1.Chondrogenic differentiation of adipose-derived stem cells induced by growth differentiation factor-5 cultured on the type I collagen scaffold

BACKGROUND:Growth differentiation factor-5 can induce adipose-derived stem cels into chondrocytes in our previous studies, but it has not been reported that the adipose-derived stem cels induced by growth differentiation factor-5 can differentiate into chondrocytes on the type I colagen scaffold. OBJECTIVE:To investigate the chondrogenic differentiation ability of adipose-derived stem cels induced by growth differentiation factor-5 cultured on the type I colagen scaffold. METHODS:Adipose derived stem cels were isolated from rabbit adipose tissue, the cels morphology was observed using inverted phase contrast microscope and the phenotypes were identified using immunofluorescence. The exogenous growth differentiation factor-5 was added to the cultural media with the type I colagen scaffold so as to induce the chondrogenic differentiation. The cels morphology was observed using hematoxylin-eosin staining and scanning electron microscope after the induction by growth differentiation factor-5 for 14 days. Meanwhile, the type II colagen and aggrecan mRNA expressions of the induced cels were measured using RT-PCR after the induction by growth differentiation factor-5 for 7, 14, and 21 days.RESULTS AND CONCLUSION:The primary cultured adipose-derived stem cels proliferated adherently with the fusiform and polygonal distribution under inverted phase contrast microscope. The positive CD44, CD49d and negative CD106 were detected by immunofluorescence. The adipose-derived stem cels induced by growth differentiation factor-5 were wel adhered to the type I colagen scaffold and strongly proliferated. The large amounts of extracelular matrix existed on the surface of the induced cels under scanning electron microscope. RT-PCR agarose gel electrophoresis indicated that the type II colagen and aggrecan mRNA expressions of the adipose-derived stem cels induced by growth differentiation factor-5 with the type I colagen scaffold were significantly increased. Growth differentiation factor-5 can successfuly induce the chondrogenic differentiation of adipose-derived stem cels cultured on the type I colagen scaffold.

Objective To study the association between clinical symptoms,psychosomatic factors and autonomic nervous function in patients with gastroesophageal reflux disease (GERD).Methods Thirty-four patients with GERD diagnosed by reflux disease questionnaire (RDQ) and endoscopy and 15 healthy control subjects were enrolled in this study.All the subjects were divided into two groups,one with normal scores of Zung's self-rating anxiety scale (SAS) and Zung's depression scale (SDS) as GERD (-) and the other with abnormal scores of SAS and SDS as GERD (+).Reflux symptom score was recorded for both groups at the same time.Autonomic nervous function was assessed by their heart rate variability (HRV) on dynamic electrocardiogram (DCG).The time domain parameters analyzed included standard deviation (SD) of average R-R interval during 24 hours (SDNN),SD of average 5-minute sinus heart rate (SDANN),mean square root of the difference of adjoining two R-R interval (rMSSD),and proportion of the heart beats with difference of R-R interval more than 50 ms from the total heart beats (PNN 50),and the frequency domain parameters analyzed included low-frequency (LF),high-frequency (HF) and ratio of LF to HF.Results Average scores of SAS and SDS were significantly higher in patients with GERD than those in healthy controls (48?9 vs 38?6 and 48?11 vs 41?6,respectively,P

7.Preliminary clinical study of 99Tcm-HL91 imaging in bone metastasis

Objective 99Tcm-4,9-diaza-3,3,10,10-tetramethyldodecan-2,11-dione dioxime(HL91),a new type of hypoxic agents,accumulates in tumor hypoxic tissue specifically.The aim of this study was to evaluate the value of 99Tcm-HL91 imaging in the diagnosis of bone metastasis.Methods Nineteen cases with bone metastasis(without any treatment)and 8 cases with benign lesions underwent SPECT imaging at 4 h after injection of 740 MBq of 99Tcm-HL91 along with 99Tcm-methylene diphosphonic acid(MDP)imaging.Regions of interest(ROIs)were drawn in tumor tissue and contralateral normal tissue respectively,and the radioactivity ratios of tumor-to-normal(T/N)were calculated.The t-test was used for data analysis with SPSS 11.0.Results There were visible uptake of 99Tcm-HL91 in 79 out of 85 focuses in 19 patients of bone metastasis;however,there was no obvious uptake of 99Tcm-HL91 in 12 focuses of 8 patients of benign lesions.Significant difference existed between the T/N values of malignant(1.877±0.288)and benign lesions[(0.735±0.236);t=13.065,P<0.05].However,the T/N value of bone metastasis from lung cancer did not differ with that from prostate cancer(1.915±0.344 and 1.825±0.175,respectively;t=1.378,P>0.05).Conclusion The results indicated that 99Tcm-HL91 was useful in diagnosing the malignant and benign bone lesions.
8.Prevalence rates of healthcare-associated infection in a psychiatric hospital in four consecutive years

