1.EFFECTS OF GLUCOSAMINE ON IMMUNE FUNCTION IN MICE

Objective:To study the effects of glucosamine(GlcNH2) on immune function in mice.Method:The effects of GlcNH2 on murine proliferation of splenocytes were carried out in vitro.After feeding mice by GlcNH2,the phagocytotic functions of mononuclear macrophage,murine delayed type hypersensitivity(DTH) caused by sheep red blood cells(SRBC),the ability of antibody production(tested by HC50),and the index of immune organs(thymus and spleen) were deteimined in vivo.Results:GlcNH2 could promote the proliferation of splenocytes,phagocytotic functions of mononuclear macrophage,DTH,the ability of antibody production and the index of immune organs.Conclusion:Glucosamine can enhance immune function in mice such as cellular immunity,humoral immunity and non-specific immunity.
2.Surgical treatment of refractory ulcerative colitis, report of 60 cases

Objective To evaluate the clinical effect of restorative proctocolectomy with ileal pouch-anal anastomosis (IPAA) for refractory ulcerative colitis (UC).Methods In this study 60 refractory UC patients received IPAA operation during the period of 1990 to 2010.Data were collected regarding early and late postoperation complications,anal continence function,and characteristics of feces.The patients' quality of life was objectively accessed using the Clevend Global Quality of Life (CGQL)index.0ne-way analysis of variance was used.Results Mean follow-up period was 2 years.Early postoperative complication rate developed in 15% (9/60),including abdominal or pelvic infection,anastomotic leak,pouch bleeding,pouch-vaginal fistula,and intestinal obstruction.Late postoperative complication rate was 12% (7/60),including pouchitis,intestinal obstruction,and male sexual dysfunction.Stool frequency per 24 hours and that at night was 3.5 ± 1.3 and 1.4 ±0.6.93% (56/60)patients differentiated gas and feces well and 3% (2/60) needed daily pads.According to Kirwan Grading Scale,anal function outcomes were Grade Ⅰ:54 (90%),Grade Ⅱ:4 (7%),and Grade Ⅲ:2 (3%).According to Bristol Stool Form Scale,characteristics of feces were Grade Ⅳ:31 (52%),Grade Ⅴ:25(42%),and Grade Ⅵ:4 (6%).Postoperative CGQL result showed a much better quality of life than preoperative CGQL (F =12.368,P < 0.05).Conclusions Refractory UC is surgically indicated and IPAA is the operation of choice with safety,satisfactory long-term outcome and improved quality of life.
3.Treatment of Asthenozoospermia Men with Shen-essence Deficiency Syndrome by Yishen Zhongzi Pill: a Clinical Study.

OBJECTIVETo observe the effect of Yishen Zhongzi Pill (YZP) on the sperm motility of Shen-essence deficiency syndrome (SEDS) patients.
METHODSTotally 144 patients were randomly assigned to the treatment group (71 cases) and the control group (73 cases). Ten cases dropped out during the therapeutic course. A total of 134 patients completed this clinical study, 67 in each group. Patients in the treatment group took YZP (6 g each time, twice daily), while those in the control group took Wuzi Yanzong Pill (6 g each time, twice daily). The therapeutic course for all was one month. Changes of the sperm motility, clinical efficacy, and Chinese medical symptoms scores were observed between the two groups before and after treatment.
RESULTSCompared with before treatment in the same group, percentages of class a spermatozoa and class a + b spermatozoa were enhanced in the two groups after treatment (P <0. 01). Besides, they were higher in treatment group than in the control group (P <0. 05). The total effective rate of the treatment group was 77. 61 % (52/67 cases), while it was 58. 21 % (39/67 cases) in the control group. It was better in the treatment group than in the control group (Z= -2. 914, P =0. 004). Compared with before treatment in the same group, scores for Chinese medical symptoms decreased in the two groups (P <0. 01). The difference of pre-post integrals was higher in the treatment group than in the control group (P <0. 01).
CONCLUSIONYZP showed favorable effect in treating asthenospermia patients with SEDS and could effectively elevate the motility of sperm.

