1.Current status and consideration of laboratory diagnosis of monogenic diseases

Highly developed molecular genetic techniques have got implemented widely in the diagnosis of birth defects and genetic diseases.Due to the special nature of genetic testing , there is still a lack of appropriate external quality assessment , but many clinical laboratories are conducting or going to conduct genetic diagnosis for monogenic diseases and prenatal diagnosis.In this paper , it will be discussed that how to ensure the quality control of the laboratory diagnosis of monogenetic diseases , based on the present application of genetic testing technologies and the difficulties in the diagnosis of monogenetic diseases.
2.Therapeutic Observation of Moxibustion with Different Acupoints for Primary Dysmenorrhea

Objective To observe the clinical efficacy of moxibustion in treating primary dysmenorrhea in adolescent female undergraduates. Method Sixty patients with primary dysmenorrhea were randomized into a treatment group and a control group, 30 in each group. The treatment group was intervened by moxibustion at Guanyuan (CV 4), while the control group was by moxibustion at Sanyinjiao (SP 6). The symptom score was observed before intervention and after 3 menstrual cycles, and the clinical efficacies were compared. Result The symptom score was changed significantly in both groups after intervention (P<0.01). After intervention, there was a significant difference in comparing the symptom score (P<0.05). The total effective rate was 100.0%in the treatment group versus 93.3% in the control group, and the difference was statistically insignificant (P>0.05). Conclusion Moxibustion is an effective approach in treating primary dysmenorrhea, and moxibustion at different points will produce different effects.
3.Bone marrow mesenchymal stem cell transplantation combined with edaravone inhibits neuronal apoptosis after cerebral infarction

BACKGROUND:Several studies have confirmed that bone marrow mesenchymal stem celltransplantation exerts a certain neuroprotective role in cerebral infarction. Edaravone is a novel potent smal-molecule hydroxyl radical scavenger, which play a protective role in the brain by eliminating free radicals and inhibiting nerve celldamage after cerebral infarction. OBJECTIVE:To explore the influence of bone marrow mesenchymal stem cells combined with edaravone on expressions of aquaporin-4, Bcl-2, and brain-derived neurotrophic factor after cerebral infarction in rats. METHODS:Eighty Wistar rats were selected to establish models of cerebral infarction by right middle cerebral artery occlusion, and then randomly divided into control group, bone marrow mesenchymal stem cells group, edaravone group and edaravone+bone marrow mesenchymal stem cells group (combination group). After 6 hours of modeling, rats from these four groups were respectively injected via the tail vein with PBS, bone marrow mesenchymal stem cells, edaravone and edaravone+bone marrow mesenchymal stem cells. After 72 hours, the rats were decapitated to take brain tissues. Consequently, expressions of aquaporin-4, Bcl-2, and brain-derived neurotrophic factor mRNA and protein were determined by RT-PCR and western blot methods. After 12, 24 and 36 hours, TUNEL method was used for the determination of cellapoptosis. RESULTS AND CONCLUSION:The expressions of Bcl-2 and brain-derived neurotrophic factor were higher in the combination group than the other three groups (P<0.05), but the expression of aquaporin-4 was lowest in the combination group (P<0.05). The number of apoptotic cells in the combination group was significantly lower than that in the other three groups. These findings suggest that the combination of bone marrow mesenchymal stem cells and edaravone can improve expressions of Bcl-2 and brain-derived neurotrophic factor in the injured site, and significantly inhibit neuronal apoptosis. Meanwhile, the combination therapy can decrease expression of aquaporin-4 and mitigate cerebral edema, which is superior to bone marrow mesenchymal stem celltransplantation and edaravone alone.
4.Investigation on common human soil-transmitted nematode infections in Quanzhou City in 2012

Objective To understand the epidemic situation of common human soil-transmitted nematode infections in Quan-zhou City in 2012,so as to provide the evidence for improving the prevention and control measures. Methods The residents aged above 3 years from 5 townships in 2 counties were investigated to detect the parasite eggs(hookworm,Ascaris lumbricoides,Trich-uris trichura)by Kato-Katz technique. One kindergarten of each county was randomly selected and 228 children were investigated to detect Enterubius vermicularis eggs by the cellophane tape anus test. Results The total hookworm infection rate was 3.60%(69/1919). For the hookworm infection rates,there was no significant difference among different areas and between different genders, but there were significant differences among different age groups and different careers. No Enterubius vermicularis eggs were detect-ed. Conclusion The effect of the prevention and control of Ascaris lumbricoides,Trichuris trichura and Enterubius vermicularis in-fections is remarkable. However,the infection rate of hookworm has a rising trend and the prevention and control of hookworm in-fection should be strengthened.

