3.Hyperbaric Oxygen Induced Autophagy in Neuropathic Rats

Objective To investigate whether hyperbaric oxygen(HBO)affected neuropathic pain through autophagy and verify the HBO effect. Methods A total of 60 male SD rats were selected for the study and randomly divided into 4 groups:control group(C group),sham operation group (S group),chronic constriction group(CCI group)and hyperbaric oxygen group(HBO group). LC3Ⅱand P62 were determined by Western blot method and the autophagy was detected by electron microscopy on the 7th day. Results CCI and HBO groups were significantly higher than C and S group in Western blot(P<0.05). There was no difference between group C and group S(P>0.05). The expression of LC3Ⅱin HBO group was significantly higher than that in CCI group(P<0.05). And the expression of P62 in CCI group was significantly higher than that in HBO group(P<0.05). The autophagy in CCI and HBO group were significantly increased comparing to the C and S groups(P<0.05). However,the difference be?tween group C and S was not significant(P>0.05). Conclusion Hyperbaric oxygen therapy can induce autophagy for anti?neuropathic pain.
4.Effect of Hyperbaric Oxygen on p38 MAPK Signal Transduction Pathway in Neuropathic Rats

Objective To investigate the effect of hyperbaric oxygen(HBO)on p38 MAPK signal pathway in neuropathic pain,and explore its mechanism. Methods The experiment was divided into two parts with thirty SD rats in each part. Each part was then randomly divided into 3 groups:sham operation group(S group),chronic constriction group(CCI group)and hyperbaric oxygen group(HBO group),with 10 rats in each group. Part one:Pain?related behavior were detected on 3 day,7 day,14 day,28 day after operation. Phosphorylative p38 was detected by Western blot method and the P2X4 receptor was detected by immunohistochemistry method on the 28th day. Part two:All the rats were treated with SB203580. Pain?related behavior were detected on 3 day,7 day,14 day,28 day after operation. P2X4 receptor was detected by immunohistochem?istry method on the 28th day. Results CCI and HBO groups were significantly lower than S group in pain?related behaviors(P<0.05). CCI group was significantly lower than that in HBO group(P<0.05). The content of phosphorylative p38 and P2X4 receptor in CCI and HBO group were increased(P<0.05),and the expression of phosphorylative p38 and P2X4 in CCI group was significantly increased than that in HBO group (P<0.05). When p38 MAPK was inhibited,pain?related behavior in HBO and CCI group increased significantly than the pain?related behavior of HBO and CCI group in Part 1(P<0.05). But the difference between group CCI and HBO was not significant(P>0.05). Though the expression of P2X4 receptor in CCI group and HBO group was significantly higher than that in S group(P<0.05),the difference between group CCI and HBO was not significant(P>0.05). Conclusion Hyperbaric oxygen therapy can affect the expression of p38 MAPK through P2X4 receptor in rats of neuropathic pain.
5.Determination and clinical significance analysis of IL-6、TNF-? and TGF-?_1 in the patients with diabetic nephropathy

Objective:To investigate the effect of IL-6?TNF-? and TGF-? 1 on the occurrence and development of DM and DN.Methods:To observe the changes of IL-6?TNF-? and TGF-? 1 amount in normal group ?DM group and DN respectively .Results:The amount of IL-6?TNF-? and TGF-? 1 in DM group and DN group are markedly higher that in normal group.The amount of IL-6?TNF-??TGF-? 1 in DN is markedly higher than that in DM group.Conclusion:IL-6?TNF-? and TGF-? 1 play an very important role in the occurrence and development in DM and DN.
6.Relationship between serum acute-phase proteins and diabetic nephropathy in type 2 diabetic patients

Objective To investigate the levels of serum acute phase protein in different stages of diabetic nephropathy and their implication. Methods This study included 30 healthy subjects and 100 patients with type 2 diabetes mellitus, diabetic patients were further divided into three subgroups according to their urinary albumin excretion rate (UAER): 39 patients with normal UAER, 30 patients with microalbuminuria and 31 patients with clinical proteinuria. C-reactive protein, ? 1-acid glycoprotein (? 1-AAG), ceruloplasmin (CER) were measured with rate nephelometry method, sialic acid was measured with MTT method. Results (1) Acute phase protein concentrations were significantly higher in patients of type 2 diabetes mellitus than those in normal controls. (2) Acute phase protein concentrations were increased with increment of UAER. (3) Some acute phase proteins (? 1-AAG, CER and SA) were correlated with serum cholesterol and triglyceride independently. Conclusion There is acute phase reaction in type 2 diabetes mellitus and the degree of acute phase reaction is correlated with the degree of diabetic nephropathy.
8.Preliminary Study of TCM Syndromes of Acute Exacerbation of Chronic Obstructive Pulmonary Disease with Malnutrition

Objective To observe the TCM syndromes of acute exacerbation of chronic obstructive pulmonary disease (COPD) with malnutrition. Methods TCM Syndromes Questionnaire of Acute Exacerbation of COPD was formulated with TCM clinical epidemiology methods. Totally 220 cases of hospitalized patients met the inclusion criteria were divided into two groups, 120 cases of malnutrition and 100 cases of non-malnutrition. The clinical data were observed and syndrome elements of malnutrition group and non-malnutrition group were analyzed to discuss the rule of syndromes in acute exacerbation period. Results In addition to respiratory system, the frenquency of symptoms such as weight loss, shortness of breath, easy cold, aversion to wind and cold, poor appetite, abdominal distension, constipation, belching, dizziness and tinnitus, tooth loose and hair loss, soreness and weakness of waist and knees significantly increased in COPD with malnutrition, mainly involving in syndromes of spleen deficiency and incoordination between spleen and stomach. In patients with acute exacerbation of malnutrition COPD, the single syndromes were 36 cases (30%), the composite syndromes were 84 cases (70%). The simple excess syndromes were 38 cases (31.67%), the simple deficiency syndromes were 23 cases (19.17%), and the intermingled deficiency and excess syndromes were 59 cases (49.17%). Conclusion Patients with acute exacerbation of COPD with malnutrition are mainly composite syndromes and the intermingled deficiency and excess syndromes. Spleen deficiency and incoordination between spleen and stomach may be the important pathogenesis of COPD with malnutrition.
9.Effects of simvastatin on expression of vascular endothelial growth factor in human umbilical vein endothe lial cells

Objective:To investigate the effect of simvastatin on the expression of vascular endothelial growth factor (VEGF)and its receptor in cultured human umbilical vein endothelial cells(HUVEC).Methods:Cells were randomly divided into simvastatin group,tumor necrosis factor-?(TNF-?)group,simvastatin+TNF-?group,interleukin-1?(IL-1?)group,simvastatin+IL-1?group and control group.The effects of TNF-?,IL-1?and simvastatin on the protein levels of VEGF and its receptor in HUVEC were studied by immunocytochemistry.Results:The protein levels of VEGF and its receptor in TNF- ?group and IL-1?group were significantly higher than those of control group,and simvastatin decreased the protein levels of VEGF and its receptor in TNF-?group and IL-1?group.Conclusion:Simvastatin can decrease the protein levels of VEGF and its receptor in HUVEC. The effect of glucose on acylcoenzyme A: cholesterol acyltransferase-1 (ACAT-1) expression in THP-1-derived macrophages was investigated.The results showed that high concentrations of glucose up-regulated ACAT-1 expression in THP-1-derived macrophages.
