1.STUDY ON THE DETECTION OF TOXOPLASMACIRCULATING ANTIGENS USING IMMUNO-PCR

Aim to explore an assay for the early diagnosis of Toxoplasma infection. Methods Free streptavidin was used to attach a biotinylated DNA to the biotinylated second antibody, through amplication of the DNA label, antigen which bound with antibodies was detected, thus Toxoplasma circulating antigens Immuno-PCR assay was estabished. detecting the CAg in serial dilutions of serum and the dynamic variation of serum CAg of Toxoplasma in experimental infected mice using Immuno-PCR and ELISA parallely to compare the sensitivity Result The detection limit of the immuno-PCR was at about 1/1000 dilution, it was at 1/5 dilution for ELISA. CAg of Toxoplasma could be detected as early as the third day after infection using Immuno - pcr, Wheareas the ELISA on the same samples, revealed no detectable CAg elevations until fifth day after infection. Conclusion In comparison with CAgdetecting by ELISA parallelly performed, this method is more sensitive than ELISA, The detection limit of the immuno-PCR was an 200 fold improvement compared with a conventional ELISA. the positive result could be detected earlier than that of ELISA,It provide scientific basis for early diagnosis of Toxoplasmosis.
2.Evaluating the Effects of Urban Resident Basic Medical Insurance on Medical Service Utilization Based on Matching DID Model

Objective: To identify the impact of Urban Resident Basic Medical Insurance (URBMI) on medical service utilization. Methods: Based on data of China Health and Nutrition Survey, matching DID model was used to compare the changes in medical service utilization between residents participated in this system and those not participated in this system in 2009. Results:In initial stage of implementation, URBMI in a certain extent promoted medical service utilization of residents participated in the system, but the impact level was still low. The promoting effect on inpatient service utilization was significant, but very limited on outpatient service utilization. Conclusion:Promoting outpatient service utilization and increasing compensation are future priorities for improving URBMI to implement the system.
3.Actualities of pharmacological effects and clinical application of propolis on oral cavity diseases

AIM: This paper summarized the actions of propolis on anti pathogen, anti inflammation, analgesia, anti ulcer, sedation, narcosis and so on. An evaluation of the pharmacological effects, and toxicity was also made. In addition, the clinical applications of propolis on oral cavity diseases were induced, such as dental caries, tooth damages, dental pulp diseases, periodontium diseases, and oral mucosa diseases. This article also prospected the distinctive advantages and application foreground of propolis as a kind of high efficiency natural medicinal material.
4.Development of soft tissue balance system and its application for quantitative knee collateral ligament release:an experimental study

Objective In order to meet the quantitative measurement of soft tissues balance required in the TKR,a new soft tissues balance navigation system have been developed and used experimentally in adjusting the soft tissue release of medial collateral ligament(MCL)and quantitative measurement of the force change in extension gap or flexion gap after various kinds of MCL release during TKA.Methods Six precise load sensors have been embedded in the front measure device,which is inserted into the cutting gaps of the 5 knee joints of cadavers to measure the force of MCL before or after various types of releases.The device has the function of changing distance between its upper and lower plates by the mechanical structure for the adjustment of different bone cutting space.The reading of the sensors representing the force are displayed in three or two-dimensional manner in real-time with accuracy of better than 0.098 N per bit.Results The preliminary test shows that the measurement system can accomplish the force measurement and the distance adjustment in the knee joint,and its application for measuring the force of medial collateral ligament shows a quantitative force change in MCL in different knee position and different way of release.Release of the posterior portion of MCL mainly produces laxity of MCL at full extension with 33.61% reduction rate of tensional force in value.Complete medial collateral ligament release causes laxity of MCL both at full extension and 90? flexion with reduction rate of 44.96% or 41.30% respectively.Conclusion The soft tissues balance measurement device are capable of clinical use for navigation of soft tissue balance in TKA and performing knee dynamics research.It has been proved quantitatively that the posterior portion of MCL plays an important role in stability for extended knee while the anterior portion of MCL mainly stabilizes the knee in flexion.
5.Investigation on the radioactivity level of natural rock in Lhasa

Objective To investigate the radioactivity level of natural rocks in Lhasa in order to obtain the radionuclide contents and availiability of the rock.Methods Radiation monitoring and sampling were carried out in the natural rock mining area in Lhasa,and the contents of radionuclides in the samples were analyzed.Both internal and external exposure indexes were calculated,and the rocks were classified according to the relevant national standards on building materials.Results The average specific activity of 40K,226Ra,232Th and 238U in the rock samples were 912,59.0,77.1,and 64.4 Bq/kg,respectively.Internal and external exposure indexes were both less than 1.Conclusions The radioactivity level in natural rocks in Lhasa were lower than the national levels.The natural rocks could be used as main building materials and Class A decoration materials.
6.Effects of fatty acid binding protein-5 silencing on proliferation, apoptosis, invasion of human glioma cells and expressions of CD44+and CD133+

