1.Effect of Tumor Promoter PMA on Cell-mediated lmmunity Ⅲ. PMA Inhibits Antigen Presentation by Spleen Adherent Cells

TNP-conjugated spleen adherent cells (SAC) from normal BALB/c mice could stimulate lymph-node T lymphocytes (LNT) of 1, 3, 5-trinitrobenzene (TNCB) -primed syngeneic mice to proliferate in vitro. The proliferative response was suppressed when phorbol myristic acetate.(PMA) was present in the culture. SAC pretreated with PMA also suppressed markedly the response. Furthermore, PMA was shown to inhibit interleukin-1 (IL-1) production and/or secretion by macrophages in response to LPS stimulation in vitro. Therefore, the suppressive effect of PMA on antigen presentation seems to be due to its inhibitory effect on IL-1 production and/or secretion.
2.Effect of Restraint and Watered Stress of Different Durations on Gastric Empting in Rats

Objective To study the effect of restraint and watered stress of different durations on the gastric empting in rats. Methods 48 male SD rats (weight ±150 g) were divided randomly into 6 groups with 8 in each. The experimental rats were restrained and watered in 23℃ water box for 1 h/d, while the controls were free. The body weight, general conditions and behavioral indexes of the rats were observed and recorded. The gastric empting rate was measured in rats with semi-solid diet after 3 d,7 d and 28 d during the statistically significant differences among different groups of different stress groups,and between the 3 d test group and 3 d control group. The gastric empting rate was higer in the 7-day test than in 7 d controlled group, lower in the 28 d test group than 28 d control group. Conclusion Different chronic stress can cause changes of gastric empting rate. With the prolonging of stress time, the empting rate is rising and then decending.
3.Observation of the efficacy of Aidi Injection combined with chemotherapy in the treatment of malignant advanced tumor

AIM: To observe the clinical effecacy of Aidi Injection (Radix Ginseng, Radix Astragali, Mylabris, Radix et Caulis Acanthopanacis Senticosi) combined with chemotherapy in the treatment of malignant advanced tumor. METHODS: Patientes (106 cases) with advanced malignant tumor were randomized into two groups: therapeutic group (56 cases) and control group (50 cases). The therapeutic group:the chemotherapy combined with Aidi Injection (60~80mL), once a day, for 10 days. The control group:the chemotherapy alone. Two groups mainly used PTX、DDP、EPI、5 FU、VP16 etc for chemotherapy. RESULTS: The therapeutic group has different degrees of improvement in life quality, short term efficacy, leukooytai drop and T cellular subfamily, etc. There was markedly difference between the therapeutic group and the control group ( P
4.Differential proteomics research on exosomes derived from tongue squamous cell carcinoma cells and normal mucosa cells.

OBJECTIVEThis study aimed to explore further the mechanisms of tongue squamous cell carcinoma (TSCC) cell recurrence, metastasis, and diffusion, as well as to establish the experimental basis for the molecular biology research on TSSC. We intend to complete our objective through differential proteomics and preliminary analysis protein expression of exosomes derived from TSCC and normal mucosa cells.
METHODSWe acquired cultured supernatant fluid in vitro in the laboratory by culturing TSCC (tongue cancer Tca8113 cell line) and human normal mucosa cells (HOK cell line). The exosomes were separated and purified through differential centrifugation. Furthermore, the different protein expressions were identified through dielectrophoresis and mass spectrometry. The functions of the different protein expressions were identified through an online database search.
RESULTSTSCC and human normal mucosa cells secrete a large amount of capsule bubble structure substances in vitro, as confirmed by electron microscopy and surface markers heat shock protein-70 and major histocompatibility complex class 1. A total of 16 oral cancer cell-derived exosomes that expressed quantity more than two times, twelve that increased their expression levels, and four that cut their expressions were identified through the differential proteomics research on the two groups.
CONCLUSIONDifferential proteins that were verified through the online database serve an important function in exosome formation and in the progress of cancer.

5.Clinical observation of dynamic blood sugar monitoring combined with insulin pump in the treatment of newly diagnosed type 2 diabetes mellitus in the elderly

