2.Correlation between serum Adropin protein and severity and prognosis of patients with hypertensive intracerebral hemorrhage

Objective To investigate the correlation between serum Adropin protein expression and the severity and prognosis of patients with hypertensive intracerebral hemorrhage.Methods Patients with hypertensive intracerebral hemorrhage who were treated in our hospital from January 2016 to January 2018 were enrolled.One hundred patients were enrolled in the study group.One hundred healthy subjects were included as control group.Serum Adropin protein levels were measured and analyzed.The relationship between the levels of Adropin protein and the disease condition and prognosis in patients with hypertensive intracerebral hemorrhage.Results The study group finally included in 97 cases.The control group finally included in 99 cases.The expression levels of Adropin protein in the study group were lower than that in the control group (P<0.05).The expression levels of Adropin protein were significantly different in patients with different severity of disease (P<0.05).The expression levels of Adropin protein in patients with hypertensive intracerebral hemorrhage with different prognosis were statistically significant (P<0.05).Univariate analysis showed that age,severity of disease,amount of bleeding,and whether it broke into the ventricle were the prognostic factors of hypertensive cerebral hemorrhage (P<0.05).Multivariate logistic regression analysis showed that Adropin protein,age,severity of disease,and whether bleeding broke into the ventricle were independent factors affecting the prognosis of patients with hypertensive intracerebral hemorrhage (P<0.05).Conclusion Serum Adropin protein is associated with the severity and prognosis of patients with hypertensive intracerebral hemorrhage.Serum Adropin protein is an independent factor affecting the prognosis of patients with hypertensive intracerebral hemorrhage.
3.Transmission of porcine endogenous retrovirus in xenotransplantation

Organ transplantation is the most effective treatment for various types of end-stage diseases. To resolve the problem of donor shortage in organ transplantation, the possibility of xenotransplantation has been gradually explored by surgeons. Pig is one of the common donor sources for xenotransplantation. As a bridge between two species, the viruses carried by pig organs may be transmitted between species and cause the risk of zoonosis. Porcine endogenous retrovirus (PERV) is integrated into the genome, which is a category of retrovirus featuring cross-species transmission. In this article, the influencing factors of transmission characteristics of PERV, the transmission risk of PERV and its recombinant virus, and the detection and transmission risk assessment of PERV in xenotransplantation test were reviewed, aiming to provide reference for alleviating severe shortage of donor organs and driving the advancement of xenotransplantation technologies.
4.Variation of three-dimensional structure of subchondral bone in early stage osteoarthritis and interventive effect of diphosphonate

Objective To observe the three-dimensional structure changes of subchondral bone in early stage knee joint instability and the effect of diphosphonate intervention so as to test the role of early three-dimensional structure changes of subchondral bone in pathogenesis of osteoarthritis (OA).Methods Sixty healthy male New Zealand white rabbits were assigned to model group (n =24),diphosphonate group (n=24) and control group (n =12) according to random number table.Rabbit right knee destabilization (anterior cruciate ligament transection) is used to induce OA.Rabbits in diphosphonate group received subcutaneous injection of 0.01 mg/kg diphosphonate (risedronate) per day.Instead,isotonic saline solution of the same volume was subcutaneously given to rabbits in model and control groups.One third of the animals in each group were killed at week 4,8 and 12 respectively.Surgical knee joint with preservation of each 2 cm bone above and below joint surface was dissected to perform Micro-CT.Bone volume fraction (BVF),trabecular thickness (Tb.Th),trabecular spacing (Tb.Sp),trabecular number (Tb.N),volumetric bone mineral density (vBMD) and tissue BMD (tBMD) were measured and analyzed statistically.Results At week 4 following operation,BVF,Tb.N and Tb.Th were lowered significantly in model group as compared to control group (P <0.01) ;BVF was lower in model group than in diphosphonate group (P < 0.05) and lower in diphosphonate group than in control group (P < 0.05) ; Tb.Sp was increased in model group as compared to diphosphonate group and control group (P <0.01) and had obvious increase in diphosphonate group as compared to control group (P <0.01) ; vBMD was significantly lower in model group than in diphosphonate group and control group (P < 0.05),but there was no statistical difference between diphosphonate group and control group.At week 12 following operation,model group presented higher BVF,Tb.Th and Tb.N (P <0.05),significantly lower Tb.Sp (P < 0.05) and significantly higher vBMD (P < 0.01) as compared to diphosphonate group and control group.Conclusions In knee joint instability,variations of subchondral bone are mainly characterized by osteoclasia in the early stage,followed by osteogenesis in later stage.Diphosphonate may improve the bone architecture of subcondral bone via inhibition of bone resorption.
5.Finite element analysis of biomechanical variation of subchondral bone in osteoarthritis and effect of diphosphonate therapy

