1.Study on the current status and influential factors of neglect of left-behind children in rural area of Anhui province

Objective To understand the status and influential factors of those neglect of left-behind children in rural area,and to provide bases for the development of intervention measures.Methods 2917 students were selected as the study subjects from Changfeng county of Anhui province with cluster sampling method and were evaluated by a Parents-Child Conflict Tactics Scales and questionnaire on influential factors.Results 1694 left-behind children,accounted for 58.1% of the total students,were surveyed in this investigation.The prevalence rates of neglect,among total children,left-behind children,non-left-behind children were 67.4%,70.2%,63.5%,respectively.The prevalence of neglect among left-behind children was higher than that among non-left-behind children (x2=14.322,P<0.000).There were no significant associations with the neglect rate of left-behind children regarding gender or age differences.Result from multivariate logistic regression analysis indicated that the neglect among the left-behind children were associated with family dysfunction(OR values of moderate and serious family dysfunctions compared to good family function were 1.628 and 2.341,respectively)and the rate of keeping in touch with parents(OR values of sometimes and seldom keeping in touch compared to regular in touch were 1.299 and 1.844,respectively).The starting age of being left-behind(OR values of starting age that being left-behind from 6 to 10 and ≤5 years relative to starting age of left-behind ≥11 years were 0.703 and 0.630,respectively)appeared to be the protection factor to the neglect of those left-behind children.Conclusion Our findings indicated that the status of neglect among the left-behind children was serious.Prevention programs on the issue should target on a number of factors,including the characteristics of the chldren them-selves,as well as on the family of the children.
2.The preoperative size measurement of breast cancer by ultrasonography and mammography

Objective To comparatively analyze the preoperative value of the size of primary breast cancer presenting as a mass by ultrasonography and by mammography.Methods The tumor size data obtained preoperatively by mammography and by ultrasonography were retrospectively analyzed in all the patients pathologically diagnosed with primary breast cancer in our hospital from July 2006 to July 2009.The Pearson correlation coefficient was calculated for the size of tumors between with pathological method and with each of the imaging modalities.Results A total of 57 patients with breast cancers took both of imaging modalities including 51 cases of infiltrating ductal cancer,4 cases of infiltrating lobular cancer,and 2 of ductal carcinoma in situ.The ranges of the maximum diameter of tumor was 0.6-11.8 cm and mean value was (2.3±2.1) cm by pathological examination,0.6-12.6 cm and mean value was (3.4±2.9)cm by mammography and 0.7-10.2 cm and mean value was (2.4±1.8)cm by ultrasonography,respectively.The Pearson correlation coefficient in the accuracy of imaging assessments was superior to those by ultrasonography (r=0.744,P<0.05) than those by mammography (r=0.544,P<0.05).Conclusion Ultrasonography is more accurate than mammography for size assessment of primary breast cancer.
3.Clinical investigation of surgery for intermittent exotropia.

OBJECTIVETo investigate the time and postoperative binocular vision of strabismus surgery for children with intermittent exotropia (X(T)).
METHODSA retrospective investigation was conducted in 80 child patients with intermittent exotropia. Pre- and postoperative angles of deviation fixating at near (33 cm) and distant targets (6 m) were measured with the prolonged alternate cover testing. The binocular function was assessed with synoptophore. Twenty-one patients took the postoperative synoptophore exercise.
RESULTS(1) A week after surgery, 96.2% of the 80 patients had binocular normotopia, while a year after surgery, 91.3% of the 80 patients had binocular normotopia; (2) Preoperatively, 58 patients had near stereoacuity, while postoperatively, 72 patients achieved near stereoacuity (P<0.05); (3) Preoperatively, 64 patients had Grade I for the synoptophore evaluation and postoperatively, 76 patients achieved Grade I. Meanwhile, 55 patients had Grade II preoperatively and 72 achieved Grade II postoperatively. For Grade III, there were 49 patients preoperatively and 64 patients postoperatively (P<0.05); (4) Patients of 5-8 years old had a significantly better recovery rate of binocular vision than those of 9-18 years old (P<0.05); (5) Patients taking postoperative synoptophore exercise had a better binocular vision than those taking no exercise (P<0.05).
CONCLUSIONS(1) Strabismus surgery can help to preserve or restore the binocular vision for intermittent exotropia; (2) Receiving the surgery at young ages may develop better postoperative binocular vision; (3) The postoperative synoptophore exercise can help to restore the binocular vision.

4.Effect of periodontitis on circulating C-reactive protein in type 2 diabetes patients.

