1.Effects of serum from burned rats on the gene expression in rat bone mesenchymal stem cells

Objective To investigate the effects of serum from burned rats on the gene expression in cultured rat mesenchymal stem cells (MSCs). Methods Bone marrow was extracted from sacrificed Wistar rats, and MSCs were then incubated for 24h in F-12 medium in the presence of normal rat serum (group N) or serum obtained from burned rats 3 days after injury (group B). Total RNA was extracted from both groups.The mRNA was isolated.An Oligo microarray containing 5705 genes was used to compare the differences of gene expression between two groups. Results There were four genes which expressed differently in two groups.In comparison with group N, the expression of steroid sensitive gene 1 was decreased, but that of fibroblast growth factor-4, dihydroxyacetonephosphate acyltransferase and a EST, which was moderately similar to Bmp2-inducible kinase, were increased in group B. Conclusion Serum from burned rats was able to change the gene expression of MSCs, and it might play the key role in wound repair.
2.Evaluation of reliability and validity of Diabetes Diet-Related Quality-of-Life Scale

Objective To evaluate the reliability and validity of Diabetes Diet-Related Quality-of-Life Scale (DDR-QOL) in Chinese version,in order to provide an efficient instrument for assessing the quality of life of diabetes in diet therapy.Methods Translating and edit the DDR-QOL scale initially,275 eligible subjects were invited to the study.DDR-QOL scale and 36 item Short Form Health Survey (SF-36) were used to collect data which were conducted for evaluating the internal consistency,reproducibility,construct validity,convergent and discriminant validity of DDR-QOL.Results The total Cronbach's α was 0.86.The factor analysis suggested good construct validity of DDR-QOL.Significant correlations were found between DDR-QOL and SF-36.The quality of life of diabetes who is on the job or not can be identified by DDR-QOL.Conclusions The Chinese version of DDR-QOL is a valid and reliable tool to assess the quality of life of diabetes who are in diet therapy.
3.Combing Application of Imaging in Mesenteric Castleman's Disease

Purpose To evaluate imaging characteristics of normal and dynamic contrast enhanced multi-slice-spiral CT (MSCT) and 18F-FDG PET/CT in the patients with the mesenteric Castleman's disease (CD).Materials and Methods A retrospective analysis was made in 11 patients who were pathologically diagnosed CD and investigated their imaging characteristics of the MSCT and 18F-FDG PET/CT.Results 6 cases showed single lesion,all of them were round or oval.5 cases presented satellite lesions.The maximum diameter was (6.11 ± 1.45)cm.3 cases showed stippled or branch calcification,and 3 cases showed slightly patchy low density.11 cases had clear margins except 1 case.All cases showed the same strong lesion enhancement as that in aorta.9 cases showed significantly enhanced tumor blood vessels around the edge or shadow.The degree of 18F-FDG uptake increased in 11 cases,SUVmax was 3.1-5.7,and the average was (4.16 ± 0.86).Conclusion Multimodality imaging performances play an important role in the diagnosis of mesenteric CD.
4.Clinical significances of growth hormone and growth hormone binding pro tein determination in patients with obstructive jaundice

Objective To evaluate the changes and clinical s ignificances of serum growth hormore (GH) and GH binding protein (GHBP) in patie nts with obstructive jaundice (OJ). Methods Serum levels of GH, GHBP, insulin-like growth factor-Ⅰ (IGF-Ⅰ), endotoxin, prealbumin concentr ation and liver function in 60 male in patients with OJ were detected 1 d preope ratively and 1, 3, 7 d postoperatively. And 40 male patients with simple gallbla dder stone hospitalized in the same period were served as control. Resu lts The levels of GH, GHBP and IGF-Ⅰ were significantly lower in pati ents with obstructive jaundice than in control group(P<0.01), which was corr elated with the levels of bilirubin, endotoxin and prealbumin (P<0.01). Conclusion The obviously downregulation of GH/GHBP axis in patients with OJ probably results from the inbibition of expression of GH receptor due t o nutritional dysfunction and endotoxemia, and further aggravate the nutritional dysfuction and endoxemia, which may be one of the important factors in the path ophysiological changes of OJ.
5.Comparison of two different workflows of crossmaching by full-automatic blood grouping instrument

Objective To provide an optimal working process for crossmatching by full-automatic blood typing instrument.Methods Two workflows were applied to crossmatch test by full-automatic blood typing instrument.One was diluting red blood cells of donor samples to concentrate of 1%,the other was detecting directly the donor′s packed red blood cells.Compared consistency and processing time of the two different workflows.Results Cross match results of two workflows were consistent well(U=0,P>0.05).The average processing times before testing and undergoing testing were not significantly different(t=0.692,t=0.562,P>0.05),whereas the average processing times after testing and throughout testing were significantly different(t=146.485,t=67.053,P<0.05).Conclusion The workflow of diluting donor′s sample before testing saved processing time and better suits hospital having a large quantity of specimens.
6.Association between IL-1B+3954/Taq Ⅰ gene polymorphisms and chronic periodontitis

