Objective To observe the clinical effect of budesonide combined with salbutamol in the treatment of children with bronchial asthma,and its influence on oxidative stress and serum interleukin-2 (IL-2),soluble IL-2 receptor (sIL-2r) levels.Methods 70 children with bronchial asthma were divided into observation group and control group according to the different treatment,35 cases in each group.The observation group was inhaled budesonide combined with salbutamol,the control group was given salbutamol inhalation therapy.The clinical efficacy of the two groups was observed,and the changes of serum oxidative stress factor,IL-2 and sIL-2r levels before and after treatment in the two groups were compared.Results In the observation group,20 cases were cured,7 cases were markedly effective,effective in 5 cases,the total effective rate was 94.3％.In the control group,17 cases were cured,6 cases were markedly effective,effective in 4 cases,the total effective rate was 77.1％.The total effective rate between the two groups had statistically significant difference (x2 =26.543,P ＜ 0.05).After treatment,the malondialdehyde (MDA),end products (AOPP) of the observation group were (16.26 ± 1.76)U/L,(42.123 ± 5.745) μmol/L,respectively,which were lower than those of the control group [(22.16 ± 2.84) U/L,(63.63 ± 6.22) μ mol/L,t =23.354,25.348,all P ＜ 0.05].The glutathione peroxidase (GSH-PX),superoxide dismutase (SOD) of the observation group were (236.46 ± 32.25) ng/L,(19.643 ± 3.587) ng/L,respectively,which were higher than those of the control group [(197.43 ± 18.42) ng/L,(18.244 ± 2.446) ng/L,t =18.943,15.123,all P ＜O.05].The IL-2,sIL-2r levels of the observation group were (5.28 ± 2.65) μg/L,(30.376 ± 2.147) mg/L,respectively,which were significantly lower than those of the control group [(9.84 ± 3.22) μg/L,(37.564 ± 1.323)mg/L],the differences were statistically significant (t =21.378,24.643,all P ＜ O.05).Conclusion Inhaled budesonide combined with salbutamol in the treatment of children with bronchial asthma is helpful to control the symptoms of asthma,and can inhibit oxidative stress,reduce serum sIL-2r and IL-2 levels,it is worthy of promotion.

Objective To investigate the levels of serum acute phase protein in different stages of diabetic nephropathy and their implication. Methods This study included 30 healthy subjects and 100 patients with type 2 diabetes mellitus, diabetic patients were further divided into three subgroups according to their urinary albumin excretion rate (UAER): 39 patients with normal UAER, 30 patients with microalbuminuria and 31 patients with clinical proteinuria. C-reactive protein, ? 1-acid glycoprotein (? 1-AAG), ceruloplasmin (CER) were measured with rate nephelometry method, sialic acid was measured with MTT method. Results (1) Acute phase protein concentrations were significantly higher in patients of type 2 diabetes mellitus than those in normal controls. (2) Acute phase protein concentrations were increased with increment of UAER. (3) Some acute phase proteins (? 1-AAG, CER and SA) were correlated with serum cholesterol and triglyceride independently. Conclusion There is acute phase reaction in type 2 diabetes mellitus and the degree of acute phase reaction is correlated with the degree of diabetic nephropathy.

Objective To study the biocompatibility of silk fibroin/poly L-lactic acid (SF/PLLA) non-woven network,a kind of new composite tissue engineering nanomaterials,and to explore its possibility as the biological implant materials.Methods The PLLA non-woven network was prepared by electrostatic spinning.Physiological saline as control,the leaching solution was prepared and injected into the mice,then the mice were observed for 2 weeks.The materials were implanted into the back of the mice,and 3-0 suture was used as control.Tissues were collected at 1,2,3,and 4 weeks after operation,dyed by HE staining and then the photos were taken.The tissue reactions in experimental group and control group were observed.The rabbit knee joint cartilage cells were cultured,and then subculture cells were seeded to the surface of materials.After cultured invitro,the adhesion and growth of the cells were observed with inverted optical microscope.The bioactivities of the rabbit knee joint cartilage cells in negative control group(DMEM culture media),experimental group(DMEM containing materials) and positive control group(DMEM containing phenol solution)were determined by MTT assay after cocultured for 24 and 48 h.Results After injection,the body status of the mice in experimental group was the same to the control group.There were little fibroblasts was and a little of lymphocytes and macrophage cells in the materials which were implanted into the back of the mice at the beginning.Then the number of the fibroblasts was increased, but the number of the lymphocytes and macrophage cells did not change obviously.The materials degraded slowly, and the material degraded obviously at 4 weeks.The inflammation of tissue around the material reduced gradually from the 2nd week.The inflammation of tissue around the material was the same to the suture,and sometimes was slighter than the suture.After sed for 24 h,there were cells attaching to the fibers of the material.More and more cells attached to the fibers.The reasult of MTT assay showed that the cytotoxicities in experimental groups were all on LevelⅠ at 24 and 48 h.Except for positive control group,the A values were increased in other groups with the extended response time.At the same time,there was no significant difference in cytotoxicity between experimental group and negative control group(P>0.05)and the A value in experimental group was higher than that in positive control group(P<0.01).Conclusion The SF/PLLA non-woven network scaffold material has good biological compatibility and safety,it could be used as implant material in tissue engineering.

