1.Effects of exercise training on bone histomorphometry of ovariectomized rats

ObjectiveTo evaluate the effects of exercise training on bone histomorphometry of ovariectomized rats.Methods46 SD rats were randomly divided into the normal control group (group A), ovariectomized group (group B) , estrogen supplement group (group C) and exercise training group (group D). The rats of group A had a sham operation, while that of other groups was ovariectomized. 3 months after operation, the rats had been treated for 3 months. The rats of group C were given estradiol, and the rats of group D were trained by exercise. The tibia sections were studied by histomorphometric analysis.ResultsThe values of the trabeclar area, trabeclar area%, trabeclar perimeter and trabeclar number of group D were significantly higher than that of group B. The values of the trabeclar separation, osteoclast number/mm2 and osteoclast number/mm of group D were the least in all groups, which had been ovariectomized. The values of the labeled perimeter % mineral apposition rate, bone formation rate of group D were superior to that of all groups.ConclusionThe exercise training can increase bone mass of ovariectomized rats, which had double effects of promoting bone formation and restraining bone absorption.
2.Analgesia of Femoral Nerve Block on Knee Stiffness in Rehabilitation Training after Surgery

ObjectiveTo compare the effect of rehabilitation training after surgical intervention of knee stiffness (SIKS) between femoral nerve block (FNB) and epidural nerve block (ENB).Methods60 patients undergoing SIKS at one knee joint were randomly assigned to two groups: 40 patients accepting a catheter for FNB in group A and 20 patients accepting a catheter for ENB in group B. All patients participated in passive rehabilitation therapy (PRT) and active rehabilitation therapy (ART) at 12 hours, 24 hours, 36 hours, and 48 hours after operation. 10 ml of lidocaine were applied via these catheters before rehabilitation therapy. The visual analogue score (VAS) for pain in PRT, the time needed from the end of PRT to the start of ART, and the incidence of side effects such as hypotension or nausea were recorded.ResultsThere was no significant difference in VAS between groups A and B. The time needed from the end of PRT to the start of ART in group A was significantly less than that of group B. The incidence of hypotension or nausea in group A was significantly less than those of group B.ConclusionFNB provides the same analgesic effect as ENB, has less time needed from the end of PRT to the start of ART, and lowers incidence of hypotension or nausea.
3.Effects of gamma-interferon gene on brain gliomas and survival of tumor-carrying rats

BACKGROUND: It has become a hot issue to treat malignant tumors with retroviral mediated cytokines. But it remains a controversy whether retroviral mediated γ-interferon(IFN-γ) is valid in killing glioma cells and prolonging the survival of tumor carriers.OBJECTIVE: To observe the effect of retrovital mediated γ-interferon on the growth of glioma and the survival of tumor-carrying rats.DESIGN: Completely ramdomized trial based on animals.SETTING: Neurosurgery department of a hospital affiliated to a university.MATERIALS: The trial was conducted in the Neurosurgery Institute of Harbin Medical Unicersity from July 2002 to July 2004. The subjects were 60 male Wistar rats provided by the Animal Center of the First Affiliated Hospital of Harbin Medical Unicersity. They were randomly assigned into 3groups: C6 control group, C6/PA317neo control group and C6/PA317IFN-γ treatment group, 20 in each group.INTERVENTIONS: Retroviral vector containing murine IFN-γ gene was transferred into retroviral package cell PA317. A high titer PA317IFN-γ cell line was obtained after G418 resistance screening. In the right basal ganglion area 1 × 106 C6 cells was injected in each of the rats. The rats in C6groups received no more intervention. Those in C6/PA317neo control group received injection of 1 × 106 PA317IFN-γ cells in the right basal ganglion 4days after C6 injection. Those in C6/PA317IFN-γ treatment group received injection of 1 × 106 PA317IFN-γ cells in the right basal ganglion 4 days after selected from each group to be kill, and the brains were taken out to make frozen sections for SABC immunohistochemical staining. The CD4 and CD8 T was measured with imaging processing software under microscope.RESULTS: The mean survival time of rats in C6 and C6/PA317neo control groups was(27.10 ± 0.82) days and (26.60 ± 0.93) days respectively( P > 0.05), while that in treatment group was(49.30 ± 4. 17) days and there were five rats outlived beyond 60 days( P < 0.01 vs the two controls). The tumor volume in the two control groups was of no difference but the tumor volume in treatment was significantly smaller than that in control groups( P < 0. 01 ) . The CD4 and CD8 lymphocytes infiltration in treatment group was more than that in control groups( P < 0.01) and there was no difference between the two controls( P > 0.05) . It was also revealed through immunohistochemical examination that in treatment group there was Brdu positive cells on the 7th day and they disappeared on the 16th day. But there was no Brdu positive cells in the control groups.CONCLUSION: Intratumor injection of PA317IFN-γ induces anti-tumor immunal reaction in rats. Retroviral mediated IFN-γ inhibits tumor growth and prolongs rat survival.
4.The effects of up-regulation of nuclear Clusterin gene on the biological behaviors of A549 cells

