1.Relationship between the expression of programmed death receptor ligand 1 and the regulatory T cell infiltration in non-small cell lung cancer

Objective To investigate the relationship between programmed death receptor ligand 1 (PD-L1) and regulatory T cell(Treg) in non-small cell lung cancer.Methods Seventy-nine patients with non-small cell lung cancer who were treated in Hainan Provincial People's Hospital from December 2013 to December 2015 were selected.The levels of PD-L1 and Treg in cancer tissues and adjacent tissues were detected,observed the relationship with clinical pathological features,and analyzed the correlation of PD-L1 level and Treg infiltration.Results High expression of PD-L1 (53.2%,42/79) and Treg (50.6%,40/79) in cancer tissues were significantly higher than those in adjacent tissues (3.8% (3/79),5.1% (4/79)),the difference was significant (x2 =9.761,9.002,P<0.05).High expression of PD-L1 (87.5%,28/32) and Treg(84.4%,27/32)were significantly higher in stage]Ⅲ and Ⅳ cancer tissues than those in stage Ⅰ and Ⅱ cancer tissues(29.8%(14/47),27.7% (13/47)),the difference was significant (x2 =8.958,7.274,P < 0.05).High expression of PD-L1 (85.3%,29/34) and Treg (88.2%,30/34) were significantly higher in lymph node metastasis cancer tissues than those in non-lymph node metastasis cancer tissues (28.9% (13/45),22.2% (10/45)),the difference was significant(x2=8.146,9.056,P<0.05).The high level of PD-L1 in cancer tissues was positively correlated with high Treg infiltration (r =0.762,P<0.05).Conclusion The expression of PD-L1 and Treg in non-small cell lung cancer tissues is related to the lymph node metastasis and stage,and the high expression of PD-L1 is positively correlated with the high infiltration of Treg.
2.The research of lymphocyte proliferation using CFSE and FCM

Aim To quantitatively analyze the lymphocyte proliferation using the time-series data of carboxyfluorescein diacetate succinimidyl ester(CFSE).Methods The model of immune suppression in mice was created by cyclophosphamide and CFSE dye was used for staining the lymphocyte of both control group and treatment group animals. The data were analyzed through mathematical model-fitting.Results The first generation of cell proliferation of the control group and treatment group were 27.17 h and 22.88 h; cell death rates in each division were respectively 20% and 40%; the half-life of cells before the proliferation was respectively 31.53 h and 43.32 h.Conclusions The mathematical fitting of CFSE data can quantitatively analyze the mechanism of the drugs attecting the proliferation of lymphocyte.In this experiment the mechanism of cyclophosphamide inhibiting lymphocyte proliferation may be due to the increase of cell death rate of each division caused by cyclophosphamide.
3.The study on college students' security and its influencing factors

Objective To develop the security scale of college students' sense to investigate college students' subjective feeling of security.According to the characteristics of the feeling tendentiousness to analysis the subject and object factors influencing the insecurity of college students.Methods The security scale of college students' sense compiled was administrated to 500 college student randomly stratified cluster sampling from four universities in Yantai,then description and analysis were analyzed by SPSS17.0,AHP and SAS9.1 to statistical.Results Through the scale of exploratory factor analysis,the Cronbach' s of the scale was 0.629.College students in four dimensions of security and total security were in above-average levels,college students' total scorc of security is significance of difference in grade,gender and whether come from the single parent(all P<0.05) ;Self security was significance of difference in the grade and the gender(P<0.01 and P<0.05) ; and interpersonal security in gender and whether come from the single parent had significance difference(P<0.05) ;Determine control and family security in these demographic variable aspects had no significant differences.Multivariate factors analysis results showed that the decision tree model was better than the logistic model (the misclassification rate of the model was lower than the decision tree model).The main factors affecting the university students' security were gender,grade,the average income and whether they,were the only children.The misclassification rate of the decision tree model was lower than the model of logistic.College students' sense of dimensions are significant with positive correlation,the dimension of the security level may affect other security of the situation.Conclusion The security scale of college students' sense has good reliability and validity and can be used in the study of college security.The factor analysis determines it can be used multiple factors analysis.The survey shows that the overall security of the crowd' s is over the average level.The gender,grade,annual per-capita income and whether the one-child are the potential factors that affect college students' insecurity.

5.The study of morbidity and mortality of acute kidney injury with the diagnosis criteria of RIFLE

Objective To evaluate the value of the RIFLE criteria of acute kidney injury (AKI) for predicting the incidence and prognosis in critically ill patients and to identify the risk factors associated with the outcomes of those patients.Methods All ICU patients admitted over three years and 6-month period were retrospectively studied at Subei People's Hospital.Based on RIFLE criteria,AKI patients were diagnosed and classified into four groups:NAKI (non-AKI),R (risk),I (injury),F (failure)groups.Results (1) AKI occurred in 404 of the 2472 patients (16.3%) during their ICU stay.The mean age of the patients was (61.4 ± 17.4) years.Infectious disease,pulmonary disease,neurological disease and trauma were the major cause of AKI.(2) Mortality in the ICU was much higher in patients with AKI than in patients without AKI (47.3% vs.23.0%,P < 0.05).The mortality rate was 35.6% in R group,48.9% in I group and 60.6% in F group.(3) The logistic regression suggested that MODS and septic shock were the independent risk factors of AKI,but continuous renal replacement therapy (CRRT) are the independent protective factor of AKI.Conclusions In these ICU patients,AKI is associated with increased hospital mortality.The RIFLE classification is a simple and useful diagnosis tool to detect and stratify the severity of AKI and aid in predicting outcome of patients.
6.Comparison of the application of ARMS-PCR and RFLP-PCR techniques in research of single nucleotide polymorphisms about MMP-2 in cervical cancer