Objective To observe the biomechanical changes of subchondral bone in the early stage of knee joint instability and effect of diphosphonate therapy so as to investigate the role of the early biomechanical changes of subchondral bone in the onset and development of osteoarthritis (OA).Methods Sixty healthy male New Zealand white rabbits were assigned to model group (n =24),diphosphonate group (n =24) and control group (n =12) according to random number table.Joint destabilization by anterior cruciate ligament transection of the right knee of the rabbits was performed to induce OA models.Rabbits in diphosphonate group received subcutaneous injection of 0.01 mg/kg diphosphonate (risedronate) per day,and isotonic saline solution of the same volume was subcutaneously given to rabbits in model and control groups.Half the animals in each group were killed by aeroembolism at postoperative 4 weeks and 12 weeks respectively.Surgical knee joint with preservation of each 2 cm bone above and below joint surface was dissected to perform gross scoring.Thereafter,two-dimensional image profile was achieved by Micro-CT examination and converted into Ansys for limit element analysis after fitting in Mimics software.Results At four weeks,bone volume fraction (BVF),elastic modulus (EM),reaction force (RF),and mean Von Mises stress were all declined in three groups,the lowest level in the model group (P < 0.01).The diphosphonate group also had lower levels than the control group,with insignificant difference.Bone mineral density (BMD) in the model group was obviously declined in contrast with the diphosphonate and control groups (P < 0.01),but there were no significant difference between the diphosphonate group and the control group.At 12 weeks,the model group showed higher level of BVF and BMD,but lower level of EM,RF and Von Mises stress in comparison with the control and diphosphonate groups (P < 0.01).EM,RF,and Von Mises stress were lower in the diphosphonate group than those in the control group as well,but the difference was statistically insignificant.The model group showed that BVF,BMD,EM,RF,and Von Mises stress at 12 weeks were improved from those at 4 weeks (P < 0.01).Conclusions Biomechanical properties of subchondral bone are affected in the early stage of knee joint instability and a notable decrease of EM is observed in the early stage,followed by an enhancement in late stage.It means that the biomechanical changes of subchondra,l bone in the early stage of knee joint instability may be connected with the bone resorption resulting from abnormal stress.On the contrary,diphosphonate may markedly improve EM of subchondral bone through inhibiting bone resorption.
6. Effect of L-tryptophan and its derivatives on cephalotaxine production in Cephalotaxu mannii suspension cells

Objective: To investigate the effects of L-tryptophan and its derivative 5-methyl-DL-tryptophan (5-MT) on suspension cells growth and cephalotaxine production of Cephalotaxu mannii. Methods: The cell suspension cultures were treated with L-tryptophan and 5-MT on day 15, respectively. Then the cell growth, cephalotaxine production, and activity of key enzymes in the metabolism pathway were determined. Results: L-tryptophan and 5-MT could enhance cephalotaxine production. The cultures treated with 5 mg/L L-tryptophan showed the highest cephalotaxine yield (2.944 mg/L). While treated with 10 mg/L L-tryptophan and 5 mg/L 5-MT, the cephalotaxine yields were 1.947 and 2.192 times of the control culture (1.343 mg/L); The biomass of suspension cultures were decreased by 9.35% and 18.04%, respectively, less than those in the control group (2.406 g/L); The activity of 3-deoxy-D-arabino- heptulosonate-7-phosphate synthase was decreased by 12.4% and 5.57% compared with the control (0.67 U/mg); The activity of phenylalanine ammonium-lyase activity increased by 37.72% and 28.02% and the total phenolics content increased by 19.14% and 9.61% compared with the control (78.21 U/mg and 0.36 mg/mL). Conclusion: L-tryptophan and its derivative 5-MT can inhibit the L-tryptophan biosynthetic pathway and promote the accumulation of cephalotaxus alkaloids.
7.A comparative study on the levels of serum cytokines and cortisol among post-traumatic stress disorder patients of Li and Han ethnicities in Hainan.

