Journal of China Medical University 2016;45(8):723-727
Objective To investigate the effect of hyperbaric oxygen(HBO)on p38 MAPK signal pathway in neuropathic pain, and explore its mechanism. Methods The experiment was divided into two parts with thirty SD rats in each part. Each part was then randomly divided into 3 groups:sham operation group(S group),chronic constriction group(CCI group)and hyperbaric oxygen group(HBO group),with 10 rats in each group. Part one:Pain?related behavior were detected on 3 day,7 day,14 day,28 day after operation. Phosphorylative p38 was detected by Western blot method and the P2X4 receptor was detected by immunohistochemistry method on the 28th day. Part two:All the rats were treated with SB203580. Pain?related behavior were detected on 3 day,7 day,14 day,28 day after operation. P2X4 receptor was detected by immunohistochem?istry method on the 28th day. Results CCI and HBO groups were significantly lower than S group in pain?related behaviors(P<0.05). CCI group was significantly lower than that in HBO group(P<0.05). The content of phosphorylative p38 and P2X4 receptor in CCI and HBO group were increased(P<0.05),and the expression of phosphorylative p38 and P2X4 in CCI group was significantly increased than that in HBO group (P<0.05). When p38 MAPK was inhibited, pain?related behavior in HBO and CCI group increased significantly than the pain?related behavior of HBO and CCI group in Part 1(P<0.05). But the difference between group CCI and HBO was not significant(P>0.05). Though the expression of P2X4 receptor in CCI group and HBO group was significantly higher than that in S group(P<0.05),the difference between group CCI and HBO was not significant(P>0.05). Conclusion Hyperbaric oxygen therapy can affect the expression of p38 MAPK through P2X4 receptor in rats of neuropathic pain.

The expression of LC3Ⅱin HBO group was significantly higher than that in CCI group(P<0.05). And the expression of P62 in CCI group was significantly higher than that in HBO group(P<0.05). The autophagy in CCI and HBO group were significantly increased comparing to the C and S groups(P<0.05). However, the difference be?tween group C and S was not significant(P>0.05). Objective To explore the effect of salubrinal (sal,an endoplasmic reticulum stress inhibitor) on radiosensitivity of human head and neck squamous carcinoma cells (HNSCC).Methods Cells were divided into two groups of sal treatment and its control. For drug treatment group, cells were treated with 10 mmol/L sal for different time (12,24,36 h) and then irradiated. The levels of a core protein GRP78 of endoplasmic reticulum stress (ERS) in HNSCC (KB,Fadu, and Detroit 562 cells)were analyzed by Western blot assay at different time (0,20 min,1 h,3 h,6 h,12 h,24 h and 48 h) after irradiation.Cell survival was measured with colony formation assay. Results Western blot assay revealed that the protein levels of GRP78 in three kinds of HNSCC significantly increased from 20 min to 1 h and peaked at 3 h after radiation (t =12.72,13.37,18.31,P < 0.05).Compared with the control group, treatment of cells with sal decreased GRP78 protein levels (t =14.25,5.34,3.12,P < 0.05) in three cell lines and also significantly enhanced radiation damage and reduced cell viability. The sensitization enhancement ratios (SER) of sal in three cell lines were 1.16,1.05 and 1.06,respectively.Conclusions Rradiosensitivity of HNSCC could be effectively enhanced by sal treatment.

Objective To examine the induction effects of bone marrow mesenchymal stem cells(BMSCs) transfected with bone morphogenetic protein 7 (BMP7) gene differentiating into chondrocytes. Methods We observed the phenotype of cells which were stained with alcian blue and HE climbing to the six pore plate with invert microscope. The glycosaminoglycan (GAG) value in culture medium was detected in control group,BMP7 transfect and culture medium induced groups after 7,14 and 21 days using standard curve method. Standard curve was described using galacturonic-acid as reference substance. The content of collagen Ⅱ was detected by ELISA method. Results HE and Alcian blue staining showed that BMP7 gene transfection group and the group induced by fluid possess the characteristics of chondrocyte. BMP7 induced BMSCs differentiation to chondrocyte which secrete specific protein called collagen Ⅱ and GAG. Content of GAG were (17.1±3.4),(39.5±5.4),(40.8±6.1)mg/L in control group,BMP7 gene transfected group and induced group, collagen Ⅱ were (89.7±14.3),(152.8±14.5),(155.5± 19.3)μg/L in these three groups separately. Comparing with control group,GAG and collagen Ⅱ of BMP7 gene transfected group and culture medium induced group increased obviously(all P < 0.05),but there was no significant difference between BMP7 gene transfeeted group and culture medium induced group (P > 0.05). Conclusion This active protein induces BMSCs differentiating into chondrocyte,in a level similar to that of inducing medium.

Objective To investigate the hMOF protein expression in non?small cell lung cancer and explore the relationship between its expres?sion and radiotherapy prognosis. Methods Immunohistochemical staining was used to detect the hMOF expression in 59 cases of non?small cell lung cancer after radiotherapy. The relationship between hMOF expression with clinicopathological and radiation prognosis was analyzed. Results Among the 59 cases of non?small cell lung cancer tissues, there were 30 cases found to be high expression with hMOF. The rate of positive expression of hMOF in non?small cell lung cancer were 50.85%. Clinical stage and hMOF expression were independent predictors for non?small cell lung can?cer. Conclusion The expression of hMOF had a positive correlation with the radiation prognosis in non?small cell lung cancer, which could be used as a prognostic indicator of radiotherapy.

Objective To observe the effects of intermittent hypoxic preconditioning on the expression of apoptosis-related Bcl-2 and Bax protein after 70% hepatectomy combined with ischemia-reperfusion injury. Methods A total of fifty-four SD rats were randomly divided into three groups (n =18).Partial hepatectomy hroup (PH Group):Rats underwent the left and middle lobectomy of liver(70% hepatectomy).Ischemia reperfusion group (IR group):The left and middle lobes of liver were resected during the occlusion of the hepatoduodenal ligament for 20 minutes.Residual liver underwent the process of ischemia-reperfusion.Intermittent hypoxia preconditioning group (IHP group):rats were exposed to hypoxic environment of 10% oxygen for 1 h/d.After 7 consecutive days, the left and middle lobes of liver were resected under the portal triad clamping. At 12,24 and 48 hours after the operation, the rats were killed and detected. The serum levels of ALT and AST were determined by automatic biochemical analyzer. The expression of Bcl-2 and Bax protein in liver tissue were measured by immunohistochemistry. Results At each time point after surgery, the serum levels of ALT and AST in IR group and IHP group were higher than that of PH group, and IHP group were lower than in IR group.Compared with IR group, the expression of Bcl2 protein significantly increased and Bax protein expression significantly decreased in IHP group. All these differences were statistically significant (P < 0.05).Conclusions Intermittent hypoxic preconditioning might protect residual liver against ischemia reperfusion injury, through increasing the expression of Bcl-2 protein and reducing the expression of Bcl-2 protein to decrease liver cell apoptosis.