Objective To study whether specific IL-1B+3954/Taq Ⅰ genotype alleles are associated with chronic periodontitis (CP) in Chinese of the Han nationality. Methods CP group consisted of 66 patients while healthy controls group consisted of 50 subjects. Anti-coagulated peripheral blood sample was obtained from each subject and genomic DNA was extracted from each sample. Single nucleotide polymorphisms (SNPs) at IL-1B+3954/Taq Ⅰ were analyzed by standard polymerase chain reaction-restriction fragment length polymorphisms (PCR-RFLP) assay. Results In the control group,higher numerical values of the A1/A1 genotype were observed for the IL-1B+3954/Taq Ⅰ SNPs (84.0%),and the A2+ genotype was present in 16% of this sample. Statistical analysis revealed no significant difference between the two groups. Conlusion There is no evidence in our study supporting the association between single nucleotide polymorphisms (SNPs) of the IL-1B+3954/Taq Ⅰ gene and prevalence and/or severity of chronic periodontitis.

Objective To investigate the isolation, purification and culture methods of human embryonic germ cells (EGCs) on feeder layer cells of human embryonic fibroblasts. Methods Primordial germ cells(PGCs) from the genital ridges of 6-11 weeks(post fertilization) human embryos were cultrued on feeder layer cells of human embryonic fibroblasts(HEFs) which were isolated from the 3-4 month embryos and routinely cultured for over 25 passages. The medium composed of growth factors and differentiation inhibitory factors. The cultures were analyzed with the expression of alkaline phosphatase, immunologic markers (SSEA-1,SSEA-4) and the transcription factor Oct-4 that have been used to routinely to characterize EGCS. Results A large-scale EG cells were obtained and meintained on feeder layers for over 8 passages. The cell surface marker showed that the EGCs possess high levels of AP activity. EGCs colonies stained strongly for SSEA-4,SSEA-1 and they expressed the transcription factor Oct-4.Conclusion EGCs have potential to maintain long term proliferation and undifferentiation state on human embryonic fibroblasts in vitro.;
8.Effect of bFGF in coordination with autografting of bone marrow mesenchymal stem cells on expression of PCNA and Ⅷ-R Ag of porcine skin defect wound

Objective To investigate the effect of bFGF and MSCs autografting on expression of PCNA and Ⅷ-R Ag of porcine skin defect wound. Methods Six male minipigs were used to produce 48 full-thickness skin wounds, each wound area about 2.54 cm~ 2 on both sides of the back (8 wounds per pig). These 48 wounds were randomly treated with bFGF, MSCs, bFGF+MSCs, and normal saline (12 wounds for each treatment). The tissue from the wounds was harvested on day 3, 7, 14 after surgery, then used for HE staining and immunohistochemical staining for proliferating cell nuclear antigen (PCNA) and factor Ⅷ related antigen (Ⅷ-R Ag). Results Granulation tissue was presented in all groups on day 3 and 7 after wound, most abundant in the wounds treated with bFGF+MSCs, as well as blood capillary formation. PCNA expression increased after skin injury, reached the peak on day 7, most in the granulation tissue, and lasted to day 14, then began to decrease. On day 3, 7, 14 after skin injury, PCNA expression was the most highest in the wounds treated with bFGF+MSCs. 10.Regulatory effect of coptisine on key genes involved in cholesterol metabolism.

To study the effect of cholesterol and 25-OH-cholesterol on cholesterol metabolism in HepG2 cells and the effect of coptisine (Cop) extracted from Coptidis Rhizoma (CR) in reducing and regulating cholesterol. In this study, TC, TG, LDL-c and HDL-c were measured by biochemical analysis; mRNA and protein expressions of LDLR, HMGCR and CYP7A1 were detected by qRT-PCR and Western blot. According to the results, cholesterol and 25-OH-cholesterol inducing could decrease in mRNA and protein expressions of LDLR and CYP7A1, so as to increase TC and LDL-c contents. However, Cop could up-regulate mRNA and protein expressions of LDLR and CYP7A1 and down-regulate that of HMGCR, so as to reduce TC and LDL-c levels. These findings suggested that Cop has potential pharmacological activity for reducing cholesterol, and may reduce cholesterol by regulating mRNA and protein expressions of key genes involved in cholesterol metabolism, such as LDLR, CYP7A1 and HMGCR. This study laid a firm theoretical foundation for developing new natural drugs with the cholesterol-lowering activity.