Objective To establish a molecular technique of internal transcribed spacer (ITS) sequencing to identify pathogenic fungi species from the fungal sinusitis tissues. Methods Total 270 sinusitis tissues samples were collected by endoscopic surgery from 2006 to 2008. The histopathology, organize spring clip culturation and ITS region (ITS region region of fungal rRNA, including ITS1-5. 8S rRNA-ITS2) sequencing were employed simultaneously. And then to evaluate the ITS sequencing as the tool for identification of pathogenic fungi directly from clinical samples. Results Of the 270 samples, histopathology positive rate was 80.0% (216/270) , organize spring clip positive rate was 80.0% (216/ 270), fungal culturation positive rate was 53.0% (143/270) , ITS region sequencing positive rate was 63. 0% [ (134 +28 +8)/270], There were 22 species and 6 genera identified by fungal culturation, and 32 species identified by ITS region sequencing. Conclusion ITS region sequencing will become a applicable tool in clinical laboratory in future.

Objective To evaluate clinical factors as predictors of radiation pneumonitis(RP)in advanced stage non-small cell lung cancer(NSCLC)patients treated with concurrent radiochemotherapy when gross tumor volume is 70 Gy. Methods Data of 84 patients with histologically proved NSCLC treated with 3DCRT or IMRT were collected. To evaluate the correlation between clinical parameters and radiation pneumonitis(RP). The clinical parameters were considered: pathological type, therapy agents, age,gender, stage, karnofsky performance status(KPS), smoking status, diabetes, chronic obstructive pulmonary disease(COPD). Results The occurrence of grade 1,2 RP was 63%, 33%, respectively. In univariate analysis, diabetes was significantly associated with RP of ≥ grade 1(x2 =4.03, P = 0.045)and ≥grade 2(x2 = 15.59 ,P =0.000). KPS was significantly associated with RP of ≥grade 1(x2 =3.98 ,P = 0.046)and ≥grade 2(x2 = 5.21, P = 0.023). In logistic multivariate analysis, diabetes was significantly associated with RP of ≥grade 1(x2 =5.50,P =0.019)and ≥grade 2(x2 = 12.92,P =0.000). KPS was significantly associated with RP of ≥ grade 1(x2 = 6.29, P = 0.012)and ≥ grade 2(x2 = 6.61, P =0.010). Conclusion The definite statistical significant risk factors of RP are diabetes and KPS.

Objective To study the expression of gonadotropin releasing hormone receptor (GnRH receptor) in the stomach of Sprague-Dawley rat exposed to light stress. Methods We established illumination stressed models that the rats were exposed to continuous light(totally 64 rats, 32 rats for experiment and control group, respectively). Then,stomachs were taken from the rats when the rats exposed to continuous light for 1 day, 2 days, 3 days, 4 days, 1 week, 2 weeks, 3 weeks, 4 weeks and their control groups, respectively.The localization and the expression of GnRHR and GnRHR mRNA in stomachs mucous membrane were detected using immunohistochemistry, Western blotting and real-time PCR. Results Immunoreactivity was displayed mostly in the parietal cells in gastric gland. There were not differences of this distribution in all groups of constant illumination and their control. The immunoreactive materials were distributed on membrane and in cytoplasm of all positive cells, but not in nuclei. The level of GnRHR in rats exposed to continuous illumination were higher from one week to four weeks compared with that in control(P<0.05), and the level of GnRHR reached the peak when rats were exposed to constant illumination for two weeks (P<0.01). The quantity of GnRH mRNA in rats exposed to continuous illumination were higher from three weeks to four weeks compared with that in control(P<0.05). Conclusion The expression of GnRHR in digestive tract was effected by illumination stress.GnRH might regulate digestive function by interaction with GnRHR, suggesting that GnRH may be acted as a hormone, not only responded to normal physiological function of digestive tract but also responded to stress activity.

