1.Clinical characters of pulmonary thromb-embolism in ≥70 elderly patients

Objective To explore the clinical characteristics and effects of thrombolytic and anti coagulation treatment in over 70 year old patients with pulmonary thromb embolism(PTE). Methods The clinical findings, diagnostic techniques, effects of thrombolytic therapy and anti coagulation in 58 over 70 year old patients with PTE were analyzed retrospectively. Results Deep venous thrombosis (DVT) in lower limbs (74 1%) was the most common thrombolism prone factors in our study. Limitation of movement and long term stay in bed due to a variety of causes were in the next place. The clinical findings were atypical, associated with a false diagnostic ratio of 46 5% in elderly patients with PTE. Undefined causal and different degree of dyspnea along with sudden and persistent hypoxemia were the main characteristics. Spiral CT and radioactive nuclear ventilation perfusion scan were important methods for diagnosis of PTE with positive finding of 95 7% and 62 5%, respectively. The effective and cure rate of thrombolytic therapy combined with anti coagulation, anti coagulation and anti platelet therapy was 91 3%, 75 8%, 0 and 47 8%, 17 2%, 0, respectively. Conclusions The most common risk factors of PTE in the elderly was DVT and the long term stay in bed or stay without active moving. The clinical symptoms were not only atypical but also variable. Thrombolytic with anti coagulation therapy is safe and effective, but anti platelet coagulation alone is not benefit.
2.Cytotoxicity of non-viral gene vector polyamide amine dendrimers-hyaluronic acid polymer

OBJECTIVE To study the cytotoxicity of polyamide amine dendrimers (PAMAM)-hyaluronic acid(HA)as a non-viral gene delivery vector in vitro. METHODS PAMAM-HA was synthesized by our laboratory. Cytotoxicities of various polymers on HeLa cells,Bel-7402 cells and HepG2 cells were evaluated by MTT assay. Apoptotic rates were determined by flow cytometry. RESULTS The cytotoxicity of PAMAM-HA polymer increased with the concentration of polymer(50-800 mg · L-1),the time of action(24-72 h), the number of generations of PAMAM(G4,G5) and decrease in the molecular mass(3850,17 200)and the graft density(5%-25%)of HA. After incubation with PAMAM G4-HA3850-5%or PAMAM G5-HA3850-5%for 24 h,the apoptotic rate of hepatoma cells—Bel-7402 cells was 4.5%and 9.9%,respectively. After incubation with complexes of PAMAM G4-HA3850-5%/DNA or PAMAM G5-HA3850-5%/DNA for 24 h,the viability of HeLa cells was more than 80%,which was lower than that of PAMAM G4/DNA和PAMAM G5/DNA. CONCLUSION Cytotoxicity of PAMAM modified by HA of different grafting density and quantity can be reduced ,suggesting the PAMAM is a promising non-viral gene vector.
3.Involvement and modulation effects of oxytocin and opioid receptor on evoked discharge of hippocampal CA1 neurons in rats

BACKGROUND: Central oxytocin (OT) may be a neurotransmitter or neuromodulator and play an important role in learning and memory, sexual behaviour, pain modulation and opiate tolerance and dependence. To research the interactions between oxytocinergic and opioidergic system in hippocampus has some significance.OBJECTIVE: To investigate the effects of OT administered intracerebroventricularly on evoked discharge of left dorsal hippocampal CA1 neurons in rats and the possible interactions between oxytocinergic and opioidergic system.DESIGN: A randomised controlled study.SETTING: Department of Physiology of Guangdong Medical College; Department of Physiology and Pathology of Medical College of Wuhan University.MATERIALS: The study was conducted in the Physiology Department of Medical College of Wuhan University from September 2002 to September 2003. A total of 36 male Sprague-Dawley rats were selected and randomly divided into six groups: control (NS), OT groups (0.2 mg/L, 2 mg/L and 20 mg/L), [d (CH2)5-OVT]+OT (2 mg/L), naloxone+OT (2 mg/L), with 6 rats in each group.METHODS: Single-unit recording was performed with glass microelectrode. The glass microelectrode was inserted by a micromanipulator into hippocampal CA1. The electrical activity was amplified by a microelectrode amplifier and then recorded by the biological experimental system,monitored at the same time with oscilloscope. When recording the neural discharge, electrical stimulation of the sciatic nerves was performed once 5minutes through a double stainless electrode. 5 μL oxytocin in dosage of 0.2, 2 and 20 mg/L were injected slowly into lateral ventricle via microlitre syringe. [d(CH2)5-OVT]+OT (2 mg/L) group: 2.5 μL [d(CH2)5-OVT](80 mg/L) was injected into lateral ventricle and then 2.5 μL oxytocin (2 mg/L). Naloxone+OT (2 mg/L) group: 2.5 μL naloxone (400 mg/L) was injected into lateral ventricle and then 2.5 μL oxytocin (2 mg/L). According to frequency of discharge, effect of oxytocin at various dosages on discharge induced by neurons in hippocampal CA1 area and [d (CH2)5-OVT]and naloxone on oxytocin was assayed. MAIN OUTCOME MEASURE: Changes of discharge frequency after stimulation.RESULTS: Data of totally 36 rats were entered the final analysis. ① OT (0.2 mg/L, 2 mg/L and 20 mg/L) administered by intracerebroventricularly could decrease the evoked discharge of hippocampal CA1 neurons in a dose-dependent manner. ② The inhibitory effects of OT (2 mg/L) could be blocked by pretreated intracerebroventricularly injection of [d (CH2)5-OVT](80 mg/L, 2.5 μL). ③ Intracerebroventricular injection of naloxone (400 mg/L, 2.5 μL) could attenuate the effects of OT (2 mg/L) significantly.CONCLUSION: OT can inhibit the electrical activities of hippocampal CA1 neurons to external electrical signal through activating the oxytocin receptor. Moreover, central opioid receptor is involving in the inhibitory effects of OT.
4.Therapeutic Effects of Artemisia Argyi Ferment Substance on Systemic Candida Albicans Infection

