1.Effects of combination treatment with SAHA and TRAIL on ER positive breast cancer cell MCF-7

Aim To investigate the effects of com-bined treatment of SAHA and TRAIL on human breast cancer ER positive cell line MCF-7 . Methods MCF-7 cells were treated with SAHA and/or TRAIL. The inhibitory rates were detected by real-time cell prolifer-ation assays. Morphology changes of MCF-7 cells were observed through time-lapse live cell imaging acquisi-tion. Results Real-time cell proliferation assays showed that the anti-tumor efficacy of SAHA was sig-nificantly enhanced in combination with TRAIL. The results of time-lapse live cell imaging acquisition dem-onstrated that, with treatment of SAHA and TRAIL, the growth inhibition of MCF-7 cells was more obvious than that of in TRAIL or SAHA treatment alone. Con-clusion The combination treatment of SAHA and TRAIL has a synergistic effect of growth inhibition on breast cancer MCF-7 cells.
2.Change of Brain Stem Auditory Evoked Potential in Newborn Infant with Hypoxic-Ischemic Encephalopathy after Hyperbaric Oxygen Therapy

Objective To investigate the therapeutic role of hyperbaric oxygen(HBO) on neonatal hearing impairment induced by hypoxic-ischemic encephalopathy(HIE),by observing the brain stem auditory evoked potential(BAEP) changes of newborn infants with HIE.Methods One hundred and six cases of HIE in newborn infants(in which 82 cases were mild and 24 cases were severe) treated with HBO and conventional therapy were chosen as HBO treatment group,while other 42 cases of HIE in newborn infants(in which 38 cases were mild and 4 cases were severe) treated with conventional therapy only were served as control group.BAEP was measured at the 3rd to 7th day on patients before treatment and 42 days after birth with or without HBO therapy respectively,and the differences of BAEP changes were compared between the both groups.Results 1.In control group,the rate of abnormality was 47.4% and 31.6%,respectively before and after therapy in mild HIE cases(P=0.240),and 75.0%,50.0% in the moderate-severe HIE cases(P=0.462).2.In HBO group,the abnormal rate was 45.1% and 25.6%,respectively in mild HIE cases before and after therapy,while 75.0% and 41.7% in moderate-severe HIE cases(Pa=0.014,0.039).3.There was obvious difference between control group and HBO group in mild HIE cases(P=0.013),while there was no significant change between the 2 groups in moderate-severe HIE cases(P=0.717).Conclusions Treatment with HBO combined with conventional therapy may improve the abnormal rate of BAEP in patients with HIE according to the present test,especially in mild HIE cases,thus earlier HBO therapy may be used to mitigate hearing impairment and improve prognosis induced by the disease.
3.Analysis of 53 Patients with Sjogren' s Syndrome and Idiopathic Pulmonary Fibrosis

Objective To explore the occurrence, clinical feature and the associatrd factors of Sjogren' s syndrome(SS) complicated with Idiopathic Pulmonary Fibrosis(IPF) to build up knowledge for early diagnosis. Methods The clinical characteristics of 160 cases damitted SS patients collected from January 2002 to August 2008 were retrospectively investigated including RF, chest - X, ESR, blood -gas analysis,pulmonary function tests,chest HRCT,ANA and so on. Results The patients with IPF accounted for 33.12% of total SS patients, including 12 cases of male. These patients presented strongly positive ANA,strongly positive RF etc,especially in the male. Conclusion The occurrence of SS - IP was related to the disease' s activity. FHRCT and pulmonary function test should taken early in order to get early diagnosis and treatment.
4.Removal of glucocordicoids influences the occurrence of microalbuminuria in recipients receiving renal transplantation

