Cell cycle checkpoints are protective mechanism responding to DNA damages originated from externalor internal factors. When cells are exposed to genotoxic stress or when nutrition crisis occurs, cell cycleprogression is usually stopped or slowed down by cell cycle checkpoints to allow for DNA repair or for handlingthe crisis. Besides, recent studies suggest that some cell cycle checkpoint proteins are also involved in regulatingphysiological DNA replication via controlling the rate of DNA replication. Cell cycle checkpoint proteins ATR,9-1-1 complex, Chk1, Cdc25A and CDK2 may participate in this process. This kind of regulation is supposed to bevery important for ensuring accurate DNA replication and maintaining genomic stability.

ObjectiveTo investigate the clinical value of continuous electronic fetal heart rate (FHR) monitoring for preterm small for gestational age (PSGA) during labor. MethodsThree hundred and three pregnant women who had inevitable preterm labor at 32-36 weeks′ gestation because of premature rupture of membrane or unknown cause underwent continuous electronic FHR monitoring during labor from Jan 2002 to May 2004. In total, 78 newborns were preterm small for gestational age (PSGA ,PSGA group) and 225 newborns were preterm appropriate for gestational age (PAGA, PAGA group). Objective To determine the effect of cetuximab(C225)on the radioresistant human esophageal squamous carcinoma eell line KYSE-150R.Methods A radioresistant human esophageal squamous carcinoma cell line KYSE-150R was established by fractionated irradiation.Morphological changes from KYSE-150 to KYSE-150R were observed by phase-contrast microscopy.Karyotype analysis was performed by G-banding.The radiosensitivities were analyzed by colony formation assays.Results The population doubling time of KYSE-150 and KYSE-150R were(23.6±0.2)h and(25.9±0.6)h (t=6.6,P<0.01),respectively.The chromosome number of KYSE-150R was increased and chromosome aberrations were observed from(69.3±1.9)h to(73.7±1.2)h(t=-8.83,P<0.01).The SF2,D0,Dq and N values of KYSE-150R were all higher than those of KYSE-150.After 5μg/ml of C225 added,the SF2,D0,Dq and N values were significantly decreased as compared to the control.After C225 treatment,the G0/G1 and G2/M phase cells were increased,while S-phase cells decreased(t=-4.478-4.308,P<0.05).Conclusion Cetuximab can enhance the radiosensitivity of radioresistant human esophageal squamous carcinoma cell line KYSE-150R.
5.Clinical significance of S-100B protein in children with different degrees of amniotic fluid meconium contamination

Objective Detection of S-100B protein in different degree meconium pollution level in serum of children with value.MethodsIn 2012 June to 2013 December in our department were simple meconium stained amniotic fluid in term newborns in 73 cases, and set up for the observation group, and according to the amniotic fluid pollution degree is divided into 47 cases of amniotic fluid of Ⅰ-Ⅱdegree pollution group and 26 cases in grade Ⅲmeconium group during the same period, selected 20 cases without amniotic fluid contamination in term healthy newborns for the control group, the groups were compared in S-100B protein content difference.ResultsⅢmeconium stained amniotic fluid were within 6h serum S-100B was significantly higher than that in control group(P<0.01), and the degree of contamination of amniotic fluidⅠ-Ⅱgroup compared with the control group, no significant difference(P>0.05).Ⅲ meconium stained amniotic fluid in children with 72h also increased.ConclusionThird degree meconium stained amniotic fluid but normal Apgar score of newborns may still exist in clinical brain injury, so pay close attention to.
6.In vitro photodynamic antibacterial activity of cationic porphyrin derivative

Objective To investigate susceptibility and antibacterial activity of cationic porphyfin derivative mediated photodynamic antimicrobial chemotherapy (CPD-PACT) against Pseudomonas aeruginosa,to provide experimental evidence for its high efficiency antibacterial activity.Methods The impacts of culture environments on minimum inhibitory concentration (MIC) were measured by double dilution method.The formation of inhibition zone was determined by diffusion plate method.The postantibiotic effect was analyzed by colony forming units.The viability and morphology of Pseudomonas aeruginosa were observed by confocal laser scanning microscopy (CLSM).Results The inoculum size of bacterial had a certain effect on the MIC.The MIC values increased as the pH of medium rose.When the calf serum content of culture medium increased,the MIC rose in light reaction and dropped in dark reaction.The diameter of inhibition zone mainly depended on the laser energy density,but not the concentration of photosensitizer.Though CPD possessed strong antimicrobial activity and persistent suppression on bacterial growth,the surviving Pseudomonas aeruginosa would soon continue to proliferate after PACT.The fluorescence images captured by CLSM showed that CPD-PACT could destroy the membrane integrity,leak the cytoplasmic component,decrease the bacterial activity and finally lead Pseudomonas aeruginosa to death.Conclusions CPD has strong inhibitory activity and obvious postantibiotic effect on Pseudomonas aeruginosa,which is suitable to be developed as an drug candidate for PACT.
7.The Effect of Different Ultraviolet on HaCaT

Objective To observe the effect of different ultraviolet on HaCaT and offer experimental evidences for further studies of the pathogenesis of photodermatitis. Methods Using different dose of UVA and UVB to irradiate HaCaT cell line respectively. 12 hours later, the morphology of HaCaT cells was observed and the sum was calculated. Results The cell linkage was loose and the refraction was weak with some dead cells. The effect of large dosage of UVB was more obvious. Conclusions the effects on keratinocyte after UV exposure are different according to the dose of irradiation, which offers experimental evidences for the further study of the pathogenesis of photodermatitis.
8.Establishment of a mouse model of chronic pulmonary infection with Pseudomonas aeruginosa and analysis of its inflammatory response

