1.Explore the Clinical Effect of Skeletal Muscle Ultrasound in the Diagnosis of Achilles Tendon Injury

Objective Explore the clinical value of ultrasonography in the diagnosis of skeletal muscle injury in the Achilles tendon .Methods Select visit from March 2016 to March 2017 period in orthopedic hospital 56 patients with Achil-les tendon injury , using a random number table group , the experimental group using diagnostic ultrasound skeletal muscles of the control group using diagnostic methods MIR , and ultimately by surgery as a diagnostic criteria between the two groups of patients with the diagnosis .Results Diagnostic accuracy rate of the experimental group were 96.42%.Skeletal muscle ul-trasound display achilles tendon rupture 12 patients , 6 patients with chronic Achilles tendon hyperplasia , swelling of the A-chilles tendon 6 patients, the diagnostic accuracy of the control group were 92.86%.Skeletal muscle ultrasound display A-chilles tendon rupture in 10 patients, 8 patients with chronic Achilles tendon hyperplasia , swelling of the Achilles tendon 3 patients.The results between the two groups of patients was statistically significant , that is P<0.05.Conclusion Ultra-sound in the diagnosis of skeletal muscle tendon injury in a significant clinical effect , but also has an affordable price , easy to operate and less damage to human advantage .It should be introduced .
2.Expression of the VEGF and mucin MUC5AC in nasal mucosa before and after chronic rhinitis - sinusitis and nasal polyps - endoscopic sinus surgery

Objective To investigate the expression of the vascular endothelial growth factor (VEGF) and mucin MUC5AC in nasal mucosa before and after chronic rhinitis - sinusitis and nasal polyps - endoscopic sinus surgery.Methods 75 cases chronic rhinitis - sinusitis and nasal polyps - endoscopic sinus surgery patients were selected as nasal polyps group and 75 cases of nasal bone fracture or epistaxis patients as the control group from January 2012 to January 2015. Took the samples of nasal polyps before surgery and the maxillary sinus mucosa specimens after surgery six weeks of nasal polyps’ patients and on the edge of the inferior turbinate mucosa specimens of the control group to detect eosinophil count by HE staining, and detect the expression of VEGF and mucin MUC5AC by immunohistochemical staining.Results The specimens eosinophils of preoperative nasal polyp group and postoperative nasal polyp group were higher than that of control group (P < 0.05), the nasal eosinophils of postoperative nasal polyp group was lower than that of preoperative nasal polyp group (P < 0.05). The expression of specimens VEGF and mucin MUC5AC area percentages in preoperative nasal polyp group and postoperative nasal polyp group were higher than that in control group (P < 0.05), the expression of nasal VEGF and mucin MUC5AC area percentages in the postoperative nasal polyp group was lower than that of preoperative nasal polyp group (P< 0.05).Conclusion Eosinophil count and the expression levels of VEGF and mucin MUC5AC of nasal mucosa in chronic rhinitis - sinusitis and nasal polyps - endoscopic sinus surgery preoperative are higher, and reduce at postoperative six weeks, VEGF and mucin MUC5AC may be involved nasal repair.
3.Design of sphygmomanometer detecting worktable and its clinical application

To design a sphygmomanometer detecting worktable to prevent the leaked mercury from flowing or doing harm to the staff after volatilization. The worktable was composed of a body, an exhaust hood, a flow-guided groove, a base, a flow-guided hole, a collecting bin, a ventilating fan and etc. A filter screen at the ventilating fan was equipped with the sulphur powder to react with the mercury vapor in the air. The worktable could reduce mercury pollution and its damage to the staff. The worktable facilitates the maintenance and detection of the sphygmomanometer, and thus is worthy popularizing clinically.