BACKGROUND:It has been demonstrated that there are different expressions of fatty acid binding proteins (FABPs) in most malignant tumors such as breast cancer, prostate cancer, liver cancer, lung carcinoma and bladder carcinoma;therefore, FABPs are closely related to the occurrence, metastasis, invasion and drug resistance of malignant tumors. OBJECTIVE:To investigate the effects of fatty acid binding protein-5 (FABP-5) silencing on the proliferation, invasion and apoptosis of U251 cel s (human glioma cel s). METHODS:siRNA molecules targeting the mRNA of FABP-5 was designed and chenical y synthesized, which was transiently transfected into U251 cel s. U251 cel s were divided into three groups:Lv-shRNA-FABP-5 was added into FABP-5-shRNA group, LV-shRNA-NC added into negative control group, and blank control group underwent normal culture. mRNA and protein expressions of FABP-5 were detected by RT-PCR and western blot assay, respectively. The cel proliferation in vitro was determined by cel counting kit-8 assay, the cel cycle, apoptosis and expressions of CD44+and CD133+were detected by flow cytometry, and the apoptosis was observed using TUNEL staining. RESULTS AND COUNCLUSION:mRNA and protein expressions of FABP-5 in the FABP-5-shRNA group were significantly lower than those in the negative control and blank control groups. Compared with the negative control and blank control groups, the cel growth rate was significantly decreased, the cel cycle arrested in the G0/G1 phase, and the cel number in the S phase was decreased, and moreover, the proportion of CD44+/CD133+expression was significantly decreased in the FABP-5-shRNA group (P<0.05). Besides, compared with the negative control and blank control groups, the apoptosis rate was significantly increased, and the cel proliferation and invasiveness were significantly decreased in the FABP-5-shRNA group (P<0.05). In conclusion, it is possible that FABP-5 directly or indirectly regulates the cel cycle and apoptosis of glioma cel s, and its expression changes share a close relationship with the invasiveness of tumor cel s.
7.Arthroscopic microfrature for articular cartilage damage of the knee

[Objective]To analyze the characteristics of articular cartilage lesions in knees and evaluate the therapeutic efficacy of microfracture technique under arthroscopy for articular carrtilage lesions.[Method]A total of seventy-nine patients with articular cartilage lesions were treated with the microfracture technique.Thirty-eight patients were males and forty-one were female,with the mean age of 38.3 years(15~57years).The averaged preoperative duration of symptoms was 11 months(3~23 months).After preparation of the lesion,used an arthroscopic awl to make multiple holes,results in microfracture holes approximately 3mm apart and the depth approximately 3 mm.[Result]A total of 131 articular cartilage lesions were found in Seventy-nine patients,for an average of 1.6 lesions per case.The main lesion was found on the medial femoral condyle in 33.6% and the patella in 22.1%;60.3% of lesions were grade Ⅳ according to Modified Outerbridge Scale;In 43.5% of the lesions the total area was from 1~2 cm2.Seventy-three patients were followed up for 17 months in average(10~33 months).At the time of the latest follow-up,79.7% of patients were satisfied with the results.According to Lysholm knee evaluation system,the mean scores was improved from 43 points preoperative to 88 points postoperative.[Conclusion]Arthroscopic microfrature is simple and effective to repair of articular cartilage lesions of knee joint.This method can release pain and improve joint function.
8.Comparison of two different prosthetic materials in the treatment of inguinal hernia in aged patients

0.05). There were 7 cased in the polypropylene mesh group had abdominal discomfort due to stiffness, which higher than that of the expanded polytetrafluroethylene patch and control groups (P 0.05). CONCLUSION: Both prosthetic patches are safe and effective when used in the repair of inguinal hernia in aged patients with low recurrence rate, especially, the expanded polytetrafluroethylene patch, which is more comfortable than polypropylene mesh.
9.Influence of thoracic sympathetic block on patients with diabetes mellitus complicated heart failure

Objective: To observe curative effect of thoracic sympathetic block on patients with diabetes mellitus (DM) complicated cardiac chamber enlargement and heart failure (HF). Methods: A total of 76 DM patients with cardiac chamber enlargement and HF were randomly divided into routine treatment group (n=36) and epidural nerve block group (nerve block group, n=40, received thoracic epidural nerve block based on routine treatment), and four weeks were a course of treatment. Changes of left ventricular ejection fraction (LVEF), left ventricular end-diastolic dimension (LVEDd), left atrial diameter (LAD), levels of tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), brain natriuretic peptide (BNP) and glycosylated hemoglobin (HbA1c) were observed in two groups. Results: There were no significant difference in all indexes between two groups before treatment (P>0.05). After treatment, total effective rate of cardiac function improvement in nerve block group was significantly higher than that of routine treatment group (95% vs. 66.7%, P<0.05). Compared with before treatment, there were significant decrease in LVEDd [(71.63±9.26)mm vs. (65.4±8.12)mm] and LAD[(45.62±7.10)mm vs. (37.14 ±6.28)mm] (P<0.05 both), significant increase in LVEF [(31.3±11.46)% vs. (42.12± 10.42) % , P< 0.01], and significant decrease in levels of TNF-α [(73.55±16.36)ng/L vs. (28.35±10.08) ng/L], IL-6 [(17.62±5.47) ng/L vs. (7.88±2.29) ng/L], BNP[(13.49±2.94) ng/L vs. (6.28±2.28) ng/L] and HbA1c [(7.9±2.6) % vs. (6.8±1.9) %] in nerve block group after treatment, P<0.05 or <0.001; and levels of LVEF, TNF-α, IL-6 and BNP in nerve block group after treatment were significantly improved than those of routine treatment group, P<0.05 or <0.01. Conclusion: Thoracic sympathetic block can significantly decrease levels of neuroendocrine and inflammatory factors, improve cardiac function, and elevate quality of life in patients with diabetes mellitus complicated heart failure.