OBJECTIVETo investigate the effect of periodontal infection on circulating C-reactive protein (CRP) in type 2 diabetes patients.
METHODS32 diabetes patients with advanced periodontitis participated in this study. They were compared to a group of 32 diabetes patients without periodontal disease, who were mathed with regard to age (+/- 3 years), gender and body mass index (+/- 1 kg/m2). The concentration of CRP on circulation was measured by ELISA.
RESULTSSignificant difference was found in the level of CRP and the percentage of subjects with elevated CRP levels > or = 3 mg/L on circulation between the two groups(P < 0.05).
CONCLUSIONPeriodontal infection results in higher circulating CRP in type 2 diabetes patients. This elevated inflammatory factor may exacerbate insulin resistance and increase the risk for great vessels complications of diabetes mellitus.

5.X-ray irradiation modulates the expressions of free ubiquitin in the serum, small intestine, and heart of C57 BL/6N mice

Objective To investigate the effect of X-ray irradiation on the expression of free ubiquitin in the serum, small intestine, and heart tissues of C57 BL/6N mice. Methods The amount of free ubiquitin protein in the serum and tissue homogenates was analyzed quantitatively with ELISA and Western Blotting assay. The mRNA expressions of free ubiquitin in the tissues were detected by RT-PCR. Results At 24 or 48 h after radiation, the free ubiquitin level in the serum and small intestine tissue increased asymptotically with increasing of radiation dose (F=183?1, 435?3, P <0?01). After 5 and 10 Gy X-ray irradiation, the concentration of serum free ubiquitin increased asymptotically with the extended response time (F=131?4, 442?9, P<0?01). Compared with the normal control group, at 24 h after 10 Gy X-ray irradiation, the expressions of free ubiquitin protein and mRNA in small intestine tissue were significantly up-regulated (t= -18?7, -10?1, P<0?01). However, there was no difference in the expressions of free ubiquitin protein and mRNA in the heart tissues between irradiated and nonirradiated groups (t = -2?0, 3?1, P >0?05). Conclusions Because of the high expressions of free ubiquitin protein in the radiosensitive mice tissues, X-ray radiation could increase the concentration of free ubiquitin in serum. The changes of free ubiquitin may be related to cellular radiosensitivity and tissue injury.
6.Mutational analysis of patients with 6-pyruvoyltetrahydrobiopterin synthesis deficiency.

OBJECTIVETo determine the gene mutation spectrum of patients with 6-pyruvoyltetrahydrobiopterin synthesis deficiency (PTPSD) in Mainland China.
METHODSThe 6-pyruvoyltetrahydrobiopterin synthesis gene lz(PTS)lz was analyzed in 55 PTPSD patients by using PCR-restriction fragment length polymorphism (PCR-RFLP) and direct DNA sequencing. The relationship between the genotype and phenotype was analyzed.
RESULTSEighteen mutations were identified and the detection rate of gene mutation was 95.28%. Four hot-spot mutations, namely P87S (40.57%), N52S(13.21%), D96N(12.26%) and IVS1nt-291A to G(10.38%) were found in this study, and the first three were associated with severe phenotype. The P87L was reported firstly in Chinese patients, and the Q13X, M80T, IVS4nt-2A to G, L93M and K131N were novel mutations.
CONCLUSIONThe P87S, N52S, D96N and IVS1nt-291A to G mutations are the hot-spots mutations of the PTS gene in Chinese PTPSD patients. Using PCR-RFLP technique to screen the mutations in the PTS gene can increase the efficiency of gene diagnosis.

7.Significance of calcineurin activation and CD40L expression in patients with active lupus nephritis.

AIMTo investigate the significance of the calcineurin (CaN) activation in active lupus nephritis patient.
METHODSPeripheral blood mononuclear cells (PBMCs) were separated from twenty-one active LN patients and 12 healthy controls. Phosphatase activity of CaN was determined using the CaN assay kit by measuring the content of released PO4. Reverse transcription-PCR was used to detect the expression of CD40L mRNA. Flow cytometry analysis was used to detect the expression of CD40L in LN PBMC.
RESULTS(1) Increased activation of CaN in spontaneous cultured PBMC in active LN group was found as compared with control group (46.08 +/- 5.58 vs 8.81 +/- 3.61, P < 0.01). In stimulated by PMA/Ionomycin , activity of CaN in active LN group was also higher than that of control (69.34 +/- 12.59 vs 37.12 +/- 11.57, P < 0.01). (2) Relative content of CD40L in PBMC in active LN groups increased significantly as compared with the control groups under spontaneous and PMA/Ionomycin-induced culture, respectively (P < 0.01). (3) FK506 reduced significantly production of CD40L in spontaneous and PMA/Ionomycin-induced PBMC of LN.
CONCLUSIONElevated activation of CaN in active LN may participate in regulation overexpression of CD40L in PBMC of LN. Through inhibiting CaN activity, FK506 may prevent abnormal activation of CD40-CD40L costimulatory pathway in lupus nephritis.

