Objective To investigate the spatiotemporal expression patterns and the relationship of ?-sarcomeric actin(?-SCA),?-smooth muscle actin(?-SMA) and intermediate filament protein desmin with the maturation of the prenatal and the neonatal mouse hearts.Methods Serial sections of the embryo mouse and the neonatal mouse hearts were immunostained with antibodies against ?-SCA,?-SMA and desmin.Results Ventricle and outflow tract of embryonic day(ED) 9 heart showed stronger expression of ?-SCA and ?-SMA,but desmin expression was lower.In the atrium,the expressions of ?-SCA and ?-SMA were restricted to the dorsal and ventral walls.In the sinus venosus,only a few weakly stained ?-SCA positive cells were detected.No desmin expression was found in the atrium and sinus venosus.The expressions of ?-SCA,?-SMA and desmin were increased to their highest level at ED 12.The higher expression of ?-SCA remained to the postnatal stages.After ED 12,the expressions of ?-SMA and desmin gradually decreased in different parts of the heart,but their expressions in the right ventricle persisted longer.After birth,desmin expression was mainly concentrated in the Z lines of I bands and intercalated disks.Conclusion The presence of spatiotemporal differences in the expression of ?-SMA and desmin reveals regional differences in cardiomyocyte maturation in various parts of the embryonic mouse heart.The right ventricle shows a relatively slow pace of maturation.The ?-SMA may contribute to a peristaltoid contraction pattern of the embryonic myocardium with a slow shortening speed,and a relatively higher level of desmin is required for the maturation of the sarcomere.

Sal-like protein 4 (SALL4), a newly discovered oncogenes, plays an important role in the formation of embryo, maintenance of embryonic stem cell pluripotency and self-renewal. SALL4 not only has a close relationship with hereditary disease, benign disease, malignant tumor, blood system tumor and germ cell tumors, but also chemoresistance and poor prognosis. SALL4 obtains biological functions by adjusting different signaling pathways, transcription factors and epigenetic modulation. Further mechanism study of SALL4 and its relationship with various diseases will provide new opportunities for the treatment of diseases.

Objective To develop a method for the direct determination of eight life elements (Ca, Mg, K, Al, P, Cu, Zn, Fe ) in serum by ICP-AES. Methods The serum samples were determined by ICP-AES after diluted in 0.1% Triton X-100. Results The RSD of the method were 2.36%-9.46% and the recovery rates were 87.3%-116.5%. Conclusion This method was quick, simple and accuracy, which is suitable for direct determination of life elements in serum samples at one time.

Eighteen cases(20 ears) of sudden Sensorineural Hearing Loss in children had been reviewed. The results showed that the course of disease was longer before the patients receieved treatment. There may be a relationship between sudden Sensorineural Hearing Loss in children and viral infection of upper respiratory or parotitis. The effect of treatment was similar to adults. DF-521 was effective and safe for sudden Sensorineural Hearing Loss in children.

Objective:To observe the curative effect of modified amniotic membrane transplantation for agricultural pterygium.Methods: A total of 276 cases (276 eyes) with early pterygium were randomized divided into traditional group (138 cases) and modified group (138 cases) as random table. The traditional group was treated with pterygium excision in combination with autologous conjunctival transplantation; the modified group was treated with modified amniotic membrane transplantation; to observe the curative effect of the two groups.Results: After all of patients were followed up for 1 year, 122 cases were cured and 16 cases were recurrence, and the recovery rate was 88.41% in traditional group; 131 cases were cured and only 7 cases were recurrence, and the recovery rate was 94.93% in modified group and was better than traditional group; the difference between the two groups was statistical significant (x2=5.02,P<0.05).Conclusion: The modified amniotic membrane transplantation method is simple in operation, mini invasive for eye tissue, and can retain conjunctival tissue in utmost; it also can improve success rate of surgery and be an effective mean for treatment of pterygium.

Objective To study the relationship between land use/cover structure and changes and risk of plague prevalence.Methods Chifeng,Aohan,Alukeerqin and Wengniute were selected as typical areas of Spermophilus Dauricus plague foci in Inner Mongolia,density of Spermophilus Dauricus,plague epidemic and land use data in 1986,1995,2000 were collected,and risk of plague prevalence was represented by the density of Spermophilus Dauricus and epizooty of Spermophilus Dauricus.The percentage of land type structure,change degree of integrated land cover,division index of grassland for plague suitable habitat were calculated with ArcGIS 9.3.The relationship between the land use,cover change and the plague epidemic were analyzed by the methods of comparative.Results From 1982 to 2000,the density of Spermophilus Dauricus was lower than 0.4/hm2 and no epizooty of Spermophilus Dauricus was observed both in Aohan Banner and Chifeng City,which indicated low risk of plague prevalence.Contrarily,from 1982 to 2000,the density of Spermophilus Dauricus was higher than 1.0/hm2,and epizooty of Spermophilus Dauricus was prevalent in Alukerqin Banner,which implied high risk of plague prevalence.In Wenguiute Banner,the density of Spermophilus Dauricus was lower than 0.3/hm2 from 1982 to 1990,after then showed a rising trend,reached higher than 1.0/hm2 in 1991,1995-1997,and epizooty of Spermophilus Dauricus was also monitored,which denoted high risk of plague prevalence;In 1986,1995 and 2000,the proportions of grassland of Wengniute Banner,Alukerqinqi Banner (51.36％,49.14％,48.28％;56.97％,57.48％,57.44％) were much higher than Chifeng and Aohan Banner (35.99％,33.24％,32.95％;38.94％,34.46％,31.31％),but those of arable land (16.81％,18.78％,20.22％;13.28％,14.92％,15.15％) were lower than Chifeng and Aohan Banner (39.73％,42.25％,42.67％;41.21％,43.99％,49.53％).The highest of comprehensively dynamic degree of land cover changes appeared in Aohan Banner,and it was 0.53 and 1.11 in 1986 vs.1995 and 1995 vs.2000,respectively,and the lowest in Alukeerqin Banner,it was 0.22 and 0.05 during those periods.The grassland division index in Alukeerqin Banner was the lowest,which was 0.29 in 1986,0.28 in 1995,and 0.29 in 2000.The grassland division index was higher and showed a upward trends in Aohan Banner,which was 0.57 in 1986,0.69 in 1995 and 0.71 in 2000.Conclusion There is a certain relation between risk of plague prevalence and land use/cover structure and changes.