6.Magnetic resonance imaging for evaluation of therapeutic effects of high intensity focused ultrasound for breast cancer

Objective:To verify the applicability of Dynamic Contrast Enhanced Magnetic Resonance Imaging(DCE-MRI)to evaluate the breast cancer therapy using the high intensity focused uhrasound(HIFU).Methods:Both DCE-MRI and conventional T_1W/T_2W images were acquired for the total 8 subjects.Two consecutive scans were performed for every patient(before operation and 2 weeks after operation)while four were performed for only 2 patients(additional for 6 months and 12 months after operation).The breast cancer therapy by HIFU was assessed hy comparing the changes of signal,the dynamic patterns of time-signal intensity curve,the early-phase contrast enhancement ratios and vessel anatomies around cancer focuses.Results: The shortened T_2 signals were observed in cancer focuses of 5 subjects while 3 still showed longer T_2 values which were differentiated by the high intensity in T_2W images.The early-phase contrast enhancement ratios of those 5 subjects decreased from 60%~80% to
7.Investigation on Anti-HCV in Blood Donors in Beijing and Some Areas of Hebei Province

Investigation on anti-HCV in different cohorts of 21844 blood donors from 18 districts and counties of Beijing, and Guan, Yongqing, Xincheng, Xushui counties of Hebei province had been performed. It was showed that the anti-HCV-positive rates of voluntary and nonpaid donors were 1.7% (13/738) and 2.9% (116/3943) respectively, and 13. 6% (2011/14757)and 48.1% (1161/2415) for paid donors and plasma pheresis donors respectively. It was also found that the anti-HCV-positive rates of paid donors from Guan and Yongqing counties were (16.4%82/499) and 13.9% (53/381) respectively, 50% (575/1148) and 56% (540/965) for plasma pheresis donors from those counties respectively. The positive rates increased with the level of ALT. More than 45.5% donors whose ALT was above 45 U had anti-HCV.
8.DEVELOPMENT OF HUMAN INTERLEUKIN-2——I. SETTING UP THE PROCESS OF PRODUCTION OF CRUDE HUMAN INTERLEUKIN-2

The methods of extraction of purified blood mononuclear cell(PBMC)from human bully coat (BC), and the conditions of serial production of high titer IL-2 by using PBMC as raw material were studied. For separation of PBMC was selected Methyl Cellulose-Ficoll Hypaque (MC-FH) which can not only be suited to its serial production but also ensure its quality. The separated PBMC's viability was more than 95%, and the phagocyte contamination was less than 5%. The combined PHA/TPA induction method was selected by investigating the various induction methods and conditions. The conditions for inducing the IL-2 by TPA/PHA method were as following., the PBMC's concentration had better be 1.25~2.5?10~6ml; 5% of normal human AB serum was substituted for calf serum; domestic universal microorganism culture kit was used instead of imported CO_2 culture kit: culture was conduced in the rotating 500ml sealed bottles at 37℃ for 72~96 hours. The titer of IL-2 produced under mentioned above conditions reached four digits, and its highest value was 2100 IU/ml and was 20 times higher than that of the titer of IL-2 induced by PHA method. The yield of IL-2 induced with 20units of buffy coat(from 4000ml whole blood) was 23?10~6IU, and the resulting amount of induced liquid was 2000~3600ml. This provided a suitable technological process for production of IL-2 using the buffy coat as raw meterial. (Original article on page 1)
9.Experimental and clinical observation of non-operative treatment of splenic rupture

The feasibility and safety of non - operative treatment of splenic rupture were assessed experimentally in rats.All the experimental animals survived except that there was one death in the group with complete laceration of all the layers of the spleen and in the group with comminuted rupture of the spleen respectively.Non-operative treatment was used to treat 7 cases of single rupture of the spleen with satisfactory results.These findings suggest that non -operative treatment is an effective and safe procedure for the management of blunt injury of the spleen when hemodynamics of the patient is stable.