8.Study on injuries and ways of coping to them among primary and middle school students in one rural area of Anhui province

Objective To describe the current epidemiological characteristics of injuries among primary and middle school students in one rural area of Anhui province and to explore the relationship between the ways of coping and related injuries.Methods Through cluster sampling methods,all students from 3 to 9 grades in 5 primary schools and 3 middle schools in Changfeng county of Anhui province were investigated with questionnaire.All participants completed an anonymous questionnaire concerning their experiences with injuries during the 12 months preceding the survey.The ways of coping to injuries were evaluated by Trait Coping Style Questionnaire.Factors associated with injuries were identified using a negative binomial regression analysis.Results Of 2917 students,the annual event-based rate of injuries was 17.4 per 100 students.Positive coping score had no significant differences between the injury and non-injury groups (33.98 ± 6.38 vs.33.66 ± 6.37) (t=0.979,P=0.328).The score of negative coping style was higher in injury group than in non-injury group (27.65 ± 7.79 vs.26.54 ± 7.62) (t=2.775,P=0.006).Statistically,the annual injury rates were significantly different in three groups on their negative styles of coping (x2=6.131,P=0.013 ).Data from the multivariable negative binomial regression analysis,after adjusted for demographic characteristics,showed no significant difference on the relationship between positive coping style and injury incidence.Those with moderate negative style of coping had lower risks compared to those with highly negative one (IRR=0.77,95% CI:0.63-0.94).Conclusion Negative ways of coping was an important risk factor for injuries.Data from our research suggested that psychological preventive measure need to be taken to improve the style of coping.It was also important to promote the related personality development in planning the strategies for future prevention on injuries.
9.Correlaion between serum Golph2 protein and hepatocellular carcinoma.

OBJECTIVETo study the correlation between hepatocellular carcinoma (HCC) and serum Golph2 protein.
METHODSGolph2 gene was cloned by RT-PCR using RNA from WBF44 cell line as template, the point mutations of the cloned sequence were corrected by PCR, then the gene (1206 bp) was cloned into pET-21 plasmid, and the resulted plasmid was transformed into E.coli DH5a. The expression of 6xHis and Golph2 fusion protein was induced by isopropylthio-beta-D-galactoside (IPTG). The expression of fusion protein was detected by SDS-PAGE and Western blot, and was purified by Ni NTA chelating agarose. The rabbit antibody against Golph2 protein was obtained by immunizing 2 rabbits with the purified Golph2 protein. The specificity and titer of the antibody was determined by Western-blot and ELISA respectively. Sandwich ELISA was used to detect the level of serum Golph2 protein.
RESULTSThere were two replacement mutation and 1 deletion mutation in the cloned sequence contrasted to NM177937 in Genbank, including 644(T-->C, L-->P) , 970 (G-->A, V-->I) and 802 G deletion. The sequence was completely reversed by PCR. The sequence of Golph2 gene cloned into expression vector was confirmed by DNA sequencing. SDS-PAGE and Western blot analysis showed that Golph2 protein was expressed in E.coli DH5a. The antiserum could bind to the 52 kD recombinant protein and serum 73 kD protein specifically. The mean A450 value of ELISA for serum Golph2 protein were significantly higher in HCC and other liver diseases than that in control groups. The sensitivity and specificity for HCC were 44.5% and 82.0%, respectively, at the cut off value is more than or equal to 0.40.
CONCLUSIONThe polyclonal antibody against Golph2 protein is specific. The level of serum Golph2 is significantly higher in patients with HCC and other liver diseases than that in healthy controls.

10.Expression, purification, characteristics and homology modeling of the HMGS from Streptococcus pneumoniae.

OBJECTIVETo understand the molecular basis for a potential reaction mechanism and develop novel antibiotics with homology modeling for 3-hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) synthase (HMGS).
METHODSThe genetic engineering technology and the composer module of SYBYL7.0 program were used, while the HMGS three-dimensional structure was analyzed by homology modeling.
RESULTSThe mvaS gene was cloned from Streptococcus pneumoniae and overexpressed in Escherichia coli from a pET28 vector. The expressed enzyme (about 46 kDa) was purified by affinity chromatography with a specific activity of 3.24 micromol/min/mg. Optimal conditions were pH 9.75 and 10 mmol/L MgCl2 at 37 degrees C. The V(max) and K(m) were 4.69 micromol/min/mg and 213 micromol/L respectively. The 3D model of S. pneumoniae HMGS was established based on structure template of HMGS of Enterococcus faecalis.
CONCLUSIONThe structure of HMGS will facilitate the structure-based design of alternative drugs to cholesterol-lowering therapies or to novel antibiotics to the Gram-positive cocci, whereas the recombinant HMGS will prove useful for drug development against a different enzyme in the mevalonate pathway.