BACKGROUND:Long-term use of corticosteroids (hereinafter referred to as hormone) after renal transplantation could obviously lead to adverse reactions. Immunosuppressive regimen with less and no hormone has been a hot focus in the study of renal transplantation al over the world. However, reduction or withdrawal of hormones has a certain risk. At present, there is no unified scheme. Because urine protein can be immediately detected after tubular injury, to monitor urine protein can find the renal dysfunction after transplantation in recipients undergoing renal transplantation, which can gain time for clinical therapy. OBJECTIVE:To discuss the influence of hormone (prednisone) removal on the occurrence of urine protein in recipients undergoing renal transplantation. METHODS:A total of 35 recipients undergoing renal transplantation after removal of prednisone received immunosuppressive regimen of cyclosporine A or tacrolimus+mycophenolate mofetil bivalent. Initial dose of prednisone was 30 mg/d, and then gradual y reduced by 5 mg per week, and withdrawn at 1 month after renal transplantation. There were 16 cases in cyclosporine A group and 19 cases in tacrolimus group. Urine protein was measured and quantified at 3, 6, 12 and 24 months after renal transplantation and 3, 6 and 12 months after addition of prednisone in both groups. Simultaneously, serum creatinine, fasting glucose, body mass increases, the rate of acute rejection, infection, patient/graft survival at 2 years after renal transplantation and urine protein at 24 hours before and after adding hormone were recorded. RESULTS AND CONCLUSION:For the two groups, urineα1-microglobulin started to rise after 6 months of removal of prednisone. Urinary microalbumin, urinaryα1-microglobulin, and urinary transferrin ascended obviously at 12 months. Urinary protein was positive in five cases of cyclosporine A group and in three cases of tacrolimus group. At 24 months, urinary microalbumin, urinaryα1-microglobulin, urinary transferrin and urinary IgG ascended obviously. Urinary protein was positive in cyclosporine A group with 11 cases and in tacrolimus group with 10 cases. 24-hour urinary protein quantity was more than 1 g in every case. On this base, we made the patients to take more prednisone for 6 months, so urineα1-microglobulin and urinary microalbumin began to descend. Each group had one case of positive urinary protein turning to negative. Twelve months after the adjustment of the prednisone, urinary microalbumin, urinaryα1-microglobulin, and urinary transferrin descended respectively. Positive urinary protein turned into negative:in cyclosporine A group with two cases and in tacrolimus group with three cases. 24-hour urinary protein quantity was around 0.7 g. Two years after renal transplantation, serum creatinine and acute rejection rates were higher in the cyclosporine A group than in the tacrolimus group (P<0.05). No significant difference in fasting glucose, body mass increase, infections, and patient/graft survival was detectable between both groups. Results suggested that removal of prednisone greatly affected urine protein in recipients undergoing renal transplantation. In particular, at 2 years after renal transplantation, urinary microalbumin, urinaryα1-microglobulin, urinary transferrin and urinary IgG ascended obviously, and the security needs further research.
5.Cloning and Sequence Analysis of Genome from the Inner Mongolia Strain of the Endogenous Betaretroviruses (enJSRV)

In order to amplify the complete genome of enJSRV from the strain of Inner Mongolia (enJSRV-NM), we used enJSRV-specific and JSRV-specific DNA probes in dot blot hybridization. Seven pairs of primers were designed based on Genbank sequences. Seven fragments were obtained by PCR and were cloned into the PMD19- T vectors. The recombinant plasmids were sequenced and analyzed. The results showed that the genome was 7 942 bp in length and contained four overlapping open reading frames corresponding to the gag, pro, pol and env genes as well as an additional open reading frame (orf-x) that overlaps the 3' end of the pol gene. The nucleotide acid sequences of the enJSRV-NM loci were compared with the sequences of South Africa enJS56A1 strain (Accession No. AF153615) and USA JSRV21 strain (Accession No. AF105220). The nucleotide acid identities were 99.2% and 92.3% respectively. Two zinc fingers were found in the NC region in the predicted amino acid sequence. However, the YXXM motif, which is a reliable molecular marker for the infectious exogenous virus, was not found in the TM region. It was found that the enJSRV-NM region was 90%-98% identical at the amino acid level to its exogenous infectious counterparts in most of the retroviral genome. This is the first nucleotide sequence of enJSRV reported in P.R China. The resource work has provided a wide range of information useful not only for expression genomics and annotation of genomic DNA sequence, but also for further research on the clinical diagnosis of OPA.
6.Clinical control study in methods of pre-labor pubic preparation

Objective To determine whether it is necessary to execute pre-labor pubic shaving.Methods Women(336 cases)who received pubic shaving through vaginal delivery from 2003 to 2006 were set as the control group.Women(103 cases)who were waiting for delivery currently in our hospital were set as the test group.The test group only received perineal cleaning without pubic shaving.If episiotomy was needed during the operation,the perineal hair Was cut by scissors.The perineum infectious rate and postpartal infection Was compared in the two groups.We interviewed the patients attitude for pubic shaving and counted the numing time used for pubic shaving.Results The perineal infection in the test group was 5.8%,which was no statistically different from that of the control group(2.4%),U=0.33,P>0.05.Totally 95% women in the test group support this method demonstrated humanization.In the telephone folow-up visit for patients in the control group they comphined itch and discomfort during the regeneration of pubes.The average numing time for pubic shaving Was(4.6±0.6)minutes.Conclusions No pubic shaving for pre-labor skin preparation was feasible.If operation was disturbed by pubes we could use scissors to cut it short.This method could reduce the skin infection and propagation of blood-derived infectious disease.It increased the comfort of parturient,saved time and proved to be a nursing method of humanization.
7.Effect of angiotensin Ⅱ on the expression of MIF in macrophages

