3.The influence of the toll-like receptor-4 signaling pathway on IFN-γ and PDGF-AB expressions in the synthesis and secretion of pulmonary artery smooth muscle cells of COPD rats

Objective To study the function of the toll-like receptor-4 (TLR-4) signaling pathway in the synthesis and secretion of pulmonary artery smooth muscle cells of rats with COPD. Methods The primary pulmonary artery smooth muscle cells (PASMCs) of rats with COPD were digested, separated and purified. Then they were randomly divided into control group, LPS group, TLR4 inhibitor group (TAK242) and LPS + TLR4 inhibitor group. RT-PCR, Western blot were used to detect the expression level of TLR-4 and NF-κB among groups. The levels of IFN-γ and PDGF-AB in supernatant with PASMCs in each group were detected by ELISA. Results LPS increased the expression of TLR-4、 NF-κB and the levels of IFN-γ and PDGF-AB. The expression of TLR4, NF-κB and the levels of IFN-γ and PDGF-AB were significantly reduced after inhibiting the expression of TLR4(P < 0.05). Conclusion TLR-4 signaling pathway involved in the inflammatory response and pulmonary vascular remodeling which increased the synthesis and secretion of IFN-γ and PDGF-AB in PASMCs. It provides a theoretical approach for the early intervention of clinical with COPD.
4.Study on Molecular Characterization of Microbial Communities in Tibetan kefir Grain

The diversity in Tibetan kefir grains and dynamics of the microbial community during the fermentation of Tibetan kefir by PCR-DGGE fingerprinting technique were studied.The results showed that bacterial community of Tibetan kefir grains was more complex than that of yeast.The bacteria communities between different Tibetan kefir grains showed 78%~84% similarity, and yeast 80%~92%.Bands B, E and N of DGGE profiles of the microbial community during fermentation of Tibetan kefir were present throughout the fermentation.Analysis of sequence date showed the majority of the DGGE bands of bacteria community corresponded to LAB, and the most intense band (band E) was completely homology to Lactococcus lactis.

7.An initial study on the mechanism of Indigo Raturalis by foam floatation technique

AIM:To study the mechanism of foam floatation in the processing of Indigo Raturalis,and provide theory to support the industrialization of Indigo Raturalis processing by foam fioatation technique. METHODS: To study the critical problems of the processing of Indigo Raturalis by foam floatation technique: the change of particle size distribution after the processing of Indigo Raturalis;The surface activity of frothing materials in Indigo Raturalis;the distinction of the foam to absorb the Indigo particles and other particles(CaCO_3);the absorbability of the Indigo particles to absorb different surface active agents. RESULTS: 1.The Indigo particles were easily absorbed by the foam,but others(CaCO_3) were not;2.There were some frothing materials with good frothing capability in the alkaline system of Indigo Raturalis,3 In the alkaline system,the Indigo particles could be absorbed by the foam of strong acid surface active agents. CONCLUSION: In the alkaline system of Indigo Raturalis,with itself surface active agents,it is effective to process Indigo Raturalis by foam floatation technique.
8.Separation of Indigo Naturalis by foam flotation technique in a continuous mode

Objective To study the process of foam flotation in separation of Indigo Naturalis in a continuous mode and to optimize the operational conditions.Methods Taking content and rate of recovery rate of indigo as index to investigate the single factors,such as the height of collecting region,the height of froth layer,flushing water rate,delivery rate,air flow rate,and aerating velocity,to study the effect of continuous foam floatation on Indigo Naturalis,and to optimize the process conditions finally.Results The flotation performance is good when the height of collecting region is 1.5 m,the height of froth layer is 30 cm,the delivery rate is 0.1 cm/s,the flushing water rate is 0.01 cm/s,the aerating velocity is 1.5 cm/s.The recovery rate of indigo is more than 75% and indgo content is over 5.0%.Conclusion Foam flotation technique is stable and can be used to the separation of Indigo Naturalis in a continuous mode.And this study is the foundation of semi-works production of Indigo Naturalis.

Objectiv e To investigate the correlation between ( CAG) n repeat polymorphism of androgen receptor(AR)geneandmalebreastcancer.Methods 40casesofmalebreastcancerand40controlswerecol-lected.DNA was extracted from peripheral blood and the AR gene CAG coding exon sequences for PCR amplifica -tion,sequencing and calculated the number of CAG repeats frquency .χ2 test and Logistic regression analysis were used assess the AR gene CAG repeat length frequency affect the number of male breast cancer risk .Results There was statistically significant difference in male breast cancer cases and controls the number of CAG repeat length frequency.Man for whom the(CAG)n≥22 repeat sequence had 3.52 times risk of male breast compared (CAG)n≤22(OR=3.52,P=0.036).Conclusion AR gene CAG repeat length is a predictor of the frequency of male breast cancer risk .Longer CAG repeats can increase the risk of male breast cancer .
10.Pharmacological Study on Periplaneta Americana against Alcohol-induced Liver Injury

This study was aimed to explore the effect of Periplaneta Americana against alcohol-induced liver injury in mice, in order to provide a theoretical basis for the industrial production of P. Americana. Kunming mice were randomly divided into six groups, which were the normal group, model group, positive group, high-, middle-, low-dose of P. Americana groups. Intragastric administration of Tiopronin Enteric-coated Tablets 100 mg·kg-1 was given to the positive group. Intragastric administrations of whole powder of medicine were given to the high-, middle-, low-dose groups with the dosage of 6.667, 3.333, 1.667 g·kg-1, respectively. The drugs were given daily for 10 con-secutive days. After 3h of the 10th day drug administration, intragastric administration of distilled water was given to the normal group, while 14mL·kg-1 of 56℃ Red Star Liquor was given to other groups. No food was given but water for 12h. Blood was collected from the orbit. The ALT, AST and GGT in blood serum of mice were measured. The liver was dissected and liver coefficient was calculated. Histopathological examination was given on liver tissues. The results showed that compared with the normal group, the level of ALT, AST in blood serum of the model group had obvious enhanced (P< 0.01), the level of GGT had obvious enhanced (P< 0.05). Compared with the model group, the level of GGT, AST of the high-, middle-dose group had obvious enhanced (P< 0.05), the level of ALT activity had obvious enhanced (P< 0.01). There were severe liver histopathological damages in mice of the high-, middle-, low-dose group. It was concluded that P. Americana had some side effects in the treatment of alcohol-induced liver injury.
