1.Clinical significance of the changes of serum IL-6, IL-8,TNF-α and CRP in perioperative patients with pancreatitis

Objective To study the variation of serum IL-6,IL-8,TNF-α and CRP in perioperative patients with pancreatitis. Methods 39 caaes in perioperative patients with pancreatitis in our hospital from January 2008 to 2010 April were selected as observation group,and 39 healthy people were selected as control group,and the serum IL-6,IL-8,TNF-α and CRP of the observation group before the treatment and after the treatment at 4h,at the first day, at the second day and the seventh day and the control group were detected, and the results were analyzed and compared. Results The serum IL-6,IL-8 ,TNF-α and CRP of the observation group before the treatment were higher than those of the control group( all P < 0. 01 ) there were significant differences, and the levels of the observation group at 4h,at the first day,and at the second day showed decreasing,the levels compared to those of the control group( all P < 0. 05 ) there were significant differences, the levels of the observation group at seventh day were compared to those of the control group(all P>0. 05) there were no significant differences. Conclusion It was regulation of the variation of serum IL-6, IL-8 ,TNF-α and CRP in perioperative patients with pancreatitis, and tit could provide evidence for the treatment.
2.Analysis of GDNF mRNA gene expression on both severed sections of the sciatic nerve in rats

Objective To investigate the gene expression of GDNF mRNA on both ends of the severed sections of sciatic nerve in rats. Methods The specimen of sciatic nerve proximal and distal to the cut off site was taken respectively before and after sciatic nerve severance. The level of GDNF mRNA on both cutting sections was observed and compared by using of semi quantitative RT-PCR method with ? Actin as a inner consult before nad after sciatic nerve cut off. Results A minimum expression of GDNF mRNA was found on sciatic nerve before the operation, however, there was an increase of 20% at 24 hours in the distal section after sciatic nerve cut off, 60% at 7th day, 85% at 14th day and 90% at 28th day. While it decreased by 10%, 38%, 45% and 52% at 1st day, 7th day, 14th day and 28th day in the proximal section respectively. Conclusion The decreasing and increasing of GDNF mRNA level at the proximal and distal sciatic nerve section is the result of the disruption of “neuron axon target”axis after sciatic nerve cut off. This may provide a theory basis in treatment of SCI by use of the foreign GDNF.
3.Prolongation of survival peirod of islets by ICCs encapsulated by semipermeable membrane xenografted in brain

Objective To identify the effect of decreasing the rejection using the biological semipermeable membrane combined with the privileged site xenotransplantation. Methods The rat ICCs encapsulated by biological semipermeable membrane were xenotransplanted into dog's brain. The pathological changes of implants and surrounding cerebral tissues were observed under light and electric microscopy. The ?-cells of implants were identified by immunohistochemistry. Results After 2 months of transplantation, The ?-cells were observed and the lymphocytes were dispersed in the grafts.The brain tissues near the grafts showed slight edema and glial hyperplasia. Conclusion The method using the biological semipermeable membrane in combination with the privileged site xenotransplantation have a beneficial effect on the inhibition of the rejection of heterogeneous ICCs implant.
4.Combined Examination of Plasma Levels of TnT and NT-proBNP for Predicting the Risk of Peri-operative Cardiovascular Events in Elder Coronary Artery Disease Patients With Non-cardiac Surgery

Objective: To investigate the combined examination of plasma levels of troponin T (TnT) and N-terminal pro-brain natriuretic peptide (NT-proBNP) for predicting the risk of peri-operative cardiovascular events in elder coronary artery disease (CAD) patients with non-cardiac surgery. Methods: A total of 200 patients from (60-82) years of age with elective non-cardiac surgery were enrolled. The patients were divided into 2 groups: CAD group,n=118 and Non-CAD group,n=82. Plasma levels of TnT and NT-proBNP were examined at 1 day before and 2 days after the operation respectively. The cardiovascular events were recorded in both groups and the best cut-off values for TnT and NT-proBNP in risk prediction were evaluated by receiver operating characteristic (ROC) analysis. Results:①CAD group had the higher levels of post-operative TnT and NT-proBNP than pre-operative; both TnT and NT-proBNP levels were higher in CAD group than those in Non-CAD group at the same time points, allP<0.05.②The total cardiovascular events in CAD group was higher than those in non-CAD group (χ2=5.85,P=0.016) and the individual event was similar between 2 groups.③In CAD group, the best cut-off value for pre-operative TnT in predicting peri-operative cardiovascular events was 18.5 ng/L with the sensitivity at 72% and speciifcity at 69.6%; for NT-proBNP was 98.0 pg/ml with the sensitivity at 90.8% and speciifcity at 74.6%.④The patients with the higher level of pre-operative TnT had the increased risk of total cardiovascular events than those with normal TnT (χ2=4.56,P=0.043), and the same as NT-proBNP (χ2=4.08, P=0.036). Likewise, the patients with higher levels of both pre-operative TnT and NT-proBNP had the increased risk of total cardiovascular events than those with normal TnT and NT-proBNP (χ2=13.32,P=0.000). Conclusion:Either plasma levels of TnT or NT-proBNP could be used as the biomarker for predicting the risk of peri-operative cardiovascular events in elder CAD patients with non-cardiac surgery, the combined examination would have the better predictive value.

