1.Research progress of transcription factor sal-like protein 4
Cancer Research and Clinic 2016;28(7):497-501
Sal-like protein 4 (SALL4), a newly discovered oncogenes, plays an important role in the formation of embryo, maintenance of embryonic stem cell pluripotency and self-renewal. SALL4 not only has a close relationship with hereditary disease, benign disease, malignant tumor, blood system tumor and germ cell tumors, but also chemoresistance and poor prognosis. SALL4 obtains biological functions by adjusting different signaling pathways, transcription factors and epigenetic modulation. Further mechanism study of SALL4 and its relationship with various diseases will provide new opportunities for the treatment of diseases.

Objective To assess the influence of pre-exposure to non-ablating ultrasound on the effect of high intensity focused ultrasound(HIFU)ablation. Methods Forty rabbits with transplanted VX2 tumors on their livers were divided into three pre-exposure groups(60 W,80 W and 100 W)and a control group(O W),with 10 rabbits in each group. Each group was pre-exposed to lower intensity focused ultrasound at the corresponding power. From each group,5 rabbits were randomly selected to be exposed to HIFU on the next day. Each group received 14scans.The rabbits were sacrificed 24 h later to take tissue samples for tfipheny tetrazolium chloride(TTC)staining to measure the amount of coagulative necrosis.Hematoxylin and eosin(HE)staining and succinate dehydrogenase (SDH)were also measured to observe the structure and activation of pre-exposed tumors. Results After HIFU exposure, the ablated volumes increased with pre-exposure acoustic intensity, and all volumes were larger than those in the control group. The ablating efficiency in the 100 W group was the highest. The pre-exposure did not itself ablate tumor tissue, but in the 80 W and 100 W groups the structure and activation of the tissues changed. Those in the 60 W group were not obviously altered. Conclusion Prior exposure to non-ablating ultrasound can highly enhance the efficacy of HIFU ablation.

Objective To observe the pathological regression of rabbit liver after the low-dose irradiation with high intensity focused ultrasound (HIFU), and to explore the role and mechanism of the irradiation in changing acoustic environment of rabbit liver. Methods Pathological study was performed in 10 New Zealand white rabbits, the livers were observed under light and electron microscope. The observation was done immediately after low-dose HIFU irradiation and in 2, 3, 5, 7 days after the low-dose irradiation with HIFU. Results Light microscope changes: Edema existed in hepatocytes after irradiation instantly, congestion and aggravated edema were found at 2-3 days, then the injured cells recovered gradually. Electron microscope changes: Detached endothelial cells of hepatic sinusoid and swelling organelles were observed after irradiation, while 2 or 3 days later, erythrocyte aggregation was found in hepatic sinusoid and cavitation was found in cytoplasm. Thereafter, organelles swelling reduced till resumed to normal. Conclusion Low-dose HIFU irradiation can make corresponding changes to the acoustic environment of rabbit liver.

Objective To analyze the MRI findings of Wallerian degeneration in pyramidal tract after cere-bral injury for improving the diagnosis accuracy. Methods The MRI findings of Wallerian degeneration in pyramidal tract in 25 cases were analyzed in association with the primary cerebral injury. Results All the primary diseases were above the tentorium of cerebellum in 25 cases,MRI in 2 cases demonstrated a continuous thin band -like long T1 or isometric T1 and long T2 abnormal changes in pyramidal tract, hyperintensify on FLAIR and DWI.2 cases showed hypointensify on T1 WI,hyperintensify on T2 WI and FLAIR,isointensity on DWI.21 cases showed hypointensify on T1 WI,hyperintensify on T2 WI and FLAIR,isointensity or hypointensify on DWI with brainstem atrophy. Conclusion MRI is valuable in the diagnosis of Wallerian degeneration in pyramidal tract after cerebral injury.

Objective To evaluate the safety and possibility of transesophageal emergency ventricular pacing with esophageal balloonelectrode catheter. Methods Sixty four patients with critical bradyarrhythmias were divided randomly to receive either transesophagealventricular pacing with esophageal balloon electrode catheter(balloon.catheter group,n=32),or transesophageai ventricular pacing withnormal esophageal electrode catheter(normal catheter group,n=32).Pulse width(ms),current output(mA),and the success rate werecompared between the 2 groups. Results Success rate in the balloon catheter group was 87.5%,which was significantly higher thanthat in the nodal catheter group(18.7%,X2=27.7,P<0.01).The success rate oftransesophageal ventricular pacing with esophagealballoon electrode catheter was 87.5%.The average current output(mA)was significantly lower in the balloon catheter group than thatin the normal catheter group (24.5+2.5 mA VS 53±3.4mA,P<0.05).Conclusions Esophageal balloon electrode catheter pacing couldbecome an available teclmiqBe for transesophageal emergency ventricular pacing.

Objective: To clone the Fab gene of a monoclonal antibody (mAb) BDI against human bladder cancer and its expression in E. coli. Methods: Fd and K genes of mAb BDI were cloned by RT-PCR and inserted into an Fab expression vector. Phage displaying Fab and soluble Fab were expressed in E. coli. The N-terminal sequence of VH region was corrected by PCR mediated mutagenesis. The antigen-binding characteristics of the Fab were tested by ELISA and immu-nohistochemistry. Results: Fd and K genes were cloned into the expressing vector p3MH and the phage displaying antibody and soluble Fab were expressed in E. coli, which showed weak binding activity to bladder cancer cells. Correction of the N-terminal sequence of the VHimproved the biding activity dramatically. The feasibility of the application of the Fab in phage antibody library screening was confirmed by a simulated panning procedure. Conclusion: The Fab gene of an anti-human bladder cancer mAb was expressed in E. coli. The importance of the N-terminal sequence on antibody binding activity was suggested.

Aiming to the fact that the distribution modules of the C3I simulation system originally developed based on TCP/IP protocol has to be compatible with the present HLA (High Level of Architecture) rules, three methods for implementing the interaction of HLA and TCP/IP subsystems are presented to make the original TCP/IP modules directly used in the HLA-based C3I system without redevelopment. The three methods are agent method, middleware method and gateway method. Moreover, the implementation of gateway method via VC++ as well as the key technology is discussed.

Objective To investigate the effects of medicine androgen deprivation therapy(ADT)on quality of life in patients with prostate cancer. Methods A total of 42 consecutive advanced prostate cancer patients without any other anti-androgen medications after ADT[A subcutaneous depot injection of LHRH-agonist(Zoladex) was instituted every 28 days]were enrolled.Levels of serum testosterone and prostatic specific antigen(PSA)were obtained just prior to ADT and after ADT.The general and disease-specific health-related quality of life were assessed. Results The average testosterone level was less than 50 mg/L after medical ADT in 3 weeks. And PSA level declined dramatically in one month.Although there were no significant differences on physical discomfort and limitations to daily activities.Urinary obstruction symptoms after ADT were disappeared and the size of prostate were reduced after one month. The appetite and vigor were worsened.Overall health status and sexual function were significantly reduced. Conclusion The ADT could make serum testosterone and PSA decline dramatically in short time, and worsen some general health-related quality of life.