5.Study on the correlation between methylation of APC genes and expression in pancreatic cancer

Objective To investigate the effect of methylation of the APC gene on expression and the correlation with clinical data in pancreatic cancer.Methods Sixty postoperative tissue samples with pancreatic cancer were collected in the First Affiliated Hospital of Zhengzhou University from August 2010 to January 2011,20 benign pancreatic disease tissues were collected as control groups.APC promoter methylation and gene expression levels were detected by Methylation Specific PCR (MSP),Real Time PCR (RT-PCR) and Western blot in 60 pancreatic carcinoma,42 metastasis and 20 benign pancreatic disease tissues,then analyze the relation between methylation of the APC gene and the clinical data.Results APC promoter methlation was observed 48.53%,46.67% and 1.16% in pancreatic carcinoma,metastasis and benign pancreatic disease tissue,respectively.Methylation of APC in pancreatic carcinoma and metastasis increased significantly compared with control tissues (x2 =12.903,14.402; P < 0.05).There were no statistically significant differences of APC expression in these tissues (P > 0.05).There was a significant correlation between methylation of APC and clinicopathological stage (x2 =6.801,P < 0.05),but no correlation with gender,age,tumor size,histological grade and metastasis (x2 =0.727,1.311,0.372,0.148,0.017 ; P > 0.05).Conclusion The methylation of APC gene is closely related with pancreatic carcinoma inogenesis and the clinicopathological stage,but do not effect the expression of APC in tissues.
6.Diagnosis of intracranial dermoid cyst with spiral CT

Objective To explore diagnosis value of spiral CT on intracranial dermoid cyst based on retrospective analysis.Methods 10 cases of intracranial dermoid cyst were enrolled in and confirmed by surgery and pathology.The retrospective analysis was based on the CT manifestations,causes,pathological and clinical features.Results Among the 10 cases,5 lesions were located in the posterior cranial fossa,3 in besides saddle,1 in up saddle and 1 in the temporal fossa.The morphology of nidus was round or round like with clear boundary.There was no edema around the nidus.Fat was found in 7 cases appeared hypodensity with insufficiency uniformity on CT,and the density was lower than that of cerebrospinal fluid with CT value ranged from-6 HU to 80 HU.There was a few calcification on the edge in 1 case.Hyperdensity irregular and lumpy hair-liked shadow was found in 2 hypodensity cases.1 lesion closed to the cranial plate was compressed thin and buckled.Conclusions Intracranial dermoid cyst has typical CT manifestations.It will be an accurate diagnosis based on clinical analysis and CT manifestations.
7.Evaluation of mycolic acid analysis in identification of Mycobacterium species through SMIS

Objective To evaluate the usefulness of mycolic acid for identification of Mycobacterium species using SMIS. Methods One hundred and eighteen clinical Mycobacterium isolates collected from Beijing Tuberculosis and Thoracic Tumor Research Institute through whole year of 2007 were analyzed. The 118 isolates contain 25 isolates of Mycobacterium tuberculosis and 93 non tuberculosis Mycobacterium identified by PNB method. Mycolic acid analysis using SMIS is evaluated for identification of a broad range of Mycobacteria in comparison with 16S rDNA , 16-23S rDNA ITS sequencing to measure the concordance rate and agreement, and verify the concordance rate and agreement among results of mycolic acid, sequencing and PNB in identifying Mycobacterium tuberculosis and non tuberculosis Mycobacterium. Results The concordance rate between mycolic acid method analysis and DNA sequencing is 92% ( 108/118), of which concordance rate in Mycobacterium tuberculosis complex and non tuberculosis Mycobacterium are 95% (35/37) and 90% (73/81) respectively, agreement of both is great( agreement Kappa value is 0. 96). Through retrospective analysis, the concordance of results between SMIS and PNB method analysis is 90% (106/118)and agreement is well( agreement Kappa value is 0. 73 ), the concordance of results between sequencing and PNB method analysis is also 90% ( 106/118 ) and agreement is well (agreement Kappa value is 0. 74 ),despite the identification results of 11 isolates by PNB method are discordant. Conclusion Mycolic acid analysis by SMIS enables rapid identification of a broad range of clinical Mycobacterium species, which could play an important role in polyphasic identification of Mycobacterium species.
8.Effects of thyroid hormone receptor β△ on apoptosis and proliferation of hepatoma RH-35 cells