Objective:To observe the up-regulation of nuclear Clusterin (nCLU)gene on the biological behaviors of human non-small cell lung cancer cell line A549 .Methods:Sense eukaryotic expression vector of nCLU was constructed by cloning the cDNA of nCLU into pIREShyg3 vector. A549 cells were transfected with pIRES-nCLU and pIREShyg3 vectors by lipofectAMINE~(TM) 2000 mediation, respectively. Stable transfected cells were selected by hygromycin B screening. CCK-8 assay was performed to evaluate the effect of nCLU over-expression on cell proliferation in vitro. The expression level of nGLU protein was examined by Western blotting. Cell cycle distribution was detected by FCM with PI staining. The alteration of migration and metastasis potential of A549 cells before and after nCLU gene transfection were assayed by cell chemotactic migration and invasion test. Results:The proliferation speed of the transfected A549 cell clones stably over-expressing nCLU was slowed down. FCM analyses revealed that the percentage of cells in G_0/G_1 phase dramatically increased from (33.54±2.10)% to (63.31±4.30)%. The cell chemotactic migration and invasion potentials were markedly reduced after nCLU gene transfection (P<0.05). Conclusion:Up-regulation of nCLU can greatly inhibited the proliferation and decreased the migration and invasion capabilities of A549 cells.
5.CT Differential Diagnosis of Thyroid Neoplasm

Objective To investigate the value of CT scans in differential diagnosis of thyroid neoplasm.Methods CT findings ofthyroid neoplasm in 29 cases including 8 cases thyroid adenoma,4 cases nodular goiter and 17 cases thyroid carcinoma identified by operation and pathology were analyzed retrospectively.Results The contour of thyroid adenoma was distinct,a completely enhanced ring in 5 cases and enhanced nodules in 3 cases were showed on contrast-enhanced scan.4 cases of nodular goiter showed homogenous cystic low density the lesions were localized and distinct in contour,the cystic wall could be seen.Thyroid carcinoma appeared unhomogenous density,the edges contour of lesion was not distinct and infiltration on adjacent tissue might be seen.Conclusion CT scan might play an important role in diagnosis and differential diagnosis of thyroid neoplasm.
6.Effect of Neostigmine on Cardiovascular Response from Neuromuscular Electrical Stimulation in Selective Posterior Rhizotomy for Patients with Cerebral Palsy

Objective To observe the effect of intravenous pre-injection of neostigmine on cardiovascular response(CVR) caused by neuromuscular electrical stimulation(NMES) in selective posterior rhizotomy(SPR) for patients with cerebral palsy.Methods 56 patients with cerebral palsy undergoing SPR at lumbarsacral segments under general anesthesia were randomly assigned to 2 groups: intravenous neostigmine 0.04 mg/kg(no more than 1 mg in total) and intravenous atropine 0.02 mg/kg(no more than 0.5 mg in total) 5 min before NMES in group N,and intravenous normal saline 0.12 ml/kg instead in group C.The systolic blood pressure(SBP) and heart rate(HR) at following time points: before skin incision(T1),before intravenous neostigmine/atropine or normal saline injection(T2),1 min after NMES(T3),and 10 min after NMES(T4).Results SBP and HR at T3 in group N were significantly lower than those in group C(P<0.01).No significant difference was found at T1,T2 and T4 between two groups(P>0.05).Cases who needed extra dose of fentanyl during NMES in group N were significantly lower than those in group C(P<0.01).Conclusion Intravenous pre-injection of neostigmine before NMES can effectively decrease the CVR caused by NMES.
7.Effect of Ropivacaine in Infiltration Anesthesia for Postoperative Analgesia on Patients with Spinal Cord Injury

