Methods The trace content of nitrite in water sample was determined by indirect UV-HPLC. The mixture of methanol/o-phthalic acid (pH value was adjusted to 8.6 by 0.10 mol/L NaOH solution)5: 95 was defined as mobile phase. The water sample was directly filtered by chromatographic column pack with ODP. Results Under the conditions of wave length of 270 nm and flow rate of 0.9 ml/ min, the linear range of this assay was 0.0~20.0?g/ml nitrite, the relative standard deviation was 3.9%. The average recovery rate and detection limit were 99.9% and 0.001?g/ ml respectively. Conclusion This method could be applied to the determination of trace amount of nitrite in drinking water, purified water and mineral water.
2.The dose rate response characterization of four kinds of dosimeters for linear accelerator

Objective To explore the dose rate response characterization of four kinds of dosimeters for clinical application.Methods Within the range of 100-600 cGy/min,the dose rate responses of the PTW 0.6 cm3 ion chamber,0.015 cm3 ion chamber,Matrixx Evolution 2D diode array and MapCHECK 2D diode array under the same measuring conditions were measured.The dose rate response of the PTW 0.6 cm3 ion chamber under different energy and working voltage were analyzed.Results All ionization chambe.r types of measured equipment showed certain dose rate dependence for 6 MV X-rays.All the differences were below 1%.The dose rate dependence disappeared for 15 MV X-rays.The 2D diode array had strong dose rate dependence and the response difference was about 2%.Conclusions It is necessary to test and analyze the dose rate response of the measured equipment in treatment technology with dose rate varying,in order to ensure the precision of daily calibration and dose verification.
3.Relationship between intrahepatic interleukin-17 expression and hepatic fibrosis in chronic hepatitis B virus infection

Objective To explore the relationship between intrahepatic expression of interleukin-17 (IL-17) and liver fibrosis in patients with chronic hepatitis B virus infection. Methods IL-17 expressions in livers with different inflammation activity grades and hepatic fibrosis stages from patients with chronic hepatitis B virus carriers (n= 30), chronic hepatitis B (CHB, n = 55), liver cirrhosis (LC, n=20) were measured by immunohistochemistry. Serum IL-17 and liver fibrosis indices of haluronic acid (HA), laminin (LN), type Ⅲ procollagen (PC Ⅲ ) and type Ⅳ collagen ( Ⅳ C) were determined by enzyme linked immunosorbent assay (ELISA). The differences between groups were compared by Kruskal-Wallis test, and the Mann-Whitney test, and the correlation analysis was done by Spearman test. Results Intrahepatic IL-17 expression in LC group was significantly higher than CHB group (x2 =25. 3982, P=0. 004), and that in CHB group was higher than chronic hepatitis B virus carriers group (x2 = 11. 5056, P= 0. 001). The inflammation activity grade and hepatic fibrosis stage were both positively correlated with IL-17 expression (r= 0.718, 0. 693, respectively; both P<0.01). IL-17 mainly located in portal area and the expression was positively correlated with serum levels of HA, LN, PCⅢ and ⅣC (r=0. 793, 0. 834, 0. 722, 0. 883, respectively; all P<0.01).Conclusion Intrahepatic IL-17 expression is closely correlated with liver inflammation activity grade and hepatic fibrosis stage.
4.Long-circulating methotrexate-loaded liposomes exhibit an antitumor effect on human osteosarcoma in vitro

BACKGROUND:Liposomes as a new drug delivery system are characterized by few adverse reactions, no immunogenicity and biodegradation. Furthermore, methotrexate-loaded liposomes can significantly reduce drug toxicity and improve anti-tumor effect. OBJECTIVE:To prepare long-circulating methotrexate-loaded liposomes and to evaluate its antitumor activity in MG-63 in vitro. METHODS:The methotrexate-loaded liposomes were prepared using the film dispersion method, and the long-circulating ones were prepared using the post-insertion method. The initial concentrations of methotrexate were 9.1, 1.82, 0.364 g/L. The ultracentrifugation method and spin column method with Sephadex G-10 or G-50 as packing were employed to separate free drugs from the methotrexate-loaded liposomes. Their recovery, size, morphology, encapsulation efficiency and drug-to-lipid ratio were evaluated. The cytotoxity of the long-circulating methotrexate-loaded liposomes purified with ultracentrifugation method and spin column G-50 method under three dose levels (0, 1, 5, 25 mg/L) were determined by MTS method. RESULTS AND CONCLUSION:According to the recovery rates of three methods, the spin column G-50 method was considered as optimal for the long-circulating methotrexate-loaded liposomes. The long-circulating liposomes were spherical or el ipsoidal under transmission electron microscope, about 200 nm in size. At the certain initial concentration of methotrexate, the encapsulation efficiency and drug-to-lipid ratio of the liposomes purified using the spin column G-50 method were remarkably higher than those purified using the other two methods. At the same mass concentration, the cytotoxity of the liposomes purified with ultracentrifugation or spin column G-50 was significantly higher than that of free methotrexate, and furthermore, the cytotoxity of the liposomes purified with spin column G-50 was higher than that of the liposomes purified with ultracentrifugation method. To conclude, the long-circulating methotrexate-loaded liposomes show a higher antitumor activity than free methotrexate in MG-63 cel s in vitro, providing the basis for further investigation of its antitumor effect on human osteosarcoma in vivo.
5.Analysis the effects of film dosimetry during processing and scanning