5.Characteristic and Resistance of Fungi Isolated from Hemopathy Patients

OBJECTIVE To investigate the distribution and resistance of fungi isolated from hemopathy patients,and to provide the laboratorial data in order to prevent and treat infection caused by fungi more effectively.METHODS Strains were isolated from the upper respiratory tract,lower respiratory tract,genitourinary tract,alimentary tract;and perianal region,perineum,and the soft tissue and skin wounds.Antimicrobial susceptibility of clinical isolates was tested by broth microdilution susceptibility test.RESULTS Candida albicans,Aspergillus,tropicalis,C.krusei,and C.glabrata were the main fungi of total 3104 detected isolates.The resistance rates of C.albicans and C.tropicalis to fluconazole,itraconazole and 5-flucytosine were no more than 1.4%.The MIC50-MIC90 range of C.tropicalis for itraconazole was narrow(0.25-0.5 mg/L),but that was wide for fluconazole(0.25-4.0 mg/L).CONCLUSIONS C.albicans,Aspergillus,C.tropicalis,C.krusei,and C.glabrata are possibly the most common pathogens causing endogenous and exogenous fungi infection.The main infection sites of Aspergillus are the upper respiratory tract and lower respiratory tract,which may be associated possibly with ward's air quality.The MIC50to MIC90 range of C.tropicalis is wider for fluconazole than for itraconazole,which is possibly correlated with overexpression of CtMDR1 in C.tropicalis caused by the widely application of fluconazole in therapy.
6.Screening and identification of HIV-1 gp41 C-helix mimotopes displayed on phages

Objective:To find small molecular leads for inhibition on early stage of HIV infection by identification and characterization of the HIV-1 gp41 C-helix mimotopes.Methods:For identification of the gp41 C-helix mimotopes,C7C phage display peptide library was biopanning by using a synthetic peptide N36 which was derived from the gp41 N-helix as target.After three rounds of screening,positive phage clones were identified by ELISA and sequenced.Results:16 of 26 phage clones were identified to bind with peptide N36,and 10 of them were sequenced.Every clone of ten clones contains at least two hydrophobic residues,which may dock into the hydrophobic pocket in the gp41 N-helix domain.9 of the 10 clones have a conservative sequence WW,which may mimic the W628 and W631 in C-helix to interact with the hydrophobic residues in the gp41 pocket.One clone expressing the conservative sequence named clone No.8(CYWWHRLHC) was selected for characterization.The binding between the clone No.8 and N36 was blocked by free peptide N36.And the binding between clone No.8 and peptide N36 was inhibited by peptide C34(IC 50=12.5 ?g/ml).Conclusion:The short circular peptides displayed on phages containing WW residues may mimic the conformational epitope of the HIV-1 gp41 C-helix to interact with the N-helix.This information may be useful for design of HIV-1 fusion inhibitors.

8.Study on Quality Standards of Pseudostellaria heterophylla

OBJECTIVE:To establish the quality standards of Pseudostellaria heterophylla.METHODS:P.cyclopeptides was identified by thin layer chromatography protosite reaction with ninhydrin reagent.The contents of moisture,total ash,acid-insoluble ash,water-soluble extract and alcohol-soluble extract were detected according to the standards stated in Chinese Pharmacopoeia.The content of pseudostellarin B was determined by HPLC.RESULTS:TLC identification of P.cyclopeptides and HPLC determination of pseudostellarin B were established preliminarily.Limitation of moisture content,ash content,extract and pseudostellarin B were defined.CONCLUSION:The qualitative method and quantitative method and physicochemical index can be used as standards for the quality control of P.heterophylla.

AIM:To prepare sustained release matrine tablet,and study its drug release test in vitro. METHODS: In combination with HPLC,we studied the cumulative matrine release using HPLC and EC as the sustained materials. RESULTS: The release of matrine was conformed to Higuchi equation. CONCLUSION: The drug release behavior could be well described in vitro,and meet the design criteria,it is easy to be produced.
10.Effects of two UDP-glucose dehydrogenases on hyaluronic acid biotransformation.

We amplified genes encoding UDP-glucose dehydrogenase, ecohasB from Escherichia coli and spyhasB from Streptococcus pyogenes. Both ecohasB and spyhasB were inserted into T7 expression vector pRX2 to construct recombinant plasmids pRXEB and pRXSB, and to express in E. coli BL21(DE3). After nickel column purification of UDP-glucose dehydrogenases, the enzymes were characterized. The optimum reaction condition of spyHasB was at 30 °C and pH 10. The specific activity reached 12.2 U/mg under optimum condition. The optimum reaction condition of ecoHasB was at 30 °C and pH 9. Its specific activity reached 5.55 U/mg under optimum condition. The pmuhasA gene encoding hyaluronic acid synthase was amplified from Pasteurella multocida and ligated with ecohasB and spyhasB to construct the coexpression vectors pBPAEB and pBPASB, respectively. The co-expression vectors were transformed into E. coli BW25113. Hyaluronic acid (HA) was produced by biotransformation and the conditions were optimized. When recombinant strains were used to produce hyaluronic acid, the higher the activity of UDP-glucose dehydrogenase was, the better its stability was, and the higher the HA production could reach. Under the optimal conditions, the yields of HA produced by pBPAEB/BW25113 and pBPASB/BW25113 in shake flasks were 1.52 and 1.70 g/L, respectively, and the production increased more than 2-3 folds as previously reported.
