1.Cardiomyocyte hypertrophy induced by PGF2α

Objecave The effects of PGF2α on the reactive oxygen species generation and cardiomyocyte hypertrophy were examined in experiments on the cultured neonatal rat cardiomyocytes.It is to study the role of ROS in the signaling pathway of cardiomyocyte hypertrophy induced by PGF2α.Methods The level of intracellular ROS wag measured by the ROS-specific probe 2',7'-dichlorofluorescin diacetate (DCF-DA).Cardiomyocyte hypertrophy was determined by total protein content of the cells and the cell diameter.Results In the cardiomyocytes treated with PGF2α(1nmol/l,10nmol/l,100nmol/l),the fluorescence intensity of intracellular DCF-DA increased by 38.99%,61.76% and 93.55% respectively compared with control group(F=195.69,P<0.01).It is indicated that PGF2α can induce intracellular ROS generation on the cultured neonatal rat cardiomyocytes in a dose-dependent manner.Compared with control group,the total protein increased by 39.51%, 69.93%and 139.06%respectively(F=74.014,P<0.01),and the cell diameter increased by 29.02%,60.79%and 127.40%respectively(F=721.02,P<0.01).It is indicated that PGF2α induce cardiomyocyte hypertrophy on the cultured neonatal rat cardiomyocytes in a dose-dependent manner.Conclusion PGF2α can induce intracellular ROS generation and cardiomyocyte hypertrophy on the cultured neonatal rat cardiomyocytes in a dose-dependent manner.
2.Study on Pharmacognosy of Chongqing Local Medicinal Material Heracleum hemsleyanum

OBJECTIVE:To establish a method for the identification of Chongqing local medicinal material Heracleum hemsley-anum. METHODS:The original plant morphology,morphological characteristics,microscopic characteristics (transverse section and powder),were identified. RESULTS:H. hemsleyanum had cone shape root,brown,many-branched with special fragrance;the microscopic characteristics of taproot a transverse showed no differences with lateral root transverse section,primary xylem was di-arch,secondary phloem was more developed than secondary xylem,secondary phloem was rich in the oil chamber. Vascular tissues contained a large number of parenchyma cells. In TLC chromatrogram,the test sample showed the same color fluorescent spots with-the substance reference of osthole and columbianadin in the corresponding locations. CONCLUSIONS:The established method is simple with high accuracy,and can be used as the basis for identification of H. hemsleyanum.
3.Effect of Extract of Ginkgo Biloba(EGb)on Cell of Neonatal Rat Cardiocyte Hypertrophy Induced by Prostaglandin F2 Alpha

Objective To investigate the effects and mechanisms of extract of ginkgo biloba(EGb)on the cell of cardiocyte hypertrophy induced by prostaglandin F2 alpha.Methods normal cells were used as negative control,and spontaneously PGF2? and EGb were used as experimental groups.The cells were isolated and cultured on cultured neonatal rat cardiocyte by PGF2?,cardiocyte hypertrophy was determined by the cell surface and the total protein of cells;the level of intracellular ROS measured by the fluorescence microscope.Results In cardiocyte hypertrophy,the cell surface,the level of intracellular ROS and the total protein of cells increased significantly in cutured neonatal rat cardiac treated with PGF2?.Compared with cells treated with PGF2?,EGb(40?g/ml,80?g/ml,100?g/ml)inhibited cardiocyte hypertrophy by PGF2?-induced,decreased in the cell surface by 19%,27%,33% and in the total protein of cells by 21%,39%,47% respectively(all P

Long non-coding RNA urothelial carcinoma associated 1 (UCA1) is initially discovered and named in bladder cancer tissue,which is highly expressed in multi types of tumor tissues,such as bladder cancer,ovarian cancer,lung cancer,suggesting that UCA1 acts as oncogene.UCA1 is confirmed to regulate tumor cell proliferation,apoptosis,invasion and migration,which plays an important role in the occurrence and development of cancers.UCA1 is expected to become a new biomarker for diagnosis,prognosis and drug susceptibility,which may be a promising therapeutic target of cancer.
6.Determination of tramadol in human plasma by High-performance liquid chromatographic