Objective To research the chronic respiratory infectious diseases caused by Pseudomonas aeruginosa need to objectively the reflection of the real environment in the body, so established a mouse model of chronic pulmonary infection with Pseudomonas aeruginosa and analyzed it's inflammatory response. Methods Establishment of chronic bronchial infection mice model that were inoculated with Pseudomonos aeruginosa-laden agarose beads, and analyzed its inflammatory response by detected the cytokines and MMP-2, and a differential cell count and a total leukocytes count were performed as well. Results Pseudomonas aeruginosa had been detected after infection, and there were changes in pathological. Pulmonary inflammation appeared in 1 d and reached near baseline levels by 7 d after inoculation. It was verified that the peak of inflammatory reaction in Pseudomonas aeruginosa infection is in 2-3 d; the mice did have detectable levels of circulating matrix metalloproteinase-2 ( MMP-2 ) after infection with Pseudomonas aeruginosa. MMP-2 concentrations in the blood serum peaked at 3 d after inoculation. It is indicated that Pseudomonas aeruginosa can initiate a certain degree of pulmonary fibrosis on the basis of the pulmonary inflammation. Conclusion In this study, chronic bronchial infection animal model affected by Pseudomonas aeruginosa was established successfully. Base on this animal model, we can do the pathogenicity and drug resistance of Pseudomonas aeruginosa further study.
9.Relationship between serum retinol-binding protein 4 and non-alcoholic fatty liver disease in schizophrenia

Objective To investigate the serum retinol-binding protein 4 (RBP4) level in olanzapinetreated schizophrenia with non-alcoholic fatty liver disease (NAFLD) via a case-control study and to explore its relationship with NAFLD.Methods Schizophrenia receiving olanzapine treatment,with or without NAFLD were enrolled as cases (n=60) or controls (n=60).The serum retinol-binding protein 4,metabolic syndrome component including body mass index (BMI),waist-to-hip ratio (WHR),fasting plasma glucose (FPG),total cholesterol (TG),total glycerin (TC) and blood pressure were assayed.TyG index,CT value ratio of liver and spleen were used for insulin resistance or NAFLD diagnosis and severity assessment respectively.Results The serum level of RBP4 was significantly higher in cases than that in controls ((70± 13)mg/L vs (52±12)mg/L ; t =4.943,P<0.01) and the same result was showed after influencing factor adjustment thru covariance analysis (F=16.797,P<0.01).Moreover,cases had significantly higher TyG index,BMI,WHR,FPG,TG and systolic pressure (t=2.383-4.300,P<0.05-0.01).Cases with severe or moderate NAFLD had significantly elevated serum RBP4 level compared with mild cases((79± 16) mg/L,(72±8) mg/L vs (63t9) mg/L,d =16.2,9.9 ; P<0.01,0.05).A significantly negative correlation between RBP4 level and the CT value ratio of liver and spleen (r=-0.244,P<0.05),and a significantly positive correlation between RBP4 level and TyG index,BMI,WHR,TG(r=0.197-0.244,all P<0.05) were observed.The partial coefficient between RBP4 level and CT value of liver and spleen was significant with influence adjusted(r=-0.453,P<0.01).Logistic regression showed that serum RBP4 level(β3=0.105,P<0.01)and TyG index,hyperlipidemia,obesity,central obesity or high blood pressure (β =1.288-9.711,P< 0.05-0.01) were closely associated with NAFLD.Conclusion A heighten serum RBP4 level may be one of NAFLD risk factors in schizophrenia treated with olanzapine.
10.Effect of TLR4/PI3K-related signaling molecules on the migration of asthmatic airway smooth musde cells induced by airway epithelial cells

ObjectiveTo explore the effect of TLR4,PI3K and NF-κB on the migration of asthmatic airway smooth muscle cell(ASMCs) induced by airway epithelial cells.MethodsPrimary ASMCs were cultured by the method of cell digestion.Cell culture supernatant of RTE cells were collected by TNF-α stimulation of epithelial cells.Detected the IL-8 and RANTES levels in the supernatant.The effect of TLR4/PI3K-related signaling molecules on the migration of asthmatic ASMCs induced by epithelial cells were detected by Modified Boyden chemotaxis chamber with anti-TLR4 antibody,Wortmannin and PDTC drugs as a tool.ResultsThe levels of IL-8 and RANTES in the supernatant of TNF-αgroups were significantly increased,and that in the 20 ng/ml group was significantly higher than other groups ( P<0.01 ).Compared with control group,the transmembrane migration of asthmatic ASMCs from other groups was significantly increased (P<0.01 ).The transmembrane migration of asthmatic ASMCs from treated groups was significantly increased than asthma group (P<0.01).The migration of asthmatic ASMCs from TNF-α+anti-TLR4 antibody group,TNF-α+Wortmannin group and TNF-α+Wortmannin+PDTC were significantly decreased than that of TNF-αgroup( P<0.01 ).The migration of asthmatic ASMCs from TNF-α+Wortmannin+PDTC were significantly decreased than that of TNF-α+Wortmannin group (P<0.05).ConclusionTLR4/PI3K-related signaling molecules involved in the transmembrane migration of asthmatic ASMCs induced by airway epithelial cells and may be one of the mechanisms of airway remodeling of asthma.