Objective To investigate the therapeutic effects and mechanism of Artemisia argyi ferment substance on systemic Candida albicans infection. Methods The model of systemic Candida albicans infection was established in immunosuppressed mice. The model mice were randomly divided into the model control,Artemisia argyi ferment substance( AAFS) at different doses(100,200,and 400 mg·kg-1 )and fluconazole group(20 mg·kg-1 ),30 mice in each. Mice in each treatment group were given therapeutic drugs by gavage for 5 consecutive days,twice daily. The survival of mice was determined 21 days after the model was set up. The serum levels of IFN-γand IL-2 were determined by ELISA. The proliferation activity of T lymphocyte in the spleen was detected by MTT assay. The number of living fungi in liver and kidney tissues was counted. Results Compared with the model control,AAFS at middle and high doses and fluconazole significantly increased the survival rate of mice,the serum levels of IFN-γand IL-2,and the proliferation activity of T lymphocyte in the spleen,but decreased the number of living fungi in tissues(P〈0. 01). Compared with low dose AAFS,middle and high doses of AAFS and fluconazole showed significantly different effect on each index(P〈0. 05 or P〈0. 01),but there was no difference among these groups(P〉0. 05). Conclusion AAFS at 200-400 mg·kg-1 has inhibitory effects on systemic Candida albicans infection in mice,the mechanism of which is related to increasing the proliferation of T lymphocyte in spleen and the levels of IFN-γand IL-2 in serum.
5.Factors affecting postmenopausal osteoporosis in Dali Bai Autonomous Prefecture

Objective:
To investigate the prevalence of postmenopausal osteoporosis (PMOP) and analyze its influencing factors among women at ages of 50 to 59 years in Dali Bai Autonomous Prefecture, Yunnan Province, so as to provide insights into the prevention of PMOP among menopausal women.
Methods:
Bai Ethnic menopausal women at ages of 50 to 59 years who received healthy examination at the Center of Healthy Examination, Dali Prefecture People's Hospital from June 2017 to May 2021 were selected as the study subjects, and subjects' demographic characteristics, living habits, history of diseases, family history of osteoporosis and history of parturition were collected using self-designed questionnaires. The height, body weight and bone density were measured, and fasting blood glucose, vitamin D3, blood lipids and liver functions were detected. The factors affecting the development of PMOP were identified using a multivariable logistic regression model.
Results:
Totally 2 000 questionnaires were allocated, and 1 584 valid questionnaires were recovered, with an effective recovery rate of 79.20%. The respondents had a mean age of ( 56.22±2.61 ) years, and mean body mass index ( BMI ) of ( 24.62±2.35 ) kg/m2. There were 497 respondents ( 31.38% ) with a family history of osteoporosis, and the prevalence of PMOP was 20.64%. Multivariable logistic regression analysis identified age ( OR=1.135, 95%CI: 1.074-1.196 ), age of menarche ( OR=1.138, 95%CI: 1.059-1.217 ), duration of menopause (OR=1.425, 95%CI: 1.228-1.622), number of parturition ( >2, OR=5.036, 95%CI: 2.972-7.101 ), smoking ( OR=2.594, 95%CI: 1.767- 3.421 ), alcohol consumption ( OR=2.051, 95%CI: 1.503-2.598 ), family history of osteoporosis ( OR=2.540, 95%CI: 1.769-3.311 ), hypertension ( OR=1.492, 95%CI: 1.406-1.578 ), diabetes ( OR=1.774, 95%CI: 1.581-1.967 ), total cholesterol ( OR=1.483, 95%CI: 1.251-1.716 ), triacylglycerol ( OR=1.801, 95%CI: 1.576-2.026 ), low-density lipoprotein cholesterol ( OR=1.614, 95%CI: 1.498-1.731 ), fasting blood glucose ( OR=1.192, 95%CI: 1.077-1.307 ), BMI ( OR=0.934, 95%CI: 0.862-0.993 ), outdoor activity ( ≥1 time/week, OR: 0.413-0.549, 95%CI: 0.329-0.637 ), age of menopause ( OR=0.909, 95%CI: 0.841-0.977 ), daily intake of calcium ( ≥600 mg, OR: 0.493-0.644, 95%CI: 0.389-0.786 ), vitamin D3 level ( ≥20 ng/mL, OR: 0.604-0.719, 95%CI: 0.523-0.853 ) and high-density lipoprotein cholesterol ( OR=0.658, 95%CI: 0.550-0.767 ) as factors affecting the development of PMOP.
Conclusions
The prevalence of PMOP in Dali Bai Autonomous Prefecture is similar to the nationwide level in China, and old age, smoking, alcohol consumption, a family history of osteoporosis and high blood lipid levels may increase the risk of PMOP.
6.Application of ultrasound in the evaluation of oropharynx in patients with obstructive sleep apnea-hypopnea syndrome