Objective To improve a HPLOUV method for the determination of Tramadol hydrochlo-ride in human plasma. Method Alkalinized samples of plasma were extracted with dichloromethane. Tramadol and internai standard were analyzed on Elite C18 column (200mm?5.0mm, 5?m) with a acetoni-trile-phsphate buffer (pH6.5) (74:26, v/v) mobile phase and detedted at 220nm. Results The Calibra-tion plots in human plasma were linear (r=0.9984, n =5) from 10 to 800 ng/ml. The limit of detection was 10 ng/ml. For 20, 100, 400 ng/ml check samples, intra-run precisions (RSD) were 3.81% -5.44% and inter-run preceisions (RSD) were 3.95% -4.41% , respectively. The average recoveries were 89% , 96% and 92% for the low, middle and high check samples, respectively. Conclusion The improved ana-lytical method for Tramadol hydrochloride was found to be sensitive, simple and rapid, suitable for applica-tion in forensic toxicological analysis and routine determination of numerous samples.
7.Application of painless ward standardized pain management for patients with inguinal hernia after surgery

Objective To investigate the effects of painless ward standardized pain management on pain control and postoperative recovery of patients with inguinal hernia after surgery.Methods From January 2015 to January 2016,44 cases of inguinal hernia in Department of General Surgery,First People's Hospital of Huainan,control group (22 cases) and observation group (22 cases) were retrospectively studied.The patients in observation group underwent painless ward standardized pain management and those in control group underwent traditional pain mnanagement model.Pain score after surgery,time for get out of bed after surgery,postoperative intestinal exhaust time,duration of night sleep,and satisfaction degree for pain-control were comparrd between two groups of patients.Results For pain score after surgery,the observation group 3 h after operation,postoperative 1 d,2 d pain score respectively (3.46 ± O.56),(2.72 ± O.21),(2.20 ± O.43) points,the control group 3 h after operation,postoperative 1 d,2 d pain score respectively (4.48 ± 1.52),(3.55 ± 0.40),(2.80 ± 0.40) points,there were significant difference between two groups (P < 0.05).For sleep time,the observation group on the day of surgery,postoperative 1 d,2 d sleep time respectively (5.23 ± 0.98) h,(6.57 ± 0.54) h,(7.50 ± 0.54) h,the control of sleep time of 2 d group on the day of surgery,postoperative 1 d,respectively (4.35 e 1.28) h,(5.17 ± 0.45) h,(6.72 ± 0.61) h,there were significant difference between two groups (P < 0.05).The satisfaction rate of pain control in the observation group was 86.3%,which was higher than that of the control group 50.0% (P < 0.05).The observation group first ambulation time,anal ventilation time,hospital stay were (10.5 ± 2.0) h,(16.8±2.1) h,(6.6±1.4) d,the control group for the first time,ambulation time,anal ventilation time,hospital stay were (15.9 ± 3.O) h,(22.6 ± 3.3) h,(7.5 ± 1.2) d,there were significant difference between two groups (P < 0.05).However,there was no difference between the two groups in the total cost of hospitalization.Conclusions Application of painless ward standardized pain management for patients with inguinal hernia after surgery can shorten time for get out of bed after surgery,postoperative intestinal exhaust time,prolong duration of night sleep,improve satisfaction degree for pain-control during hospitalization.
8.Effect of Rational-emotive Uherapy on Rehabilitation of Patients with Depression after Stroke

Objective To observe the effect of rational-emotive therapy(RET)on rehabilitation of patients with depression after stroke.Methods 62 hemiplegic patients with depression after stroke were randomly divided into the RET group and control group with 31 cases in each group.All patients were evaluated with Hamilton Rating Scale for Depresstion(HAMD)and Barthel Index(BI)before and after treatment.Results After treatment,the scores of HAMD and BI of the patients in the RET group was superior to those in the control group(P<0.01).Conclusion The rational-emotive therapy is effective to improve not only depressive symptoms but also the quality of living.
9.Analysis of treatment effect of chronomodulated chemotherapy plus concurrent intensity-modulated radiotherapy in patients with nasopharyngeal carcinoma (NPC)