Objective To compare the advantages and disadvantages of the amplification refractory mutation system -polymerase chain reaction (ARMS-PCR) with restriction fragment length polymorphism polymerase chain reaction(RFLP-PCR) technology, then explore its application in detection and analysis of single nucleotide polymorphism (SNP). Methods By using ARMS-PCR and RFLP-PCR the polymorphisms of the MMP-2 in cervical cancer and the normal-control group were analyzed. The research sites were-1306 C/T.The final result was verified by DNA sequencing. Results The results of amplified used ARMS-PCR showed that the genotypes of the SNP site in MMP-2 gene promoter were CC, CT, TT, and the DNA fragments were corresponding with the results of DNA sequencing. Conclusion ARMS-PCR technology has the advantages of lower cost time-saving simple operation and reliable results, while has a high value for research of SNP.
7.Influence of Climate and Geographical Environment on Quality of Desmodium Styracifolium Herba Harvested at Different Time

Objective To study the influence of climate, geographical environment and harvesting time on contents of schaftoside in Desmodium styracifolium Herba. Methods Content of schaftoside in Desmodium styracifolium Herba was determined by using HPLC. ODS-C18 column (4. 6 mmí250 mm,5 μm) was used, the mobile phase was methanol-water (3268), the detection wavelength was 272 nm, the flow rate was 0. 5 mL·min-1, and the column temperature was 35℃. Results The climate type, amount of precipitation, average temperature, duration of sunshine, geographical environment of different province had significant impacts on the content of schaftoside in Desmodium styracifolium Herba. Conclusion Subtropical monsoon climate, temperature from 25 ℃ to 32 ℃, sunshine time of 10 h per day and the average annual rainfall of 1 942 mm are suitable for growth of Desmodium styracifolium. The content of schaftoside in samples cultivated from Guangxi Province is higher than that cultivated from Guangdong Province and Hainan Province. The schaftoside content of sample cultivated in July is higher than that cultivated in other months.
8.Effects of bisphenol A on reproductive function in young male mice

Objective To observe the effects of bisphenol A (BPA) on the reproductive function in F1 mice. Methods Pregnancy mice were divided into four groups randomly. From the beginning of pregnance,the mice in experimental groups were given BPA with 30,120,360 mg?kg-1?d-1 until the new mice were born. The mice in control group were given the same volume of corn oil. When the young mice were 30 d, the sperm malformation rate,the activities of lactic acid dehydrogenase(LDH) and glucose-6-phosphate dehydrogenase(G-6-PD) of testicle,the organ coefficient, the histopatholgical changes of testicle in male mice were examined. Results The form of the epithelial cells of seminiferous tubules was disordered. The number of mature spermatozoa was decreased. This changes were more and more with the increase of BPA dose. Compared with control group,organ coefficients in low and large dose groups reduced,LDH activity and G-6-PD activity in middle and large dose groups decreased,LDH activity in low dose group increased. 9.Histological observation of periodontal tissue regeneration guided with collagen membrane combined with recombinant human bone morphogenetic protein-2

Objective To explore the effect of collagen membrane combined with recombinant human bone morphogenetic protein-2(rhBMP-2) on periodontal tissue regeneration and repair in periodontal defect models of rats.Methods Eighteen male healthy Wistar rats,which were made experimental periodontal bone defects in the inferior incisors,were randomly divided into three groups:control group,collagen membrane group(Co),collagen membrane and rhBMP-2 group(Co/rhBMP-2).Two rats from each group were sacrificed at 4,6,8 weeks after operation.The mandibles were removed,and examined under light microscope.Results In Co/rhBMP-2 group: 4 weeks after operation,a large amount of bone formed in the defects;6 weeks later,new bone filled in the defects;8 weeks later,alveolar bone emerged,newly-formed periodontal ligament and cementum localized at the root edges,no gingival epithelium was observed.In Co group:4 weeks after operation,new bone formed at the edge of the defects;6 weeks later,a larger amount of bone and periost were observed;8 weeks later,a small amount of periodontal ligament and cementum localized at the root edges.In control group: 4 weeks after operation,there was a small amount of newly-formed bone at the edge of the defects;6 weeks later,collagen fibers were found at the edge of defects;8 weeks later,a small amount of periodontal tissue reached its original height and gingival epithelium proliferated obviously.Conclusion Collagen membrane combined with rhBMP-2 which has not only conductive effect but also effect of osteoinduction,can prevent the long-epithelium growing,maintain the growth gap,and promote periodontal tissue to regenerate.
10.The chemical component dynamic changes in compatibility of Carthamus tinctorius L. with Glycyrrhiza uralensis Fisch

AIM: To study the dynamic change of chemical components in the compatibility of Carthamus tinctorius L.with Glycyrrhiza uralensis Fisch during decoction together. METHODS: By means of HPLC analysis and phytochemical isolation technique,focusing on the differences between chemical components in co-decoction and individual decoction.Two chemical substances which showed quantitative changes during co-decoction had been isolated and identified as Licochalcone A and Licochalcone B,respectively. CONCLUSION: From the method described in this article,it could be concluded that the chemical profiles of TCM drug-pairs prepared by co-decoction are different from that of extract mixture combined by the individual extract of Carthamus tinctorius L.and Clycyrrhiza uralensis Fisch,respectively.