Objective To explore the feasibility and application value of sonography in the evaluation of oropharynx in patients with obstructive sleep apnea-hypopnea syndrome (OSAHS). Methods The oropharynx of forty-four patients with OSAHS and forty-four healthy subjects selected as the control group were separately examined by ultrasonography.The parameters including anterior-posterior diameter and the lateral diameter of oropharyngeal airway of the end-deep inspiratory and end-deep expiratory,thickness and wide of tongue base,lingual wide,lingual height,distance from mandibular plane to hyoid bone and distance from tongue base to mandibular plane were obtained for statistical analysis and compared between the two groups.Correlation analysis of above parameters plane with body mass index (BMI) were carried out. Results The anatomy of the oropharynx was well shown on sonography.When compared with the control group,the lateral diameter of oropharyngeal airway,thickness of tongue base,lingual height,lingual wide and distance from mandibular plane to hyoid bone also showed significant statistically differences between the OSAHS patients group and the control group (all P <0.01).The wide of tongue base,thickness of tongue base,lingual wide,lingual height,distance from mandibular plane to hyoid bone,and distance from tongue base to mandibular plane in OSAHS patients group were positively correlated with BMI( r =0.533,0.415, 0.555,0.349,0.378,0.419;all P <0.05).The wide of tongue base and lateral diameter of oropharyngeal airway in the control group were negatively correlated with BMI ( r = -0.324,-0.405,-0.317;all P <0.05).Conclusions Ultrasonography can be used to assist in the assessment of OSAHS,which can provide an important reference for clinical diagnosis and therapeutic effect evaluation of OSAHS.
7.Analysis of etiological surveillance results of influenza in Chuxiong prefecture from 2017 to 2022

Objective:The result of influenza etiology surveillance in Chuxiong prefecture were analyzed to master the epidemic characteristics of influenza virus and provide scientific basis for influenza prevention and control.Methods:Real-time PCR assay and MDCK cell culture method were used to detect and separate influenza virus nucleic acids from influenza-like cases (ILI) and suspected influenza cases from 2017 to 2022 come from the sentinel hospitals.Results:From 2017 to 2022, a total of 7 302 cases of ILI and 1 079 cases were detected positive, with a positive rate of 14.78%. The positive rate in the six years was 14.71%、15.44%、20.11%、5.04%、10.90% and 21.08%, and the positive rate in 2020 was the lowest. Among them, the positive rate of influenza A was higher than that of influenza B, and higher in spring and winter than in summer and autumn., One H9N2 human infected with avian influenza virus was detected positive.The outbreak cases were mainly influenza B, followed by influenza A H3N2, and students were susceptible to it. The proportion of influenza-like cases in the 0~5 year old group was the highest (64.16%), and the protortion in 60~year old group was the lowest (0.05%).Conclusions:From 2017 to 2022, influenza A was predominant in Chuxiong prefecture, with high prevalence in winter and spring. The influenza strains presented diversity and each strain circulated alternately. The implementation of COVID-19 prevention and control measures can effectively reduce the infection rate of influenza virus .In addition, there are human cases of avian influenza infection in Chuxiong prefecture. So the surveillance of influenza-like cases should be strengthened an improved, and to expand the monitoring of human and avian influenza.
8.Assessment of quality deviation of pork and salmon due to temperature fluctuations during superchilling.

Superchilling is an emerging technology for meat preservation; however, the temperature changes during the process have been commonly ignored. Thus, the effects of temperature fluctuations on meat quality during superchilling are yet to be evaluated. In our study, pork loins and salmon fillets were stored for several days (0, 8, 15, 23, and 30 d) under different temperature fluctuations based on -3.5 ℃ as the target temperature. The results showed that after 15 d of superchilling storage, the values of total volatile basic nitrogen, total viable count, and lipid oxidation were significantly (P<0.05) altered in the ±2.0 ℃ fluctuation group compared with the constant temperature group. On the contrary, there was no significant difference in these parameters between the ±1.0 ℃ fluctuation group and the constant temperature group after 30 d of storage. In addition, irregular temperature changes significantly accelerated the modulation of various indicators. In brief, temperature fluctuations and irregular temperature changes accelerated the destruction of muscle structural integrity, increased the water loss, gradually widened the water loss channels, and thereby reduced the edibility by accelerating the spoilage of meat.