Objective To evaluate the therapeutic effects,acute and late side-effects of cisplatin plus 5-fluorouracil (PF) schemed chronomodulated chemotherapy combined with concurrent intensitymodulated radiotherapy(IMRT) for nasopharyngeal carcinoma.Methods A total of 48 nasopharyngeal carcinoma patients treated with chronomodulated chemotherapy plus concurrent IMRT were retrospectively studied.The radiation doses prescribed to the gross target volume GTVnx of nasopharyngeal,to the positive neck lymphnodes GTVnd,to the clinical target volume CTV1,and to CTV2 were 73.92-77.88,69.96,60.06-66.00 and 50.96-56.00 Gy respectively.The Kaplan-Meier survival analysis was used to calculate the local-regional progression-free rate.The acute and late side-effects were graded according to RTOG radiation morbidity scoring criteria.Results The complete remission (CR) rate was 41.6%(20/48) ; the partial remission (PR) rate was 47.9% (23/48) ; the stable disease (SD) rate was 4.2%(2/48) ; and the local tumor control rate was 89.6%.Most of the patients had grade 1 to 2 acute side effects and grade 0 to 1 late side-effects.No grade 4 acute and late toxicity was observed.1,2,4-year survival rates were 93.8%,79.2% and 64.5%,respectively.The DVH analysis showed that IMRT improved the total dose and dose per-fraction to the target volume with considerable dose reduction of the normal structures at the same time.Conclusions PF chronomodulated chemotherapy plus concurrent intensity-modulated radiotherapy for NPC could improve therapeutic effects with satisfactory tolerance of the side effects.
10.The study on dual-energy lung perfusion imaging in the diagnosis of pulmonary embolism using dualsource CT

Objective To explore the diagnostic values of dual energy lung perfusion in the diagnosis of pulmonary embolism by using dual-source CT (DSCT). Methods Thirty patients with clinically suspected pulmonary embolism underwent dual-energy scanning with dual-source CT. The scanned data were integrated into three groups including 140, 80 kV and coefficient of 0.3. According to the CT pulmonary angiography (CTPA) of the fusion data, the patients were divided into pulmonary embolism group and normal group. The thin-slice reconstruction of data was analyzed using dual-energy perfusion imaging analysis software. The lung field was divided into upper, middle and lower part to make quantitative analysis of lung tissue perfusion. Paired t-tests were used in the normal patients to compare bilateral lungs, and independent samples t-tests were applied to compare the embolism group and normal group, while minimum intensity projection images (MinIP) were utilized in the assessment of lung ventilation. Results Dual energy CT showed symmetrical homogeneous perfusion in 16 normal cases, without significant perfusion defects. Quantitative analysis showed that left and right lung perfusion were (27 ± 7) and (28 ± 8 ) HU respectively, and no significant difference was found between the two sides ( t=-1.73, P >0.05 ).Perfusion of the left upper, middle and lower lung was ( 23 ± 6), (24 ± 6), and (28 ± 8) HU respectively, while the perfusion of right upper, middle and lower lung was (26 ±8), (27 ±8), and (28 ±9) HU respectively, showing no statistical significant difference between the two sides (t=-1.91, -1.96,-1.73 ,P>0.05 ). Angiography of pulmonary embolism group(14 cases)showed filling defects in the pulmonary trunk, segments and sub-segments. Pulmonary perfusion imaging showed low perfusion or defectsin lung field that dominated by embolic vessels. Quantitative analysis showed that the perfusion of the whole lung and the middle and lower lung were (22 ±5), (22 ±8), and (21 ±8) HU in the embolism group,which were significantly different from the normal group (t=-2. 10, -2.32, -2.63, P<0.05).Minimum intensity projection images showed a good consistency of abnormal ventilation zone area and perfusion abnormalities. Conclusions Pulmonary perfusion status, especially pulmonary embolism, can be analyzed by dual energy CT scanning. It helps to early discover and precisely locate the embolism.