Objective To evaluate islet cells functional recovery and clinical effects of dynamic blood sugar monitoring combined with insulin pump in newly diagnosed type 2 diabetes mellitus in the elderly.Methods 300 cases with newly diagnosed type 2 diabetes mellitus were randomly divided into treatment group (n=150) and control group (n=150) accompanying with three months follow-up.Body mass indexes (BMI),levels of hemoglobin A1c (HbA1c),fasting plasma glucose (FPG),2-h postprandial blood glucose,fasting insulin,2-h postprandial insulin,fasting C-peptide,2-h postprandial C peptide,and the percentage of patients who took oral hypoglycemic drugs alone without insulin could well control the blood glucose were compared between the two groups before versus after treatment.Results The levels of BMI,HbA1c,FPG,2-h postprandial blood glucose,fasting insulin,2-h postprandial insulin,fasting C peptide,2-h postprandial C peptide in the two groups were significantly decreased after treatment as compared with the pretreatment (P < 0.05).The improvement of blood sugar spectrum was better in treatment group than in control group(P<0.05) and the percentage of patients who took oral hypoglycemic drugs alone without insulin could well control the blood glucose was significantly higher in treatment group than in control group(21% vs.3%,x2 =22.475,P<0.05).Conclusions The clinical effects of dynamic blood sugar monitoring combined with insulin pump on newly diagnosed type 2 diabetes mellitus in the elderly is definite and can improve the recovery of islet cells function.It is worthy of clinical promotion.
6.Clinical and imaging features of vertebrobasilar dolichoectasia in patients with ischemic stroke: a retrospective case series study

Objective To investigate the clinical and imaging features in patients with vertebrobasilar dolichoectasia (VBD) through a comparative study in patients with ischemic stroke with or without VBD.Methods The patients with acute cerebral infarction were divided into either a VBD group or a non-VBD group according to magnetic resonance angiography.The VBD group was further divided into an anterior circulation infarction subgroup and a posterior circulation infarction subgroup.The cardiovascular risk factors,the diameter of basiar artery (BA),bifurcation height,and horizontal displacement were compared in all groups.Results A total of 269 patients with acute cerebral infarction were included,28 had VBD,accounting for 10.41% of the patients with acute cerebral infarction during the same period.The proportion of male patients (78.6% vs.66.8% ;x2 =4.392,P =0.036),age (70.38 ± 10.58 years vs.62.86 ± 12.20 years; t =2.870,P =0.009),and the proportion of hypertension (89.3% vs.47.7% ; x2 =17.367,P =0.000) in the VBD group were significantly higher than those in the non-VBD group.The multivariate logistic regression analysis showed that the advanced age (odds ratio [OR] 1.248,95% confidence interval [CI] 1.137-1.371; P=0.000),hyperglycemia (OR 1.599,95% CI 1.181-2.164; P =0.002),hypertension (OR 1.251,95% CI 1.020-1.534; P =0.032) and increased triglyceride level (OR 1.876,95% CT 1.021-3.445; P =0.043) were the independent risk factors for VBD,while female gender (OR 0.133,95% CI 0.024-0.735; P =0.021) was the independent protective factor for VBD.Of the 28 cerebral infarction patients with VBD,9 had anterior circulation infarction and 19 had posterior circulation infarction.There were significant differences in BA diameter ([5.40 ± 0.49] cm vs.[6.00 ± 0.77] cm; t =2.046,P =0.041),and the proportions of high score in bifurcation height (x2 =6.768,P =0.037) and horizontal displacement (x2 =5.241,P =0.042) between the 2 groups (all P <0.05).The multivafiate logistic regression analysis showed that the BA bifurcation height was an independent risk factor for posterior circulation infarction (OR 1.347,95% CI 1.069-2.457; P =0.038) in patients with VBD.Conclusions VBD accounted for 10.41% of the patients with acute cerebral infarction during the same period.Advanced age,hyperglycemia,hypertension and increased triglyceride level were the independent risk factors for VBD.Female gender was the independent protective factors for VBD,and the BA bifurcation height was an independent risk factor for VBD occurring posterior circulation infarction.

8.Free-hand cervical pedicle screw fixation for upper cervical fracture and instability

Objective To evaluate the clinical effect of the free-hand cervical pedicle screw fixation in treatment of the upper cervical fracture and instability.Methods A retrospective review was performed on 15 patients with upper cervical fracture and instability treated with cervical pedicle screw fixation and fusion from September 2006 to January 2009.There were 11 males and 4 females,at average age of 41.2 years(range,18-60 years).Of all,there were five patients with atlas fracture and dislocation(including three simple anterior arch fractures and two Jefferson fractures),three with axis fracture and dislocation,one with dens fractures plus nonunion,two with C2,3 fracture and dislocation and four with atlantoaxial instability without fracture.The main clinical complaints included local neck pain and/or tetraplegia.Halo traction was recommended to restore the cervical sequence preoperatively in all patients.All 15 patients were treated by cervical pedicle screw-rods internal fixation and bone graft fusion.During the operation,the point and angle of the implanted pedicle screws were determined by preoperative X-ray and CT scan and the bony channel drilled with free-hand before implantation of the Summit or Vertex pedicle screws(22-26 mm long)and posterior interlaminar autologous or allogeneic bone fusion.Patients could get out of bed with neck collar at days 1-2 after operation.Results A total of 64 cervical pedicle screws were implanted in all 15 patients,with no vertebral artery injury,spinal cord injury or cerebrospinal fluid leakage.Postoperative X-ray and CT scan confirmed satisfactory internal fixation.The clinical symptoms were improved significantly.Fourteen patients were followed up for 12-36 months,which showed bony fusion,with no looseness or breakage of the screws.Neurologic impairment was improved in all patients,with no complications associated with the cervical pedicle screw.Conclusions Cervical pedicle screw internal fixation can reestablish the upper cervical vertebrae stability and help to recover the spinal cord and nerve function and hence is a reliable method for upper cervical fracture and/or instability.
9.Aorta-pulmonary septal defect and aortic origin of the right pulmonary artery with interruption of the aortic arch: a clinical analysis of 5 cases