[Objective]To investigate the clinical effect of Chinese herbal medicine washout combined with lactic acid bacteria vaginal capsule on refractory vaginitis. [Method] Divide 500 patients into 3 types: downward flow of damp-heat, damp toxin implication, internal heat due of Yin deficiency, which treated with different decoctions with lactic acid bacteria vaginal capsule. Fol ow up for 3m after treatment course, observe recurrence; make vaginal secre-tion tests before and after treatment. [Result] In downward flow of damp heat type, 139 cases were cured, 11 effective, the total effective rate 100%; on damp toxin implication type, 183 cases were cured, 13 effective, total effective rate 100%;for internal heat due to Yin deficiency, 141 were cured, 9 effec-tive and total effective rate 100%. There’s no recurrence after 3m. [Conclusion] The said method can effectively regulate vaginal micro-ecological envi-ronment and reduce recurrence of refractory vaginitis, with safe clinical application and definite cure effect.

: ObjectiveTo study the effect of acupuncture on resistin and adiponectin and its gene expression in type 2 diabetic rats.MethodsThe rats of type 2 diabetes model were randomly divided into acupuncture group, rosiglitazone group and model control group, meanwhile 8 normal rats were assigned as the normal control group. The changes of total cholesterol(TC), triglycerides(TG), high density lipoprotein-cholesterol(HDL-C) and fasting plasma glucose (FPG) were measured. The expression of resistin gene and adiponectin gene in the adipose tissue was determined with RT-PCR. The level of serum resistin and adiponectin in type 2 diabetes rats were observed before and after acupuncture.ResultsThe levels of TC, TG, FPG resistin lowered obviously, and the levels of HDL-C adiponectin raised significantly. The expression of resistin gene lowered obviously. The expression of adiponectin gene significantly raised in acupuncture group, compared with the model control(P＜0.01, P＜0.05).ConclusionAcupuncture can decrease resistin gene expression, raise adiponectin gene expression, accordingly release insulin resistance.

Objective To study the protective effects of choline on myocardial ischemia rat heart and its potential mecha -nisms .Methods Ischemia hypoxia environment was simulated with low value of pH (pH 6 .8) and lack of oxygen .Calcium currents were recorded by whole cell patch under the voltage clamp configuration .The alternations in[Ca2 + ]induced by KCl was detected by laser scanning confocal microscope in ventricular myocytes ,then disccuss the effects of choline on calcium and calcium store in cells . Results The normalized peak currents of ICa-L in ventricular myocytes were larger in pH 6 .8 group than those in pH 7 .4 group , which can be attenuated by choline .The(Ca2 + )i induced by KCl in ventricular myocytes were significantly increased in pH 6 .8 Ty-rode solution and its increasing can be suppressed by choline .4-DAMP can inhibit the suppressing effect of choline .Conclusion The possible mechanism of M 3 receptor involved in the protection of ischemic myocardium by inhibiting myocardial cells in ICa-L ,in-hibiting intracellular calcium overload .

Objective To investigate the effect of methylation of the APC gene on expression and the correlation with clinical data in pancreatic cancer.Methods Sixty postoperative tissue samples with pancreatic cancer were collected in the First Affiliated Hospital of Zhengzhou University from August 2010 to January 2011,20 benign pancreatic disease tissues were collected as control groups.APC promoter methylation and gene expression levels were detected by Methylation Specific PCR (MSP),Real Time PCR (RT-PCR) and Western blot in 60 pancreatic carcinoma,42 metastasis and 20 benign pancreatic disease tissues,then analyze the relation between methylation of the APC gene and the clinical data.Results APC promoter methlation was observed 48.53％,46.67％ and 1.16％ in pancreatic carcinoma,metastasis and benign pancreatic disease tissue,respectively.Methylation of APC in pancreatic carcinoma and metastasis increased significantly compared with control tissues (x2 =12.903,14.402; P ＜ 0.05).There were no statistically significant differences of APC expression in these tissues (P ＞ 0.05).There was a significant correlation between methylation of APC and clinicopathological stage (x2 =6.801,P ＜ 0.05),but no correlation with gender,age,tumor size,histological grade and metastasis (x2 =0.727,1.311,0.372,0.148,0.017 ; P ＞ 0.05).Conclusion The methylation of APC gene is closely related with pancreatic carcinoma inogenesis and the clinicopathological stage,but do not effect the expression of APC in tissues.
