2.The roles of motility in colonization of Helicobacter pylori in human stomach
Chinese Journal of Laboratory Medicine 2008;31(7):733-736
Helicobacter pylori is able to colonize the human stomach and dwell in the human stomach for decades or for whole lifetime. A number of potential virulence factors contribute to Helicobacter pylori to colonize this unusual niche. Motility is an essential colonization factor based on the fact that nonmotile variants of Helicobacter pylori can't infect gnotobiotic piglets. Motility is not a colonization factor based on rapid loss of motility of Helicobacter pylori in the gastric lumen in vivo. The exact roles of Helicobacter pylori motility are not yet known. The aim of this article is to discuss correlation between colonization and motility of Helicobacter pylori.
3.Comparative analysis of clinical characteristics and outcome of neonates transferred by different ways
Chinese Journal of Emergency Medicine 2011;20(5):477-480
Objective To explore the clinical characteristics and outcome of neonates transferred by different ways. Method A total of 1 027 neonates transferred to neonatal intensive care unit(NICU)of our hospital from July 2009 to July 2010 were analyzed prospectively. They were divided into three groups: twoway transport(TWT),hospital-aided one-way transport(HAT)and self-help one-way transport(SHT).The conditions at admission and discharge and pre-hospital care of the neonates were compared between the groups. The data were statistically analyzed with square tests, variance analysis and Mann Whitney U test. Results The median ages of TWT,HAT, and SHT groups at admission were 0.46 d,0.33 d and 5 d respectively. The TWT and HAT neonates had smaller gestational age and birth weight than SHT ones, had more severe conditions and cost more, and the differences were significant(P<0.05).Significant differences existed in all emergency treatment except in warming measures(x2=96.855~103.863,P<0.05).The cure percentage at discharge was the highest in SHT group, and differences in treatment results of three groups were significant(Z=-1.987 to-5.347,P<0.05).Conclusions The TWT and HAT neonates have smaller gestational age, ages at admission and birth weight than SHT ones, have more severe conditions, cost more and lower cure percentage at discharge than SHT ones.

BACKGROUND: Previous studies have shown that bone marrow mesenchymal stem cells (BMSCs) may be a promising candidate for cell-replacement therapies for neurodegenerative diseases including Alzheimer's disease (AD). But, it is still unclear whether BMSCs have therapeutic effects on AD.OBJECTIVE: To investigate the influence of BMSC transplantation on ethology of rats with AD, and to find a potential strategy for the development of effective therapies for the treatment of AD.DESIGN, TIME AND SETTING: The randomized controlled study was performed at the Laboratory of the Department of Physiology and Pathophysiology, Medical College of Chinese People's Armed Police Force in March 2009.MATERIALS: A total of 40 healthy male Wistar rats aged 24 months were used in this study, weighing approximately 450 g.METHODS: The natural senile AD rat model was chosen by maze test. The AD rats were randomly divided into four groups with ten animals in each group. In the control group, rats were injected bilaterally with physiological saline into the hippocampus. In the BMSC transplantation group, rats received BMSCs. In the normoxic differentiation and transplantation group, rats received injection of BMSCs induced from neuron-like cells under normoxic condition. In the hypoxic differentiation and transplantation group, rats received injection of BMSCs induced from neuron-like cells under hypoxic condition.MAIN OUTCOME MEASURES: The learning and memory ability of the AD rats were detected by Y type maze test 8 weeks later.Memory ability was tested 48 hours after learning test.RESULTS: The learning and memory scores decreased in control group and increased in BMSC transplantation group, there were all not statistics significance compared with that before transplant treatment (P > 0.05). The learning and memory scores were all higher than before in normoxic differentiation and transplantation group and hypoxic differentiation and transplantation group. There was significant difference between control group and the other groups (P < 0.01).CONCLUSION: BMSC transplantation may improve the learning and memory ability of AD rats. Our results demonstrate that BMSCs not only play an important role in improving cognitive disturbance of AD rats. Oriented differentiated BMSC transplantation was superior to non-differentiated BMSCs. Transplantation of hypoxic differentiated BMSCs has obtained optimal outcomes.

Objective To establish a radioresistant human cell line from esophageal squamous car-cinoma cells and detect the marker expression of cancer stem cells (CSCs). Methods A radioresistant hu-man esophageal squamous carcinoma cell line KYSE-150R was established by fractionated irradiation. Mor-phological changes from KYSE-150 to KYSE-150R were observed by phase- contrast microscopy. Karyotype analysis was performed by G-banding. The radiosensitivity of these two cell lines was assessed by colony for-marion assays. The cell cycle distribution was assayed by flow cytometry. CSCs markers of β-catenin and In-tegrin-β_1 were measured by Western Blot. Results The population doubling time of KYSE-150 and KYSE-150R were (23.62±0.23) hrand (25.90±0.55) hr, respectively (t =6.62,P=0.00). The numbers of chromosomes in KYSE-150R cells were increased and chromosome aberrations were observed. The SF_2, D_0, D_q and N values of KYSE-150R were all higher than those of KYSE-150. The ratio of D_0 values was 1.21. After irradiation, the number of S-phase cells of KYSE-150 increased from 45.35%±4.03% to 55.09%± 1.70% (t = -3.86,P=0.02) and G2/M phase cells decreased from 9.91%±3.83% to 1.15%±0.32% (t = 3.95, P = 0.02). However, no apparent change of cell cycle distribution for KYSE- 150R was observed. The expression levels of CSCs markers, β-catenin and Integrin-β_1 in KYSE-150R were about 2 times of those in KYSE-150. Conclusions The new cell line KYSE-150R is more radioresistant than its parental cell line KYSE-150. The CSCs in KYSE-150R is more than those in KYSE-150, which may suggest that CSCs is re-lated with the radioresistance.