Objective To investigate whether angiotensin Ⅱ (AngⅡ) can regulate the expression of MIF in macrophages via the NF-κB pathway. Methods Western Blot, real time RT-PCR and ELISA were used in the present study. Results Western blot result showed that the expression of NF-κBp65 gradually increased with the increase of the concentration of AngⅡ. Results of real time RT-PCR and ELISA revealed that MIF mRNA expression and the content of MIF were significantly higher in AngⅡ group than those in the control group. PDTC could reverse the effect of AngⅡ on MIF mRNA expression and MIF secretion in macrophages. Conclusion AngⅡ can promote MIF mRNA expression and MIF secretion in macrophages via the NF-κB signaling pathway.

Objective To study the association between clinical symptoms,psychosomatic factors and autonomic nervous function in patients with gastroesophageal reflux disease (GERD).Methods Thirty-four patients with GERD diagnosed by reflux disease questionnaire (RDQ) and endoscopy and 15 healthy control subjects were enrolled in this study.All the subjects were divided into two groups,one with normal scores of Zung's self-rating anxiety scale (SAS) and Zung's depression scale (SDS) as GERD (-) and the other with abnormal scores of SAS and SDS as GERD (+).Reflux symptom score was recorded for both groups at the same time.Autonomic nervous function was assessed by their heart rate variability (HRV) on dynamic electrocardiogram (DCG).The time domain parameters analyzed included standard deviation (SD) of average R-R interval during 24 hours (SDNN),SD of average 5-minute sinus heart rate (SDANN),mean square root of the difference of adjoining two R-R interval (rMSSD),and proportion of the heart beats with difference of R-R interval more than 50 ms from the total heart beats (PNN 50),and the frequency domain parameters analyzed included low-frequency (LF),high-frequency (HF) and ratio of LF to HF.Results Average scores of SAS and SDS were significantly higher in patients with GERD than those in healthy controls (48?9 vs 38?6 and 48?11 vs 41?6,respectively,P
10.Therapeutic effects of multi-dose activated charcoal on the acute dichlorvos poisoning in rats

Objective To assess the therapeutic effects of activated charcoal on the acute dichlorvos poisoning in rats. Method Thirty male clean grade Wistar rats were randomly (random number) divided into three groups: control group (group A, n = 10), single dose activated charcoal group (group B, n = 10) and multi-dose activated charcoal (group C, n=10). The rats of group A were suffered from 35 mg/kg dichlorvos exposure by oral without activated charcoal and senna. The rats of group B received 35 mg/kg dichlorvos exposure by oral with 175 mg/kg activated charcoal given immediately after dichlorvos exposure and 35 mg/kg senna given half an hour later. In the group C, 35 mg/kg dichlorvos was given to rats by oral with 175 mg/kg activated charcoal given immediately after dichlorvos exposure and 35 mg/kg senna given half an hour later and then every four hours. Blood samples were collected from the carotid artery at different intervals after exposure. DDVP concentration and total blood acetyl-cholinesterase activity were detected. Differences in serum DDVP concentration, Cmax, AUC (0→∞ ), MRT and acetylcholinesterase among three groups were calculated by using ANOVA. Results Serum DDVP levels in single dose group and in multi-dose group were significantly different from those in control group (P < 0.05). The DDVP levels in multi-dose group were significantly different from those in single dose group 4 hours after exposure (P < 0.05). The AUC and Cmax in activated charcoal treatment groups were significantly different from those in control group (P < 0.05). There were no significant differences in MRT among three groups. Fours hours after exposure to dichlorvos,the levels of serum acetylcholinesterase in rats of group B and group C were significantly different from that in rats of group A (P < 0.05), and there was no significant difference in acetylcholinesteras between group B and group C (P > 0.05). Another four hours later, no differences in acetylcholinesterase were found a-mong three groups (P > 0.05). Conclusions The peak concentrations of dichlorvos in blood are lower in group B and group C, and the blood acetylcholinesterase inhibition is quelled by activated charcoal. Therefore, the effects of multi - dose of activated charcoal is better than that of single dose of activated charcoal.