BACKGROUNDPost-traumatic stress disorder (PTSD) is a mental disorder with delayed or chronic onset caused by unusual, threatening, or disastrous psychological trauma, and it is an important manifestation of post-disaster mental and behavioral disorders. Studies have shown that IL-6 is a cytokine associated with PTSD occurrence. This study aimed to explore the role of cytokine and ethnicity in the pathogenesis of PTSD by examining levels of serum cytokines IL-2, IL-6, IL-8, TNF-α, and cortisol in PTSD patients of Li and Han ethnic groups.
METHODSLevels of serum cytokines IL-2, IL-6, IL-8, TNF-α, and cortisol were examined by enzyme-linked immunoabsorbent assay (ELISA) method and assessed by PCL-C scale among 30 PTSD patients of Han ethnicity and 30 of Li ethnicity, and compared with 30 normal controls of Han and Li ethnicity, respectively.
RESULTSPTSD patients of Li ethnicity scored higher than PTSD patients of Han ethnicity, and normal controls of Li and Han ethnic groups in each of the re-experiencing, avoidance/numbing, and hyperarousal symptoms. The differences reached statistical significance (P < 0.05). In PTSD patients of Li ethnicity compared to patients of Han ethnicity and normal controls of either Li or Han ethnicity, the levels of IL-2, IL-6, IL-8, and TNF-α were higher, and the level of cortisol was lower.
CONCLUSIONSThere are ethnic differences in re-experiencing, avoidance/numbing, and hyperarousal symptoms among PTSD patients. The levels of serum cortisol and cytokines are strongly associated with the race.

8.Identification and verification of key salt-tolerant amino acid sites of banana MaNHX5.

In order to improve the salt tolerance of banana NHX genes, we cloned a MaNHX5 gene from Musa acuminata L. AAA group and predicted the key salt-tolerant amino acid sites and mutant protein structure changes of MaNHX5 by using bioinformatics tools. The 276-position serine (S) of MaNHX5 protein was successfully mutated to aspartic acid (D) by site-directed mutagenesis, and the AXT3 salt-sensitive mutant yeast was used for a functional complementation test. The results showed that after the mutated MaNHX5 gene was transferred to AXT3 salt-sensitive mutant yeast, the salt tolerance of the mutant yeast was significantly improved under 200 mmol/L NaCl treatment. It is hypothesized that Ser276 of MaNHX5 protein plays an important role in the transport of Na+ across the tonoplast.

Objective To investigate residents' psychological stress factors and research the correlation between post-traumatic stress disorder (PTSD) and platelet 5-HT concentrations so as to provide scientific bases for diagnosis and treatment of PTSD and psychological intervention for people in the disaster area. Methods A questionnaire survey of 5 500 residents who have accepted psychological help was conducted by the emphatic investigation method. 10. Psychological impacts of the COVID-19 epidemic on Chinese people: Exposure, post-traumatic stress symptom, and emotion regulation

Objective: To examine the effects of coronavirus disease-2019 (COVID-19) exposure, expressive suppression/cognitive reappraisal, and demographic variables on post-traumatic stress symptoms (PTS) among Chinese. Methods: Participants were recruited by social media through WeChat and 6 049 Chinese (aged from 17 to 63 years; median=24) from 31 provinces were included in the study. PTS symptoms, expressive suppression, and cognitive reappraisal were assessed after the outbreak of COVID-19. A regression mixture analysis was conducted in Mplus 7. Results: A regression mixture model identified three latent classes that were primarily distinguished by differential effects of COVID- 19 exposures on PTS symptoms: (1) Class 1 (mildly PTS symptoms, 80.9%), (2) Class 2 (moderate PTS symptoms, 13.0%), and (3) Class 3 (high PTS symptoms, 6.1%). The results demonstrated that the young, women and people with responsibilities and concerns for others were more vulnerable to PTS symptoms; and they had more expression inhibition and less cognitive reappraisal in three latent classes. Conclusions: The findings suggest that more attention needs to be paid to vulnerable groups such as the young, women and people with responsibilities and concerns for others. Therapies to encourage emotional expression and increase cognitive reappraisal may also be helpful for trauma survivors.
