Journal of Clinical Hepatology 2015;31(5):787-789
Pancreatic encephalopathy (PE)is one of the severe complications of severe acute pancreatitis (SAP).Early diagnosis mostly depends on the history of disease as well as clinical symptoms and signs.PE progresses rapidly and is often complicated by multiple organ dysfunction, and it may finally develop into multiple organ failure with a high fatality rate if not treated in time.It is currently known that de-myelination is one of the important pathological features of this disease, with fat -soluble demyelination of cerebral gray matter and white matter,as well as inflammatory changes such as hemorrhage and edema. The target antigen of demyelinating lesions, however,is myelin basic protein (MBP).This paper reviews the changes in MBP levels in the demyelinating lesions of the central nervous system among PE pa-tients,with the purpose of providing clues for the early diagnosis and prognostic study of demyelinating lesions in PE.

Objective:To explore the inhibition of microRNA (miRNA, miR)-5590-3p on the expression of transforming growth factor beta typeⅡreceptor (TGFBR2) gene and its effect on the invasion and proliferation of gastric cancer cell line HS-746T.Methods:The gastric cancer cell line was cultured in vitro. Real-time quantitative polymerase chain reaction (qRT-PCR) was used to analyze the expression of miR-5590-3p in gastric cancer tissues and cell lines. The miR-NC and miR-5590-3p mimic were transfected into gastric cancer cell line HS-746T by lipofection, and named as miR-NC group and miR-5590-3p group, respectively. qRT-PCR was used to measure transfection effects. Transwell assay and cell counting kit-8 (CCK-8) assay were used to detect cell invasion and proliferation after transfection. The bioinformatics software predicts and the dual luciferase reporter gene system validates the target gene of miR-5590-3p. qRT-PCR and western blot were used to measure the expression of TGFBR2 and its downstream proteins in the transfected cells. Results:The expression of miR-5590-3p in gastric cancer tissues was significantly lower than that in adjacent tissues ( P<0.01). The expression of miR-5590-3p in gastric cancer cell lines was significantly lower than that in normal gastric mucosal epithelial cells ( P<0.05), and lowest in HS-746T cells ( P<0.01). After transfection, the expression of miR-5590-3p in miR-5590-3p group (11.76±0.21) was significantly higher than that in miR-NC group (1.06±0.21), with statistically significant difference ( P<0.01). The number of invasive cells in miR-NC group and miR-5590-3p group were (101.20±15.47) and (26.53±6.53), respectively, and the invasion ability of miR-5590-3p group was significantly decreased ( P<0.01). Compared with the miR-NC group, the cell proliferation ability of the miR-5590-3p group was significantly decreased ( P<0.05). Bioinformatics software showed that the target gene for miR-5590-3p is TGFBR2. The dual luciferase reporter gene system confirmed that miR-5590-3p can target the TGFBR2 gene ( P<0.01). Western blot results showed that compared with miR-NC group, the expression of TGFBR2 in HS-746T cells in miR-5590-3p group was significantly decreased ( P<0.01), the expression of ZEB-1 and ZEB-2, and the expression of CDK1 and cyclin B proteins were decreased as well. Conclusions:miR-5590-3p can inhibit the invasion and proliferation of gastric cancer cell HS-746T by targeting and regulating the expression of TGFBR2 gene.

Journal of Huazhong University of Science and Technology (Medical Sciences) 2008;28(6):683-7
In this study, in vitro chemosensitivity testing was conducted on primary cultured breast cancer cells from 96 patients with breast cancer, and the results showed that the cells from a few patients with primary breast cancer developed multidrug resistance (MDR) prior to the first chemotherapy exposure. All the cells from the recurrent cancer patients had MDR. The findings suggested that patients having MDR would benefit from high-dose chemotherapy (HDC) regimens. In vitro chemosensitivity screening, which was aimed at improving the therapeutic efficacy and minimizing side effects, helps in choosing individualized treatment for breast cancer.

Chinese Journal of Clinical and Experimental Pathology 2017;33(1):63-67
Purpose To investigate the expression of programmed death ligand-1 (PD-L1) in breast cancer tumor cells and stromal tumor-infiltrating lymphocyte (sTIL),and to study the relationship between the expression of PD-L1 and the clinicopathological characteristics of the patients.Method The protein expression of PD-L1 was detected by immunohistochemistry of EliVision two-step method in 68 cases of non special type of invasive breast cancer, and the relationship between the expression of PD-L1 protein and the immunohistochemistry subtypes and clinical parameters was analyzed. Results The total expression rate of PD-L1 was 35.3% in breast tumor tissue, specially in triple negative breast cancer (TNBC) which occupy the highest positive rate. The expression rates of PD-L1 in tumor tissue of the luminal subtype,HER-2 over-expression subtype and TNBC subtype were 16.1%,37.5% and 61.9% respectively, and the difference was statistically significant. The total expression rate of PD-L1 in sTIL was 51.5%,and the highest expression rate was 81.0% in TNBC.There were significant differences of PD-L1 expression in sTIL of the luminal subtype,HER-2 over-expression subtype and TNBC subtype.Expression of PD-L1 in tumor tissue and sTIL had a significant positive correlation. Conclusion PD-L1 expressed in TNBC was significantly higher than other types of breast cancer, which suggest the blocking of signal pathway of PD-1/PD-L1 may expected to become a new immunotherapy for breast cancer, especially for TNBC subtype.