ObjectiveTo explore the effects of different flushing and scanning processing on optical density (A) responding and set up the clinic quality assurance protocol based on silver halide emulsions radiographic films.MethodsSetting different flushing temperature and choosing different batch's film and developer and fixer or fixer in the same batch with different analyzing dose were performed to analyze the effect on A value; The effect of light uniform,the stabilize work time and the noise of different scanning resolution were discussed.ResultsThe A value at the same dose level would enhance as the temperature increased;the responding curve of dose and A value with different batches of films varied a lot;the responding curve of dose and A value with different batches of films and developer and fixer solution had marked variations;the responding of dose and A with the same batches would show some low.The heterogeneity of the scanner would achieve 0.03 ; the A value of the same dose would gradually steady while the time ofscanning was more than 10 minutes:the affect of noise would increase as the dose and resolution ratio increased.ConclusionsThe best processing temperature is 29-31 ℃.Different batches of film couldn't be confounded.A new calibration must be obtained when the film dosimetry to evaluate dose distribution is used.A 5 - 10 min warm up for stabilize work and the best setting resolution/depth lever are 72 dpi/16 bit for scanning films are determined.
6.Effects of individual nostalgia on positive feelings and sadness of spouses of postoperative patients with esophageal cancer

Objective To investigate the effect of individual nostalgia therapy on positive feelings and sadness of the spouses of postoperative patients with esophageal cancer. Method From June 2014 to June 2015, 60 hospitalized patients and their 60 spouses were set as control group. Routine health education and guidance were conducted once or two times a week for 7 weeks. Another 60 cases of hospitalized patients and their 60 spouses during July 2015 and June 2016 were set up as observation group. The patients were treated with the same nursing intervention as in the control group, and their spouses were subjected to individual nostalgia treatment once a week for 7 weeks. Before the intervention (postoperative day 2) and 7 weeks after the intervention, the positive aspects of caregiving (PAC) and the Margit-Meuser caregiver grief ineventary short form (MM-CGI-SF) were used to investigate their positive feelings and sadness. Result The score on spouse's positive feelings in the observation group after intervention was significantly higher than those of the control group and that of the observation before intervention (P < 0.01). Conclusion Individual nostalgia can effectively improve the feelings of the spouses of esophageal cancer patients and reduce their grief.
7.Adverse Drug Reactions in Hematological System Induced by Antituberculosis Drugs:Literature Review of 441 Cases

OBJECTIVE:To evaluate the abnormality in hematological system induced by antituberculosis drugs so as to promote the rational use of drugs in the clinic.METHODS:The 441 case reports on abnormality in hematological system induced by antituberculosis drugs retrieved from Chinese medical science periodicals from Jan 1990 to Apr 2007 collected in CHKD periodicals knowledge base in China hospital digital library were analyzed statistically.RESULTS:A total of 15 antibuberculosis drugs were involved in the ADR cases,leading the list were rifampicin,ciprofloxacin and isoniazide,which resulted in a total of 211 ADR cases.Leukopenia was the chief clinical manifestation.CONCLUSIONS:It is essential for clinical practitioners master the distribution patterns of ADR in hematological system caused by antituberculosis drugs so as to promote medication safety.
8.Nitric oxide and endoplasmic reticulum stress

Nitric oxide (NO) is a multifunctional biomolecule involved in a variety of physiological and pathological processes, including regulation of blood vessel dilatation and function as a neurotransmitter. However, a large amount of NO is toxic to the host and causes several diseases such as cardiovascular system diseases, septic shock, and diabetes mellitus. Endoplasmic reticulum (ER) stress pathway was first identified as a cellular response pathway induced by the accumulation of unfolded proteins in ER to preserve ER functions. Later it was found that ER stress pathway is also activated by various cellular stresses to protect cells, but when stresses are severe, apoptosis is induced to remove damaged cells. It is reported that NO disturbs ER functions, then ER stress-mediated apoptosis pathway is activated. CHOP/GADD153, which belongs to C/EBP transcription factor family, is induced in this process and mediates apoptosis. ER stress pathway induced by NO is involved in the pathogenesis of various diseases.

10.Bone formation induced by purified BMP and by recombinant human BMP-2

Objective: To study the biodegradable of coral PLA composite artifical bone combined with bBMP or rhBMP as a new kind of bone substitute material. Methods: The composites were implanted into the muscle pouches of mice after combined with rhBMP-2 or bBMP respectively. Ectopic osteoinductive activity of rhBMP-2 or bBMP was examined and compared by histology and histo-morphometry.Results: rhBMP-2 and bBMP had different osteoinductivety. rhBMP-2 appeared to induce less bone and more angioid tissue and marrow. While bBMP seemed to have opposite effects. Conclusion: bBMP is more osteoinductive than rhBMP-2.