Objective To study the effects of TRβ△ on apoptosis and proliferation of liver cancer cell line RH-35 from rat in vitro. Methods RH-35 cells were transfected by empty vector pcDNA3. 1 and expression plasmid pcDNA3. 1-TRβ△, then exposure to 10 nmol/ L T3 . RH-35 cells apoptosis and proliferation were observed by flow cytometry and MTT colorimetric assay; Levels of catenin β-1(CTNNB1), senescence marker protein-30(SMP-30) and BCL2-antagonist/ killer ( BAK ) mRNA evaluation were detected by quantitative real-time RT-PCR (RT-qPCR). Results In the presence of T3 , overexpression of TRβ△ significantly inhibited the proliferation, increased the percentage of apoptotic, down-regulated CTNNB1and SMP-30 expression, up-regulated BAK expression in RH-35 cells( P < 0. 05). Conclusion TRβ△ could inhibit the proliferation of RH-35 cells and promote their apoptosis, which may be related to upregulation of BAK genes expression and downregulation of CTNNB1 and SMP-30 gene expression, and these effects could be regulated by T3 .
9.Nonclinical safety evaluation of Insulin NPH, a biosimilar of Humulin NPH

Objective To investigate the toxic reaction,toxic organs or target tissues of protamine recombinant human insulin (Insulin NPH),and provide basis for clinical trials by single dose toxicity test in mice,repeated toxicity and immunogenicity of Beagle's dogs,and systemic active allergy in guinea pig.Methods ① Using maximum dose method,mice in single dose toxicity test were sc injected with normal saline (NS),vehicle,and Insulin NPH (2092-2488 IU/kg),the toxic reactions after injection were monitored.② In repeated toxicity study,Beagle's dogs were sc administrated with vehicle,the original (Humulin NPH,1.5 IU/kg)and different doses of Insulin NPH (0.5,1.0 and 1.5 IU/kg) for 30 d continuously,followed by a 14-d recovery.During the administration and recovery period,general observation,local irritation,body weight,anus temperature,blood glucose,and electrocardiogram (ECG) were checked,moreover,hematology,serum biochemistry and urine were detected.Also,organic weights and histopathological examination were conducted.Binding antibodies in dog serum were measured by indirect ELISA method in immunogenicity test.③ In systemic active allergy study,cavies were sc injected with low-and high-dose (4 and 12 IU/kg) Insulin NPH,normal saline and vehicle.Besides,ova as positive control was also included.After five times of sensitization test with above doses,the excitation reactions of iv injection with tripled sensitizing doses were observed.Results No obvious toxicity was observed in mice after injected with 165 times of usual clinical dose of Insulin NPH.Repeated toxicity study of Beagle's dogs revealed that 1.0 IU/kg was the no-toxic-effect dose (NOAEL) for Insulin NPH,which was equivalent to 2 times of clinical dose.No bindingantibodies were found in immunogenicity test.There was no obvious allergic symptom in the active systemic allergy study of guinea pig.Conclusion Under the experimental conditions,no serious toxicity of Insulin NPH is found.
10.Osteoblast is necessary for IGF-I to promote bone resorption by osteoclast

Objective To study whether osteoblast is necessary for IGF-Ⅰ to promote bone resorption by osteoclast.Methods Mouse MC3T3 osteoblast cells and mature osteoclasts induced by RANKL were cultured in vitro.These osteoblasts and osteoclasts were subjected to treatment with recombinant human insulin-like growth factor-1 (rhIGF-Ⅰ),and the activation of IGF-Ⅰ receptor was verified by Western blotting.Thereafter,osteoclasts were cultured individually or co-cultured with osteoblast,in the absence or presence of rhIGF-Ⅰ.Osteoclast proliferation and apoptosis were observed by MTT colorimetric assay and flow cytometry.Cathepsin K gene expression was detected by real-time PCR; bone adsorption activity of osteoclast was determined by resorption pits formation on calf cortex slice with toluidine blue staining.Results Western blotting result confirmed that rhIGF-Ⅰ could effectively activate IGF-Ⅰ receptors either in osteoblast or osteoclast.In co-cultured group,in the presence of rhIGF-Ⅰ osteoclast showed inhibited apoptosis,enhanced proliferation and up-regulated cathepsin K expression (P < 0.05).The functional experiment revealed that osteoclasts collected from IGF-Ⅰ treated co-cultured group resulted in more resorption pits formation (P < 0.05); rhIGF-Ⅰ did not show any significant effect on the individually cultured osteoclasts.Conclusion Osteoblast is necessary for osteoclast induced bone resorption resulting from IGF-Ⅰ treatment.