Objective To investigate the prevalence rates of healthcare-associated infection (HAI) and antimicrobial use in a psychiatric hospital.Methods The annual cross-sectional survey on HAI among all hospitalized psychiatric patients from 0:00 to 24:00 of June 20, 2012-2015 were conducted, HAI during 4 years were analyzed.Results A total of 2 584 hospitalized patients were surveyed, 47 patients had 48 episodes of HAI, the prevalence rate and case prevalence rate of HAI were 1.82% and 1.86% respectively;prevalence rates in 2012-2015 were 2.09%, 1.56%, 1.48%, and 2.11% respectively(x2=1.23, P=0.75).The top three departments of high prevalence rates were male section of medical rescue department(3.80%), male section of acute intervention department(3.31%), and department of geriatric psychiatry(2.78%).The top three infection sites were upper respiratory tract (31.91%), lower respiratory tract (29.79%), and gastrointestinal tract(14.89%);28 strains of pathogenic bacteria were detected, the top three pathogens were Escherichia coli (28.58%), Klebsiella pneumoniae (17.86%), and Staphylococcus aureus (14.29%);antimicrobial use rates in 2012-2015 were 10.91%, 5.78%, 5.41%, and 5.06% respectively, which decreased year by year(x2=23.13,P<0.01).Conclusion Prevalence rates of HAI in four consecutive years maintained a low level in this hospital, antimicrobial usage rates decreased year by year.
9.Rehabilitating the non-motor symptoms of Parkinson's disease using repetitive transcranial magnetic stimulation

Objective To research the clinical rehabilitative effect of repeated transcranial magnetic stimulation (rTMS) for the non-motor symptoms of Parkinson's disease.Methods Fifty-five PD patients were randomly divided into an rTMS treatment group (n =29) and a sham stimulation group (n =26).The treatment group received a course of 0.5 Hz rTMS treatment,while the sham stimulation group had the same treatment but with no energy output.Both groups were evaluated using a non-motor symptoms questionnaire (NMSQuest),the Hamilton depression scale (HAMD),the Pediatric Daytime Sleepiness Scale (PDSS),the mini-mental state examination (MMSE) and the scale for outcomes in PD for autonomic symptoms (SCOPA-AUT) before treatment,immediately after,and a month after treatment.Results Compared with before treatment,the average NMSQuest score of the treatment group declined significantly,though 1 month after treatment the improvement was no longer significant.After treatment and 1 month later the average NMSQuest score of the sham stimulation group increased gradually.The effect in the treatment group was therefore significantly better than in the sham stimulation group.After treatment and 1 month later the average HAMD score of the treatment group was significantly lower than before treatment while the average HAMD score of the sham stimulation group had increased gradually.The improvement in depression in the treatment group was therefore significantly better than in the sham stimulation group.After treatment,sleep disorders in the treatment group had been significantly alleviated,but one month later the improvement was no longer significant compared with before treatment.PDSS scores in the sham stimulation group declined gradually,but the improvement in sleep disorders among the treatment group was significantly better than among the sham stimulation group.Cognition improved significantly among the treatment group right after treatment,but 1 month later it had worsened while the MMSE scores of the sham stimulation group decreased gradually.The difference between the two groups was statistically significant.The average SCOPA-AUT scores of the two groups were not significantly different from each other or from the scores before treatment.Conclusion Repeated TMS can improve most non-motor symptoms of PD.The improvement in depression is the most significant.A short course of rTMS has no obvious rehabilitative effect on the autonomic function disorders of PD patients.