Objective To investigate apoptosis of HepG2 ceils after transfecfion with LIGHT gene and interferon-γ. Methods LIGHT gene and interferon-γ were transfected into HepG2 cells by liposome mediated method. The HepG2 cells were divided into group A (transfected with LIGHT gene or interferon-γ), group B (transfeeted with LIGHT gene and interferon-γ) and group C (non-transfection group). The apoptosis rate of the HepG2 cells and the expression of Bcl-2 and Caspase-8 were detected 12, 24, 48 hours after transfeetion. Results (1) The apoptosis rates of HepG2 cells at hour 12, 24 and 48 after transfeetion were 18.8% ± 3.5%, 25.7%± 2.8% and 36.4% ±3.6% in group A, 23.8% ±2.4%, 31.1% ±2.1% and42.5% ±4.5% in group B, and 8.7% ± 2.1%, 9.3% ± 1.6% and 10.9% ± 1.2% in group C. There was a significant difference in apoptosis rate among the 3 groups (F = 15.69, 53.33, 48.28, P < 0.01). (2) The expression of Bcl-2 in HepG2 cells at hour 12, 24 and 48 after transfection was 16.4% ± 5.0%, 13.4% ± 3.5% and 8.6% ± 2.3% in group A, 14.7%±3.8%, 9.1% ±2.0% and 4.6% ±2.0% in group B, and 25.3% ±6. 3%, 19.8% ±4.4% and 10.1% ±3.8% in group C. There was a significant difference in the expression of Bcl-2 among the 3 groups (F = 6.19, 12.29, 5.81, P <0.05). (3) The expression of Caspase-8 at hour 12, 24 and48 after transfection were 19.3% ±2.4%, 27.2% ±1.9% and 33.7% ±3.0% in group A, 22.7% ±2.2%, 30.9% ±3.1% and 38.2% ±3.2% in group B, and 1.2% ±0.8%, 1.8% ±0.6% and 3.2% ±1.5% in group C. There was a significant difference in the expression of Caspase-8 among the 3 groups (F =71.54, 112. 78, I01.61, P < 0.01). Condusions LIGHT gene can signiticanfly promote cell apoptosis through regulating the expression of Bcl-2 and Caspase-8. Interferon-γ enhanced the effect of LIGHT gene on the apoptosis of HepG2 cells.

Objective To analyze the effectiveness of chronic disease management among suburb residents to secure an optimal health management model. Methods Since 2002, physical examinations had been conducted for rural residents aged≥40. Hypertensive patients detected on health check received medical evaluation and risk factor intervention. Results The incidence of high blood pressure decreased from 26.57% in 2002 to 11.99% in 2007-2008. The mortality of cerebrovascular disease declined from 68.00% to 5.26%. The incidence, disability rate, and death rate of hypertension related-stroke were also significantly reduced. Conclusion Primary healthcare center-based chronic disease screening, along with the health management network, can largely decrease the incidence, disability rate, and mortality of stroke associated with high blood pressure.

Objective To establish the genetic test technique of trisomy 21 concurrently conducts with prenatal diagnosis for hereditary hearing loss. Methods Fifty-four pregnant women who underwent prenatal diagnosis for hearing loss of their fetuses in Chinese People's Liberation Army General Hospital from March 2009 to May 2010 were enrolled in this study. All probands from the deaf families have confirmed the causative mutation for hearing loss in Genetic Testing Center in Chinese People's Liberation Army General Hospital. The mean age of 54 pregnant women is 31 years at pregnancy of 18 - 26 weeks, 5 cases ＞ pregnancy of 23 weeks, 9 cases ≥ 35 years. All subjects did not conduct the serologic tests for trisomy 21before. Fifteen to twenty ml amniotic fluid was drawn from 49 cases at pregnancy of 18 - 23 weeks and 5 cases ＞ pregnancy of 23 weeks. One to two ml umbilical blood was drawn from 5 cases ＞ pregnancy of 23 weeks. For 9 cases ≥ 35 years, amniotic fluid cell culture and karyotyping analysis were conducted concurrently. A multiple quantitative fluorescent ( QF) PCR and six microsatellite markers were applied to as trisomy 21. Results (1) Fifty-four fetuses were successfully conducted prenatal genetic diagnosis for hearing loss (included GJB2 and SLC26A4). Ten fetuses copied the exactly same genotypes as the probands. The other 44 cases fetuses did not copy the same genotypes as the probands and won't develop hearing loss. The hearing test showed normal hearing for the neonates. (2) All the 54 fetuses were excluded of trisomy 21 by QF-PCR and were verified after birth. Five fetuses with advanced maternal age were performed karyotyping analysis and showed normal. The diagnostic results of QF-PCR can be obtained in 1 - 3 days without misdiagnosed. Conclusions QF-PCR is an efficient, rapid and accurate technique for detection of trisomy 21 without increasing sample amount. It can be used for fetuses who were undertaken hearing loss gene test or other prenatal gene test.