Objective To observe the effect of ropivacaine in infiltration anesthesia before the end of surgical procedures for postoperative analgesia on patients with spinal cord injury. Methods 60 patients with spinal cord injury undergoing internal fixation of spine fractures under combined general anesthesia were assigned in 3 groups, who received 0.25% ropivacaine in infiltration anesthesia (group RL, n=20),0.5% ropivacaine in infiltration anesthesia (group RM, n=20), or no infiltration anesthesia (group C, n=20) before the end of surgical procedures.The time for post-anesthesia recovery (PART), agitation score (AS), and Visual Analogue Score (VAS) for pain 2 h, 6 h, 24 h, and 48 h post operation were recorded, and related side effects were observed. Results AS within 6 h postoperation was significantly less in group RL and RM than in group C (P<0.05), as well as VAS, PART, mean of arterial pressure and heart rate 2 h, 6 h, and 24 h post operative (P<0.05).There was no significant difference in side effects including postoperative nausea and vomiting, and pruritus among these groups (P>0.05). Conclusion Ropivacaine in infiltration anesthesia would provide better postoperative analgesia, decrease the incidence of postoperative agitation, and increase the safety of patients with spinal cord injury in the postoperative period.
8.Postoperative Analgesic Effects of Flurbiprofen Axetil Applied Preoperatively on Patients with Spinal Cord Injury

Objective To observe the post-operative analgesic effect of flurbiprofen Axetil applied preoperatively on patients with spinal cord injury(SCI).Methods46 SCI patients undergoing spine surgery under general anesthesia were randomly divided into the flurbiprofen axetil group(group F)and control group(group C)with 23 cases in each group.Before cutting skin,flurbiprofen axetil(1 mg/kg)was injected intravenously in the group F,and normal saline(1 ml/kg)was infected intravenously in the group C.The scores of Visual Analogue Scale(VAS)for pain at postoperative 2,4,8,24,and 48 hours,and the incidence of side effects within 48 hours after operation were recorded.ResultsVAS scores at the time of 2,4,8,and 24 h after operation in the group F were significantly lower than that in the group C(P<0.05).But VAS scores of two groups at 48 h were not significantly different(P>0.05).The incidences of side effects of two groups were also not significantly different(P>0.05).ConclusionFlurbiprofen axetil applied preoperatively can release pain of SCI patients after spine surgery,and does not increase the incidence of side effects.
9.Comparison of Total Intravenous Anesthesia and Combined Inhalation and Intravenous Anesthesia in Lower Limbs Orthopedics for Patients with Cerebral Palsy

Objective To compare the characteristics of total intravenous anesthesia (TIA) with manual assisted ventilation and combined inhalation and intravenous anesthesia (CIIA) with mechanical ventilation in lower limbs orthopedics for patients with cerebral palsy.Methods 50 patients with cerebral palsy undergoing scheduled lower limbs orthopedics were randomly divided into the TIA group and CIIA group with 25 cases in each group. The patients of the TIA group were treated with manual assisted ventilation. Those of the CIIA group were treated with fast induction, endotracheal intubation, inhalated and intravenous anesthetics maintaining the anesthesia and mechanical ventilation. The recovery time after operation and post-operative side effects including nausea, vomiting, and psychomotor agitation of all patients were observed.Results The recovery time of the patients in the CIIA group was significantly longer than that in the TIA group ( P<0.01), and post-operative psychomotor agitation was significantly higher than that in the TIA group ( P<0.05). However, there was no significant difference in nausea or vomiting found between two groups ( P>0.05).Conclusion Patients treated with TIA is easier to recovery and has fewer side effects than those treated with CIIA.
10.Change of glucose in plasma in selective posterior rhizotomy at lumbar and sacral regions on cerebral palsy children

Objective To investigate the carbohydrate metabolism during selective posterior rhizotomy (PCR) at lumbar and sacral regions on cerebral palsy children (CPC).Methods62 CPC who received SPR at lumbar and sacral regions under combined intravenous and inhalation anesthesia were tested with the level of glucose (Glu) in plasma at different time during the operation.Results and ConclusionThere is no significant difference of Glu at different time during the operation, however, the Glu level shows an elevating trend at 3 and 10 minutes after posterior root electric stimulation.