Objective Reviewing the experience in the diagnosis and treatment of cases with aorta-pulmonary septal defect and aortic origin of right pulmonary artery complicated with interruption of the aortic arch. Methods Reviewed clinical data from medical records for5 patients (4 boys and 1 girl, age from 1 to 18 years on admission) who had been treated in Wuhan Asia Heart Hospital between 2005 and 2009. Results All cases had type A interruption of the aortic arch ( according to Celoria and Patron's Classification) , type Ⅱ aorta-pulmonary septal defect( according to Mori Classification) and aortic origin of right pulmonary artery complicated with interruption of the aortic arch. Large patent ductus arteries were observed in all cases,and no cardiac abnormality such as ventricular septal defect was found. All patients received echocardiography and spiral CT examination, and 3 of them over 10 years of age received cardiac catheterization and angiography for the evaluation of the pulmonary artery pressure. Two patients, at the age of 14 and 18 years, had severe pulmonary hypertension and declined to receive the operation. One of the 2 cases had hemoptysis and was given prostacyclin ( Iloprost solution for inhalation, a drug for pulmonary artery hypertention ) for reducing the pulmonary pressure. The patient now has an improved cardiac fu0ction without recurrent hemoptysis. One 14-year-old case gave up the operation because of the financial problem and failed to communicate with us after discharge. Radical surgery was performed in the remaining three cases, one had serious infection in the lung and died 11 days after the operation, in one case ( 2 years old) pulmonary artery hypertension has been persisted even though drug therapy was given for a long time and was found at a poor cardiac function 18 months after the operation. One patient recovered well under routine drug therapy and has been followed-up. Conclusion Aorta-pulmonary septal defect and aortic origin of right pulmonary artery with interruption of the aortic arch ( with intact ventricular septum) are different from the classic triad of inter ruption of the aortic arch, consisted of the interruption of the aortic arch, patent ductts arteriosus and ventricular septal defect.It should be considered as a special kind of cardiovascular malformation syndrome. Echocardiography can be used for the preliminary diagnosis of aorta-pulmonary septal defect and aortic origin of right pulmonary artery with interruption of the aortic arch. The final diagnosis and decision for surgery should be based on the combined information from cardiac catheterization,cardiovascular angiography and spiral CT examination. Pulmonary artery pressure and resistance are the most important factors,because they have great effects on the duration of surgery and the outcome. A radical operation is necessary for all cases, but the mortality rate is high. The prognosis would be great if only the patient can survive successfully during the peri-operative period, a period with a poor cardiac function, potential lung infection and the pulmonary artery hypertension.
10.Sensitive genes screening of mice liver injury induced by hepatotoxicant and their biological function analysis

Objective:To scan the common differentially expressed genes in mice liver tissue following subacute exposure to hepatotoxicants,including alcohol,carbon tetrachloride,BCG & LPS,the alcohol extracts of Rhizoma Dioscoreae Bulbiferae and Senecio scandens Buch.-Ham.,and apply to the rapid toxic evaluation of TCM and other xenobiotics.Methods:The gene expression profiles of mice liver tissue,after respective administration of alcohol,CCl4,BCG associated with LPS,the alcohol extracts of Rhizoma Dioscoreae Bulbiferae and Senecio scandens Buch.-Ham.to healthy mice,were analyzed by use of mouse genome cDNA microarray.The correlations between the common differential expressed genes and the liver injury were analyzed based on the biological functions of those differentially expressed genes.Results:Among all of the five drug administration groups,there were 7 known function genes(BC05200,NM-019410,NM-173019,AB028272,AK088925,AK030862 and AK088816)and 1 unknown function gene(BC069871)differentially expressed.Beside AK088816,the other 7 genes were all down-regulate expressed.Conclusion:The common differentially expressed genes participate in the process of saccharometabolism,apoptosis,cell growth cycle,cytoskeleton and signal transmission,protein folding and protein ubiquitin.The abnormal expression of the common genes were closely correlated to the liver,and might be important for rapid hepatoxicity elucidation of traditional Chinese medicine and other xenobiotics.