6.A clinical study on relationship between hypertriglyceridemia and acute pancreatitis

Objective To evaluate the relationship between hypertriglyceridemia and acute pancreatitis.Methods Data was analyzed from 41 patients with acute pancreatits from Apr.1998 to Oct.2000 in our hospital.Of the 13 patients(GroupⅠ) with plasma TG level being higher than 11.3 mmol/L, 8 accompanied with gallstone(61.5%)and 5 without gallstone(38.5%)of the 28 patients (GroupⅡ) with plasma TG level being lower than 11.3 mmol/L,19 accompanied with gallstone(47.3%)and 9 without gallstone(52.7%)RANSON score,morbidity of complications and the level of ALT and AST were compared between two groaps.The correlation between TG and RANSON score was analysed.Results There were significant difference between the two groups on RANSON score,morbidity of complications and level of ALT and AST(P0.05).Conclusion There is an close relationship between hypertriglyceridemia and acute pancreatitis,the high level of plasma TG plags a key a role in acute pancreatitis,patients with acute pancreatitis with hypertriglyceridemia are more likely to have higher morbidity of complications and liver function aggravating.
7.Intelligent optimization of the structure of the large section highway tunnel based on improved immune genetic algorithm

As in the building of deep buried long tunnels, there are complicated conditions such as great deformation, high stress, multi-variables, high non-linearity and so on, the algorithm for structure optimization and its application in tunnel engineering are still in the starting stage. Along with the rapid development of highways across the country, It has become a very urgent task to be tackled to carry out the optimization design of the structure of the section of the tunnel to lessen excavation workload and to reinforce the support. Artificial intelligence demonstrates an extremely strong capability of identifying, expressing and disposing such kind of multiple variables and complicated non- linear relations. In this paper, a comprehensive consideration of the strategy of the selection and updating of the concentration and adaptability of the immune algorithm is made to replace the selection mode in the original genetic algorithm which depends simply on the adaptability value. Such an algorithm has the advantages of both the immune algorithm and the genetic algorithm, thus serving the purpose of not only enhancing the individual adaptability but maintaining the individual diversity as well. By use of the identifying function of the antigen memory, the global search capability of the immune genetic algorithm is raised, thereby avoiding the occurrence of the premature phenomenon. By optimizing the structure of the section of the Huayuan tunnel, the current excavation area and support design are adjusted. A conclusion with applicable value is arrived at. At a higher computational speed and a higher efficiency, the current method is verified to have advantages in the optimization computation of the tunnel project. This also suggests that the application of the immune genetic algorithm has a practical significance to the stability assessment and informationlzation design of the wall rock of the tunnel.

The abdominal CT findings of cirrhosis of advanced schistosomiasis in 262 cases were analyzed.All the cases had the calcification in liver in varying degrees,hypodense structure in junction areas and blood vessel in centre,calcification in portal vein system,calcification of colon and so on.The characteristics of CT performance can provide the evidence for defferential diagnosis.

Objective To observe the regulation of sry-related high-mobility-group box-containing 2 (Sox2) on epidermal growth factor receptor(EGFR)and on the sphere formation rate of glioma stem cells .Methods Promoter reporter plasmids of epidermal growth factor homologous receptor ErbB2 ,ErbB3 ,ErbB4 were respectively constructed .Sox2 expression plasmid was co-transfected together with the reporter plasmid into U251 cells .Then the luciferase activity was analyzed by dual-luciferase reporter assay sys-tem to test the regulation of Sox2 on ErbB2 ,ErbB3 ,ErbB4 promoters .Sphere formation assay was used to observe selfrenew of the cancer stem cells after transfection of Sox 2 .The expression of EGFR and ErbB2 proteins in the spheres was examined by Western blot .Results Sox2 could dose-dependently increase the ErbB2 ,ErbB3 ,ErbB4 promoter drived luciferase activity .Sox2 promotes the sphere-forming rate of U251 cells and upregulates the expression of EGFR and ErbB2 in the spheres .Conclusion Sox2 could up-regulate the expression of EGFR and promote selfrenew of glioma stem cells .
10.Comparative study of GDNF and methyl prednisolone on intracellular free calcium and containing water capacity after spinal cord injury in rats

Objectives:To compare the effect of glial cell line-derived neurotrophic factor(GDNF) and methylprednisolone(MP) on intracellular free calcium([Ca2+]i) and capacity of containing water(CCW) after spinal cord injury(SCI). Methods:Forty-two SD rats were divided into four groups: control, GDNF,MP and GDNF+MP.GDNF and MP were supplied through subarachnoid cavity, tail vein and both of the above after T12 segments spinal cord injury by modified Allen crush method. Animals were executed and specimens were collected at 24 h, 72 h and 168 h after SCI.[Ca2+]i and CCW were measured using Fura-2 method. Results:Both [Ca2+]i and CCW of injured spinal cord in GDNF,MP, and GDNF+MP groups were lower than that of the control group at 24 h and 72 h after SCI(P<0.01).Levels of [Ca2+]i and CCW in GDNF group was much higher than that of MP and MP+GDNF groups(P<0.01),but there was no difference between MP group and MP+GDNF group(P>0.05) 24 hours after SCI. Seventy-two hours after SCI,[Ca2+]i in GDNF group was higher than that in MP group, MP group was higher than MP+GDNF group(P<0.05).CCW of GDNF group exceeded that of MP and MP+GDNF group(P<0.01).[Ca2+]i in GDNF group was higher than that in MP group, and MP group was higher than MP+GDNF group(P<0.01) 7 days postoperatively, but for CCW, there were no differences among GDNF,MP and MP+GDNF groups(P>0.05). Conclusions: Both GDNF,MP and GDNF + MP groups might reduce [Ca2+]i and CCW of injured spinal cord. The curative effect of MP for SCI preponderate over that of GDNF, the effect of MP could be enhanced by adding GDNF.
