1.db/db mouse and diabetic retinopathy
Chinese Journal of Experimental Ophthalmology 2014;32(1):75-79
Diabetic retinopathy (DR) is one of the most common microvascular complications of diabetes, the incidence increased year by year,so the research of pathogenesis and interventions of DR is in the ascendant. The mechanism of DR mainly include glucose toxicity products and its effects on cell signaling pathways, leading to vascular disease and neuropathy.Research shows that,DR may occur with actived polyol pathway, increased, oxidative stress, the formation of advanced glycation end products (AGEs),protein kinase C pathway and vascular endothelial growth factor (VEGF)expression.db / db mice as an animal model of spontaneous type 2 diabetes, which is very similar to human type 2 diabetes,is more widely used in studies of DR.This article reviews some progresses of db / db mouse in the DR,and provide clues to further elucidate the pathogenesis of DR and development effective preventive measures.

Objective To assess the effectiveness and safety and biocompatibility of balloon-expanding stainless steel stents (SSS) covered with polyurethane membrane (PUM), expanding poly teflon ester membrane(ePTFEM)and biomembrane (BM) in the treatment of experimental saccular aneurysms of carotid arteries in canine models. Methods 36 experimental saccular aneurysms of carotid arteries in 20 canines were created successfully by sugery. Experimental aneurysms were treated with covered stents including twelve PUM-SSS, twelve ePTFEM-SSS and twelve BM-SSS for each twelve canines repectively. They were placed endovascularly in the common carotid arteries covering the orifice of aneurysms. Follow up angiography was performed immediately after the procedure and 2, 4 and 12 weeks afterwards under the control of conventional anticoagulation. The animals were then sacrificed for histopathologic and statistical investigation. Results 36 stents were placed successfully in the target arteries. The complete patency rates of PUM-SSS, ePTFEM-SSS and BM-SSS were 25.0%, 41.7% and 91.7% respectively. Histological analysis indicated that all treated aneurysmal pouches were filled with thrombus. Stent wires were found to be located deep within the vessel wall and encased by extension of the tunica intima. Endothelialization of BM-SSS groups already began at the 2nd week which was earlier than that of PUM-SSS and ePTFEM-SSS. Various degrees of degenerative cells were seen under the transmission electron microscopy without surface erosion of stents. Conclusion Placement of covered stent endovascularly is expedient, safe, and effective. BM-SSS provides the best mechanical behavior, physiochemical stability, anticoagulative ability and biocompatibility.

Objective To study the availability by making experimental saccular aneurysm models of carotid arteries in canine similar to human intracranial aneurysms. Methods Twenty healthy canines with experimental saccular side-wall aneurysms of carotid arteries were created successfully by surgery. Results Forty experimental saccular side-wall aneurysms of carotid arteries were created successfully with 36 aneurysms and parent arteries maintaining patency with each other and four spontaneously occluded confirmed by angiography.Model successfurll rate reached 90%.Conclusions Experimental saccular side-wall aneurysms of carotid arteries in canines were one of best models created for simulating human intracranial aneurysms.

Objective:To explore the correlation betWeen deceleration capacity of heart rate (DC)and heart rate var-iability (HRV)in patients With essential hypertension (EH)complicated coronary heart disease (CHD).Methods:DC and HRV Were monitored in 81 patients With EH + CHD (EH + CHD group)and 80 healthy volunteers (healthy control group)respectively. Results:Compared With healthy control group, there Were significant decrease in DC [(11.99±4.93)ms vs.(6.62±1.83)ms],standard deviation of normal to normal RR intervals calculated o-ver the 24 h period [SDNN,(129.94±15.22)ms vs.(63.66±12.13)ms],root-mean square of differences be-tWeen successive normal to normal intervals [rMSSD,(55.52±9.03)ms vs.(28.25±8.81)ms]and adjacent nor-mal RR interval difference > 50ms stroke accounted for a percentage of 24h total RR interval [PNN50,(13.99± 5.63)% vs.(7.29±1.11)%]of HRV in EH + CHD group,P <0.05 all;Pearson correlation analysis indicated that DC Was positively correlatedWith SDNN,rMSSD and PNN50 in EH+CHD patients (r =0.571,0.662,0.763, P <0.05 all). Conclusion:There is positive correlation betWeen deceleration capacity of heart rate and heart rate variability in patients With hypertension complicated coronary heart disease,Which can be jointly used as evidence for diagnosis and treatment.

We detected the seroprevalence of Toxoplasma gondii (T.gondii) in the wild birds in northeast China. The wild bird's blood was collected from the cutaneous ulnar vein and the serum was isolated and used for detection of anti T.gondii antibody by modified agglutination test (MAT).Results showed that totally 179 birds' serum samples were collected.Twenty serum samples (11.17%) were positive with T.gondii antibody, which belonged to 9 orders,17 families and 31 species. The seroprevalence against T.gondii was about 5.26% (1/19) in Columbiformes,9.09% (9/99) in Passeriformes,14.29% (3/21) in Falconiformes,15.00% (5/22) in Piciformes,16.67% (1/6) in Coraciiformes, and 25.00% (1/4) in Anseriformes.Based on their feeding behavior, the seroprevalence was 12.00% (3/25) in carnivorous wild birds,10.60% (15/141) in omnivorous wild birds, and 15.38% (2/13) in the wild birds feeding on aquatic animals or plants. These wild birds also can be sorted as migratory and sedentary (non-migratory) according to their migration habits, and the serum positivity was 11.67% (14/120),and 10.71% (6/59) respectively. The seroprevalence against Toxoplasma gondii in wild birds in northeast China is about 11.17%,which indicates a common infection of Toxoplasma gondii in wild birds.

Objective To determine the levels of miR-193a-3p in serum from patients with renal clear cell carcinoma, and eval-uate the potential of miR-193a-3p asa molecular biomarker of renal clear cell carcinoma. Methods Serum samples were taken from 107 untreated hospitalized patients with renal clear cell carcinoma (TNM Ⅰ grade 76 cases,Ⅱ grade 16 cases,Ⅲ grade 2 cases,Ⅳ grade 8 cases and unknown 5 cases)in Nanjing General Hospital from 2010 to 2014 year and 107 age-and gender-matched healthy individuals to determine the level of miR-193a-3p by quantitative reverse-transcription PCR (qRT-PCR). The early diagnostic value of miR-193a-3p for renal clear cell carcinoma was assessed by ROC curve. Results qRT-PCR confirmed that serum miR-193a-3p expression levels was significantly elevated in cancer than in normal controls (3.294± 1.526 vs 1.944±0.600,P =0.000).TNM Ⅰ grade (3.411±1.676 vs 1.944±0.600,P =0.000),Ⅱ grade (2.926±0.927 vs 1.944±0.600,P =0.001),Ⅳ grade(2.926±0.894 vs 1.944±0.600,P =0.000)is significantly higher than that in con-trol group. The area under the ROC curve (AUC)of miR-193a-3p was 0.820 (95% CI,0.756 ~ 0.883),demonstratinga high sensitivity (80.3%)and specificity (93.5%)for the early diagnosis of renal clear cell carcinoma. Conclusion MiR-193a-3p content was significantly elevated in serum from renal clear cell carcinoma and has a high accuracy in diagnosing ear-ly cancer, which holds potential as molecular biomarker for renal clear cell carcinoma.
